We induced the activation of melanocytes in the epidermis of C57BL/6 mice by ultraviolet B (UVB) irradiation and observed the effect of red ginseng (RG) on the formation, and decrease of UVB-induced epidermal mel-anocytes. C57BL/6 mice were irradiated by UVB $80mJ/cm^2$ (0.5 mW/sec) daily for 7 days, and RG was intraperitoneally or topically applied pre- or post-irradiation. For the estimation of change of epidermal melanocytes, light microscopic observation with dihydroxyphenylalanine (DOPA) stain was performed. Split epidermal sheets prepared from the ear of untreated mice exhibited 11-16 $melanocytes/mm^2$, and one week after UV irradiation, the applied areas show an increased number of strongly DOPA-positive melanocytes with stout dendrites. But intraperitoneal or topical treatment with RG before each irradiation interrupted UVB-induced pigmentation and resulted in a marked reduction in the number of epidermal melanocytes as compared to radiation control skin. The number and size of DOPA-positive epidermal mel-anocytes were also significantly decreased in intraperitoneally injected or topically applicated group after irradiation with RG at 3rd and 6th weeks after irradiation. The present study suggests the RG as inhibitor of UVB-induced pigmentation and depigmenting agent.
Melanin is a natural skin pigment produced by specialized cells called melanocytes via a multistage biochemical pathway known as melanogenesis, involving the oxidation and polymerization of tyrosine. Melanogenesis is initiated upon exposure to ultraviolet (UV) radiation, causing the skin to darken, which protects skin cells from UVB radiation damage. However, the abnormal accumulation of melanin may lead to the development of certain skin diseases, including skin cancer. In this study, the antioxidant and antimelanogenic activities of the cell-free supernatant (CFS) of twenty strains were evaluated. Based on the results of 60% 2,2-diphenyl-1-picrylhydrazyl scavenging activity, 21% 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) scavenging capacity, and a 50% ascorbic acid equivalent ferric reducing antioxidant power value, Limosilactobacillus fermentum JNU532 was selected as the strain with the highest antioxidant potential. No cytotoxicity was observed in cells treated with the CFS of L. fermentum JNU532. Tyrosinase activity was reduced by 16.7% in CFS-treated B16F10 cells (but not in the cell-free system), with >23.2% reduction in melanin content upon treatment with the L. fermentum JNU532-derived CFS. The inhibitory effect of the L. fermentum JNU532-derived CFS on B16F10 cell melanogenesis pathways was investigated using quantitative reverse transcription polymerase chain reaction and western blotting. The inhibitory effects of the L. fermentum JNU532-derived CFS were mediated by inhibiting the transcription of TYR, TRP-1, TRP-2, and MITF and the protein expression of TYR, TRP-1, TRP-2, and MITF. Therefore, L. fermentum JNU532 may be considered a potentially useful, natural depigmentation agent.
We calculated dose rate using radiative transfer equations to consider radiative processes distinctly. The dose rate at Pohang(36°02'N, 129°23'E) was calculated using measured ozone and meteorological data and two-stream approximations(quadrature, Eddington, delta Eddington, PIFM(practical improved flux method), discrete ordinate, delta discrete ordinate) are used in solving equation. The purpose of this study is to determine the most compatible radiative transfer approximation for simulating the radiative and photochemical processes of atmosphere through comparision between calculated and measured values. Dose rate of the biologically effective irradiance in the region 0.28-0.32 U m showed the highest value when quadrature and Eddington was used and lower value on condition that delta scaling was applied. Correlation coefficient between dose rate at surface using radiation transfer equation and measured UV-B at Pohang was 0.78, 0.79 and 0.81 when delta Eddington, PIFM and delta discrete ordinate were used. Also, in case of above approximations were used, MBE(Mean Bias Error) was within -0.3MED/30min and RMBE(Relative Mean Bias Error) was below 10% between 1200 LST and 1400 LST Approximations which are compatible in estimating radiative process are delta Eddington, PIFM and delta discrete ordinate. Especially, in case that radiative process is considered more detail, delta discrete ordinate increased the number of stream is proper.

This study was done to ivestigate the fabrics thatminimized harmfulness of UVB(ultraviolet B) and that might product Vit. {{{{ {D }`_{3 } ^{ } }} by UVB. Twelve female subjects wearing in three different types i.e fabric A(UVB 100% protection) fabric B(UVB 50% protection) and bikinii were exposed to outdoor environment (Air Temp : 25℃, 42% R,H Air velocity : 0.13m/s UV does :6KJ/m2) Blood samples were taken 24 hours before the after the experiment in order to examine concentration of vit.{{{{ {D }`_{3 } ^{ } }} in the blood. During the experiment axillary temperature skin temperature of 7 areas(forehead Chest Upper arm, Hand Thigh Lower leg, Foot) were measured. The more irradiated areas by UVB were the more the concentration of serum 25(OH){{{{ {D }`_{3 } ^{ } }} were significantly. Mean skin temperature was significantly low levekl in wearing the fabric of UVB 50% protection (p<0.001) Axillary temperature was significantly high level in wearing the fabric of UVB 50% protection (p<0.001). Therefore the fabric of UVB 50% protection intercepts the radiation and has advantage to give off body heat over other fabrics
The interaction of bilobalide (BB) and ginkgolides B (GB) with bovine serum albumin (BSA) was investigated by fluorescent technique and UV/vis absorption spectroscopy. The results showed that BB and GB could intensively quench the fluorescence of BSA through a static quenching procedure. The binding constants (Ka) and the average binding distance between the donor (BSA) and the acceptor (ginkgolides) were obtained ($r_{BB}$ = 5.33 nm and $r_{GB}$ = 4.20 nm) by the theory of non-radiation energy transfer, and then the thermodynamic parameters such as ${\Delta}S^0$ (0.17-0.32 kJ/mol), ${\Delta}G^0$ (-20.76 ~ -17.79 kJ/mol) and ${\Delta}H^0$ (32.47-76.52 kJ/mol) could be calculated, respectively. All these results revealed that the interaction of BB and GB with BSA were driven mainly by hydrophobie force. The synchronous fluorescence spectroscopy was applied to examine the effect of two ginkgolides on the configuration of BSA. The configuration alteration of BSA could be induced by the hydrophobicitv environment of tyrosine with the increase of the drug concentration.
LRP15 is a novel gene cloned from lymphocytic cells, and its function is still unknown. Bioinformatic data showed that LRP15 might be regulated by DNA methylation and had an important role in DNA repair. In this study, we investigate whether the expression of LRP15 is regulated by DNA methylation, and whether overexpression of LRP15 increases efficiency of DNA repair of UV-induced DNA damage in HeLa cells. The results showed (1) the promoter of LRP15 was hypermethylated in HeLa cells, resulting a silence of its expression. Gene expression was restored by a demethylating agent, 5-aza-2'-deoxycytidine, but not by a histone deacetylase inhibitor, trichostatin A; (2) overexpression of LRP15 inhibited HeLa cell proliferation, and the numbers of cells in the G2/M phase of the cell cycle in cells transfected with LRP15 increased about 10% compared with controls; (3) cyclin B1 level was much lower in cells overexpressing LRP15 than in control cells; and (4) after exposure to UV radiation, the LRP15-positive cells showed shorter comet tails compared with the LRP15-negative cells. From these results we conclude that the expression of LRP15 is controlled by methylation in its promoter in HeLa cells, and LRP15 confers resistance to UV damage and accelerates the DNA repair rate.

UV induce oxidative stress and increase matrix metalloproteinase (MMP) expression, resulting in skin aging. Thus, preventing skin damage from ultraviolet B (UVB)-induced skin damage can attenuate skin aging. Spirulina is comprised of prokaryotes a powerful antioxidant. This study aimed to investigate the photoprotective effects of spirulina-derived phycocyanin (PC) against UVB radiation using human skin fibroblast. As a results, PC showed no toxicity at concentrations of 5-40 ㎍/mL in terms of fibroblast viability. Survival rate of UVB-irradiated fibroblast incresased to 73.5% from 50.5% with PC treatment. UVB treatment increased MMP-1 and MMP-9 expression whereas PC treatment decreased it. The results indicate that PC might reduce or prevent skin aging by reducing UVB irradiation-induced skin wrinkles and free radicals.

Portulaca oleracea L. is an edible plant widely consumed in daily diet throughout Europe, Asia and America. In this study, protective effects of P. oleracea L. extracts against oxidative stress and matrix metalloproteinase (MMP) activity induced by ultraviolet B (UVB) radiation were investigated using HaCaT immortal human keratinocytes. In this context, the mRNA and protein productions of MMPs (MMP-1, -2, and -9) and type I procollagen, which are major markers of photoaging induced by UVB radiation in HaCaT keratinocytes, were evaluated. Furthermore, UVB-induced reactive oxygen species (ROS) generation and mRNA and protein expression levels of superoxide dismutase-1 (SOD-1), oxygenase-1 (OH-1), and nuclear factor-erythroid 2-related factor-2 (Nrf-2), all of which are associated with the antioxidant balance, were investigated. As shown by the results, UVB radiation induced ROS formation and led to increased production of MMPs and decreased collagen production in human keratinocytes, which resulted in skin photoaging or photodamage. The treatment with P. oleracea L. extracts downregulated MMP (MMP-1, -2, and -9) production and upregulated type I procollagen expression in UVB-induced HaCaT cells. Furthermore, treatment with the extracts decreased UVB-induced ROS generation and increased the expression of antioxidant enzymes, such as SOD-1 and OH-1, through the Nrf-2 pathway. Taken together, these results suggest that P. oleracea L. extracts could be a potential cosmeceutical agent for the prevention of skin photoaging or photodamage.

In this study, the solution plasma process was utilized with the aim of degrading synthetic dyes in water at atmospheric pressure. The experiments were conducted in a batch-type reactor consisting of a symmetric wire-wire electrode configuration with rhodamine B (RhB) as the target synthetic dye. The effects of the plasma treatment time and initial dye concentration on the RhB degradation were investigated by monitoring the change in absorbance of RhB solutions. The RhB solutions turned lighter in color and finally colorless with prolonged plasma treatment time, indicating the destruction of dye molecules. The RhB solutions were found to have degraded, following the first-order kinetic process. However, for high initial RhB concentrations, another kinetic process or factor seems to play a dominant role at the initial degradation stage. The fitted first-order rate constant decreased as the initial concentration increased. This result suggests that the degradation behavior and kinetic process of the RhB solution strongly depends on its initial concentration. The RhB degradation is considered to be due to a combination of factors, including the formation of chemically oxidative species, as well as the emission of intense UV radiation and high-energy electrons from the plasma. We believe that the solution plasma process may prove to be an effective and environment-friendly method for the degradation or remediation of synthetic dye in wastewater.
Ultraviolet B (UVB) radiation may activate or deteriorate cultured human epidermal melanocytes, depending on the doses and culture conditions. In this study, we examined whether apoptosis of melanocytes can be induced by physiologic doses of UVB irradiation. PI staining for DNA condensation and flow cytometric analyses demonstrated the apoptotic cell death of melanocytes after UVB irradiation. The level of p53 and Bax revealed a dose-dependent increase with increasing dose of UVB, but the level of Bcl-2 remained unchanged. Confocal microscopic examination showed that Bax moved trom a diffuse to a punctate distribution after UVB irradiation. However, there were no changes in the pattern of Bcl-2. We next examined the downstream targets of apoptosis. Our results showed that a precursor form of caspase-3 disappeared with increasing doses of UVB. We also observed cleavage of poly(ADP-ribose) polymerase (PARP) after UVB irradiation. In addition, UVB irradiation resulted in a remarkable activation of c-Jun N-terminal kinase (JNK). These results indicate that UVB may induce apoptosis via JNK activation in human melanocytes.
