Popping Pretreatment for Enzymatic Hydrolysis of Waste Wood (팝핑 전처리를 이용한 폐목재의 효소당화)

  • We have investigated pretreatment of waste wood using milling refinery combined with poping method, which can save energy for pretreatment and enzyme loading for enzymatic hydrolysis. The chemical analysis of holocellulose of non and popping treated waste wood showed 65.9% and 58.8%, and the lignin, organic extracts and ash were increased by 3%, 4% and 0.7% after pretreatment, respectively. The reducing sugar yields of pretreated waste wood were increased four times more than non-pretreated one and the synergistic effect of cellulase and xylanase were evaluated compare with individual enzyme treatment. Especially, enzyme cocktail (cellulase 50 U and xylanase 50 U) treatment was very efficient in 1% substrate (50 mg). Also, glucose and xylose conversion rate of pretreated waste wood by GC analysis were 45.9% and 38.7%, respectively.

Conservation Status of Rare and Endangered Plant Species in T$\v{o}$kyusan National Park (덕유산 국립공원내 회귀 및 멸종위기식물의 보전실태)

  • The present conservation status on the rare and endangered plants in Tokyusan National Park were surveyed and re-evaluated by the new criteria which prepared by I.U.C.N. The species such as Lonicera vidalii, Clematis chiisanensis, Cypripedium macranthum and Allium taquetii were grouped as Critical, Eranthis stellata, Abies koreana, Taxus cuspidata, Tricyrtis dilatata, Paeonia japonaca, Stewartia koreana, Rhododendron tschnoskii, Buplerum euphorbioides, Lilium cernum and Oreorchis patens were grouped as Endangered, Adonis amurenis and Disporum ovale were grouped as Vulnerable. The above mentioned species were mainly located at the vicinity of mountain trails and projected developing sites. The potentials for the habitat destructions, mainly, due to human activities are expected to be serious in the future unless the proper management plans prepared for the specific plants and the specific habitats.

Effects of Nitrogen Fertilization on Growth of Populus sibirica and Ulmus pumila Seedlings and Soil Properties in a Semi-Arid Area, Mongolia (몽골 반건조지에서 질소 시비가 백양나무와 비술나무 묘목의 생장 및 토양 특성에 미치는 영향)

  • This study was conducted to investigate the effects of different levels and types of nitrogen fertilizer on seedlings and soil chemical properties in a semi-arid area, Mongolia. 2-year-old Populus sibirica and 4-year-old Ulmus pumila seedlings were planted in May 2014. Six treatments with three levels of nitrogen (low-level: urea $5g\;tree^{-1}$; medium-level: urea $15g\;tree^{-1}$, ammonium sulfate $33g\;tree^{-1}$, urea $15g\;tree^{-1}$ with potassium phosphate $10g\;tree^{-1}$; high-level: urea $30g\;tree^{-1}$) were applied and for the medium-level of nitrogen, different types of fertilizer were treated. Survival rate, root collar diameter (RCD) growth rate, leaf nitrogen concentration of seedlings, and soil chemical properties were determined in August 2014. The seedling survival rate of both species decreased as the level of nitrogen increased. This result can be explained by water stress caused by nitrogen fertilization in arid regions. The RCD growth rate of P. sibirica was significantly decreased by the treatment of high-level of nitrogen due to excessive nitrogen fertilization, and was increased by the treatment of ammonium sulfate due to sulfur which might promote nitrogen uptake. The leaf nitrogen concentration of P. sibirica did not change by the treatment of low-level of nitrogen, and was increased by the treatment of medium-level of nitrogen. There were no significant differences in the RCD growth rate and the leaf nitrogen concentration of U. pumila among the six treatments. None of soil chemical properties was affected by nitrogen fertilization. Overall, the low-level of nitrogen showed no effect on seedlings and soil chemical properties, except on survival rate of U. pumila and the high-level of nitrogen was considered excessive fertilization. Continuous monitoring of medium-level nitrogen fertilization including the ammonium sulfate, which increased early growth of seedlings, would be needed to elucidate the effect of fertilization on seedling growth and soil properties in a semi-arid region.

Production, Purification and Characterization of a Melanin Bleaching Enzyme from Trametes velutina JS18 (Trametes velutina JS18 유래 멜라닌 탈색 효소의 생산, 정제 및 특성)

  • The JS18 strain was isolated from an old tree forest and produced extracellular enzymes that decolorize synthetic melanin. Phylogenetic analysis, based on the internal transcribed spacer (ITS) sequence, indicate that JS18 belongs to the Trametes velutina species. JS18 demonstrated laccase activity but no manganese peroxidase or lignin peroxidase activity. Batch culture indicated that the melanin decolorization activity of JS18 strain originated from the laccase. Syringic acid and CuSO4 induced maximum laccase production, yielding 98 U/ml laccase activity after cultivation for 7 days at 25℃. T. velutina secretes an extracellular laccase in GYP medium, and this enzyme was purified using (NH4)2SO4 precipitation, Hi-trap Q Sepharose columns and gel filtration. The molecular weight of the purified enzyme was estimated to be 67 kDa using sodium dodecyl sulfate polyacrylamide gel electrophoresis. This enzyme produced 80% of its melanin decolorization activity within the first 24 h of evaluation in the presence of 1-hydroxybenzotriazole (HBT), while only about 4% of the melanin was decolorized in the absence of the mediator. The greatest decolorization was observed at 1.5 mM/l HBT, which decolorized 81% of the melanin within the first 24 h. The optimum pH and temperature for this decolorization were found to be 5.0 and 37℃, respectively. Our results suggest the possibility of applying HBT induced T. velutina JS18 laccase-catalyzed melanin decolorization.

Anomaly detection and attack type classification mechanism using Extra Tree and ANN (Extra Tree와 ANN을 활용한 이상 탐지 및 공격 유형 분류 메커니즘)

  • Anomaly detection is a method to detect and block abnormal data flows in general users' data sets. The previously known method is a method of detecting and defending an attack based on a signature using the signature of an already known attack. This has the advantage of a low false positive rate, but the problem is that it is very vulnerable to a zero-day vulnerability attack or a modified attack. However, in the case of anomaly detection, there is a disadvantage that the false positive rate is high, but it has the advantage of being able to identify, detect, and block zero-day vulnerability attacks or modified attacks, so related studies are being actively conducted. In this study, we want to deal with these anomaly detection mechanisms, and we propose a new mechanism that performs both anomaly detection and classification while supplementing the high false positive rate mentioned above. In this study, the experiment was conducted with five configurations considering the characteristics of various algorithms. As a result, the model showing the best accuracy was proposed as the result of this study. After detecting an attack by applying the Extra Tree and Three-layer ANN at the same time, the attack type is classified using the Extra Tree for the classified attack data. In this study, verification was performed on the NSL-KDD data set, and the accuracy was 99.8%, 99.1%, 98.9%, 98.7%, and 97.9% for Normal, Dos, Probe, U2R, and R2L, respectively. This configuration showed superior performance compared to other models.

Kansas Vegetation Mapping Using Multi-Temporal Remote Sensing Data: A Hybrid Approach (계절별 위성자료를 이용한 미국 캔자스주 식생 분류 - 하이브리드 접근방식의 적용 -)

  • To address the requirements of gap analysis for species protection, as well as the needs of state and federal agencies for detailed digital land cover, a 43-class map at the vegetation alliance level was created for the state of Kansas using multi-temporal Thematic Mapper imagery. The mapping approach included the use of three-date multi-seasonal imagery, a two-stage classification approach that first masked out cropland areas using unsupervised classification and then mapped natural vegetation with supervised classification, visualization techniques utilizing a map of small multiples and field experts, and extensive use of ancillary data in post-hoc processing. Accuracy assessment was conducted at three levels of generalization (Anderson Level I, vegetation formation, and vegetation alliance) and three cross-tabulation approaches. Overall accuracy ranged from 51.7% to 89.4%, depending on level of generalization, while accuracy figures for individual alliance classes varied by area covered and level of sampling.

Characteristics, Protection and Management of Bird Community in Geum River Estuary (금강 하구지역 조류군집의 특성 및 보호와 관리)

  • This study was conducted to clarify the characteristics, and proper way of protection and management of bird community in Geum river estuary from Nov. 1997 to Oct. 1998. Water was the most dominant factor of habitats. And there were cultivated area, forest, constructed area, reed bed and tidal fiat in study area. Total 104 bird species were recorded in Geum river estuary. Maximum 46 species in Apr. and 51,002 individuals in Feb. were observed. Dabbling ducks used islands and shallow area of river, and tidal flat. Gulls and waders were distributed in estuary and geese were in islands of the upstream and cultivated area of Sipjadeul. For the maintenance, protection and management of bird community in Geum river estuary, diversity of natural habitats, such as, tidal flat, cultivated area, islands and reed bed, should be maintained.

Genetic Transformation of Irpex lacterus and Phlebia tremellosa to an Antibiotic Resistance (아교버섯과 기계충버섯의 형질전환)

  • White-rot fungi which degrade lignin can also degrade diverse recalcitrant compounds such as polymeric dyes, explosives, pesticides, and endocrine disrupting chemicals. Lignin degrading enzymes are involved in the degradation reactions, and introduction of foreign genes into a white-rot fungus is required in order to increase the degrading capacity. Genetic transformation experiment has been carried out in Irpex lacteus and Phlebia tremellosa to an antibiotic resistance. The transformation yields were 50-70 transformants/${\mu}g$ DNA and 15-25 transformants/${\mu}g$ DNA in I. lacteus and P. tremellosa, respectively. The stable replication of the plasmid was confirmed by PCR using the plasmid-specific primers, and many mutants were generated during this integration in both fungi.

Phytophagous Insects Associated with Composite(Campanulales : Dicotyledoneae) (국화과(초롱꽃목 : 쌍자엽식물아강)의 잡초가해 곤충)

  • Phytophagous insects were surveyed from field populations of Compositae weeds in the order Campanulales. Forty five insect species including unidentified ones of 43 genera in 5 orders and 17 families were recorded from 16 species in 12 genera of Compositae weeds. The hemipterans and coleopterans were the most abundant insects representing 13 and 13 species, repectively. Aphids and leaf beetles were important phytophagous insects on Compositae weeds. The potential biological control insects were Uromelan gobonis and Aethes cnicana on Cirsium japonicum var. ussuriensis and Cryptosiphum artemisiae on Artemisia lavandulaefolia. The leaf beetles, Chrysolina aurichalcea, Longitarsus succineus, Basilepta fulvipes, and Hamushia konishii were often collected from A. lavandulaefolia. Aphids and leaf beetles showed host specificities. The roles of hemipterans collected from the flowers of A. lavandulaefolia and Erigeron canadensis were questionable.

Taxonomic study on the capitulum morphology of Korean Artemisia (Compositae) (한국산 쑥속(국화과)의 두상화서 형태에 의한 분류학적 연구)

  • This study was intended to investigate the capitulum morphology and to evaluate its taxonomic importance within the 23 taxa of Korean Artemisia L. The Korean Artemisia was classified into sterile subg. Dracunculus and fertile subg. Artemisia by the fertility of the disk florets, which is the traditional diagnostic character of subgenera. There are sections in subg. Artemisia: sect. Absinthium with a densely, sparsely hairy receptacle, and sect. Abrotanum and sect. Artemisia with a glabrous receptacle. However, A. fukudo and A. sacrorum belonging to sect. Abrotanum, and A. viridissima belonging to sect. Artemisia were observed to have sparsely hairy receptacles. Therefore, the presence of hair on a receptacle, which is now regarded as a key character distinguishing sections, has to be reevaluated. The whole shape and size of the capitulum, the characteristic of the stigma apex, the hair on the involucral bract, and the shape of the central or peripheral floret are thought to be the most valuable characters to consider in recognizing species.
