Antioxidants Stimulated by UV-B Radiation in Rice Seedling

  • In order to investigate low molecular antioxidants synthesized by enhanced UV-B radiation, we used the seedlings of two rice varieties. Woonjangbyeo, UV-tolerant, and Hwajoongbyeo, UV-susceptible, were subjected under supplemental UV-B irradiation. When rice seedlings were irradiated with UV light for short period, biosynthesis of total phenolic compound, ascorbate and glutathione were momently reduced. With an increase of UV-B radiation, however, those were slightly synthesized. The content of lipid peroxides in UV-challenged rice leaves was considerably increased after 12 hrs of UV-B treatment. Lipoxygenase activity under supplemental UV-B radiation was differently responded on rice varieties.

Effects of Hexaconazole on Growth and Antioxidant Potential of Cucumber Seedlings under UV-B Radiation

  • The present study was conducted to determine the effect of hexaconazole (HEX), a triazole fungicide, on the growth, yield, photosynthetic response and antioxidant potential in cucumber (Cucumis sativus L.) plants subjected to UV-B stress. UV-B radiation and HEX were applied separately or in combination to cucumber seedlings. The growth parameters were significantly reduced under UV-B treatment, however, this growth inhibition was less in HEX treated plants. HEX caused noticeable changes in plant morphology such as reduced shoot length and leaf area, and increased leaf thickness. HEX was quite persistent in inhibiting shoot growth by causing a reduction in shoot fresh and dry weight. HEX noticeably recovered the UV-B induced inhibition of biomass production. Significant accumutation in anthocyanin and flavonoid pigments in the leaves occurred as a result of HEX or UV-B treatments. HEX permitted the survival of more green leaf tissue preventing chlorophyll content reduction and higher quantum yield for photosystemII under UV-B exposure. HEX treatment induced a transient rise in ABA levels in the leaves, and combined application of HEX and UV-B showed a significant enhancement of ABA content which activates $H_2O_2$ generation. UV-B exposure induced accumulation of $H_2O_2$ in the leaves, while HEX prevented UV-B induced increase in $H_2O_2$, indicating that HEX serves as an antioxidant agent able to scavenge $H_2O$ to protect cells from oxidative damage. An increase in the ascorbic acid was observed in the HEX treated cucumber leaves affecting many enzyme activities by removing $H_2O_2$ during photosynthetic processes. The activities of antioxidant enzymes including catalase(CAT), ascorbate peroxidase(APX), superoxide dismutase(SOD) and peroxidase(POD) in the leaves in the presence of HEX under UV-B stress were higher than those under UV-B stress alone. These findings suggest that HEX may participate in the enhanced tolerance to oxidative stress. From these results it can be concluded that HEX moderately ameliolate the effect of UV-B stress in cucumber by improving the components of antioxidant defense system.

딸기 시설재배에서 UV-B 램프와 멀칭 종류에 따른 흰가루병과 점박이응애 억제 (Suppression of Powdery Mildew and Two-Spotted Spider Mite by UV-B Radiation and Mulching Type of Strawberry Cultivation in the Greenhouse)

  • 딸기에 발생하는 흰가루병과 점박이응애는 수확기 문제되는 주요 병해충이다. 딸기 수확기 병해충방제를 위해 적용 약제를 처리하지만 최근에는 소비자의 요구에 따라 친환경적 병해충 방제 방법이 요구되고 있다. 딸기 정식 후 자외선 램프(ultraviolet B, UV-B)를 이용한 친환경 병해충 방제효과를 조사하기 위해 수확기 토경과 수경재배로 2년 동안 시험을 수행하였다. 설향, 킹스베리, 두리향 품종을 정식하였으며 UV-B 램프+멀칭 종류(흑색, 녹색, 빛반사시트[light reflection sheet, LRS])별 4처리로 시험을 수행하였다. 처리는 UV-B+흑색 또는 녹색 멀칭, UV-B+흑색 또는 녹색+LRS, 무처리로 하였다. UV-B 램프 처리에 의한 흰가루병은 모든 품종에서 65% 이상의 방제효과를 보였다. 점박이응애 성충밀도는 UV-B+흑색 또는 녹색+LRS 멀칭 처리가 다른 처리보다 가장 낮았다. 따라서 딸기 수확기에 UV-B 램프 처리는 식물체에 미치는 영향도 적으며 흰가루병과 점박이응애 밀도를 낮추는 데 효과적인방제방법이될수있다.

UV-B 반복노출에 따른 항산화효소의 변성 및 활성저하와 RGP렌즈의 차단효과 (Denaturation and Inactivation of Antioxidative Enzymes due to Repeated Exposure to UV-B and Inhibitory Effect of RGP Lens)

  • 목적: 본 연구는 안구에 존재하는 항산화효소인 superoxide dismutase(SOD)와 catalase(CAT)가 UV-B에 반복적으로 노출되었을 때 유발되는 구조변성 및 활성저하의 상관관계를 밝히며, 자외선 차단 RGP렌즈의 사용으로 효소의 변성과 활성저하가 효율적으로 차단되는가를 알아보기 위하여 수행되었다. 방법: SOD와 CAT의 표준품으로 각각의 효소용액을 제조하고 하루 30분, 1시간 및 2시간씩 312 nm의 UV-B에 1, 2, 3, 4 및 5일 동안 반복적으로 노출시켰다. 이 때 UV-B에 직접 노출시킨 항산화효소의 구조 및 활성변화를 자외선 차단기능이 있는 RGP렌즈로 UV-B를 차단시킨 경우와 비교하였다. UV-B 반복노출에 따른 SOD와 CAT의 구조변성은 전기영동분석으로 확인하였으며, 이들 효소활성은 분석키트를 이용하여 비색분석법으로 측정하였다. 결과: UV-B에 반복노출된 SOD는 일일 30분 조사조건으로 반복노출되었을 때에도 전기영동분석에서 효소다중화(polymerization)가 관찰되었으나 활성의 변화는 10% 이내로 나타났다. 반면 UV-B에 반복노출된 CAT은 전기영동 시 효소밴드크기나 진하기가 감소하여 구조변성이 나타났음을 확인할 수 있었으며, 반복노출시간이 긴 경우 CAT은 전기영동분석에서 효소밴드를 보임에도 불구하고 그 활성은 완전히 소실되었다. 또한 UV-B 조사로 인한 CAT의 변성은 63.7%의 UV-B 차단기능을 가진 RGP렌즈의 사용으로 어느 정도 억제되었으나 완전히 억제되는 것은 아니었다. 결론: 이상의 결과로 UV-B 반복노출에 따른 항산화효소의 구조변성은 그 종류에 따라 효소활성의 감소정도와 반드시 일치하는 것은 아님을 알 수 있었다. 따라서 자외선으로 인한 항산화효소의 손상을 막기 위하여서는 콘택트렌즈를 착용한 상태에서 자외선 노출시간을 최소화하거나, FDA Class I 차단제에 해당하는 UV 차단율을 가지는 콘택트렌즈를 착용 또는 이에 상응하는 UV차단율을 가지는 선글라스를 덧착용할 것을 권장한다.

Accumulation of Chlorogenic Acid as a near UV-shielding Compound in Cauliflower Grown under Enhanced UV-B Radiation

  • Since solar radiation contains wavelength essential for photosynthesis accompanying with near-UV light, UV-B effects on biological parameters and acclimation mechanisms are influenced by photosynthetically active radiation (PAR). Therefore, to elucidate near-UV shielding mechanism in higher plants, we cultivated cauliflower under usual solar radiation and increased UV-B from fluorescent lamps, two- or three-fold excess over continuously estimated UV-B dose in PAR during daytime, using computer regulated systems. Increased UV-B radiation had little effect on growth expressed as fresh weigh and leaf area. Water soluble low molecular weight compounds showing absorption in near UV region were enhanced according to the irradiated UV-B dose. One of compounds in cauliflower leaves was identified as chlorogenic acid. This was found to have no near-UV photosenSitizerable activity and is known to have an ability to scavenge a wide species of active oxygen. Another pro-oxidant compound that generates superoxide anion radical under near-UV irradiation was not induced by increased UV-B during cultivation, and identified as lumazine, a degradation product from folic acid.

질소, 인산, 칼륨시비에 따른 오이의 자외선 감수성 변화 (Effect of Fertilization of UV-B Sensitivity of Cucumber Plant)

  • UV-B 조사에 의한 오이의 생장 및 생리 기능에 미치는 영향을 조사하기 위하여 비료처리에 의한 오이 제1 엽의 UV-B의 영향을 조사하였다. UV-B 조사에 의한 가시장해는 처리 1주일부터 나타났으나 생장 저해 현상은 볼 수 없었다. 그러나 UV-B 조사에 의한 생장 저해 현상은 비료 첨가에 의해 나타났으며 가장 큰 저해 효과는 질소 시용에 의해 나타났다. 이 결과는 UV-B가 질소대사에 관여하고 있음을 시사한다. 따라서 질소대사에 관여하고 있는 주 효소인 질산 환원효소 활성을 조사한 결과 UV-B 조사 7일째에 비료 처리구에서 현저히 증가하는 경향을 나타냈다. 또한 UV-B 조사에 의한 엽면적의 생장감소는 세포의 신장, 분열 생장에 관여하고 있는 식물호르몬 활성에 영향을 미치는 것으로 보여 오이의 제 1엽에서의 BA 처리에 의한 사이토카이닌의 감수성을 조사했다. UV-B 조사에 의한 사이토카이닌의 감수성은 UV-B를 조사하지 않은 구에 비해 억제되었다. 이 결과에 의해 UV-B 조사가 식물 생장에 관여하고 있는 사이토카이닌의 활성을 억제시켜 엽면적의 감소가 발생된 것으로 생각되었다.

Action Spectra of Apoptosis Induction and Reproductive Cell Death in L5178Y cells in UV-B Region

  • It is important to determine the action spectrum of UV-B radiation contained in the sunlight to estimate the risk of skin cancer. We have investigated action spectra for induction of apoptosis and reproductive cell death in L5178Y cells using the Okazaki Large Spectrograph at NIBB. L5178Y cells were exposed to light at different wavelengths in UV-B or UV-A region. Frequencies of apoptosis induction and reproductive cell death were determined by counting cells with chromatin condensation, and by the colony formation assay, respectively. The measured sensitivity spectra for the two end-points were in very good agreement. Sensitivity decreased steeply with increase of wavelength in UV-B region and remains nearly constant in UV-A region. The action spectra were also slightly steeper than that for the minimum erythematic dose (MED), but very similar to the light absorption spectrum of DNA in UV-B region. On the other hand, the spectra for both endpoints were similar to MED spectrum but not DNA spectrum in the UV-A region. Also different time-course and morphological difference of apoptosis were found between UV-B (long time, fragmentation) and UV-A (short time, shrinkage) region. These results suggest that DNA damage induced by UV-B light triggers apoptosis and reproductive cell death, but other damaged targets (membrane, protein and so on) trigger these effects in UV-A region.

UV-B 조사시 옥수수 잎의 산화적 스트레스에 대한 Nitric Oxide의 보호효과 (Protective Effect of Nitric Oxide against Oxidative Stress under UV-B Radiation in Maize Leaves)

  • The effect of nitric oxide (NO) on antioxidant system and protective mechanism against oxidative stress under UV-B radiation was investigated in leaves of maize (Zea mays L.) seedlings during 3 days growth period. UV-B irradiation caused a decrease of leaf biomass including leaf length, width and weight during growth. Application of NO donor, sodium nitroprusside (SNP), significantly alleviated UV-B stress induced growth suppression. NO donor permitted the survival of more green leaf tissue preventing chlorophyll content reduction and of higher quantum yield for photosystem II than in non-treated controls under UV-B stress, suggesting that NO has protective effect on chloroplast membrane in maize leaves. Flavonoids and anthocyanin, UV-B absorbing compounds, were significantly accumulated in the maize leaves upon UV-B exposure. Moreover, the increase of these compounds was intensified in the NO treated seedlings. UV-B treatment resulted in lipid peroxidation and induced accumulation of hydrogen peroxide ($H_2O_2$) in maize leaves, while NO donor prevented UV-B induced increase in the contents of malondialdehyde (MDA) and $H_2O_2$. These results demonstrate that NO serves as antioxidant agent able to scavenge $H_2O_2$ to protect plant cells from oxidative damage. The activities of two antioxidant enzymes that scavenge reactive oxygen species, catalase (CAT) and ascorbate peroxidase (APX) in maize leaves in the presence of NO donor under UV-B stress were higher than those under UV-B stress alone. Application of 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3- oxide (PTIO), a specific NO scavenger, to the maize leaves arrested NO donor mediated protective effect on leaf growth, photosynthetic pigment and free radical scavenging activity. However, PTIO had little effect on maize leaves under UV-B stress compared with that of UV-B stress alone. $N^{\omega}$-nitro-L-arginine (LNNA), an inhibitor of nitric oxide synthase (NOS), significantly increased $H_2O_2$ and MDA accumulation and decreased antioxidant enzyme activities in maize leaves under UV-B stress. This demonstrates that NOS inhibitor LNNA has opposite effects on oxidative resistance. From these results it is suggested that NO might act as a signal in activating active oxygen scavenging system that protects plants from oxidative stress induced by UV-B radiation and thus confer UV-B tolerance.

Are antioxidants induced by UV-B exposure in a leaf of spinach transported into another leaf\ulcorner

  • Seedlings of spinach (Spinacia oleracea L. "King of Denmark" and "Minsterland") were grown with or without supplemental UV-B under field conditions for five or nine days when their fifth or sixth leaves were expanding. Except two leaves which were just expanded and expanding, all other leaves of each seedling were removed before tested. One of these two leaves was exposed to UV-B irradiation and the other leaf was covered with lumiror film (no transmission below 320 nm) to prevent it from UV-B exposure. l,l-diphenyl-2-picrylhydrazyl scavenging activities (antioxidative activity) in leaves without covers were increased according to UV-B doses they were exposed to. And removal of UV-B exposure with lumiror decreased foliage antioxidative activities. This increase or decrease of activities in leaves did not dependent on the UV-B doses their paired leaves were exposed to in both two cultivars. The results demonstrated that spinach foliage antioxidants induced by UV-B exposure were not transported into another leaf.

제주도 고산지역 자외선복사의 월변화 특성과 원인 고찰 (A Study on the Monthly Characteristics of Solar UV Radiation in Gosan, Jeju)

  • The monthly-mean irradiance of ultra violet (UV)-B and UV-A observed from 2005 to 2014 and 2012 to 2014, respectively, at noon in Gosan, Jeju, South Korea are analyzed. We compare cloudiness, total ozone, visibility, and relative humidity with an emphasis on the four months from May to August (MJJA), which shows the largest UV radiation. While the incoming UV-B radiation at the top of the atmosphere in Gosan is the largest in June due to the small solar zenith angle, the observed surface UV-B shows an unexpected smaller value in June than those in May, July or August. In June, the meteorological conditions affecting Gosan are completely dominated by cloudiness and thus, frequent overcast seems to determine the minimum UV-B. Another important UV-determining factor is the total ozone, which exhibits a monotonic decrease during MJJA without agreeing to the characteristic feature of UV. The ratio of UV-B to UV-A is not generally influenced by cloudiness. Thus, the ratio is a useful indicator of atmospheric turbidity showing larger values for increasing visibility, except in June. A simple model has been used to estimate surface UV by using the observed ozone and visibility in the cloudless condition. The result shows that UV has the lowest value in June with small variation during MJJA. Model estimation also shows that the different characteristic features observed in July between surface UV-B and UV-A is the result of less absorption of UV-B by ozone than that of UV-A by a smaller amount of total ozone.
