Clincal Studies on the Gangrenous Mastitis Caused by Mircrococus pyogenes var aureus in Goats (Micrococcus pyogenes var aureus에 의한 유산양(乳山羊)의 괴저성유방염(壞疽性乳房炎)에 관한 임상학적(臨床學的) 연구(硏究))

  • Six cases of gangreous mastitis of goats infected spontaneously were observed clinically and pathologically in Daegu and Daejeon district. and with strain isolated purely from the infected goats, the artificial infection to the animal was examined, the sensitivity of strain to the antibiotics was tested and clinical treatment was carried out. The results obtained are summarized as follows: 1. In the six cases approximately same clinical findings were observed as the previously published literatures on gangrenous mastitis of cattle, sheep and goats. 2. The micrococcus pyogenes var aureus was highly virulent strain which was the causative organism for the gangrenous mastitis by inoculating in the udder. 3. The gangrenous mastis was probably occured by the formation of thrombosis in veins of udder. 4. In the sensitivity test, the micrococcus pyogenes var aureus resited for penicillin in 2 cases among the 6 strains, but sensitived for streptomycin, chloromycin, oxyteracycline, erythromycin, achromycin, neomycin and kanamycin in other 4 and in all case. 5. The treatment for gangrenous mastitis may be extirpated the gangrenous region surgically in the case of unilaterally or locally affected, treated by muscle injection or teat-operation in the case of severely or diffusely affected and infused antibiotics up to teat canal or treated by mammary tissue injection in the case of slightly affected.

Effects of insufficient dietary amino acids on the milk production of dairy cows milked three times daily (사료 내 아미노산의 부족이 하루 착유횟수를 2회에서 3회로 증가시킨 젖소의 유생산 반응에 미치는 영향)

  • An experiment was conducted to examine how the response of dairy cows to a change from twice to thrice-daily milking is affected by deficiencies in the dietary supplies of three amino acids, His, Met and Lys. The amino acid deficiencies were obtained by feeding the cows a diet of grass silage and a cereal-based supplement containing feather meal as the sole protein supplement. Taken overall, the results show that when cows were given the feather meal diet, even though dietary ME was in considerable excess, a deficiency of specific amino acids prevented any increase in milk yield in response to increasing the frequency of milking from twice to thrice daily. The results of half-udder milking showed that when cows consumed the diet deficient in amino acids, milking one half of the gland more frequently reduced the secretion of protein and lactose by the control gland. Neither MBF nor the ratio of 3-MH/creatinine in urine was affected by thrice-daily milking. The present results go a stage further in showing that, against a background of insufficient dietary amino acids, the stimulus of thrice-daily milking is not sufficient to induce a measurable change in the partition of amino acid use between body and udder.

Detection of QTL on Bovine X Chromosome by Exploiting Linkage Disequilibrium

  • A fine-mapping method exploiting linkage disequilibrium was used to detect quantitative trait loci (QTL) on the X chromosome affecting milk production, body conformation and productivity traits. The pedigree comprised 22 paternal half-sib families of Black-and-White Holstein bulls in the Netherlands in a grand-daughter design for a total of 955 sons. Twenty-five microsatellite markers were genotyped to construct a linkage map on the chromosome X spanning 170 Haldane cM with an average inter-marker distance of 7.1 cM. A covariance matrix including elements about identical-by-descent probabilities between haplotypes regarding QTL allele effects was incorporated into the animal model, and a restricted maximum-likelihood method was applied for the presence of QTL using the LDVCM program. Significance thresholds were obtained by permuting haplotypes to phenotypes and by using a false discovery rate procedure. Seven QTL responsible for conformation types (teat length, rump width, rear leg set, angularity and fore udder attachment), behavior (temperament) and a mixture of production and health (durable prestation) were detected at the suggestive level. Some QTL affecting teat length, rump width, durable prestation and rear leg set had small numbers of haplotype clusters, which may indicate good classification of alleles for causal genes or markers that are tightly associated with the causal mutation. However, higher maker density is required to better refine the QTL position and to better characterize functionally distinct haplotypes which will provide information to find causal genes for the traits.

  • A total of 251 clinical isolates (human origin, 43 strains and bovine udder origin 208 strains) of the Staphylococcus that fermented mannitol aerobically were tested for their ability to produce coagulase, DNase, and thermostable nuclease. Of these, 158 isolates coagulated human or bovine plasma, produced DNase, and thermostable, nuclease and were identified as St. aureous, 146 of which produced a 1+ to 3+ clot. The remaining 12 isolated produced a -clot in citrate treated plasma but produced 1+ to 3+ clot in ethylenedi-aminetetraacetic acid (EDTA) treated plasma. It was found that 7 coagulase positive isolates failed to produced thermostable nuclease. In these organisms, we found out of the clot formation is not by coagulase activity but utilization of citrate, because EDTA treated plasma is not coagulated. Among 93 isolates which did not coagulate citrate-or EDTA treated plasma and thermostable nuclease negative, 28 strains produced DNase were identified as St. epidermidis, and other strains were not identification further. It was found that thermostable nuclese production appears to be a consistent property of St. aureus and the test is easy to perform, is rapid became quite distinct within 2 to 4 hour, and is not influenced by as many factors and variations as the coagulase test.

A Study on The Lesions of The Mammary Glands of Slaughtered Holstein Cows in Gwangju Area (광주지역에서 도태되는 홀스타인 소 유방의 병변 조사)

  • Macroscopic investigations were made on the lesions of a total of 101 udders from slaughtered Holstein cows in Gwangju area. Incidence of supernumerary teats was 9.9%. Gross lesions were found in the surface of teats of 29.4% of cows; these lesions comprised papilomatosis (16.0%), fronds (8.9%) and injury at the tip (3.2%). Approximately 95% of the cows had slight to severe teat orifice keratosis. On the mucous membranes of the teat cisterns, fibrosis (22.7%) and congestion (8.9%) were observed. Gross lesions were found in the mammary glands of 68% of cows; these lesions comprised fibrosis (18.8%), serous exudate (4.9%), fatty tissue (4.9%), pus (4.0%), congestion (2.0%).

A Pathological and Bacteriological Study on the Mammary Glands of Korean Native Cows Slaughtered in Gwangju Area (광주지역에서 도태되는 한우 유방의 병리학적 및 세균학적 연구)

  • Pathological and bacteriological studies were made of a total of 111 udders from slaughtered Korean native cows originating from various locations in Chonnam province. Incidence of supernumerary teats was 8.1%. Gross lesions were found in 21.4% of mucous membranes of total teat cisterns; these lesions comprised epithelial hyperplasia (12.6%), congestion (4.1%), slight fibrosis (3.8%) and severe fibrosis with congestion (0.9%). Gross lesions in the glandular tissues were found in 35.1% of cows; these lesions comprised slight fibrosis (27.9%), moderate fibrosis (6.3%) and severe fibrosis with pus (0.9%). Mastitis was found only in one cow. One hundred and twenty-five strains of microorganisms were isolated from mammary gland fragments of the cows. Pathogens most commonly isolated were Staphylococcus spp.; most of them were minor mammary pathogens. Major mammary pathogens were Streptococcus spp. (9.6%) and E. coli (2.4%). Most of the isolates were susceptible to many of 13 antimicrobial agents used, but Streptococcus spp. and Gram-negative rods showed resistance to 7 and 8 agents, respectively. 97.5% of the isolates showed susceptibility to amoxicillin, but only 55.8% to erythromycin.

Study of plasma β-carotene concentration in dairy cows (젖소의 혈장 베타카로틴 농도 조사)

  • This study was carried out to determine the effects of ${\beta}-carotene$ on the control of mastitis in dairy cows during the dry period. The relationship between the levels of plasma ${\beta}-carotene$ and the status of udder health in Holstein dairy cows were investigated. Blood samples were collected from 117 cows to compare the levels of plasma ${\beta}-carotene$ in lactating cows. The levels of plasma ${\beta}-carotene$ were $1.82{\mu}g/ml$ in healthy cows(n = 65) and $1.12{\mu}g/ml$ in mastitic cows(n = 52), respectively(p < 0.01). In the experiment to compare the level of plasma ${\beta}-carotene$ in the cows at different stages of lactation, the plasma ${\beta}-carotene$ levels were $1.73{\mu}g/ml$ in lactating cows(n = 22), $1.29{\mu}g/ml$ in nonlactating cows(n = 35) and $0.43{\mu}g/ml$ in cows after calving(n = 16)(p < 0.05).

Lysozyme Activity in Buffalo Milk: Effect of Lactation Period, Parity, Mastitis, Season in India, pH and Milk Processing Heat Treatment

  • Lysozyme activity in buffalo milk in relation to the period of lactation, parity of animal, weather conditions and udder infections was studied. Effect of storage and heat processing of milk on lysozyme activity was determined. Lysozyme activity was higher in buffalo milk than in cow milk. Buffalo colostrum showed lysozyme activity 5 times of that in mature milk. Lysozyme activity in buffalo milk was not influenced by the parity of animal and the stage of lactation, however, it increased during extreme whether conditions (winter and summer). Lysozyme in both cow and buffalo milk exhibited maximum activity at pH 7.4. Buffalo milk lysozyme was fully stable while the cow milk lysozyme was partly inactivated by pasteurization (low temperature-long time as well as high temperature-short time treatments). Lysozyme in buffalo milk was more stable than in cow milk during storage and heat treatment. A 10 to 50-fold increase in milk lysozyme activity was observed in mastitic cows. An assay of lysozyme activity in milk can be used to diagnose mastitis in cattle but not in buffaloes. Some buffaloes exhibited 1000 fold greater lysozyme activity and moderately raised somatic cell count in milk, but there was no sign of mastitis in these animals. A possible role of milk lysozyme in prevention of mastitis in buffaloes is discussed.

  • The accuracy of the gamma function to describe lactation curve of Yankasa ewes was investigated using milk production data obtained from 168 ewes with either complete or incomplete 84 day lactation records. Ewes were daily hand milked after over-night separation from lamb and thorough disinfection of the udder. The daily milk yield on the average was estimated from the weekly total yield. The effect of type of birth, parity, season of birth and lactation length on the components of lactation curve (a, b, c and s) were analysed by least-square procedures. All factors except parity significantly (p<0.01) influenced the curve parameters. The goodness of fit of the function did not differ between the two classes of varying duration of lactation; incomplete lactation however, showed a lower persistency and as expected lower level of production. Correlations ($r_p$) between lamb weight gain and milk yield using the weight records from lamb weighed at weekly intervals indicated a high and positive relationship in the early lactation that decreased in intensity as the lactation advanced.

  • In order to clarify the morphological changes in the mammary glands of cows affected with coliform (Escherichia coli) mastitis, histopathological observations were undertaken on the mastitis of the lactating rabbits which was experimentally induced with E. coli or its endotoxin isolated from cases of acute and chronic matitis in dairy cattle. In the bacterial suspension-infused groups the affected quarters of udder showed cloudy swelling, hyperemia and hemorrhage to local necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells, and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue, and destruction of alveoli. Later, proliferation of firoblasts, plasma cells, lymphocytes, eosinophils and histiocytes appeared in the glandular tissue and necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. Granuloma-like inflammatory changes could be observed in the glandular tissue on the 7th days after infusion. The inflammatory response in the group infused with E. coli strain isolated from the natural case of acute mastitis was rapid and severe as compared with that of chronic mastitis. In the endotoxin-infused group the morphological changes were similiar to those of the bacterial suspension-infused groups.
