Separation and flux characteristics in cross-flow ultrafiltration of bovine serum albumin and bovine hemoglobin solutions

  • The flux behavior in the separation of equimolar bovine serum albumin (BSA) and bovine hemoglobin (HB) in aqueous solutions by cross-flow ultrafiltration (UF) was investigated, in which polyacylonitrile membrane with a molecular weight cut-off (MWCO) of 100 kDa was used. BSA and HB have comparable molar mass (67,000 vs. 68,000) but different isoelectric points (4.7 vs. 7.1). The effects of process variables including solution pH (6.5, 7.1, and 7.5), total protein concentration (1.48 and 7.40 ${\mu}M$), transmembrane pressure (69, 207, and 345 kPa), and solution ionic strength (with or without 0.01 M NaCl) on the separation were examined. It was shown that the ionic strength had a negligible effect on separation performance under the conditions studied. Although BSA and HB are not rigid bodies, the flux decline in the present cross-flow UF did not result from the mechanism of cake filtration with compression. In this regard, the specific cake resistance when pseudo steady-state was reached was evaluated and discussed.

Selective Separation of Trypsin by Affinity Polymer and Ultrafiltration Membranes (친화성 고분자 및 한외여과 분리막을 이용한 트립신의 선택적 분리에 관한 연구)

  • The fed-batch process which combinded high selectivity of affinity chromatography and membrane process was developed. The mixture of trypsin and chymotrypsin, having almost the same molecular weight and the chemical structure, were used as model enzymes. The water soluble polymer having more affinity for trypsin and celluose acetate membrane gelated in 50vol.% ethanol for removing free enzymes and retentating trypsin-affinity polymer complex simutaneously were used in this system. The membrane pore size was controlled by ethanol concentration in the gellation bath, and the affinity polymer was prepared by polymerization of acrylamide with N-acryloyl-m-aminobenzamidine at $4^{\circ}C$. The trypsin could be effectively concentrated by utilizing an affinity polymer and a prepared UF-50 ultrafiltration membrane. As a result, 86% purity trypsin was recovered by the current purification process.

  • Maillard reaction products like melanoidins present in industrial fermentation wastewaters are complex compounds with various functional properties. In this work, novel ultrafiltration (UF) mixed matrix membrane (MMM) composed of polysulfone (PSF) and nanocomposites was prepared through a phase inversion process for the recovery of melanoidins. Nanocomposites were prepared with acid functionalized multiwalled carbon nanotubes (MWCNTs) as the reinforcing filler for chitosan-thermoplastic starch blend. Higher nanocomposites content in the PSF matrix reduced the membrane permeability and melanoidins retention indicating tighter membrane with surface defects. The membrane surface defects could be sealed with dilute polyvinyl alcohol (PVA) solution. The best performing membrane (1% nanocomposites in 18% PSF membrane sealed with 0.25% PVA coating) resulted in uniform melanoidins retention of 98% and permeability of 3.6 L/m2 h bar over a period of 8h. This demonstrates a low fouling PSF membrane for high melanoidins recovery.

  • Pressurized membrane module systems, including hollow fiber type of Microfiltration (MF) and Ultrafiltration (UF) membranes are being increasingly used in drinking water treatment due to their high removal efficiency of pathogen. However, when fibers are damaged in pressurized membrane system, the pathogen will be able to penetrate the membrane. Therefore, it is essential to guarantee the regulatory requirements for water quality by an effective on-line or off-line condition integrity monitoring methods. Recently, pressure decay test (PDT) which is one of membrane integrity tests has been reflected to drinking water treatment plants using pressurized membrane module. In this paper, three different method were used to perform PDT and three different sensitivity values were analyzed through experiments. Three types of direct integrity test methods were applied to pressure feed side, filtrate side and bidirectional pressure decay test. The results of these experiments show that the sensitivity was increasing when the volume of pressurized gas was decreasing. The sensitivity is inversely proportional to the gas volume. Furthermore, it is desirable to increase pressure difference between feed side and permeate side in order to achieve higher sensitivity in the PDT by membrane damage.

Cadmium and zinc removal from water by polyelectrolyte enhanced ultrafiltration

  • The efficiency of two metal ions (cadmium, zinc) removal from aqueous solutions by ultrafiltration (UF) and Polymer Enhanced Ultrafiltration (PEUF) processes were investigated in this work. The UF and PEUF studies were carried out using an ultrafiltration tangential cell system equipped with 5.000 MWCO regenerated cellulose. A water-soluble polymer: the polyacrylic acid (PAA) was used as complexant for PEUF experiments. The effects of transmembrane pressure, pH, metal ions and loading ratio on permeate fluxes and metal ions removals were evaluated. In UF process, permeate fluxes increase linearly with increasing pH for different transmembrane pressure, which may be the consequence of the formation of soluble metal hydroxyl complexes in the aqueous phase. In PEUF process, above pH 5.0, the Cd(II) retention reaches a plateau at 90% and Zn(II) at 80% for L = 5. Also, cadmium retention at different L is greater than zinc retention at pH varying from 5.0 to 9.0. In a mixture solution, cadmium retention is higher than zinc for different loading ratio, this is due to interactions between carboxylic groups of PAA and metal ions and more important with cadmium ions.

Lake Water Treatment using Ceramic Ultrafiltration Membrane System with Periodic Water-backflushing

  • We treated lake water by ceramic ultrafiltration membranes and found the optimal backflushing period and trans-membrane pressure (TMP) of periodic water-backflushing system. The optimal filtration time interval at fixed BT = 3 sec was 30 for A002 membrane in all viewpoints of $J/J_0,\;R_f$, and $V_T$, and we could acquire the highest $V_T$ value in the membranes used here. However, the highest $V_T$ was acquired at FT = 60 sec for M9, and at FT = 90 sec for C005 membrane. Then the lower TMP reduced the membrane fouling during filtration of lake water, and could maintain the higher permeate flux compared with the initial flux. However, the largest value $V_T$ could be obtained at the highest TMP condition for M9 membrane at fixed FT = 60 sec and BT = 3 sec. The quality of treated water in our UF ceramic system was Turbidity = $0.20{\sim}4.88NTU$, $COD_{Mn} = 0.00{\sim}2.58 mg/L$, $TDS = l8{\sim}71 mg/L$, and $NH_3-N = 0.004{\sim}1.689 mg/L$.

  • Preparation of asymmetric polyimide membranes by the phase inversion process was investigated to develop ultrafiltration, reverse osmosis and pervaporation membranes for organic solutions, using a commercially available solvent-soluble polymaide. The influences of the various factors such as the composition of a cast solution, casting conditions, gelating solutions and others on membrane structure and performance were studied in detail, and it was made clear that a wide variety of asymmetric polyimide membranes ranging from UF to RO for organic solutions could be prepared from the aromatic polyimide used. It was also found that the chemical stability and separation performance of the asymmetric polyimide membranes could be improved by annealing in a liquid or a vacuum at above 200$\circ$. The membrane annealed at 300$\circ$ in a vacuum exhibited the separation factor $\alpha(H_2O/EtOH)$ of 900 with the flux of 1.0 kg/$m^2\cdot h$ at 60$\circ$C for an aqueous ethanol solution of 95 vol%.

  • 1. 서론 : 한외여과(ultrafiltration; UF)는 장치 및 조작이 간단하고, 열의 사용없이 물질을 분리 정제를 할 수 있기 때문에 생물제품 등을 비롯한 거대분자들의 분리에 널리 활용되고 있다. 그러나 한외여과 공정의 운전시 가장 큰 문제점은 막에 의해 분리된 물질들이 필연적으로 막표면에 가역적으로 누적되는 농도분극(concentration polarization)과 이 누적된 물질이 막과의 상호작용에 의해 막표면에 비가역적으로 침적되는 막오염(membrane fouling)현상을 유발시킨다는 것이다. 일반적으로 한외여과는 막세공과 분리 대상물의 크기차에 의한 sieve effect에 의해 분리가 이루어지는데 막오염이 발생되면 세공의 일부 또는 전체가 막히게 되어 결국에는 투과선속(permeate flux)이 감소하고, 막의 세공분포가 달라져 막 본래의 분리성능에 변화가 초래된다.(생략)

  • The phase inversion is a significant phenomena in the preparation of membranes and wet spinning. In both these processes, a viscous dope solution is precipitated in a nonsolvent bath and a porous structure is formed under certain conditions. Such structure could have been considered as a diffusion controlled process. The membrane formation of the polymer solution includes both phase separation and gelation. We have studied the influence of variables on the final structure and tried to control the porosity and ultrafiltration(UF) performance of the polyacrylonitrile(PAN) and its copolymer.

  • Ceramic membranes have attracted a great attention because they have excellent resistance to most organic solvents and can be used over a wide temperature range. Especially, titania (titanium oxide, TiO$_2$) shows excellent chemical resistance and can be used both acidic and alkali solutions, and therefore, titania is one of the most promising materials for the preparation of porous membranes; titania membranes having pore sizes in the range of nanofiltration (NF) to ultrafiltration (UF) membrane have been prepared by the sol-gel process (Tsuru 2001).(omitted)
