Antioxidative Materials in Domestic Meju and Doenjang 2. Separation of Lipophilic Brown Pigment and Their Antioxdative Activity (재래식 메주 및 된장중의 항산화성 물질에 관한 연구 2. 지용성 갈변물질의 분리와 항산화력)

  • Lipophilic brown pigments produced during the fermentation of domestic Meju and Doenjang were fractionated by column and thin -layer chromatography (TLC). Each of the fractions was tested for the antioxidant activity and then characterized by spectroscopic analysis. The lipophilic brown pigments were separated into chloroform -soluble and methanol-soluble parts in which Meju resulted the higher content of chloroform-soluble part than that of methanol-soluble part ; however, Doenjang exhibited the opposite result to that of Meju. More strong antioxidant activity was found in the methanol-soluble part than the chloroform-soluble part. Four and five fractions were separated from chloroform-soluble and methanol-soluble parts respectively. by silicagel TLC. The fraction that exhibited the high antioxidant activity showed a strong absorption at 260nm caused by amino compounds in UV spectrum The other fractions which did not have antioxidant activity absorbed at 240nm by carbonic acid and it ester,. IR spectrum of each fraction commonly showed absorption at 3400cm-1 , 2800cm-1 , 1700cm-1, 1600cm-1, 1400cm-1 , 1300cm-1 and 1100cm-1. Especially , the fraction which had a strong antioxidant activity showed absorption at 2800cm-1, 1400cm-1, 1600cm-1 suggesting that the fraction contain Schiff's base and primary amine structure.

  • Electroluminescence(EL) devices based on organic thin films have been attracted lots of interests in large-area light-emitting display. In this stuffy, an emissive layer was fabricated using Langmuir-Blodgett(LB) technique in organic light-emitting (OLEDs). This emissive organic material was synthesized and named PECCP[poly(3.6-N-2-ethylhexyl carbazolyl cyanoterephthalidene)] which has a strong electron donor group and an electron acceptor group in main chain repeated unit. This material has good solubility in common organic solvents such as chloroform. THF, etc, and has a good stability in air. The Langmuir-Blodgett(LB) technique has the advantage of precise control of the thickness down to the molecular scale, In particular, by varying the film thickness it is possible to investigate the metal/polymer interface. Optimum conditions for the LB film deposition are usually determined by investigating a relationship between a surface pressure $\pi$ and an effective are A occupied by one molecule on the subphase. The LB films were deposited on an indium-tin-oxide(ITO) glass at a surface pressure of 10 mN/m and dipping speed of 12 mm/min after spreading PECCP solution on distilled water surphase at room temperature, Cell structure was ITO/PECCP LB film/Alq$_3$/Al. We considered PECCP as a hole -transport layer inserted between the emissive layer and ITO. We also used Alq$_3$ as an emissive layer and an electron transport layer. We measured current-voltage(I-V) characteristics, UV/visible absorption, PL spectrum and EL spectrum of the OLEDs.

  • High performance liquid chromatographic method with diode-array detection (HPLC-DAD) has been performed for the simultaneous determination of four marker constituents, paeoniflorin, trans-cinnamic acid, schisandrin and glycyrrhizin in traditional herbal medicinal preparation, So-Cheong-Ryong-Tang (SCRT). The presence of paeoniflorin, trans-cinnamic acid, schisandrin and glycyrrhizin in this decoction was ascertained by retention time, spiking with each authentic standard, UV spectrum and ESI mass spectrum. All four compounds showed good linearity ($r^2$ > 0.998) in a relatively wide concentration ranges. The RSD for intra-day and inter-day precision was less than 3% and the limits of detection (LOD) were less than 30 ng. The mean recovery of each compound was 94.1-113.0% with RSD values less than 3.0%. These results suggest that the developed HPLC method is simple, effective and could be readily utilized as a quality control method for commercial SCRT products.

New Azobenzene Crown p-tert-Butylcalix[4]arenes as Switchable Receptors for $Na^+ and K^+$ ions: Synthesis and Isomerization Studies

  • Two new azobenzene crown ether calix[4]arenes, 10 and 11, were synthesized by two pathways. In the first pathway,two ethoxy nitrobenzene groups were attached to t-butylcalix[4]arenes in a 1,3 position. Subsequent reduction ofthe nitrobenzene group s by metallic zinc in an alkaline solution afforded 10 and 11 in8% and 12%,respectively. In the second pathway,an azobenzene containing two glycolic units was prepared prior connect-ing to t-butylcalix[4]arenes. The yields from the second approach (5%, 8% for 10 and 11, respectively) were lower than those from the former approach. Single crystals of 10 suitable for X-ray crystallography was ob-tained by recrystallization in methanol.Both the X-ray structure and the 1H-NMR spectrum of 10 indicated that the stereoisomer of the azobenzene moiety was trans and the calixarene platform was in cone conformation. 1H NMR spectroscopy suggested that 10 underwent an observable cis-trans isomerization in CDCl3 under room light and upon UV irradiation with cis:trans ratios of 33:67 and 36:64,respectively. Compound 6 which was the precursor of 11showed fluxional behavior and was found to have mixed conformations ofcone and partial cone with a ratio of 47:53 at -30 $^{\circ}C.$ 1H NMR spectrum of 11 suggested that 11 was initially isolated as cis azobenzene with calix[4]arene in cone conformation and underwent conformational interconversion through calix[4]arene annulas in a similar fashion to 6 upon exposing to light. The complexation studies of 10 with picrate salts of Na+ and K+ using 1H NMR spectroscopysuggested that Na+ preferred to bind the cis form of 10 while K+ preferred to bind the trans form. The stereoisomer of the azobenzene unit in 11 changed partially from cis to trans upon complexing with K+.

Analysis of Scutellaria baicaleinsis Georgi (Scutellariae Radix) by LC-DAD and LC-ESI/MS

  • In this study, baicalin, as a marker substance of Scutellariae Radix, was quantitatively analyzed by a high performance liquid chromatography-photodiode array detector (HPLC-DAD). We identified wogonoside, baicalein, and wogonin in the Scutellariae Radix by a high performance liquid chromatography-electrospray ionization-mass spectrometer (HPLC-ESI-MS). The baicalin was separated on a Xterra C18 column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of 38% acetonitrile in 0.68% phosphoric acid. The baicalin spectrum in the Scutellariae Radix extracts was coincided by comparing with UV-visible spectrum (200-550 nm) of baicalin standard in the library. The amount of baicalin in Scutellariae Radix was 10.46%, which is higher than KFDA's guideline. The marker substances of Scutellariae Radix showed a strong base peak $[M]^+$ in the positive detection mode following as; baicalin (m/z; $271[MH^+-sugar]^+$, $447[M+H]^+$), wogonoside (m/z; $285[MH^+-sugar]^+$, $461[M+H]^+$), baicalein (m/z; $271\;[M+H]^+$), wogonin (m/z; $285[M+H]^+$). These results are consistent with the fragment pattern and molecular weight of standard components from literature.

  • Various fabrication methods have been used to synthesize sapphire which has qualities of jewelry well beyond the industrial class. Among them, the flux sapphire of Chatham Company which has as high value as jewelry was selected in order to compare natural and synthetic sapphire. First, the WD-XRF (Wavelength dispersive x-ray fluorescence spectrometer) was used to analyze the chemical composition of natural and synthetic sapphire. Although natural sapphire had very diverse chemical compositions, flux sapphire had small quantities of Mo, Pt and Pb elements in addition to the similar chemical ingredients to natural one. Pt is decisive proof of flux sapphire. Next, by investigating spectroscopic characteristics using UV-VIS Spectrophotometer after heat treatment at high temperatures of $1300^{\circ}C$ and $1500^{\circ}C$, the variation of 690 nm absorbance related to $Cr^{3+}$ was detected in the natural sapphire while those of the 690 nm absorbance (related to $Cr^{3+}$) as well as absorbance of 376 nm and 388 nm ($Fe^{3+}$) were seen in the flux sapphire. It was found that the difference in the absorbance variation of flux sapphire is greater than that of natural sapphire after heat treatment. The chemical composition and spectrum analysis were utilized to compare the natural sapphire and the flux synthetic sapphire.

Extraction and Analysis of Astaxanthin from Haematococcus pluvialis Using Sonication (초음파처리를 이용한 Haematococcus pluvialis로부터의 아스타잔틴의 추출 및 분석)

  • The extraction and quantitative analysis conditions for astaxanthin from Haematococcus pluvialis, and the structural characteristics of H. pluvialis extract, H. pluvialis hydrolysate and synthetic astaxanthin were investigated using UV/visible and FT-IR spectrometers. Astaxanthin was dissolved in methanol, and then treated to enhance the solubility by sonication for 45 min. With sonication pretreatment, the solubility of astaxanthin increased up to 1.5 times compared to that without sonication. The extracts were hydrolyzed by cholesterol esterase for the analysis of H. pluvialis extract containing astaxanthin ester. A HPLC method using reverse phase C18 column with methanol-water (95:5, v/v) as mobile phase was developed to analyze astaxanthin. After hydrolysis, the absorption spectrum of H. pluvialis hydrolysat was changed to similar pattern to synthetic astaxanthin, confirming the extraction and analysis condition of astaxanthin from H. pluvialis.

Fabrication of a Schottky Type Ultraviolet Photodetector Using GaN Layer (GaN를 이용한 Schottky diode형 자외선 수광소자의 제작)

  • We fabricated a planar ultra-violet photodetector whose ohmic and schottky contacts were respectively formed with evaporated Al and Pt on the GaN layer. To examine the applicability of the device to the UV sensor, we investigated its electrical and optical characteristics. The GaN layer on the sapphire waver had $7.8{\times}10^{16}cm^{-3}$ of doping concentnation and the $138 cm^2/V{\cdot}s$ of electron mobility and it absorbed the spectrum of the light below 325 nm wavelength. It had the responsivity of 2.8 A/W of at 325 nm, and the signal to noise ratio(SNR) of $4{\times}10^4$, and the noise equivalent power(NEP) of $3.5{\times}10^9$W under 5 V reverse bias. These results confirmed that the GaN schottky diode had a solar blind properly when it was applied to the UV photodetector.

  • Pure $TiO_2$and 10 mol % Pb-doped $TiO_2(Pb_xTi_{1-x}O_2$(x = 0.1)) powder and thin films have been prepared by the sol-gel method. Titanium isopropoxide and ethanol are used for pure $TiO_2$, lead acetate trihydrate and titanium triisopropoxide monoacethylacetonate are used for Pb-doped $TiO_2$, respectively. Films are coated on p-type Si(100) wafer and ITO glass substrates by the sol-gel spin-coating method. The powder and multi-coated films are annealed at different temperature (400~$800^{\circ}C$) for phase formation and crystallization. TGA/DTA, XRD analysis, SEM and UV-visible transmission spectroscopy have been used to study the characteristics of the powder and films. XRD results show that the films are polycrystalline, anatase type and oriented predominantly to the A(101) plane. A slight shift in the d-spacing for the Pb-doped film indicates the incorporation of the Pb into $TiO_2$lattice. A shift of the absorption wavelength in the transmission spectrum towards longer wavelength has been observed about $Pb_xT_{1-x}O_2$(x = 0.1) thin film, which indicates a decrease in the bandgap of $TiO_2$upon Pb-doping.

Separation of Antioxidant Compounds from Edible Marine Algae (식용 해조류에서 항산화 물질의 분리)

  • To isolate new antioxidants from twelve edible seaweeds, mixed methanol and chloroform extract of marine algae was fractionated into several subfractions and their antioxidant activities were measured by using AOM and 1, 1-diphenyl-2-picrylhydrazyl (DPPH). Especially the aqueous-methanol soluble fractions of brown and red algae showed a considerable antioxidant effect. Their antioxidant activities were stronger than synthetic antioxidants such as BHA, BHT, under the same concentration. Further fractionation of the aqueous-methanol soluble fractions using silica gel column chromatography yielded five subfractions. Among them methanol fraction exhibited high DPPH quenching activities. Also, it was confirmed to be benzene-derivative substances of two compounds by UV, HPLC, GC/MS analysis. Its each molecular weight was about 181, 238. These results suggested the existence of two effective natural antioxidant compounds in three edible marine algae.
