Accumulation of Chlorogenic Acid as a near UV-shielding Compound in Cauliflower Grown under Enhanced UV-B Radiation

  • Since solar radiation contains wavelength essential for photosynthesis accompanying with near-UV light, UV-B effects on biological parameters and acclimation mechanisms are influenced by photosynthetically active radiation (PAR). Therefore, to elucidate near-UV shielding mechanism in higher plants, we cultivated cauliflower under usual solar radiation and increased UV-B from fluorescent lamps, two- or three-fold excess over continuously estimated UV-B dose in PAR during daytime, using computer regulated systems. Increased UV-B radiation had little effect on growth expressed as fresh weigh and leaf area. Water soluble low molecular weight compounds showing absorption in near UV region were enhanced according to the irradiated UV-B dose. One of compounds in cauliflower leaves was identified as chlorogenic acid. This was found to have no near-UV photosenSitizerable activity and is known to have an ability to scavenge a wide species of active oxygen. Another pro-oxidant compound that generates superoxide anion radical under near-UV irradiation was not induced by increased UV-B during cultivation, and identified as lumazine, a degradation product from folic acid.

용수재이용을 위한 하수처리 유출수의 UV 소독 효율 연구 (Feasibility Study of UV-Disinfection for Water Reuse of Effluent from Wastewater Treatment Plant)

  • The feasibility study of UV-disinfection system was performed for disinfection of effluent from wastewater treatment plant. Three low-pressure UV lamps of 17, 25, and 41 W were examined with various flow rates. Low-pressure UV lamps of 17W were examined with various turbidity, DOM (dissolved organic matter), and SS (suspended solid). The pilot plant was a flow-through type UV-disinfection system, and the range of exposure time varied from 5 to 40 seconds, turbidity from 0 to 40 NTU, DOM from 0 to 30 mg/L, and SS from 10 to 40 mg/L. The 41W lamp demonstrated complete disinfection showing no survival ratio in all the experimental conditions, and generally 17W and 25W lamps also showed high removal ratio over 97%. For the same UV dose (UV intensity times exposure time), high intensity-short exposure conditions showed better disinfection efficiency than low intensity-long exposure conditions. While the effects of turbidity and DOM were not apparent, the effects of SS was significant on the disinfection efficiency which indicates that SS control before UV-disinfection appears to be necessary to increase removal efficiency. Considering characteristics of effluent from existing wastewater treatment plants, cost-effectiveness, stable performance, and minimum maintenance, the flow-through type UV-disinfection system with high intensity and low-pressure lamps was thought to be a competitive disinfection system for wastewater reclamation.

  • The objective of this research was to examine the significance of change to indicator microorganisms (TC, FC, and E. coli) in the paddy field and repair of UV disinfected secondary effluent. Average concentrations of Microorganisms were maintained by more than about 1,000 MPN/100mL in paddy field after irrigation. Microorganism repair was evaluated in relation to UV dose in photoreactivating light and dark. In addition, a significant inverse relationship was found between UV dose and repair of indicator microorganisms when the research of wastewater was conducted in the condition of low and high UV dose.

복숭아 저장성 증진을 위한 저선량의 UV-C 조사 효과 (Application of Low Dose UV-C Irradiation for Shelf-Life Extension of Peach (Prunus persica L. Batsch))

  • 국내산 복숭아의 저장성 증진을 위하여 저선량(0~3.0 $kJ/m^2$)의 UV-C를 조사하고, 저장기간 동안 부패율, 이화학적 변화 및 기호도의 변화를 측정하였다. 부패율의 경우, 대조군은 4일 이후부터는 부패된 복숭아를 보이기 시작하였으며, 저장 8일에는 50.00%의 부패율을 나타내었다. 저선량의 UV-C 처리군은 대조군보다 낮은 부패율을 보였으며, 특히 1.0 및 2.0 $kJ/m^2$의 UV-C 처리군이 가장 낮은 부패율을 나타내었다. 무게 변화의 경우, 대체적으로 대조군 및 UV-C 처리군 모두 저장기간 동안 큰 차이를 보이지는 않았다. pH 변화에서는 UV-C 처리에 의한 차이를 보이지 않았으며, 저장기간 중 pH가 증가하였으나 모든 실험군에서 차이가 없는 것으로 나타났다. 경도의 경우, UV-C 처리 직후에는 모든 실험군에서 차이가 없었으나, 저장 중 대조군의 변화가 가장 많았으며, UV-C 처리군의 경도 변화는 적은 것으로 나타났다. 색도 변화(명도, 적색도 및 황색도)에서는 UV-C 처리 및 저장 중 모든 실험군에서 큰 차이를 보이지는 않는 것으로 나타났다. 맛, 향, 색에 대한 기호도의 변화에서는 UV-C 처리에 의한 변화를 보이지 않았으며, 저장 중 기호도는 감소하였으나 모든 실험군에서 차이가 없는 것으로 나타났다. 조직감 및 종합적 기호도에서는 UV-C 처리에 의한 변화를 보이지 않았으나 저장 중 대조군의 기호도값이 가장 많이 감소하였고, UV-C 처리군의 변화는 적은 것으로 나타났다. 전반적으로 대조군보다는 저선량의 UV-C 처리군이 기호도가 높은 것으로 나타났다.

Low-dose of Ultraviolet radiation-, Ethyl methanesulfonateor Bleomycin-lnduced Adaptive Response in Chinese hamster ovary Cells

  • The adaptive response and cross-adaptive response to sister chromatid exchanges (SCEs) and DNA single-strand breaks (SSBs) in Chinese hamster ovary (CHO)-K$_1$ cells treated with ultraviolet radiation (UV), ethyl methanesuffonate (EMS), or bleomycin (BLM) were investigated. Two assays were used in this study; SCEs and alkaline elution. The pretreatment with low conditioning dose of 2 mM EMS or 1 J/m$^2$ UV decreased the yield of SCEs induced by subsequent treatment with 8 mM EMS, 5 J/m$^2$ UV or 5 $\mu$g/ml BLM. And the pretreatment with low conditioning dose of 1 $\mu$g/ml BLM decreased the yield of SCEs induced by subsequent treatment with 5 $\mu$g/ml BLM or 5 J/m$^2$ UV. The rejoining of DNA SSBs in cells subsequently treated with 2 J/m$^2$ UV, 50 mM EMS or 400 $\mu$g/ml BLM is higher than that only treated with 2 J/m$^2$ UV, 50 mM EMS or 400 $\mu$g/ml BLM. These results suggest that there are the adaptive response and cross-adaptive response to SCEs, and is the adaptive response to the rejoining of DNA SSBs in CHO cells.

Effect of Low Dose Mutagens on Adoptive Response and Plasma Membrane Glycoconjugates in Sarcoma 180 Cells

  • The present investigation was performed to elucidate the effect of pretreatment with low dose ultraviolet radiation (UV) and ethyl methanesulfonate (EMS) on cell survival by trypan blue dye exclusion method and plasma membrane glycoconjugates by lectin-cytochemistry in sarcoma 180 (S180) cells. Pretreatment with 2 J/$m^2$ UV or 2 mM EMS increased the percentage of survival of cells subsequently treated with high dose UV (10 or 20 J/$m^2$) or EMS (10 or 20 mM), respectively. Staining intensity of concanavalin A (Con A) of the cells pretreated with 2J/$m^2$ UV or 2 mM EMS and subsequently treated with 10 or 20 mM EMS was stronger than that of the cellstreated with 10 or 20 mM EMS. These results suggest that there is an adaptive response on cell survival to EMS or UV in S180 cells. And the results show a change in mannose-containing glycoconjugates of plasma membrane in S180 cells pretreated with EMS or UV and subsequently treated with EMS.

Compositional changes in mycosporine-like amino acids induced by UV radiation: marine dinoflagellate Scrippsiella sweeneyae

  • The compositional changes in mycosporine-like amino acids (MAAs) were investigated in the marine dinoflagellate Scrippsiella sweeneyae exposed to four different spectral compositions and five relative intensities of UV-B (280-320 nm) to UV-A (320-400 nm) + photosynthetically available radiation (PAR: 400-700 nm). Neither dose nor wavelengths of UVR significantly affected the growth rates. UVR caused a significantly increase in cell volume. Cell volume in the >280nm treatment was more than two times greater at 6.8 % of UVR intensity. Production of UVR induced MAAs was dependent on the dose of UVR. However. the induction of MAAs was related to the cell growth. Greater induction of MAAs was observed at shorter wavelengths. The composition of MAAs varied with increasing light intensity of UVR.

Effects of Ginseng Protein on Relative Survival and Chromosome Aberration of UV Irradiated Cells

  • A ginseng protein fraction which has been reported to have radiation protective effect was purified from Korean ginseng and its effects on relative survival and chromosome aberration were studied in UV irradiated CHO-K1 cells. When the protein fraction $(100\;{\mu}g/ml)$ was added to the cells before UV irradiation at 4\;J/$m^2$,, the survival rates were increased to 53.8% from 40.6% in control. Addition of the protein $(100\;{\mu}g/ml)$ after UV irradiation at 4 and $8\;J/m^2$ raised the rates to 85.4 and 24.0% from 79.2 and 11.5% in control, respectively. When the ginseng protein $(800\;{\mu}g/ml)$ was added to the cells exposed to UV light at 10, 20, $30\;J/m^2$, the frequencies of chromosome aberration (CA) were reduced significantly to almost same level regardless of the UV dose increment and there was no significant difference between pre- and post-treatment. When the concentration of ginseng protein was increased from 200 to $800\;{\mu}g/ml$, at UV dose of 10, 20, $30\;J/m^2$ each, the CA frequencies were decreased consistently as the dose of ginseng protein increased, at all UV doses tested. Similar effects were observed in both cases of pre- and post-treatment. The data suggest that the protein may reduce cell damage caused by UV light, especially damage to DNA molecule, or play a role in repair processes of damaged DNA, to increase cell survival and reduce chromosome aberrations.

Inactivation of various bacteriophages by different ultraviolet technologies: Development of a reliable virus indicator system for water reuse

  • There is an urgent need to identify more reliable indicator systems for human pathogenic viruses in water reuse practice. In this study, we determined the response of different bacteriophages representing various bacteriophage groups to different ultraviolet (UV) technologies in real wastewater in order to identify more reliable bacteriophage indicator systems for UV disinfection in wastewater. Bacteriophage ${\varphi}X174$ PRD1, and MS2 in two different real wastewaters were irradiated with several doses of both low pressure (LP) and medium pressure (MP) UV irradiation through bench-scale UV collimated apparatus. The inactivation rate of ${\varphi}X174$ by both LP and MP UV was rapid and reached ${\sim}4{\log}_{10}$ within a UV dose of $20mJ/cm^2$. However, the inactivation rates of bacteriophage PRD1 and MS2 were much slower than the one for ${\varphi}X174$ and only ${\sim}1{\log}_{10}$ inactivation was achieved by the same UV dose of $20mJ/cm^2$. Overall, the results of this study suggest that bacteriophage MS2 could be a reliable indicator for human pathogenic viruses for both LP and MP UV disinfection in wastewater treatment processes and water reuse practice.

잔류 의약품류의 제거 및 미생물학적 안전성을 고려한 하수 재이용 기술로서의 UV 및 UV/$H_2O_2$ 공정의 적용성 (Applicability of UV and UV/$H_2O_2$ Processes in the Control of Pharmaceuticals and Personal Care Products and Microbiological Safety for Water Reuse)

  • 하수 재이용에 대한 관심이 급부상하고 있는 가운데, 재이용수의 미량물질 등에 의한 화학적 리스크와 더불어 미생물학적 안전성에 대한 고려가 요구되고 있다. 본 연구에서는 벤치스케일의 연속식 실험장치를 이용, 미량물질 중 최근 주목을 받고 있는 의약품류를 대상으로, UV처리공정의 의약품류 제거효과 및 미생물학적 안전성의 달성 가능성을 고찰하였다. 시험수로 이용한 하수 2차 처리수중에서는 항생물질, 해열진통제 등 38종의 의약품류가 수 ng/L에서 수백 ng/L의 범위로 검출되었으며, 이를 대상으로 하여 UV 및 UV/$H_2O_2$ 처리실험을 행한 결과, UV 단독처리에 의한 의약품류의 효과적인 제거에는 상당량의 UV조사량이 필요할 것으로 예상되었다. 반면, UV/$H_2O_2$ 공정의 경우 $H_2O_2$의 첨가농도를 약 1 mg/L에서 6mg/L까지 증가시킴에 따라 각 의약품류의 제거율은 점점 증가하는 것으로 나타났다. 한편, $923\;mJ/cm^2$의 UV 조사량과 6.2 mg/L의 $H_2O_2$를 병용한 UV/$H_2O_2$ 처리는 Naproxen(>89%)외 모든 의약품류의 농도를 90% 이상까지 감소시킬 수 있었다. 또한 이 운전조건은 현행 하수처리장 유출수중의 대장균군에 대한 규제농도($3,000/cm^3$)를 기준으로 하였을 때, 4~5 log의 불활성화를 달성할 수 있을 것으로 예상되어, 하수 재이용에 요구되는 California Title 22의 criteria를 만족시킬 수 있을 것으로 판단되었다.
