The Study for Efficacy, Effect and Stabilization of Trichosanthes Kirilowii Root, Prunella Vulgaris Leaf and Clematis Chinensis Root as a New Whitening Ingredients (새로운 미백제인 천화분근, 하고초엽, 위령선근의 효능, 효과 및 안정화에 대한 연구)

  • Numerous novel ingredients have been introduced for the higher functionality of whitening cosmetics. Through the preliminary research, we have found Trichosanthes kirilowii root, Prunella vulgaris leaf and Clematis chinensis root have high whitening efficacy. But they are insoluble. Moreover the discoloration of and decrease in content take place when they are exposed to light, heat or oxygen. From Trichosanthes kirilowii root, Prunella vulgaris leaf and Clematis chinensis root, efficacious ingredients were ethanol-extracted by heating to 75∼85$^{\circ}C$ for 6∼8 h. These extracts have the inhibitory activity of tyrosinase and B16 melanin formation, thus enhancing whitening effect. We made liposomes using propylene glycol (PG)/hydrogenated lecithin/middle chain triglycerides (MCT)/glycerin/water and microfuidizer to stabilize extracts. The stability against heat and light was enhanced by 3∼5 times compared with untreated extracts. Particle size analyzer, freeze fracture transmission electron microscopy (FF-TEM), chromameter and HPLC are used for the analysis.

Enhancement of Skin-Whitening and UV-Protective Effects of Centella asiatica L. Urban by Utrasonification Process (초음파 병행을 통한 병풀의 미백 및 자외선 차단 활성 증진 효과)

  • Enhancement effect of ultrasonification process on UV-protection and skin-whitening activities using Centella asiatica L. Urban extract was investigated. Cytotoxicity of the extracts measured on human skin fibroblast cells, CCD-986sk, and then, ultrasonification associated extracts showed 5~9% lower cytotoxicity then normal crude extracts on 1.0 mg/$m{\ell}$ of highest sample concentration. The associated extrats showed highest inhibition activity of hyaluronidase on 1.0 mg/$m{\ell}$ of concentration as 54.2%. Also, the associated extract reduced expression of MMP-1 on UV-irradiated CCD-986sk cells down to 100.2% from 136.1%, and revealed high inhibitory potency on tyrosinase as 74.6% by adding 1.0 mg/$m{\ell}$ of concentration. Ultrasonification associated extract showed strong inhibition effect of melanin production on Clone M-3 cells as 84.2% by adding 1.0 mg/$m{\ell}$ of concentration. From the preliminary observations, we considered that the extracts from C. asiatica could be potent natural materials for skin-whitening and anti-aging agent, and could enhance the activities by ultrasonification process.

  • The Rhodiola Sachalinensis 5 g were mixed and extracted with methanol 150 $m\ell$ at the room temperature for 12 h. The effluents were collected and grouped into the two. Un this experimental condition, the mobile phase composition were linearly changed as follows; water/methanol : 90/10 - 30/70 (vol. %, for 5 min), 30/70 - 10/90 (vol. %, for 15 min) and an analytical column (3.9 ${\times}$ 25 em, 15 $\mu\textrm{m}$ particle size, and 300 ${\AA}$ pore size) was utilized. The performance of the extracted Rhodiola Sachalinensis as a whitening agent was not favorable, so it classifies the Rhodiola Sachalinensis extractions with two fractions and collects each fraction for whitening agent assay. For the in-vivo melanin production ratio assay that used melanin-a cell in 10 ppm concentration, it was 58.6%, the first fraction of the effluents collected between 1.0 and 4.0 min, while it was 60% between 10.4 and 17.6 min for the second fraction, which were more efficient than that of arbutin, 45.6%.

  • High-resolution quadrupole-Orbitrap mass spectrometry (HRMS), with high-resolution (> 10,000 at full-width at half-maximum) and accurate mass (< 5 ppm deviation) capabilities, plays an important role in the structural elucidation of drug metabolites in the pharmaceutical industry. ML106, a derivative of imidazobenzimidazole, decreased melanin content and tyrosinase activity in a dose-dependent manner. Here, we investigated the phase 1 metabolic pathway of ML106 using HRMS in human liver microsomes (HLMs) and recombinant cDNA-expressed cytochrome P450 (CYP). After the incubation of ML106 with pooled HLMs and recombinant cDNA-expressed CYP in the presence of NADPH, five phase 1 metabolites, including three mono-hydroxylated metabolites (M1-3) and two di-hydroxylated metabolites (M4 and M5), were investigated. The metabolite structures were postulated by the elucidation of protonated mass spectra using HRMS. The CYP isoforms related to the hydroxylation of ML106 were studied after incubation with recombinant cDNA-expressed CYP. Here, we identified the phase 1 metabolic pathway of ML106 induced by CYP in HLMs.

Study of Enzyme Immobilization on Composite of CTA and PCL Membrane for Biosensor (바이오센서용 CTA와 PCL 혼합막에의 효소고정화 기법의 개발)

  • The disposable glucose bio-sensor using composite of CTA and PCL membrane was developed for measurement of glucose. The most effective membrane was composed of CTA/PCL(80/20, w/w) and glutaraldehyde one-step immobilization method ($10{\mu}m$ thickness) for glucose sensor gave the best result among various methods, considering oxygen permeability and electronic sensitivity. A scanning electron micrograph of the cross-section of a typical asymmetric CTA/PCL composite membrane showed that the membrane fused with a dense layer covered with a GOD-glutaraldehyde. Glucose oxidase immoblilized on the membrane showed the linearity between difference of absolute amperometric values and glucose concentrations within 7mM when the GOD immobilized electrode was used. About 35% of activity was remained after 8 days when the tyrosinase was immobilized on CTA/PCL (80/20) membrane.

The Processed Radish Extract Melanogenesis in Humans and Induces Anti-Photoaging Effects in Ultraviolet B-Induced Hairless Mouse Model

  • The radish skin and radish greens are an edible part of the radish. But they are removed before eating the radish and used as a byproduct or an animal feed material because of their tough and rough texture. Melanin is a pigment that gives colour to our skin. But increased production of melanin can turn into benign or malignant tumours. These days due to global warming, the amount of Ultra violet (UVB) rays has been extensively increased with sunlight. Due to this, a phenomenon called exogenous photo aging is widely observed for all skin colour and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases (MMP's) that serves as degradation enzymes for extracellular matrix proteins mainly collagen is increased, causing depletion in collagen and resulting in early wrinkles formation. Therefore in our study we used the murine melanoma cell line B16/F10 to study the melanogenesis inhibition by Heated radish extract (HRE) in vitro and we used HRM-2 hair less mice exposed to artificial UVB for checking the efficacy of Heated radish extract in vivo. Furthermore, we prepared a 3% Heated radish extract (HRE) cream and checked its effects on human skin. Our results have clearly demonstrated that Heated radish extract (HRE) have potently suppressed the tyrosinase activity and melanin production in B16/F10 cells. It had also reduced the expression of components involved in melanin production pathway both transcriptionally and transitionally. In in vivo studies, HRE had potently suppressed the expression of MMP's and reduced the wrinkle formation and inhibited collagen degradation. Moreover, on human skin, ginseng cream increased the resilience, skin moisture and enhanced the skin tone. Therefore in light of these findings, we conclude that HRE is an excellent skin whitening and antiaging product.

Enhancement of Ganodermanondiol and Anti-melanogenesis Effect of Ganoderma lucidum by Rhus verniciflua Extract Supplementation (옻나무 추출물 첨가에 따른 영지버섯의 가나도마난디올 생합성 증대 및 멜라닌 생성 저해효과)

  • This study was carried out to investigate the effect of lacquer tree (Rhus verniciflua) extract on ganodermanondiol (GN), tyrosinase and melanin biosynthesis inhibitor, biosynthesis in Ganoderma lucidum mycelia. In HPLC analysis, GN contents were significantly increased in G. lucidum mycelial extracts supplemented with of 1, 5, 10, and 15% lacquer tree extracts (LTE). In addition, G. lucidum mycelial extracts supplemented with LTEs which had no cytotoxicity activity against B16F10 cells, significantly inhibited melanogenesis in B16F10 cells. GN biosynthesis was facilitated by LTE. Taken together, we propose that G. lucidum mycelial extracts supplemented with LTE can be used as an effective ingredient of skin care products in the future.

Oxidation-Deficient Silkworm Hemolymph as a Medium Supplement for Insect Cell Culture

  • Hemolymph is oxidized and darkens visibly during the collection from silkworm due to the activity of tyrosinase in it. Toxic quinones are produced by the oxidation and consequently inhibit the cell growth. Heat treatment can be used to prevent the oxidation ; however, the oxidation may occur during the collection of hemolymph before it is heat-treated. It makes the hemolymph collection difficult especially on a large-scale preparation. Hemolymphs collected from 257 different strains of silkworms were examined to select the slowly oxidized hemolymphs. Hemolymphs collected from mutant strains such as Lemone, TBO, Cre, Y4, and wEb showed relatively slow color changes. Oxidation rates of the hemolymphs were measured by the absorbance change using a spectrophotometer. The hemolymph of wEb showed the slowest oxidation. The absorbance of this mutant hemolymph reached the saturation value at 20$^{\circ}C$ in 450 min, whereas the total oxidation time of the wild-type (Baekokjam) hemolymph at the same temperature was 120 min. We tested if this mutant hemolymph is useful as a medium supplement for insect cell culture. Cell growth rate and final cell concentration in the medium supplemented with the wEb hemolymph were almost same as those in the medium supplemented with the wild-type hemolymph. Hemolymph is collected on a small scale by clipping the abdominal leg; however, this method is not appropriate fro large scale preparation. Centrifugation after chopping the silkworm hemolymph by a blending mixer is a more appropriate procedure for large scale collection. Slowly oxidized wEb hemolymph resulted in higher cell concentration than the wild-type hemolymph when hemolymph was collected by the large scale preparation method.

Identification of Biologically Active Compounds from Panax ginseng C. A. Meyer (한국산 인삼의 polyphenol 화합물의 생리활성 효과)

  • The polyphenol compounds of Korea ginseng radix were extracted with 60% acetone for 4 days at room temperature and purified using Sephadex LH-20 column chromatography, MCI gel column chromatography, Bondapak $C_{18}$, column chromatography, TLC and HPLC. As a result in three compounds were isolated from Korean ginseng. In the inhibitory activities of angiotensin converting enzyme, compound Ⅱ showed the highest value of 31.86% inhibition at 157 ppm. Compound I showed 19.4% inhibition at 157 ppm. In the inhibitory activities of xanthine oxidase, compound I, II showed complete inhibition at 666 ppm but compound III didn't have inhibitory activity. In the inhibitory activities of tyrosninase, compound III showed 6.1% inhibition at 300 ppm and 28.6% at 400 ppm.

  • This study aimed to provide consumer perception and evaluation of natural Cynanchi Atrium Fermented Original Cosmetics for Skin Whitening that can improve skin by using oriental medicinal ingredients. The product was applied three times a day for two weeks by a total of 60 users who agreed to participate in a clinical trial assessing the usability and skin sensitivity to extracts that had already completed component safety testing. This study subjects reported which images were most reminiscent of Korea. They preferred products that bore Korean images because they felt trust and recognized that the quality of the cosmetics was excellent. The responses also showed that many consumers were willing to buy a product, even though it was luxurious, elegant, and expensive. The cognitive evaluation of Original Cosmetics was high and showed that the quality was likely to be good. The usability and CMF(Color, Material, Finishing) evaluation of Original Cosmetics showed good product the product is long-lasting and tightened and moisturized skin naturally. They are think about that this product it has moisturizing effects and good application for use, so it is light and soft to use. And had a whitening effect. It was intended to assist in the development of products that meet consumers' desires and expectations for safe, healthy, and beautiful skin by developing a skin whitening Original Cosmetics prototype that can inhibit the production of melanin and tyrosinase activity safely and effectively.
