Development of a novel genetic assay for telomere recombination in Saccharomyces cerevisiae (효모에서 텔로미어 재조합을 관찰하기 위한 새로운 유전학적 연구방법의 개발)

  • Stable maintenance of telomere is required for cell proliferation and survival. Although telomerase is the primary means for telomere maintenance, recombination is another important pathway to maintain telomeres. In this study, we developed a genetic assay for telomere recombination using the internal $TG_{1-3}$ repeats present in subtelomeric regions of yeast. The recombination frequencies were dependent on the presence of the internal $TG_{1-3}$ repeats. PCR amplification of the regions near URA3 and CAN1 markers using genomic DNA isolated from $FOA^rCan^r$ colonies indicated that each isolate had lost the chromosome end including the markers. In addition, the recombination frequencies increased with longer internal $TG_{1-3}$ repeats. Our results suggest that the $FOA^rCan^r$ colony formation is the consequence of recombination between the internal and terminal $TG_{1-3}$ repeats.

  • Needle type and growth characteristics of Cryptomeria japonica planted in Cheju, Korea, were investigated and compared with those of the same species distributed in Japan. Needle types were classified into nine lines, and the stands were composed of more than six lines. The trees of the same needle type as Ura-Sugi (Cryptomeria japonica) in Japan showed the best growth in height.

Characterization of the cloned RNA1 gene of Saccharomyces cerevisiae (Cloning된 효모의 RNAI 유전자의 특성에 관하여)

  • The RNAI mutation of Saccharomyces cerevisia is a recessive and temperature sensitive lethal mutation which interferes with the production of mRNA, rRNA, and tRNA. However, the precise role of RNAI gene have not been revealed until yet. We have cloned rna1-1 mutant gene from rna1-1 mutant yeast strain(R49 ; trpl, ura3-52, rna1-1). The 3.4kb BglII fragment of wild type RNAI clone(81-2-6) contains whole RNAI gene. The genomic southern blotting with BglII digested R49 genomic DNA as a probe shows the unique and identical band with wild type 3.4kb BglII fragment. Therefore, We prepared partial BglII genomic library(3~4kb BglII fragments) into BamH I site of pUC19. The rna 1-1 mutant clone was screened with Digoxigenin(DIG)-lableled probe by high density colony hybridization. The 5'-flanking region of rna1-1 gene was sequenced by dideoxy chain termination method. The 5'-flanking sequence of RNAI gene contains three TATA-like sequence ; TAATA, TATA and TTTTAA at position of -67, -45, and -36 from first ATG codon respectively. The 5'-flanking region of wild type RNA I gene from ATG codon to -103nt was deleted with Bal31 exonuclease digestion, generating $pUC{\Delta}$/RNA I. After constructing $pYEP{\Delta}RNA$ I (consists of -103nt deleting RNA I gene, URA3 gene, $2{\mu}m$ rep. origin), pYEPrna1-1(consists of Xba I fragment of pUCrna1-1. URA3 gene, $2{\mu}m$ rep. origin), and pYEPRNAI. each plasmid was transformed into host strain(trpl, ura3-52, rna1-1) by electroporation, respectively. Yeast transformant carrying $pYEP{\Delta}RNA$ I did not complement the thermal sensitivity of rna1-1 gene. It means that TATA-like sequences in 5'-flanking region is not TATA sequence for transcribing RNAI gene and there may be other essential sequence in upstream region for the transcription of RNAI gene.

  • The objective of this study is to evaluate corrosive wear resistance of metals used for bearings and gears in seawater. Sliding wear test of ferrous and copper materials against $Al_2O_3$ were carried out in artificial seawater using an electrochemical potentiostat. As the results, the wear rate and the coefficient of friction of the copper materials are lower than those of the ferrous materials. The corrosive wear of stainless steel is remarkably affected by normal load and sliding speed in view of tribological characteristics including adhesion and corrosion products.

  • The synthesis and evaluation of a novel calix[4]arene-based fluorescent chemosensor 1 for the detection of I. is described. The fluorescent changes observed upon addition of various anions show that 1 is selective for I. over other anions. Addition of I. results in ratiometric measurements with 1 : 1 complex ratio.

  • Three kinds of acrylic polymers containing silk protein were synthesized, which are (1) blending of silk fibroin (SF) fiber and polyacrylonitrile (PAN); (2) graft-copolymer of PAN onto SFs; (3) random-copolymer synthesized by copolymerization of acrylonitrile (AN) and silk fibroin peptide (SFP) with vinyl groups, and their solubility, thermal property, and moisture absorption was investigated, respectively. These polymers have difference solubility and attributable to their structure. Their excellent thermal stabilities and better moisture absorptions were indicated.

  • A series of novel spinning acrylic polymer containing silk protein were synthesized by copolymerization of acrylonitrile (AN) and silk fibroin peptide (SFP) modified by acryloyl chloride (AC) with vinyl groups. The viscosity values of these copolymers showed that the copolymers have good spinnability, which are synthesized under the condition of putting a micro amount of metal ions into synthesizing solvent. The fiber based on the poly (AN-co-SFP) was prepared and characterized by SEM, FTIR measurement of its shell and core flakes, and moisture absorption. The fiber appeared a smooth surface and could be assumed to have excellent adhesive between SFP and PAN. Furthermore, these fibers showed a shell-core structure and excellent moisture absorption.

  • For transformation of Pleurotus ostreatus, two novel vectors, pPhKM1 and pPhKM2, were constructed, using the regulatory sequences of the P. sajor-caju $\beta$-tubulin gene(TUB1) and the ble gene encoding phleomycin binding protein. pPhKM1 contains ble fused to the TUB1 promoter and the Schizophyllum commune GPD terminator. pPhKM2 contains ble fused to the promoter and terminator regions of P. sajor-caju TUB1. To confirm phleomycin-resistance activity, each vector was cotrans-formed with pTRura3-2 into the P. ostreatus homokaryotic $ura^-$ strain. The transforming DNA was stably integrated into the genomic DNA. Subsequently, phleomycin resistance was conferred on wild-type dikaryotic P. ostreatus by transformation with pPhKM1 or pPhKM2. This transformation system generated stable phleomycin-resistant transformants.

  • 본 논문에서는 휘어지거나 굴곡진 array인 3차원 conformal array의 beam pattern을 보정하고자 기존의 2차원에서 3차원으로 확장한 interpolation technique과 compressive sensing을 이용하여 3-D uniform rectangular array(3-D URA)에 적용하는 방법을 연구하였다. 시뮬레이션 결과는 compressive sensing이 interpolation technique보다 우수한 특성을 보여준다.

  • Based on input and output data pairs of nonlinear systems, this paper proposes a simple and effective Just-In-Time (JIT) method, called Direct JIT Control, for nonlinear control design. It uses an inverse model of controlled plant to compute an initial control action, and then adapts the initial control action by adding a fine-tuning control action, depended on the errors between the real outputs and the expected reference signals. Meanwhile, the proposed JIT method accomplishes the adaptation of the inverse model just simply by means of the refreshment of input and output data pairs.
