Isolation and Identification of ura5 Gene in Entomopathogenic Fungus, Metarhizium anisopoliae (살충성곰팡이 Metarhizium anisopliae의 ura5 유전자의 분리동정)

  • About 250 bp ura5 gene (Orotate phosphoribosyl transferase) fragment was cloned from genomic DNA of entomopathogenic fungus Metarhizium anisopliae by using PCR method. Entire nucleotide sequences of cloned DNA fragment were determined and analysed as compared with other fungus ura5 genes. The amino acid sequence deduced from the nucleotide sequence showed 85.5% homology to ura5 protein of Trichoderma reesei. Using this 250 bp PCR fragment we have isolated full ura5 gene of M. anisopliae by genomic Southern hybridization and the isolated 4.4 kb DNA fragments were mapped by restrictional enzyme.

Cloning and Characterization of the Orotidine-5'-Phosphate Decarboxylase Gene (URA3) from the Osmotolerant Yeast Candida magnoliae

  • We determined the nucleotide sequence of the URA3 gene encoding orotidine-5'-phosphate decarboxylase (OMPDCase) of the erythritol-producing osmotolerant yeast Candida magnoliae by degenerate polymerase chain reaction and genome walking. Sequence analysis revealed the presence of an uninterrupted open-reading frame of 795 bp, encoding a 264 amino acid residue protein with the highest identity to the OMPDCase of the yeast Kluyveromyces marxianus. Phylogenetic analysis of the deduced amino acid sequence revealed that it shared a high degree of identity with other yeast OMPDCase homologs. The cloned URA3 gene successfully complemented the ura3 null mutation in Saccharomyces cerevisiae, revealing that it encodes a functional OMPDCase in C. magnoliae. An enzyme activity assay and reverse transcription polymerase chain reaction indicated that the expression level of the C. magnoliae URA3 gene in S. cerevisiae was not as high as that of the S. cerevisiae URA3 gene. The GenBank accession number for C. magnoliae URA3 is JF521441.

Construction of Yeast Strain Suitable for Bioethanol Production by Using Fusion Method (융합법을 이용한 바이오에탄올 생산에 적합한 효모균주의 구축)

  • To construct useful yeast strain for bioethanol production, we improved yeast harboring various phenotypes by using yeast protoplast fusion method. In this study, S. cerevisiae BYK-F11 strain which have ethanol tolerance, thermotolerance and ${\beta}-glucanase$ activity and P. $stipitis{\Delta}ura$ strain which has xylose metabolism pathway were fused by genome shuffling. P. $stipitis{\Delta}ura$ strain was constructed for protoplast fusion by URA3 gene disruption, resulting in uracil auxotroph. By protoplast fusion, several fused cells were selected and BYKPS-F8 strain (fused cell) showing both karyotypes from two parent strains (S. cerevisiae BYK-F11 and P. $stipitis{\Delta}ura$ strain) among 22 fused cells was finally selected. Sequentially, various phenotypes such as ${\beta}-glucanase$ activity, xylose utility, ethanol tolerance, thermotolerance and ethanol productivity were analyzed. The BYKPS-F8 strain obtained ${\beta}-glucanase$ activity from BYK-F11 strain and 1.2 fold increased xylose utility from P. $stipitis{\Delta}ura$ strain. Also, the BYKPS-F8 strain showed thermotolerance at $40^{\circ}C$ and increased ethanol tolerance in medium containing 8% ethanol. In this fused cell, 7.5 g/l ethanol from 20 g/l xylose was produced and the multiple phenotypes were stably remained during long term cultivation (260 hr). It was proved that novel biological system (yeast strains) is easily and efficiently bred by protoplast fusion among yeasts having different genus.

  • A ura5 gene encoding Orotate Phosphoribosyl Transferase (OPRTase) of Metarhizium anisopliae was cloned by PCR methods and sequenced. The sequenced ura 5 gene encodes a polypeptide of 234 amino acid residues. This deduced amino acid sequence showed high similarity to other fungi OPRTase and there was no intron sequence between ATG starting codon and TGA ending codon.

  • 역할기반 접근제어(RBAC)는 역할(Role)과 역할계층(Role hierarchy)을 통해 사용자 및 접근권한 관리를 효율적으로 수행할 수 있도록 해준다. 그러나 시스템에 수많은 사용자, 역할, 권한이 존재하는 경우 한사람의 보안 관리자가 이들을 모두 관리하는 컷은 불가능하므로 역할을 관리하는 관리역할을 두어 시스템을 효율적으로 관리할 수 있는 방법(ARBAC)이 제안되었다. ARBAC는 URA(User Role Assignment), PRA(Permission Role Assignment), RRA(Role Role Assignment)로 구성되어있다. 본 논문에서는 URA99 모델을 기반으로 사용자-역할 관리를 위하여 관리도구를 구현한다. 구현된 관리도구는 오라클의 저장 프로시저를 사용하고 자바를 기반으로 한 EJB 컴포넌트로 구현한다.

Polyethyleneimine Derivative for Nucleic Acid Model

  • Water-soluble polyethyleneimine (PE) derivatives containing nucleic acid bases and hydrophilic amino acids such as homoserine (Hse) and serine were prepared by the activated ester method as nucleic acid models. From spectroscopic measurements, the polymers were found to interact with DNA accompanied by an induction of conformational change. Hypochromicity in UV spectra indicated that a stable polymer complex was formed between poly (A) with PEI­Hse-Ura by complementary hydrogen bonding with equimolar nucleic base units (adenine:uracil=1:1). The induced conformation of DNA by the interaction with the polymer containing uracil and homoserine (PEI-Hse-Ura) was concluded to be a super triple helical structure. The formation of the polymer complex, DNA: PEI-Hse-Ura, was found to be affected by the presence of metal ions such as $Ca^{2+}\;and\;Cu^{2+}$.

DOA-based Beamforming for Multi-Cell Massive MIMO Systems

  • This paper proposes a direction-of-arrival (DOA)-based beamforming approach for multi-cell massive multiple-input multiple-output systems with uniform rectangular arrays (URAs). The proposed approach utilizes the steering vectors of the URA to form a basis of the spatial space and selects the partial space for beamforming according to the DOA information. As a result, the proposed approach is of lower computational complexity than the existing methods which utilize the channel covariance matrices. Moreover, the analysis demonstrates that the proposed approach can eliminate the interference in the limit of infinite number of the URA antennas. Since the proposed approach utilizes the multipaths to enhance the signal rather than discarding them, the proposed approach is of better performance than the existing low-complexity method, which is verified by the simulation results.
