Antimicrobial Effect of Oleanolic Acid and Ursolic Acid against Streptococcus downei (Oleanolic acid 및 Ursolic Acid의 Streptococcus downei에 대한 항균작용)

  • Oleanolic acid (OA) and ursolic acid (UA) are triterpenoid compound present in many plants. This study examined the antimicrobial activity of OA and UA against Streptococcus downei. The antimicrobial activity was evaluated by the minimal inhibitory concentration (MIC) and time kill curves. The MIC values of OA and UA for S. downei isolated from the Korean population were $8{\mu}g/ml$. OA and UA had a bactericidal effect on S. downei ATCC $33748^T$ above $2{\times}MIC$, $16{\mu}g/ml$ and $8{\mu}g/ml$, respectively. The results suggest that OA and UA can be used in the development of oral hygiene products for the prevention of dental caries.

Synergistic effect of xylitol and ursolic acid combination on oral biofilms

  • Objectives: This study was designed to evaluate the synergistic antibacterial effect of xylitol and ursolic acid (UA) against oral biofilms in vitro. Materials and Methods: S. mutans UA 159 (wild type), S. mutans KCOM 1207, KCOM 1128 and S. sobrinus ATCC 33478 were used. The susceptibility of S. mutans to UA and xylitol was evaluated using a broth microdilution method. Based on the results, combined susceptibility was evaluated using optimal inhibitory combinations (OIC), optimal bactericidal combinations (OBC), and fractional inhibitory concentrations (FIC). The anti-biofilm activity of xylitol and UA on Streptococcus spp. was evaluated by growing cells in 24-well polystyrene microtiter plates for the biofilm assay. Significant mean differences among experimental groups were determined by Fisher's Least Significant Difference (p < 0.05). Results: The synergistic interactions between xylitol and UA were observed against all tested strains, showing the FICs < 1. The combined treatment of xylitol and UA inhibited the biofilm formation significantly and also prevented pH decline to critical value of 5.5 effectively. The biofilm disassembly was substantially influenced by different age of biofilm when exposed to the combined treatment of xylitol and UA. Comparing to the single strain, relatively higher concentration of xylitol and UA was needed for inhibiting and disassembling biofilm formed by a mixed culture of S. mutans 159 and S. sobrinus 33478. Conclusions: This study demonstrated that xylitol and UA, synergistic inhibitors, can be a potential agent for enhancing the antimicrobial and anti-biofilm efficacy against S. mutans and S. sobrinus in the oral environment.

  • SCE-UA(Shuffled Complex Evolution-University Arizona)기법은 최적해 탐색 알고리즘으로 개념적 강우유출 모형(conceptual rainfall runoff model)의 보정을 위한 도구로 개발되었다. SCE-UA기법은 메타휴리스틱 방법의 일종으로 최적해를 구하기 위하여 여러번 목적함수 값을 계산해야 한다. 이 때 목적함수 계산 횟수와 해의 수렴과 관련된 제어 매개변수가 존재하며, 사용자가 적절한 값을 입력해주어야 한다. 이 연구에서는 SCE-UA와 관련된 제어 매개변수의 기능에 대해서 검토하였다. 그리고 집합체 수의 변화에 따라서 검사함수인 Ackley function의 전역해를 얼마나 잘 탐색하는지 검토하였다. 검토 결과 랜덤 시드에 따라서 전역해 탐색 결과가 달라졌으며, 집합체의 수가 증가할수록 목적함수 계산 횟수는 증가하는 경향을 나타내었다. 검사함수의 차원(결정 변수의 수)이 증가하면 전역해의 탐색률이 감소하며, 집합체의 수가 많아지면 전역해를 더 잘 찾는 경향이 나타나지만, 목적함수 계산 횟수는 더 많아지게 되는 것을 확인할 수 있었다. 2차원인 경우 집합체의 수가 7개 이상일 때 탐색 성공률은 90% 이상이 되었지만, 10차원인 경우 집합체의 수가 시험 최대값인 20개일 때의 전역해 탐색률은 37%에 그쳤다. 이 연구의 결과는 SCE-UA 기법의 설정 매개변수에 관한 기본 개념을 이해하고, 사용자가 설정 매개변수 선정 시에 활용할 수 있을 것이다.

  • A global optimization method known as the Shuffled Complex Evolution method from the University of Arizona(SCE-UA) was used for calibrating a Tank model. The model was calibrated with error-free synthetic data, and the SCE-UA method was found to effectively search optimal parameters. Historical data from an agricultural watershed was used to calibrate and validate the model parameters. The simulated results were in good agreement with the observed.

Preparation of Water-Soluble Solid Forms Containing Ursolic Acid Using Supercritical Fluid (초임계유체를 이용한 우르솔산 함유 고체 가용화 제제의 제조)

  • In this work ursolic acid (UA), a poorly water-soluble compound, was inclusion complexed with 2-hydroxypropyl-$\beta$-cyclodextrin (HP-$\beta$-CD) by various methods such as kneading, solvent evaporation and two types of supercritical fluid processes. The solubility and characteristics of these UA/HP-$\beta$-CD complexes were investigated by scanning electron microscopy, x-ray diffraction and HPLC. The water solubilities of the two complexes obtained from solvent evaporation and ASES processes were observed to increase up to 6$\sim$240 folds and 12$\sim$56 folds, respectively, compared with that of unprocessed UA. The stability of UA/HP-$\beta$-CD complex samples in cosmetic formulations was examined at various temperatures for one month. The UA/HP-$\beta$-CD complex prepared by solvent evaporation was found to be most stable among all the cosmetic formulations tested in our experiments.

Relationship between Albuminuria and Uric Acid to High-Density Lipoprotein Cholesterol Ratio in Korean Adults (대한민국 성인에서 알부민뇨와 요산 대 고밀도 지단백 콜레스테롤 비율의 관련성)

  • This study assesses the urine microalbumin to urine creatinine ratio (uACR) with uric acid and the association with the high-density lipoprotein cholesterol (UA/HDL-C) ratio in Korean adults. Data from the 2019 Korean National Health and Nutrition Examination Survey was procured and included 5,845 adults aged 20 years or older. Several key findings were obtained in the present study. After adjusting for the related variables (age, gender, hypertension, obesity, type 2 diabetes mellitus, and chronic kidney disease), the uACR was found to be positively associated with the quartiles of the UA/HDL-C ratio (P=0.002). Moreover, after adjusting for related variables, the odds ratio (OR) of albuminuria (uACR≥30.0 mg/g) was not significant in quartile 2 (Q2) (OR, 1.015; 95% confidence interval [CI], 0.757~1.361) and Q3 (OR, 1.090; 95% CI, 0.811~1.465) but was significantly higher in Q4 (OR, 1.416; 95% CI, 1.044~1.921) as compared to Q1 of the UA/HDL-C ratio. We conclude that since the UA/HDL-C ratio is positively associated with albuminuria in Korean adults, an increased UA/HDL-C ratio could be applied as a useful measure to assess albuminuria in Korean adults.

Research about Designation of Restricted Area Dedicated for Remote Piloted Aircraft Flight Test (무인항공기 비행시험 전용 제한구역 설정에 관한 연구)

  • Global market of unmanned aircraft(UA) is rapidly expending based on the versatile and efficient utility of the UA. Domestically many industries, universities and research institutes are trying to do research and development of the UA in various angle of aspect. In spite of these effort, all the participants of research and development of the UA has been suffering the difficulty of acquiring the airspace around vicinity of Goheung airfield for the flight test of UA. Although the current procedure of execution of the flight test of UA is set after acquiring the airspace by applying the NOTAM(Notice To Air Man) to the Ministry of Land, Infrastructure and Transport(MLIT) at least 7 days before the flight test and commencing with the publication of the NOTAM by MLIT, if the flight test is carried out as planned, changing or reapplying the NOTAM expends mort time and makes difficulty often. Therefore it is needed that a restricted airspace for the flight test of UA is established and make all the executioner of flight test uses the exclusive airspace without limitation. This research proposes the restricted airspace with short term and long term establish requirement of airspace separately. The short term requirement has been established with the airspace of 10 nm radius and 8,000 ft altitude in which the requirements of flight test can be carried out more than 90% without needs of supplement of the additional airspace. The long term has been established within the airspace of 30 nm radius which is the maximum Radio Line Of Sight(RLOS) and 8,000 ft altitude with exclusion of current air way, airport control area, approaching corridor to the airport, existing restricted area(RA) and Military Operating Area(MOA) for the purpose of minimizing inconvenience of the other airspace user. Once establishing the exclusive airspace for the flight test of UA, research and development of industries, universities and research institutes will be more vigorous and contributes to the national economy.

Effects of Combined Treatment with Ultrasound and Ascorbic Acid on the Storage Qualities of Fresh-cut 'Jonathan'Apples (초음파와 ascorbic acid의 병용처리가 신선절단 '홍옥' 사과의 저장 중 품질에 미치는 영향)

  • The effects of ultrasound treatment, in combination with ascorbic acid, on the quality of fresh-cut 'Jonathan' apples was investigated. Prepared apple slices were ultrasonicated in distilled water (US) or in 1%(w/v) ascorbic acid solution (UA) and the other samples were just dipped in 1%(w/v) ascorbic acid solution (AA). All samples were stored at$10^{\circ}C$ for 12 days. UA-treated samples showed high $L^{*}$ and hue values and low $a^{*}$, $b^{*}$, chroma, and ${\Delta}E$ value. Both control and US-treated samples showed considerable browning. A significant inhibition of polyphenol oxidase activity was observed after UA treatment. The level of total phenolics in UA-treated samples was higher on the day of treatment compared with other samples. Total soluble solids, pH, titratable acidity, and gas concentrations were similar in all samples. This study demonstrated that the simultaneous treatment of ultrasound and ascorbic acid was effective in preventing enzymatic browning of fresh-cut 'Jonathan' apples and maintaining total phenolics contents.

Ursolic acid improves the indoxyl sulfate-induced impairment of mitochondrial biogenesis in C2C12 cells

  • BACKGROUND/OBJECTIVES: Patients with chronic kidney disease (CKD) have a high concentration of uremic toxins in their blood and often experience muscle atrophy. Indoxyl sulfate (IS) is a uremic toxin produced by tryptophan metabolism. Although an elevated IS level may induce muscle dysfunction, the effect of IS on physiological concentration has not been elucidated. Additionally, the effects of ursolic acid (UA) on muscle hypertrophy have been reported in healthy models; however, it is unclear whether UA ameliorates muscle dysfunction associated with chronic diseases, such as CKD. Thus, this study aimed to investigate whether UA can improve the IS-induced impairment of mitochondrial biogenesis. MATERIALS/METHODS: C2C12 cells were incubated with or without IS (0.1 mM) and UA (1 or 2 μM) to elucidate the physiological effect of UA on CKD-related mitochondrial dysfunction and its related mechanisms using real-time reverse transcription-polymerase chain reaction, western blotting and enzyme-linked immunosorbent assay. RESULTS: IS suppressed the expression of differentiation marker genes without decreasing cell viability. IS decreased the mitochondrial DNA copy number and ATP levels by downregulating the genes pertaining to mitochondrial biogenesis (Ppargc1a, Nrf1, Tfam, Sirt1, and Mef2c), fusion (Mfn1 and Mfn2), oxidative phosphorylation (Cycs and Atp5b), and fatty acid oxidation (Pdk4, Acadm, Cpt1b, and Cd36). Furthermore, IS increased the intracellular mRNA and secretory protein levels of interleukin (IL)-6. Finally, UA ameliorated the IS-induced impairment in C2C12 cells. CONCLUSIONS: Our results indicated that UA improves the IS-induced impairment of mitochondrial biogenesis by affecting differentiation, ATP levels, and IL-6 secretion in C2C12 cells. Therefore, UA could be a novel therapeutic agent for CKD-induced muscle dysfunction.

Enhancement of Platelet Aggregation by Ursolic Acid and Oleanolic Acid

  • The pentacyclic triterpenoid ursolic acid (UA) and its isomer oleanolic acid (OA) are ubiquitous in food and plant medicine, and thus are easily exposed to the population through natural contact or intentional use. Although they have diverse health benefits, reported cardiovascular protective activity is contentious. In this study, the effect of UA and OA on platelet aggregation was examined on the basis that alteration of platelet activity is a potential process contributing to cardiovascular events. Treatment of UA enhanced platelet aggregation induced by thrombin or ADP, which was concentration-dependent in a range of $5-50{\mu}M$. Quite comparable results were obtained with OA, in which OA-treated platelets also exhibited an exaggerated response to either thrombin or ADP. UA treatment potentiated aggregation of whole blood, while OA failed to increase aggregation by thrombin. UA and OA did not affect plasma coagulation assessed by measuring prothrombin time and activated partial thromboplastin time. These results indicate that both UA and OA are capable of making platelets susceptible to aggregatory stimuli, and platelets rather than clotting factors are the primary target of them in proaggregatory activity. These compounds need to be used with caution, especially in the population with a predisposition to cardiovascular events.
