Evaluation of ISSR and RAPD Markers for the Detection of Genetic Diversity in Mulberry (Morus spp.)

  • The present study was carried out to evaluate the ISSR and RAPD markers for their efficiency as genetic marker systems to establish the relationships between 18 mulberry genotypes. A total of 36 from 56 (64%) RAPD primers and 12 from 48 (25%) ISSR primers produced reproducible amplification patterns. A high proportion of polymorphic bands ranging from 44 to 91% was observed respectively with RAPD and ISSR markers. The average Resolving Power (Rp) of ISSR primers was higher than RAPD primers. The ISSR primers, UBC 825, 868 and 873, and RAPD primers, UBC 712, 720 and 729, possessed the highest Rp values and could in each instance distinguish all the 18 genotypes. Similarity matrix values were estimated based on Jaccards coefficient, considering 109 polymorphic ISSR and 212 polymorphic RAPD bands and two dendrograms were constructed. The dendrograms obtained with ISSR and RAPD markers distinguished the eight exotic genotypes from the ten indigenous (Indian) genotypes. A significant correlation value (r=0.959; p=0.001) for the cophenetic matrix between the RAPD and ISSR matrices was observed. The results indicated that the ISSR and RAPD markers could assist in the differentiation of genotypes and permit the determination of genetic distances that might be exploited by mulberry breeders in improvement programs.

Characterization of a novel posttranslational modification in polypyrimidine tract-binding proteins by SUMO1

  • Polypyrimidine tract-binding protein 1 (PTBP1) and its brain-specific homologue, PTBP2, are associated with pre-mRNAs and influence pre-mRNA processing, as well as mRNA metabolism and transport. They play important roles in neural differentiation and glioma development. In our study, we detected the expression of the two proteins in glioma cells and predicted that they may be sumoylated using SUMOplot analyses. We confirmed that PTBP1 and PTBP2 can be modified by SUMO1 with co-immunoprecipitation experiments using 293ET cells transiently co-expressing SUMO1 and either PTBP1 or PTBP2. We also found that SUMO1 modification of PTBP2 was enhanced by Ubc9 (E2). The mutation of the sumoylation site (Lys137) of PTBP2 markedly inhibited its modification by SUMO1. Interestingly, in T98G glioma cells, the level of sumoylated PTBP2 was reduced compared to that of normal brain cells. Overall, this study shows that PTBP2 is posttranslationally modified by SUMO1.

  • The seismic design recommendations of the Jordan Code for Loads and Forces (JC) are evaluated, based on comparisons with analytical studies and the Uniform Building Code. It was established that the overall safety ensured by the implementation of these recommendations is not consistent with the established seismic risk in Jordan and the intended objectives of the code. A new zoning map is proposed with effective peak ground acceleration values. The different period formulae of the code were studied and were found to grossly underestimate the fundamental period when compared with analytically derived values or other codes' formulae. Other factors including the dynamic, soil, importance and behavior factors are discussed. It was determined that the JC's lateral load distribution formulae clearly lead to smaller internal forces than both dynamic analysis and UBC loads, even when those loads are normalized to give the same base shear. The main reason for this is attributed to the limited allowance for a backlash force in the JC.

  • Aluminum can to can recycling was divided into two stpes. The first step was composed of the processes such as collection of used beverage cans (UBC), shredding, magnetic separation, De-laquiring, melting and casting. The second one was remelting and casting, heat treating, hot and cold rolling, annealing, and can making. In this study, the effect of alloying elements on the microstructure and texture of the secondary ingots made by Al UBC was investigated. In aluminum can to can recycling, the second phase particles appeared in the solidification stage must be controlled by heat treatment. The optimum heat treatment condition was $615^{\circ}C$ for 5hrs. the texture in hot rolled sheet was depressed with increasing Mn content, on the other hand, Si and Fe elements promoted the texture development. The textures of can-body sheet should be controlled in the hot rolling and annealing stage because can was formed from cold rolled sheet without heat treatment.

Seismic Performance of a Knee-Braced Moment Resisting Frame (Knee brace가 설치된 모멘트저항골조의 내진성능)

  • In this study the seismic performance of a three-story knee-braced moment-resisting frame (KBMRF), which is typically employed to support pipelines for oil or gas, was investigated. Nonlinear static pushover analyses were performed first to observe the force-displacement relationship of KBMRF under increasing seismic load. The results show that, when the maximum inter-story drift reached 1.5% of the story height, the main structural members, such as beams and columns, still remained elastic. Then nonlinear dynamic time-history analyses were carried out using eight earthquake ground motion time-histories scaled to at the design spectrum of UBC-97. It turned out that the maximum inter-story drift was smaller than the drift limit of 1.5 % of the structure height, and that the columns remained elastic. Based on these analytical results, it can be concluded that the seismic performance of the structure satisfies all the requirements regulated in the seismic code.

  • This study does by purpose that propose shear resisting force equation of reinforced masonry wall that is reinforced by GFRP(glass fiber reinforced polymer) based on result that is noted through cyclic loading of masonry wall and a shaking table experiment of mock that reflect identifying marks of masonry building which is constructed in domestic. It was Rocking mode to dominate failure of masonry wall in the experiment results, and the equations of UBC show the most resemblant value with experiment results. Through this study, propose the shear force equation of GFRP strengthened masonry wall as following. $$V_n=0.02A_n{\sqrt{f'_m}}+0.022b_gh_g(1+2{\alpha})^3{\sqrt{f_g}}(N/mm^2)$$.

  • The information material in the library or in the information service center is at once its own property and the national resources, so that every citizen in the country has the right to the equal use of it for the development of the country. Library co-operation enables every citizen to use easily and equally the materials in the library. In the informational society of today, owing to the information explosion, the increase in the various user's demand of the information, and the rise of the cost of information material, even the richest and highly advanced country cannot be self-sufficient. Under these circumstances, the international co-operation among libraries seems to be the best way to solve the difficulties of selfsufficiency. Therefore, this paper aims to study the background of IFLA founded in 1927 in order to promote the international library co-operation and its development up to 1970's since its establishment, and, by introducing IFLA's programs of UBC, ISBD, and UAP, to consider the significance of IFLA's contribution to the development of library and librarianship.

  • BPES (Blepharophimosis/Ptosis/Epicanthus inversus Syndrome) is an autosomal dominant disorder caused by mutations in FOXL2. Affected individuals have premature ovarian failure (POF) in addition to small palpebral fissures, drooping eyelids, and broad nasal bridge. FOXL2 is a member of the forkhead family transcription factors. In FOXL2-deficient ovaries, granulosa cell differentiation dose not progress, leading to arrest of folliculogenesis and oocytes atresia. Using yeast two-hybrid screening of rat ovarian cDNA library with FOXL2 as bait, we found that small ubiquitin-related modifier (SUMO)-conjugating E2 enzyme UBE2I protein interacted with FOXL2 protein. UBE2I also known as UBC9 is an essential protein for processing SUMO modification. Sumoylation is a form of post-translational modification involved in diverse signaling pathways including the regulation of transcriptional activities of many transcriptional factors. In the present study, we confirmed the protein-protein interaction between FOXL2 and UBE2I in human cells, 293T, by in vivo immunoprecipitation. In addition, we generated truncated FOXL2 mutants and identified the region of FOXL2 required for its association with UBE2I using yeast-two hybrid system. Therefore, the identification of UBE2I as an interacting protein of FOXL2 further suggests a presence of novel regulatory mechanism of FOXL2 by sumoylation.

Early Identification of Citrus Zygotic Seedlings Using Pollen-specific Molecular Markers (화분 특이적 마커를 이용한 감귤 교잡종 실생묘의 조기 동정)

  • This study was carried out to develop molecular techniques to allow the selection of zygotic seedlings in the early stage of the plant development. We identified 37 pollen-specific molecular markers from RAPD analysis and successfully used them for identification of the zygotic seedlings from various hybrid crosses. Three Satsuma mandarin cultivars ('Morita unshiu', 'Nangan 20' and 'Miyagawawase') were used as mother parents and seven cultivars ('Ponkan', 'Lee', 'Kinokuni', 'Shiranuhi', 'Tamnaneunbong', 'Shinyegam', and 'Sunburst' mandarins) served as pollen parents. PCR analysis showed that 2 primers could identify zygotic hybrid seedlings. Among them, an UBC-27 primer was used to identify the zygotic seedlings from hybrid crosses of "'Nangan 20' ${\times}$ 'Kinokuni'" mandarin, "'Nangan 20 ${\times}$ Ponkan'" mandarin and "'Miyagawawase ${\times}$ Sunburst'" tangerine. In total 29 out of 40 seedlings (73%), 9 out of 47 seedlings (19%), and 13 out of 45 (29%) were identified as zygotic seedlings, respectively. These results can show that the pollen-specific markers selected in this study can be used effectively for early identification of zygotic seedlings from Citrus hybrid crosses.

Intraspecific Relationship of Eleutherococcus senticosus Max. by RAPD Markers (가시오갈피 수집종의 RAPD 변이분석)

  • To evaluate the intraspecific variations among the Kasiogalpi(Eleutherococcus senticosus Max.) collections. randomly amplified DNA polymorphisms were examined. Twenty primers from 90 primers applied were selected. The range of polymorphism was $7.1{\sim}90.9%$ in 113 randomly and specifically amplified DNA fragments. Collections were divided into two major groups at the similarity coefficient value of 0.65. A considerable degree of genetic diversity was also detected among plants within the same collections. Deokyu (1, 2, 3, 4, 6), Bukhaedo (7, 8) and Odae (9, 10) col1ections showed higher degree of genetic similarity with a value of $0.65{\sim}0.86$, while Deokyu 5 showed much lower genetic similarity than other col1ections.
