The Melanin Synthesis Inhibitory Effect of Aspergillus fumigatus Fermented Extract (Aspergillus fumigatus 발효 추출물의 멜라닌 합성 억제 효과)

  • This study was carried out to investigate the melanin synthesis inhibitory effect of Aspergillus fumigatus fermented extract. In this study, we revealed the effects of A. fumigatus fermented extract on melanin contents, mushroom tyrosinase activity, and expression levels of mRNA and proteins of melanogenesis-related gene in B16F10 melanoma cells. A. fumigatus fermented extract inhibited both melanin contents and tyrosinase activity. In addition, the expression level of mRNA or proteins of melanogenesis was down-regulated in the A. fumigatus fermented extract treated B16F10 cells with dose-dependent manner. Moreover, when the clinical test was conducted, it was confirmed that the use of the fermented extract of A. fumigatus for 8 weeks improved skin brightness 1.586 times brighter and skin melanin 1.331 times better compared to the control product. Taken together, our results suggest that A. fumigatus fermented extract has melanogenesis inhibitory effect and whitening activity, thus it showed the possibility for using as a functional whitening cosmetic resource.

Inhibitory Effect on Melanogenesis of Radix Glycyrrhizae Water Extract (감초 물추출물의 멜라닌 형성 억제효과)

  • This study was conducted to evaluate the effects of Glycyrrhizae Radix water extract, known as depigmenting agent, on melanin biosynthesis in cellular level. The inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis was identified by mushroom tyrosinase assay, To determine whether Glycyrrhizae Radix water extract suppress melanin synthesis in cellular level, B16 mouse melanoma cells were cultured in the presence of different concentrations of Glycyrrhizae Radix water extract. The maximum concentration of Glycyrrhizae Radix water extract that was not inhibitory to growth of the cells was 2 mg/ml. At that concentration, melanin synthesis was significantly inhibited without cytotoxicity after 5 days, compared with untreated cells. The treatment with Glycyrrhizae Radix water extract reduced tyrosinase and DOPAchrome tautomerase activity in a dose-dependent manner. These results suggest that the inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis is due to the suppression of tyrosinase and DOPAchrome tautomerase activity.

Potential Biological Activities of Magnoflorine: A Compound from Aristolochia debilis Sieb. et Zucc

  • Magnoflorine, an important compound in Aristolochia, was usually used as an anxiolytic chemical. In this study, the magnoflorine was isolated from Aristolochia and the biological activities such as antioxidant, ${\alpha}$-tyrosinase inhibitory, anti-inflammatory, and anticancer activities were investigated. The magnoflorine showed significant antioxidant activity as a 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenger, $50{\mu}g/mL$ of the magnoflorine scavenged about 70.8% of all the free radicals. And it was good at ${\alpha}$-tyrosinase inhibiting, $100{\mu}g/mL$ of the magnoflorine inhibited 36.5% of the tyrosinase. High dosage of magnoflorine inhibited the inflammation production nitric oxide (NO), and the magnoflorine protected the murine macrophage cells (RAW 264.7) from LPS-induced apoptosis. The cell viability of human colon cancer calls (HT-29) was around 100% when treated with different dose of magnoflorine, it's suggesting that magnoflorine had no anticancer effect.

Effect of Ailanthi Radicis Cortex Extracts on Melanogenesis

  • Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. Melanogenesis is controlled by the intra- and extracellular environments. In the present study, to develop a new whitening agent, it was investigated the antioxidant activity and the inhibitory effect of Ailanthi Radicis Cortex extract on tyrosinase activity and on melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from Ailanthi Radicis Cortex was higher than that of arbutin. The ethylacetate fraction showed scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also in ethylacetate fraction ($40.0{\pm}5%$ at the concentration of $400{\mu}g/ml$). This study demonstrates that the Ailanthi Radicis Cortex extract might be used to be a potential agent for skin whitening.

Inhibitory Efficacy of Smilax china L. on MITF, TRP-1, TRP-2, Tyrosinase Protein and mRNA Expression in Melanoma Cell (B16F10) (멜라노마 세포(B16F10)에서 청미래 덩굴 뿌리 추출물의 MITF, TRP-1, TRP-2, tyrosinase 단백질 및 mRNA 발현 억제 효과)

  • The purpose of this study was to assess the whitening effects of an extract from Smilax china L., which is a vine shrub belonging to the lily family. With regard to the whitening effects, 70% ethanol and water extracts from Smilax china L. showed more than 77.6% and 40.2% tyrosinase inhibition at a concentration of $1,000{\mu}l$. Furthermore, the 70% ethanol extract showed cytotoxicity of 89% at a concentration of $100{\mu}g/ml$ in melanoma cells. Western blot showed that the inhibitory effect of the 70% ethanol extract on MITF, TRP-1, TRP-2, and tyrosinase protein expression decreased by 89.9%, 46.2%, 57.6%, and 55.8%, respectively, at a concentration of $50{\mu}g/ml$. Moreover, reverse transcription-PCR showed that the inhibitory effect of the 70% ethanol extract on MITF, TRP-1, TRP-2, and tyrosinase mRNA expression decreased by 78.5%, 58.0%, 78.8%, and 70.8%, respectively, at the same concentration of $50{\mu}g/ml$ concentration. Further, realtime PCR showed that the 70% ethanol extract-induced decrease in MITF, TRP-1, TRP-2, and tyrosinase quantitative mRNA expression rate was concentration-dependent. The findings suggest that the extract from Smilax china L. has great potential as a cosmetic ingredient with whitening effects.

Inhibitory Effect of Water Extract of Adenophorae Radix on the Melanogenesis (사삼 물 추출액의 멜라닌 형성 억제 효과)

  • Recently many efforts were focused to understanding the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Radix Trichosanthis on the basal Melanogenic activities of Bl6/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Radix Trichosanthis alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Radix Trichosanthis also suppressed the increase of ${\alpha}$-MSH(10 nM) or forskolin(20 ${\mu}$M)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Radix Trichosanthis also inhibited the increase of forskolin(20 ${\mu}$M) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Radix Trichosanthis showed less intensity than B16 melanoma cells stimulated with ${\alpha}$-MSH or forskolin. These results suggest that Radix Trichosanthis inhibits melanogenesis and abrogates ${\alpha}$-MSH and cAMP-induced melanogenesis in B16 melanoma cells.

Antioxidant and Whitening Effects of Agrimonia pilosa Ledeb Water Extract (짚신나물 물 추출물의 항산화 활성 및 미백효과에 관한 연구)

  • This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

Effects of Paeoniae radix alba(PRA) on Skin whitening and Elasticity using Melanoma cells (백작약(白芍藥)이 피부 미백 및 주름에 미치는 영향)

  • Objective : Paeoniae radix alba(PRA) can enrich the blood and regulate menstruation, astringe yin and arrest sweating, calm the liver and arrest pain. This study was designed to investigate effects of PRA on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of PRA on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. More than $1,000\;{\mu}g/ml$ of PRA treated group showed lowered proliferation rates significantly compared to non-treated control group. 2. All of treated groups were lower levels of melanin synthesis respectively. 3. PRA did not show inhibitory effect on tyrosinase activities in vitro. But, PRA suppressed tyrosinase activities in B16F10 cells significantly. 4. PRA suppressed elastse type 1 activities in dose-dependent manner in vitro. But, PRA slightly suppressed elastase type 4 activities in vitro, and PRA also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that PRA can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin whitening or elasticity.

Inhibitory Effect of the Methanol Extract of Fructus Forsythiae on the Melanogenesis (연교 메탄올추출물의 멜라닌생성 억제효과)

  • The aim of this study was to investigate the effect of Fructus Forsythiae on the melanogenesis. To determine whether Fructus Forsythiae methanol extract suppress melanin synthesis in cellular level, HM3KO human melanoma cells were cultured in the presence of various concentrations of Fructus Forsythiae methanol extract and the effects on cell proliferation, tyrosinase activity and melanin contents were examined. Treatment with Fructus Forsythiae methanol extract inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis, in a dose-dependent manner. And also suppressed melanin contents as a dose dependent manner without cytotoxicity morphological change. It was observed that the color of cell pellets was totally different from the control. These results suggest that the inhibitory effect of Fructus Forsythiae methanol extract on melanogenesis is due to the suppression of tyrosinase in HM3KO cells and Fructus Forsythiae is a candidate for an efficient whitening agent.

  • The inhibitory effect of extract of Atractylodis Rhizoma alba on melanin biogenesis was studied by using B16/F10 melanoma in culture. Atractylodis Rhizoma alba significantly inhibited tyrosinase activity, and melanin contents with or without $\alpha$-MSH and forskolin in vitro. Melanin contents and tyro-sinase activity have decreased in a dose-dependent manner. These results show that extract of Atractylodis Rhizoma alba could be developed as skin whitening components of cosmetics.
