RSSCT를 이용한 GAC의 상수원수 내 용존유기물질 제거 (Removal of Dissolved Organic Matters in Drinking Water by GAC adsorption using RSSCT)

  • Granular activated carbon (GAC) has been identified as a best available technology (BAT) by the United States Environmental Protection Agency (USEPA) for removal disinfection by-product (DBP) precursors, such as dissolved organic carbon (DOC) and dissolved organic nitrogen (DON). Rapid small-scale column test (RSSCT) were used to investigate four types of carbon (F400, Norit1240, Norit40S, and Aquasorb1500) for their affinity to absorb natural organic matter (NOM). DOC, $UV_{254}$, and Total dissolved nitrogen (TON) concentrations were measured in the column effluent to track GAC breakthrough. DOC and $UV_{254}$ breakthrough occurred at around 3500 bed volumes (BVs) of operation for all GACs investigated. The $UV_{254}$ breakthrough curves showed 33% to 48% at 8000 BVs, when the DOC was 48% to 65%. All GACs showed greater removal in DOC than $UV_{254}$. The NORIT1240 GAC was determined to have the highest adsorption capacity for DOC and $UV_{254}$. The removal of nitrate (NOTN) had not broken through over BVs. The initial TON breakthrough curves were started around 50%, when the DOC breakthrough was only 10 % at 500 BVs. The curves were gradually increased after 3500 BVs and approximately 69% through 81% of TON breakthrough occurred at 8000 BVs. All of the GACs were able to remove TON, in the case of this investigation the majority of the TON was present as DON. Because nitrate nitrogen was seldom removed and ammonium nitrogen ($NH_3-N$) was not detected in the effluent from RSSCTs even though raw water. The carbon usage rate of DOC was from 2 to 6 times less than that of TON. The NORIT1240 GAC demonstrated the best performance in terms of DOC removal, while the F400 GAC was best in terms of TON removal. Excitation emission matrix(EEM) analysis was used to show that GAC adsorption successfully removed most of Humic-like DOC and Fulvic-like DOCs. However, soluble microbial product(SMP)-like DOC in the absence of raw water were detected in the NORIT40S and Aquasorb1500 GAC. The authors assumed that this results is due probably to the part of GAC in the RSSCT which was converted into biological activated carbon(BAC). To compare with organics removal by GAC according to preloading, the virgin GACs had readily accessible sites that were adsorbed DOC more rapidly than preloaded GACs, but the TDN removal had not showed differences between those GACs.

코치닐을 활용한 우피의 천연염색 (Natural dyeing of cow leather with cochineal)

  • The introduction of natural dyes into modern dye houses is very promising green chemistry concept that should be popularized more to reduce the dependency of leather dyeing on some toxic and non-biodegradable synthetic dyes. In this study, the properties of dyeing of cochineal on cow leather were evaluated. The proper dyeing conditions were identified with dye uptake (K/S values) depending on the colorant concentration, dyeing duration, dyeing temperature, and dye bath pH. For the proper mordanting conditions, color changes for different mordants were observed as $La^{*}b^{*}$ and H V/C values. Color fastness (light, rubbing, and dry cleaning) was also examined. Additionally, antibacterial properties and UV protection were examined. The results were as follows: The optimized dyeing conditions were 300% o.w.f., $40^{\circ}C$, 40 min., and pH 5. The cow leather color was red in the absence of mordanting, while it was red purple after being mordanted with Al and Cu, and purple mordanted with Fe. The K/S value of cow leathers increased in the order of the dyeing using $AlK(SO_4)_2$ > $CuSO_4$ > $FeSO_4$. The colorfastness to light and rubbing were reduced compared to original (untreated) cow leather. However dry cleaning fastness was very satisfactory, with a 4~5 rating. The dyed and pre-mordanting dyed cow leather showed excellent antibacterial properties.

Fabrication and Processing Method of Ophthalmic Hydrogel Tinted Lens Containing Indium Tin Oxide-Composited Materials

  • In this study, a multifunctional ophthalmic lens material with an electromagnetic shielding effect, high oxygen permeability, and high water content is tested, and its applicability is evaluated. Metal oxide nanoparticles are applied to the ophthalmic lens material for vision correction to shield harmful electromagnetic waves; the pyridine group is used to improve the antibacterial effect; and silicone substituted with urethane and acrylate is employed to increase the oxygen permeability and water content. In addition, multifunctional tinted ophthalmic lens materials are studied using lens materials with an excellent antibacterial effect (2,6-difluoropyridine, 2-fluoro-4-pyridinecarboxylic acid) and functional (UV protection, high wettability) lens materials (2,4-dihydroxy benzophenone, 2-hydroxy-4-(methacryloyloxy)benzophenone). To solve problems such as air bubbles generated during the polymerization process for the manufacturing and turbidity of the lens surface, polymerization conditions in which the defect rate is minimized are determined. The results show that the polymerization temperature and time are most appropriate when they are $110^{\circ}C$ and 40 minutes, respectively. The optimum injection amount of the polymerization solution is 350 ms. The turbid phenomenon that appears in lens processing is improved by 10 to 95 % according to the test time and conditions.

곰보배추 추출물로 염색한 면직물의 자외선 차단율, 소취성 및 항균성 (Study of UV Protection, Deodorization and Antimicrobial Properties of Cotton Fabrics Dyed with the Liquids Extracted from Salvia Plebia R. Br.)

  • This study examined the stainability and functionality of cotton dyed with the extract of Salvia Plebia R. Br. as a research preceding the development of health functional materials and fashion products with a healing motif. The CIELAB measurements of the cotton fabric dyed with the extract of Salvia Plebia R. Br. showed 73.32 for $L^*$, 1.7 for $a^*$, and 37.78 for $b^*$, while the Munsell measurements exhibited 2.63Y in color, 7.18 of brightness, and 5.49 of chroma. The degree of staining resulting from color fastness to laundering, and that resulting from color fastness to perspiration (acid and alkaline), as well as the level of color fastness to crocking (wet and dry) all stayed between relatively higher levels of 4 and 5. The fabric dyed also exhibited SPF 50+, an outstanding sun blocking performance. The deodorization rate was also excellent, rising from 97% to over 99% when the time elapsed increased from 30 minutes to 120 minutes. The fabric dyed also showed a 99.6% antimicrobial activity against staphylococcus aureus ATCC 6538, and a 71.4% antimicrobial activity against Klebsiella pneumoniae ATCC 4352. The aforementioned findings indicate that fabrics dyed with the extract of Salvia Plebia R. Br. contain the potential to be developed for use as health-related materials and fashion products that promote healing.

대장균 세포 내 다양한 외부 스트레스에 대한 DPS 단백질의 생리적 기능 (Physiological Function of a DNA-Binding Protein from Starved Cells in Combating Diverse External Stresses in Escherichia coli)

  • 대장균에서 DNA 결합 단백질로 확인된 DNA-binding Protein from Staved cells (DSP)는 DNA를 보호하는 중요한 기능을 한다는 것을 보여주었다. 이 연구의 목표는 야생형 대장균과 dps 유전자 결손 대장균(${\Delta}dps$ E.coli)의 특성 비교를 통해 여러 종류의 스트레스에 대해 대장균에서 DPS의 기능적 역할을 설명하는 것이다. 다양한 스트레스 상태에서 자외선 흡광도계(UV-spectrophotometer)를 이용하여 야생형 대장균과 dps 유전자 결손 대장균의 세포성장을 측정하였으며, 각각의 대장균 세포 성장 속도를 비교함으로써 우리는 대장균에 존재하는 DPS 단백질의 기능적 역할을 확인하였다. 야생형 대장균에 비해 dps 유전자 결손 대장균은 영양분 결핍, 산성화, 열충격, 다양한 활성산소종 스트레스들에 민감한 현상을 나타내었으며, 이것은 DPS가 다양한 극단적인 스트레스에 중요한 기능을 한다는 것을 제안하였다. 결론적으로 대장균의 DPS는 다양한 환경적인 스트레스로부터 DNA와 강하게 결합하여 유지함으로써 세포를 보호하고 세포성장에 결정적인 기능을 한다는 것을 증명하였다.

Lactobacillus plantarum에 의한 흰 민들레 발효물의 항산화 및 광노화 억제 효과 (Antioxidation and Anti-photoaging Effects of White Taraxacum Coreanum Extract by Lactobacillus plantarum)

  • 본 연구는 김치로부터 β-glucosidase 생산하는 Lactobacillus plantarum SM4를 분리하여 흰 민들레 발효를 수행하고 미백과 주름개선 효과를 가지는 생리활성 평가를 위해 총 폴리페놀 함량, 플라보노이드 함량, 항산화 활성, 타이로시네이즈 활성저해, 콜라게네이즈 활성저해, 자외선 차단율을 분석하였다. 흰 민들레 발효물의 총 폴리페놀 함량이 41.8±0.26 mg GAE/g DW로 열수추출물 21.4±0.67 mg GAE/g DW에 비해 2배 가량 증가하였으며 항산화 활성은 65.6±4.7%로 4배 증가하였다. 미백과 주름개선의 지표인 타이로시나아제 및 콜라게네이즈 활성저해에 있어 흰 민들레 발효물이 87.9±4.73%와 66.7±3.48%로 열수추출물에 비해 각각 2.4배와 1.5배 증가한 결과를 보였다. 흰 민들레 발효물과 열수추출물의 자외선 차단 효과를 비교하였을 때, UVA(320 nm) 차단율이 각각 64.7%와 15.2%로 측정되어 흰 민들레 발효물이 열수추출물에 비해 더 높은 효과를 나타내어 김치로부터 분리한 L. plantarum SM4가 생산하는 β-glucosidase를 발효를 통해 피부 미백, 주름 방지, 자외선 차단 효과가 증진됨을 확인하였다.

고효율 바이오 에탄올 생산을 위한 당화효소 개발 및 동시당화발효 공정 연구 (Development of Glucoamylase & Simultaneous Saccharification and Fermentation Process for High-yield Bioethanol)

  • 화석연료의 사용은 개발 한계성 및 고갈 그리고 지구온난화 등과 같은 심각한 문제를 지니고 있어 이를 극복하기 위해 환경 친화적이고 재생 가능한 바이오에탄올이 대두 대고 있다. 현재, 공정의 최적화와 미생물 개발을 통한 생산 단가를 낮추기 위한 많은 연구가 진행되고 있다. 본 연구에서는 쌀보리, 현미, 옥수수, 절간의 국내산 원료를 바탕으로 각각의 원료의 최적의 전처리 조건을 탐색하여 원료별 특성을 파악하여 동시당화공정발효를 이용하여 에탄올을 생산하였다. 또한 에탄올 생산성과 수율을 높이기 위하여 당화효소를 UV-mutation을 통해 개발한 SGA와 기존의 당화효소인 GA를 비교하였다. 그 결과 모든 원료에서 SGA가 GA보다 우수한 당화력을 나타내어 에탄올의 생산성과 수율을 향상 시켰다. 특히 가장 큰 차이를 보인 현미의 경우 에탄올 최대 농도가 1.25배 증가하여 18.4 g/L가 향상되었다. 또한 최대 농도의 95%도달시간을 비교해 본 결과 GA는 88시간, SGA는 35.98시간을 기록하였다. 이는 SGA의 당화력이 매우 우수함을 입증해준 결과라 할 수 있다.

구아바 잎 추출액을 이용한 직물의 천연염색 III - 한지면직물의 염색성과 기능성 - (Natural Dyeing of Fabrics with Guava (Psidium guajava L.) Leaf Extract III - Dyeability and Functional Property of Hanji Cotton Fabrics -)

  • The natural dyeing of hanji cotton fabrics with guava leaf extract was investigated. The temperature and time of dyeing were $40^{\circ}C$ and $90^{\circ}C$ for eighty minutes, respectively. In addition, the dyebath has been set at pH 5. Sn pre-mordanted fabrics showed the highest K/S value. Regardless of dyeing temperature, K/S values were high when Al, Cu, Fe-mordanted fabrics were dyed in post-mordanting and Sn-mordanted fabrics in pre-mordanting. The dyeing equilibrium was shown at the fourth time of repeated dyeing. In the processing of hanji cotton fabrics, K/S value was high when hanji cotton fabric was treated with soybean milk at $90^{\circ}C$. Similar K/S value of dyeing was shown when fabrics were processed with chitosan, regardless of dyeing temperature. High K/S value of dyeing was seen when fabrics processed with gallnut tannin at 40. Fabrics dyed with acidic water extract showed yellowish color. Fabrics dyed with alkaline water extract showed reddish color, and fabrics dyed with ethanol extract showed greenish yellow color. In case of Sn-mordanted fabrics, the washing fastness level was between 3 to 4, and other dyed fabrics showed low fastness. The dry cleaning fastness also showed very excellent result with level 4-5. The rubbing fastness of the fabrics was better in dry rubbing than in wet rubbing. For the light fastness, all dyed fabrics showed low fastness. For antibacterial activity, the dyed fabrics with guava leaf extract showed 99.9% of high antibacterial activity. Hanji cotton fabric maintained certain deodorization in the state of raw fabric. All dyed fabrics showed higher UV protection rate than control fabric.

For antibacterial activity, the powder of guava leaf extract and the dyed fabrics with guava leaf extract showed 99.9% of high antibacterial activity. All dyed fabrics showed higher deodorization and UV protection rate than control fabric.

  • The natural dyeing of silk fabrics with guava leaf extract was investigated. Mixed components of flavonoid and tannin seem to be the components of guava leaf extract. The temperature and time for dyeing of silk fabrics with guava leaf extract were $40^{\circ}C$ and $90^{\circ}C$ for eighty minutes, respectively. The dyeing equilibrium was shown at the fourth time of repeated dyeing. The highest K/S value was recorded at pH 3. Fe-mordanted fabrics showed the highest K/S value. High K/S values were shown in the post-mordanting of dyed fabrics at $40^{\circ}C$ and the pre-mordanting of dyed fabrics at $90^{\circ}C$. After repeated dyeing and mordanting, various color change occurred with mordant treatment. Surface colors were changed to YR color in alkaline water extract and to Y color in acidic water and ethanol extract, respectively. The washing fastness was level 3-4 for Sn-mordanted fabrics and level 4 for Fe-mordanted fabrics, and the dry cleaning fastness also showed very excellent result with level 4-5. The rubbing fastness was satisfactory with level 3-4. The light fastness was above level 4 only when Fe-mordanting was conducted, and the rest of dyed fabrics was not fast enough. In the antibacterial activity, the powder of guva leaf extract showed 99.9% of high antibacterial activity and Staphylococcus aureus showed 99.6% in dyed fabric. All dyed fabrics showed higher deodorization and UV protection rate than control fabric.
