Effect of Ulmus davidiana Planch (Ulmaceae) on T-lymphocyte-producing cytokines such as IL-2, IL-6, and $IFN-{\gamma}$ production in collagen-induced arthritis of rats

  • Objective : The effect of Ulmus davidiana Planch(UD), which has long been known to have anti-inflammation and protective effects on damaged tissue, inflammation and bone among other functions, on the development of type II collagen (CII)-induced arthritis (CIA) in rats was studied. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA)or saline during the experiment. When compared with rats treated with saline as control, UDHA at doses of more than $20{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-2, interleukin-6, $IFN-{\gamma}$ when the cells were obtained from rats 14 days after immunization and cultured in vitro with CII. Results : When rats were injected intraperitoneally, UD -treated group and control group rats did not differ significantly when low doses of UD was given to rats. Conclusion : The recommended dose of UD in the management and treatment of rat CIA will be more than $20{\mu}g/100\;g$, which is two-firth of human therapeutic dose. From the results, it was concluded that the effect of UDHA is dependent of dosage.

Suppressive effects of a water extract of Ulmus davidiana Planch (Ulmaceae) on collagen-induced arthritis in mice

  • Objective: Since Ulmus davidiana Planch (Ulmaceae) has been known to have anti-inflammatory and protective effects on damaged tissue, inflammation and bone among other functions, this study was undertaken to address whether the water extract of the bark of Ulmus davidiana Planch (Ulmaceae) (UD) could modulate the expression of inducible inflammatory cytokines in mice. The present study was also done in order to assess the therapeutic effects of UD in collagen-induced arthritis (CIA) in mice. Methods : DBA/1 mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with UD orally at 100 mg/kg once a day for 3 weeks. Paws were evaluated macroscopically for redness, swelling and deformities. The levels of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the ankle were examined. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Results : Administration of UD significantly suppressed the progression of CIA and inhibited the production of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the paws. The erosion of cartilage was dramatically reduced in mouse knees after treatment with UD. In the serum of UD-treated mice, the levels of IL-4 and IL-10, anti-inflammatory cytokines, were increased. Conclusion : From the results, it was concluded that administration of UD has therapeutic effects on CIA including protection of cartilage and RA for a potential therapy.

Anti-Obesity and Lipid Metabolism Effects of Ulmus davidiana var. japonica in Mice Fed a High-Fat Diet

  • The root bark of Ulmus davidiana var. japonica (Japanese elm) is used in Korea and other East Asian countries as a traditional herbal remedy to treat a variety of inflammatory diseases and ailments such as edema, gastric cancer and mastitis. For this study, we investigated the lipid metabolism and anti-obesity efficacy of ethyl alcohol extract of Ulmus davidiana var. japonica root bark (UDE). First, HPLC was performed to quantify the level of (+)-catechin, the active ingredient of UDE. In the following experiments, cultured 3T3-L1 pre-adipocytes and high-fat diet (HFD)-fed murine model were studied for anti-obesity efficacy by testing the lipid metabolism effects of UDE and (+)-catechin. In the test using 3T3-L1 pre-adipocytes, treatment with UDE inhibited adipocyte differentiation and significantly reduced the production of adipogenic genes and transcription factors PPARγ, C/EBPα and SREBP-1c. HFD-fed, obese mice were administered with UDE (200 mg/kg per day) and (+)-catechin (30 mg/kg per day) by oral gavage for 4 weeks. Weight gain, epididymal and abdominal adipose tissue mass were significantly reduced, and a change in adipocyte size was observed in the UDE and (+)-catechin treatment groups compared to the untreated control group (***p < 0.001). Significantly lower total cholesterol and triglyceride levels were detected in UDE-treated HFD mice compared to the control, revealing the efficacy of UDE. In addition, it was found that lipid accumulation in hepatocytes was also significantly reduced after administration of UDE. These results suggest that UDE has significant anti-obesity and lipid metabolism effects through inhibition of adipocyte differentiation and adipogenesis.

  • 느릅나무 근피와 점액질의 화학 조성을 분석하였고, 근피의 MeOH추출물을 각종 유기용제 및 수층으로 분획하고 각 분획물의 항균성을 조사하였다. 원소조성은 수종간의 차이는 없었지만, 목부의 질소 및 리그닌 함량은 사부보다 높았으며 회분 및 추출물양은 목부보다 사부가 높게 나타났다. 근피의 점액질은 냉수에 의해 매우 느리게 용출 되었으며, 높은 산성당 함량을 갖는 pectin 유사 물질이었다. 느릅나무 근피로부터 약 7% 정도의 메탄올 추출물이 얻어졌고 이 중 78%가 수용성 물질이었으며 Staphylococcus aureus와 Salmonella typhimurium에 대하여 항균성을 나타내었다.

느릅 분말을 첨가한 요구르트의 이화학적 및 관능적 품질 특성 (Physicochemical, Microbial, and Sensory Properties of Yogurt with Ulmus davidiana var. japonica During Storage)

  • This study evaluates changes in yogurt with Ulmus davidiana var. japonica (U. davidiana) in terms of its physicochemical, microbial, and sensory properties during storage. The pH value and mean microbial counts of this yogurt with 0.2 to 0.8% of U. davidiana added ranged from 4.1 to 4.3 and from $1.4{\times}10^8$ to $2.3{\times}10^8cfu/mL$, respectively, during storage at $7^{\circ}C$ for 16 d. In terms of its color, the L value was significantly higher in the control and U. davidiana yogurt 0.2 to 0.4% up to 4 d, than other yogurt samples, and the b value was influenced by the addition of U. davidiana over 0.6% throughout the storage. According to a sensory test, there was a significant difference in the yellowness score between U. davidiana yogurt and the control, but earthy, and bitterness, and viscosity scores were similar. Overall acceptability was not significantly influenced after 8 d of storage in all yogurt samples, including the control. According to these results, concentrations (0.2 and 0.4%) of U. davidiana yogurt had no significantly adverse effects on its physicochemical, microbial, and sensory properties.

Anti-Inflammatory Activity of Constituents Isolated from Ulmus davidiana var. japonica

  • Twenty six compounds (1-26) were isolated from the root barks of Ulmus davidiana var. japonica. The anti-inflammatory activity of the isolated compounds were evaluated agai nst the generation of inflammatory chemical mediators in bone marrow-derived mast cells. Among them, compounds 10, 11, 13, 15 and 19 inhibited not only cyclooxygenase-2 dependent prostaglandin $D_2$ generation but also 5-lipoxygenase dependent leukotrien $C_4$ generation in a concentration-dependent manner. In addition, compounds 11, 12, 13, 15 and 19 also inhibited $\beta$-hexosaminidase release, a marker of mast cell degranulation reaction, from bone marrow-derived mast cell. These results suggest that the anti-inflammatory activity of U. davidiana might in part occur by both the inhibition of eicosanoid generations and the degranulation reaction of mast cells.

유백피 추출물의 항세균 작용 (Antibacterial Activity of Ulmus pumila L. Extract)

  • 유백피 추출물의 항세균 효과를 측정하기 위해 각각 다른 농도의 추출물의 함유한 배지에서 MIC를 측정하고, 증식에 미치는 영향을 조사한 결과는 다음과 같았다.이 추출물은 실험에서 사용된 세균 중 S.aureus, S.epidermidis, S.faecalis, B.subtilis, M.luteus와 같은 그람 양성균 및 E.coli, S.typhi, P.vulgaris, P.aeruginosa와 같은 그람 음성균에서 높았고, K.pneumoniae에 대해서는 항균력을 나타내지 않았다. 이 추출물 중의 항세균 효과를 가진 물질은 $121^{\circ}C$에서 20분간 멸균한 후에도 항세균 효과에는 별다른 변화가 없었다.

  • A sesquiterpene ortho-naphtpoquinone was isolated from the 80% aqueous methanolic extract of root bark of Ulmus davidiana Planch whose stem and root bark have been used as an oriental medicine for the treatment of edema, mastitis, gastric cancer and inflammation. On the basis of spectral data obtained from UV-vis, IR, HR-EIMS and NMR spectrometry, including the pulse field gradient (PFG) HMQC and HMBC techniques, the structure of this compound was determined as 2,3-dihydro-3-hydroxy-3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione which has been known as mansonone 1. This compound has been isolated only from the heart-wood of Mansonia altissima Chev, a Sterculiacea from West Africa. Thus, this compound was isolated for the second time from natural source in this study. This is the first report to present the carbon chemical shift assignment of mansonone I.

Nitric oxide and $ProstaglandinE_2$ Synthesis Inhibitory Activities of Flavonoids from the Barks of Ulmus macrocarpa

  • Eight phenolic compounds (1-8) which were isolated from the barks of Ulmus macrocarpa were evaluated for their inhibitory activities on nitric oxide (NO) and prostagrandin $E_2$ (COX-2) production in $interferon-{\gamma}\;(INF-{\gamma})$ and lipopolysaccharide (LPS)-activated RAW 264.7 cells in vitro. NO and COX-2 levels were moderately reduced by the addition of compounds (1-8). Among them 3,4,5,6,7 and 8 inhibited NO production in a dose dependent manner with an $IC_{50}$ of 92.2, 97.3, 36.1, 43.5, 32.8, 39.4 and 37.1 ${\mu}g/ml$, respectively (positive control, L-NMMA; 36.4 ${\mu}g/ml$), and 3,4,5,6,7 and 8 reduced the COX-2 level in a dose dependent manner with an $IC_{50}$ of 43.2, 24.8, 24.8, 33.4, 44.8 and 22.7 ${\mu}g/ml$, respectively (positive control, indomethacin; 23.4 ${\mu}g/ml$). These results suggest that the phenolic compounds may be developed as potential anti-inflammatory and cancer chemopreventive agents.

초미세 느릅 분말의 항산화 활성 연구 (Antioxidant Activities of Powdered and Ultra-fine Powdered Ulmus Davidiana var. Japonica)

  • The antioxidant activity was examined on ultra-fine powdered (UFP) Ulmus davidiana (U. davidiana) var. japonica. The average particle size of UFP or commercial powdered (CP) was $1-3{\mu}m$ or $100{\mu}m$. The extraction was performed using either ethanol (EtOH) or hot-water. Contents of phenolic compound of CP and UFP U. davidiana extracts in EtOH was 40.38 and 65.61 mg/g, respectively. In DPPH, EtOH extract of UFP U. davidiana showed a significantly greater activity than hot-water extract at 40 and $200{\mu}g/mL$. At $200{\mu}g/mL$, the activity was over 90% in all groups. The reducing power of UFP U. davidiana var. japonica in EtOH extraction was 74.3%, which was significantly greater than in other samples (p<0.05). In addition, reducing powder was significantly higher in UFP-EtOH than in other samples at all concentrations except for $0.32{\mu}g/mL$. The above results suggest that EtOH extraction of U. davidiana showed slightly higher DPPH radical scavenging activity, and ultra-fine powder of U. davidiana extracts may show higher antioxidative activities based on reducing power.
