Depletion of Cytoplasmic Tail of UL18 Enhances and Stabilizes the Surface Expression of UL18

  • Background: Human cytomegalovirus UL18, a MHC class I homologue, has been considered a natural killer (NK) cell decoy. It ligates LIR-1/ILT2 (CD85j), an NK inhibitory receptor, to prevent lysis of infected target cells. However, precise role of UL18 to NK cell cytotoxicity is yet elusive. Difficulty in clarifying the function of UL18 lies in complication in detecting UL18 mainly due to low level expression of UL18 on the surface and gradual loss of its expression. Methods: To overcome this hurdle, cDNA of cytoplasmic tail-less UL18 was constructed and expressed in swine endothelial cell (SEC). The expression level and its stability in the cell surface were monitored with FACS analysis. Results: Surface expression of UL18 is up-regulated by removing cytoplasmic tail portion from UL18F (a full sequence of UL18). SECs transfected with a cDNA of UL18CY (a cytoplasmic tail-less UL18) stably expressed UL18 molecule on the surface without gradual loss of its expression during 6 week continuous cultures. In the NK cytotoxicity assay, UL18 functions either inhibiting or activating NK cell cytotoxicity according to the source of NK cells. We found that there is individual susceptibility in determining whether the engagement of NK cell and UL18 results in overall inhibiting or activating NK cell cytotoxicity. Conclusion: In this study, we found that cytoplasmic tail is closely related to the regulatory function for controlling surface expression of UL18. Furthermore, by constructing stable cell line in which UL18 expression is up-regulated and stable, we provided a useful tool to clarify exact functions of UL18 on various immune cells having ILT2 receptor.

A comparison study of Wumul-maru components in upper class Chosun dynasty (조선시대 상류주택의 사랑대청 우물마루에 관한 비교 연구)

  • The purpose of this study was to compare with the characteristics of Wumul-maru components in upper class Chosun dynasty. Physical trace method was used for this study. The samples were taken from the Sarang-taechong of 6 traditional Korean houses, Yunkyungdang, the ancient Chusa estate, Sunkyojand, Chunghyodang, Yangjindang, Unjoru. The major findings were summarized as follows; 1) Regarding to the scale characteristics, the number of changguit'ul(long board) was 1, chungguit'ul(middle-length board) 5, tongguit'ul(center board) 8m and maruno. 98, and the size of changguit'ul was 253$\times$4478mm, chungguit'ul 182$\times$2396mm, tongguit'ul 179$\times$2310mm, and marunol 247$\times$574mm. 2) Regarding to the proportion characteristics, the proportion of changguit'ul was 1:18.2, chungguit'ul 1:16.7, tongguit'ul 1:12.9, and marunol 1:2.62 and the proportion of components was 1:3:4:17.

  • Recently, I presented Gagok(歌曲) Collection Yeong-eon永言 to learned circles. Yeong-eon is very similar to Yukdang-version六堂本 CheongGuYeongeon靑丘永言. Compared with Cheong- Yuk, it is the same age or little bit early time of Cheong-Yuk in culture Icon. This paper paid attention to the considerable difference between Yeong-eon and Cheong-Yuk in the way of the distribution of Yisakdaeyap二數大 葉's Woo mode羽調 and Ke-myeon mode界面調. There was the way of gathering ‘real name’+‘namelessness’ in Yisakdaeyap, which is the feature of the 18th century Gagok Collection. I found this way just put on the 19th century Gagok Collection way which is the distribution of Yisakdaeyap's 'Woo mode and Ke-myeon mode' in CheongYuk. Then I proved in this paper that the way of gathering Yisakdaeyap in Cheong Yuk didn't correspond to an actual singing in the early 19th century when 'Woo mode and Ke-myeon mode' was fixed. In case of Yeong-eon, however, it was not written any writers' names at all, when it was researched retroactively, I knew it was distributed evenly both the works of 'real name' and 'namelessness' in Yisakdaeyaps 'Woo mode and Ke-myeon mode'. Consequently, I found Yeong-eon is the good Gagok Collection for an actual singing at that time. In addition, there was discord in the mode or key distribution among many Gagok Collections. I found this issue of the application had kept on make Gagok Collections edit during 2 centuries. Because the actual Gagok enjoyment in the specific time is connected the way of the mode application directly.

The Detection of ICD p24 Antigen Predicts Bad Prognosis in HIV-1 Infected Patients (인면역결핍바이러스 감염자에서 ICD-p24 항원 탐지가 CD4+T 세포수 및 예후에 미치는 영향)

  • In order to evaluate the effect of viral load on the prognosis of human immunodeficiency virus-1 (HIV-1)-infected individuals, immune complex dissociated (ICD) serum p24 antigen (p24) by acid treatment was retrospectively measured for 50 HIV-infected patients for 60 months. Among them, 27 patients were p24 positive (p24+) above 25pg/ml for $40.4{\pm}12$ months and 23 patients were negative (p24-). Follow-up periods from HIV diagnosis were $63.0{\pm}19$ months (range; 40-112) for the p24+ and $68.4{\pm}19$ months (range; 38-106) for the p24-, respectively (P>0.05)Mean CD4+T cell counts in the p24+ group decreased from $473{\pm}$277/ul (median;373) to $157{\pm}150/ul$ (median; 111) for $60{\pm}16$ months (5.3/month P280/ul (median; 476) to $432{\pm}285/ul$ (median;382) for $63{\pm}19$ months (2.5/month, P<0.01). From CD4+T cell count >200/ul, the patient who progressed to AIDS of <200/ul were 13 of 23 (56%) in the p24+ and 4 of 22 (18%) in p24-, respectively (p<0.01). And the number of death in two groups were 6 (22%) and 1 (4%), respectively (p<0.01). Presumed survival in two groups were about 12 and 24.5 years. These data suggest that viral load itself be very important for the prognosis of HIV-infected patients.

User Scheduling Algorithm for Cell Capacity Maximization in Full Duplexing Wireless Cellular Networks (전이중 무선 셀룰라 네트워크에서 셀 용량 최대화를 위한 사용자 스케쥴링 방식)

  • In this paper, we consider a full duplexing (FD) wireless cellular network where a central base station (BS) works in the FD mode while the downlink (DL) and uplink (UL) users work in the time division duplexing (TDD) mode. Since this FD system induces the inter-user interference from UL user to DL user, the main challenge for maximizing the system performances is user scheduling that makes a pair of DL user and UL user to use the same radio resource simultaneously. We formulate an optimization problem for user pairing to maximize the cell capacity and propose a suboptimal user scheduling algorithm with low complexity. This scheduling algorithm is designed in a way where the DL user with a better signal quality has a higher priority to choose its UL user that causes less interference. Simulation results show that the FD system using the proposed user scheduling algorithm achieves the optimal performance and significantly outperforms the conventional TDD system in terms of the cell capacity.

A Study on the Wumul-maru Components of Sarang-taechong in traditional Houses (전통주택 사랑대청의 우물마루 구성요소에 관한 연구)

  • The purpose of this study was the wumul-maru components of Sarang-taechong with themselves in traditional houses. Physical trace method was used for this study. The samples were taken from the Sarang-taechong of 6 traditional Korean houses, Yunkyungdang, the ancient Chusa estate, Sunkyojang, Chunghyodang, Yangjindang, Unjoru. The major findings were summarized as follows; 1) Regarding to the changgui't'ul(long board); the number was from 0(minimum) to 3(maximum), the size was $244.3{\times}4407.1mm$ on average, and the proportion was 1:18.9. 2) Regarding to the tongguit'ul(center board); the number was from 5(minimum) to 18(maximum), the size $188.7{\times}2374mm$ on average, and the proportion was 1:12.9. 3) Regarding to the marunol; the number was from 42(minimum) to 155(maximum), the size was $247{\times}574mm$ on average and the proportion was 1:2.29.

Primary Orbital Lymphoma : A Retrospective Analysis of Results of Radiation Therapy (원발성 안와 림프종의 방사선치료 성적에 관한 후향적 분석)

  • Purpose : This study evaluated the treatment outcomes, patterns of failure, and treatment related complications of primary lymphoma patients who received definitive radiation therapy. Materials and Methods : A retrospective analysis was undertaken for 31 patients with primary orbital lymphoma at the Asan Medical Center between February 1991 and April 2001. There were 18 males and 13 females with ages ranging from 3 to 73 years (median, 44 years). The involved sites were 9 conjunctivae, 12 eyelids and 10 other orbits. The histological types were 28 MALT lymphomas (low-grade B-cell lymphoma of mucosa-associated lymphoid tissue type), 1 diffuse large B-cell lymphoma, 1 anaplastic large cell lymphoma and 1 lymphoblastic lymphoma. The Ann Arbor stages were all IE $(100\%)$. Ann Arbor stage III or IV patients were excluded from this study, Bilateral orbital involvement occurred in 6 cases. Radiation therapy was given with one anterior port of high energy electrons $(6\~16\;MeV)$ for the lesions located at the anterior structures like the conjunctivae or eyelids. Lesions with a posterior extension or other orbital lesions were treated with 4 or 6 MeV photons with appropriately arranged portals. In particular, lens blocks composed of lead alloy were used in conjunctival or eyelid lesions. Twelve patients received chemotherapy. The median follow-up period was 53 months. Results : The 5-year overall, cause-specific, and disease-free survival was $91\%,\;96\%,\;and\;80\%$, respectively. The complete response rate 6 months after radiation therapy was $100\%$. Local recurrences were observed in 2 patients at 16 and 18 months after completion of radiation treatment. They were salvaged with additional radiation therapy. Two patients developed distant metastases. A MALT lymphoma patient with a lung relapse was successfully salvaged with radiotherapy, but the other lymphoblastic lymphoma patient with bone marrow relapse expired. There were no severe complications but 5 patients developed radiation-induced cataracts and 2 patients developed dry eye. Conclusion : Most primary orbital lymphomas consisted of MALT lymphomas. Radiation therapy was a successful treatment modality for orbital lymphoma without any severe complications. In cases of local relapses, radiation therapy is also a very successful salvage treatment modality.

Effects of Cryo-extenders for Spermatozoa Sorted by Percoll on In Vitro Fertility of in Miniature Pigs (Percoll 분리된 미니돼지 정액의 체외 수정 능력에 있어서 동결보존액의 영향)

  • The objective of this study was to evaluated the efficiency on sperm cryosurvival and ability of in vitro fertilization using Triladyl and Lactose Egg-Yolk(LEY) as extenders for cryopreservation of separated sperm by 65% percoll in miniature pig. Sperm viability was measured with SYBR-14/PI double stained sperm by flow cytometry. Ability on embryo cleavage rate and blastocyst development were observed by in vitro fertilization after frozen-thawing of sperm separated by 65% percoll. The experimental groups were designed that separated sperm by 65% percoll with Triladyl (ST) or LEY(SL) and unseparated sperm with Triladyl(UT) or LEY(UL) for cryopreservation. As a results, the viability was significantly(p<0.05) higher in ST(55.1%), SL(63.1%), UL(58.8%) than UT(38.2%) group. Sperm viability in SL(63.1%) group was significantly(p<0.05) higher than other experimental groups. On the other hand, embryo cleavage rate was significantly(p<0.05) higher in ST(79.1%), SL(83.2) than UT(74.1) and UL(75.7%) groups at 96h after in vitro fertilization. Blastocyst development was also significantly(p<0.05) higher in ST(21.5%), SL(20.9%) than UT(17.0%) and UL(18.8%) groups. In conclusion, cryopreservation of miniature boar sperm separated by 65% percoll were beneficial to viability and capacity on in vitro fertilization.
