Dietary effect of green tea extract on hydration improvement and metabolism of free amino acid generation in epidermis of UV-irradiated hairless mice (자외선 조사와 병행된 녹차추출물 식이공급이 무모생쥐의 표피보습 개선 및 유리아미노산 생성 관련 대사에 미치는 영향)

  • Purpose: Ultraviolet (UV) irradiation decreases epidermal hydration, which is maintained by reduction of natural moisturizing factors (NMFs). Among various NMFs, free amino acids (AA) are major constituents generated by filaggrin degradation. This experiment was conducted to determine whether or not dietary supplementation of green tea extract (GTE) in UV-irradiated mice can improve epidermal levels of hydration, filaggrin, free AAs, and peptidylarginine deiminase-3 (PAD3) expression (an enzyme involved in filaggrin degradation). Methods: Hairless mice were fed a diet of 1% GTE for 10 weeks in parallel with UV irradiation (group UV+1%GTE). As controls, hairless mice were fed a control diet in parallel with (group UV+) or without (group UV-) UV irradiation. Results: In group UV+, epidermal levels of hydration and filaggrin were lower than those in group UV-; these levels increased in group UV+1% GTE to levels similar to group UV-. Epidermal levels of PAD3 and major AAs of NMF, alanine, glycine and serine were similar in groups UV- and UV+, whereas these levels highly increased in group UV+1% GTE. Conclusion: Dietary GTE improves epidermal hydration by filaggrin generation and degradation into AAs.

Effects of different UV-8 levels on the growth, photosynthesis and pigments in cucumber(Cucumis sativus L.) (UV-B 강도 변화가 오이(Cucumis sativus L.)의 생장, 광합성 및 색소에 미치는 영향)

  • To investigate the effects of different UV-B levels on plant growth, cucumber plants were subjected to three levels of biologically effective ultraviolet-B(UV--$B^{BE}$ radiation [daily dose : 0.03(No UV-B), 6.40(Low UV-B) and 11.30 (High UV-B) kJ $m^{-2}$, UV--$B^{BE}$] in the growth chambers for 3 weeks during the early growth period. High and low levels of UV-B irradiation drastically decreased both dry weight and leaf area, but increased specific leaf weight of cucumber. Plants subjected to UV-B resulted in 30% and 20% reduction of photosynthesis rate by high and low UV-B, respectively. However, respiration rate was not affected by the UV-B. With increasing UV-B intensity, total chlorophyll contents were decreased linearly, while the contents of flavonoid were increased linearly. These results suggest that the present levels of UV-B may affect the growth of cucumber plant compared with a UV-B-free condition. It is likely that the growth of cucumber will be affected by enhanced UV-B due to ozone depletion in the near future.

Characterization of Expression of UV-Inducible Gene (UV100 and UV150) in Caenorhabditis elegans (Caenorhabditis elegans에서 분리한 자외선 유도유전자 (UV100과 UV150)의 발현 및 특성에 관한 연구)

  • The present study intends to characterize the DNA damage-inducible responses in Caenorhabditis elegans. To study UV-inducible responses in C. elegans, two UV-inducible cDNA clones were isolated from C. elegans by using subtration hybridization method. To investigate the expression of isolated genes, UV100 and UV150, the cellular levels of the transcript were determined by Northern blot analysis after UV-irradiation. The transcripts of isolated gene increased rapidly and reached maximum accumulation after UV-irradiation. Compared to the message levels of control, the levels of maximal increase were approximately 2 folds to UV-irradiation. These results implied that the effects of damaging agents are complex and different regulatory pathways exist for the induction of these genes. To study the function of UV100 and UV150 gene in response to UV irradiation, we carried out a RNAi experiment and investigated the UV sensivity. This result indicated that UV100 gene involved in stage-specific repair pathway or regulated by development.

Effect of UV-B on fatty Acid Composition, Lipid Peroxidation and Polyamine in Kidney Bean(Phaseolus vulgaris L.) (UV-B가 강낭콩(Phaseolus vulgaris L.)의 지방산 구성, 지질과산화 및 polyamine 함량에 미치는 영향)

  • To investigate the effects of UV-B on fatty acid composition, lipid peroxidation and biochemical defense responses of plant, kidney bean (Phaseolus vulgaris L.) was subjected to enhanced UV-B irradiation [daily dose : 0.02.(No UV-B) and 11.36 (enhanced UV-B) $kJ\;m^{-2};UV-B_{BE}$] for 3 weeks. UV-B drastically inhibited both height and dry weight of kidney bean. The content of malondialdehyde significantly increased by about 50% after 3 weeks of UV-B irradiation. The ratio of unsaturated to saturated fatty acids of kidney bean was increased by UV-B irradiation. Three major polyamines of kidney bean leaves : putrescine, spermidine and spermine, were observed. All of the polyamine contents were increased with UV-B irradiation. These results suggested that enhanced UV-B radiation caused oxidative stress on lipids and biochemical protection responses might be activated to prevent from damaging effects of oxidative stress generated by UV-B irradiation.

Effects of Antioxidants on UV-B Susceptibility in Soybean (항산화 물질이 UV-B에 대한 콩의 감수성에 미치는 영향)

  • To determine whether the enhanced UV-B causes oxidative stress, and to test the relationship between plant growth response and biochemical defense response to UV-B, two soybean plants, Keunolkong, a highly UV-B susceptible cultivar, and Danyeubkong, a less UV-B susceptible cultivar, were subjected to the enhanced UV-B [daily dose : 0.06 (control) and 11.32 (enhanced UV-B) kJ $m^{-2}$ ; $UV-B_{BE}$] for 3 weeks. Contents of malondialdehyde and total carotenold were increased in Keunolkong compared with Danyeubkong by UV-B. In control plants, ascorbate level of Danyeubkong was 3 times higher than that of Keunolkong. The ratio of dehydroascorbate/ascorbate was highly increased in Keunolkong by UV-B . The activities of antioxidative enzyme such as superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase and glutathione reductase were increased in both cultivars by UV-B. This results indicate that enhanced UV-B caused oxidative stress in both two cultivars, especially in Keunolkong. Susceptibility of two soybean cultivars to UV-B is closely related to the levels of antioxidants such as carotenoid and ascorbate.

The Analysis of the UV cut-off effect of tinted lens and the Characteristic of UV tester (착색렌즈의 UV 차단효과와 UV TESTER 평가 분석)

  • We obtained the UV cut-off effect of tinted CR-39 Ophthalmic lens with the respect of concentration of green, brown, blue color. First, we analyzed the UV spectrum of tinted CR-39 lens measured by Spectrophotometer and then compared the UV data by Spectrophotometer with UV data by UV tester used in an optician shop. The result obtained show that the UV cut-off effect is increased with color concentration of tinted CR-39 lens, and the transmittance of 0%~5% at 400nm by Spectrophotometer is correspondence with transmittance of 0% by UV tester.

The effect of UV blocking lens on the denaturation of RNase A induced by UV-A (UV-A로 유발된 RNase A의 변성에 대한 UV 차단렌즈의 작용)

  • The aim of this study was to find the proper UV-A blocking percentage that could protect the denaturation of ribonuclease A (RNase A), one of protein enzymes in eye, induced by UV-A. RNase A was irradiated at 365 nm for 1, 3, 6, 24, 48, 72, 96 hr and the extent of denaturation was monitored by polyacrylamide gel electrophoresis. Furthermore, it was investigated whether blocking of UV-A by 20, 50, 80 and 99% eyeglass lens could protect the denaturation of RNase A or not. The denaturation of RNase A was induced by only 1 hr UV-A irradiation and the extent of denaturation became severe depending on UV-A irradiation time. The mild denaturation of RNase A induced by irradiation for 1 hr could be sufficiently protected by 20% UV-A blocking lens. When RNase A was irradiated for 3 hr, more that 50% blocking of UV-A needed to prevent the denaturation. Even though 99% UV-A blocking lens was used, the denaturation of RNase A induced by 6 hr irradiation could not be prevented perfectly. However, 99% UV-A blocking lens could dramatically decrease the severe denaturation of RNase A induced by irradiation for 96 hr.

Color and COD Removal of Rhodamine B Using Ozone, Photocatalyst and Ozone-Complex Process (오존, 광촉매 및 오존-복합 공정을 이용한 Rhodamine B의 색도와 COD 제거)

  • The effect of advanced oxidation processes such as $O_3$, $UV/TiO_2$, $O_3/UV$ and $O_3/UV/TiO_2$ on decolorization and COD removal of Rhodamine B(RhB) wastewater were considered. The results showed that the higher the $O_3$ concentration was, the higher the decolorization observed and the optimum $TiO_2$ dosage was 0.4 g/L in $UV/TiO_2$ and $O_3/UV/TiO_2$ process. $O_3/UV$ process showed the higher initial decolorization rate constant and the shorter termination time for decolorization than those of the $O_3$ process. The decolorization rate constants in various systems followed the order of $O_3/UV/TiO_2>O_3/UV>O_3{\gg}UV/TiO_2$. The decolorization rate of the RhB solution in every processes was more rapid than the mineralization rate identified by COD removal. The latter took longer time for further oxidation. The COD removal rate constants in four systems followed the order of $O_3/UV/TiO_2>O_3/UV>UV/TiO_2{\geqq}O_3$. Among four processes, combined photocatalysis and ozonation$(O_3/UV/TiO_2)$ was the most prospective process for removing color and COD such as dye wastewater.

Effect of Ultraviolet (UV-B) on Antioxidants and Antioxidative Enzymes in Garden Balsam(Impatiens balsamina L.) (자외선(UV-B)이 봉선화(Impatiens balsamina L.)의 항산화제 및 항산화 효소에 미치는 영향)

  • To investigate the effects of ultraviolet(UV-B) on growth and biochemical defense responses of plant, garden balsam (Impatiens balsamina L.) was subjected to enhanced UV-B irradiation [daily dose: 0.02 (No UV-B) and 11.34 (enhanced UV-B) kJ $m^{-2}$ ; $UV-B_{BE}$] for 3 weeks. Enhanced UV-B drastically inhibited leaf area as well as dry weight of garden balsam. The content of malondialdehyde was significantly increased by about 50% after 3 weeks of UV-B irradiation. The ratios of dehydroascorbate/ascorbate and oxidized glutathione/reduced glutathione were also considerably increased by UV-B irradiation. Three major polyamines of garden balsam leaves: putrescine, spermidine and spermine were observed. All polyamine contents were increased with UV-B irradiation. The enzyme (superoxide dismutase, ascorbate peroxidase etc.) activities of garden balsam were increased by the UV-B enhancement. Based on the results, enhanced UV-B caused oxidative stress in garden balsam and biochemical protection responses might be activated to prevent from damaging effects of oxidative stress generated by UV-B irradiation.

Isolation and Characterization of UV-Inducible Gene UV150 and UV200 in Eukaryotic Cells (진핵세포에서 DNA 상해에 반응하는 유전자 (UV150과 UV200) 기능연구 분리 및 특성 연구)

  • 본 연구는 DNA 상해유도기작을 규명하기 위하여 하등 진핵생물인 분열형 효모 Schizosaccharomyces pombe로부터 subtraction hybridization방법을 이용하여 자외선 유도 유전자인 UV150과 UV200을 분리하고 그 유전자 구조와 발현양상을 조사하였다. 분리한 유전자의 발현양상을 Northern hybridization 방법으로 살펴본 결과 자외선 조사 1시간 후부터 발현이 증가되었다. 또한 알킬화제인 Methyl Methanesulfonate (MMS) 처리에 의해서도 발현이 증가되었다. 이 결과 다른 UV-inducible유전자와는 다르게 분리한 UV150유전자는 UV에 UV200유전자는 MMS에 의하여 발현이 증가됨을 알 수 있었다. 유전자의 기능을 알기 위하여 URA4 유전자를 이용하여 null-mutant 세포주를 제조하여 그 특성을 살펴본 결과 분리한 UV150 유전자는 세포의 성장에 필수적인 유전자임을 알 수 있었다.
