  • The present study was conducted to investigate the epidemiological properties of salmonellosis of poultry farms in Kyongbuk province during the relied from November 1998 to November 1999 And antibiotic susceptibility and biochemical characteristics of 120 Salmonella cultures isolated from chicken samples were also investigated. The results obtained through this study were summarized as follows, 1. Among 667,200 chickens of 31 flocks in 17 layer farms and 9 broiler farms, 61,350 chickens of 31 flocks were died with salmonellosis. The death rate of 25 farms varied from 0.1% to 75.0%, and the average death rate was 9.2%. 2. According to etiological agents, fowl typhoid was shown the most predominant outbreak among the salmonellosis during a year, which accounted for 8R.0%(22/25) of the total case 3. The serotypes of 120 Salmonella isolates were identified as 7 strains(5.8%) of S pullorum, 10 strains(8.4%) of S typhimurium and 103 strains(85.8%) of S gallinarum. 4. Most outbreak of fowl typhoid were prevalent on the layer chicken farms(77.1%), and the summer season(45.5%) also appeared the most hazardous season during the year. 5. It seemed that the Hyline breed(70.6%) was the most susceptible among the layer chicken breeds, and followed by Isabrown(23.5%), Tetra(5.9%) in order. 6. In layers, 76.4% of fowl typhoid occurred commonly from 14 to 40 weeks including the early laying peroid, but in broiler farms, all cases was outbreak within first second weeks. 7 All the strains of S pullorum were resistant to lincomycin(Lm), penicillin(Pm), and steptomycin(Sm), but sensitive to amikacin(Ak), ampicillin(Am), cephalothin(Ce), ciprofloxacin (CiP), chloramphenicol(Cm), colistin(Co), enrofloxacin(Enr), furazolidone(Fu), gentamicin(Gm), kanamycin(Km), neomycin(Nm), polymyxin(Po), and teracycline(Tc). All the strains of S typhimurium were resistant to Lm(100%), Pm(100%), Po(90%), and Sm(90%), but were sensitive to Ak, Am, Ce, CiP, Cm, Co, Enr, Fu, Gm, Km, Nm, and Tc. 8. Minimum inhibitory concentration(MIC) of 103 strains of S gallinarum were also evaluated and their patterns were much more variable than others. All the strains of S gallinarum were sensitive to Ak, Am, Ce, Cip, Cm, Enr, Fu, Km, and Nm, but resistant to Lm(100%), and Sm(100%), 99(96.1%) to Co, 83(80.6%) to Pm, and 83(80.6%) to Po, 55(53.4%) to Gm, and 33(32.0%) to Tc. 9. The multiple drug resistance patterns of 120 Salmonella strains were CoLmPmPo Sm pattern(34.2%), CoGmLmPmPoSmTc(20.8%), CoGmLmPmPoSm(13.3%), CoGmLmSm(7.5%), LmPmSm(7.5%), LmPm(6.7%), CoLmSmTc(3.3%),, CoGmLmSmTc(1.7%), GmLmSmTc(1.7%), CoGmLmPoSm(0.1%), LmPmPo(0.1%), CoLm Sm(0.1%), and LmSm(0.1%), in order.

추백리 혈청검사 양성 산란계로부터 Salmonella속균 분리 (Isolation of Salmonella from the layer chickens reacting in pullorum-typhoid agglutination test)

  • To investigate the specificity of rapid slide agglutination test for pullorum-gallinarum diseases and to obtain a basic data for avian salmonellosis control, salmonella isolation was peformed for the layer chickens positively reacted in pullonlm-typhoid agglutination test. The biochemical, serological and antimicrobial properties of the isolates were examined. The results obtained through this study were summarized as follows; 1. Of 2,384 chickens tested by the agglutination test, 606 chickens (25.4%) were positive reactors. 154 of 606 reactors and 49 of the non-reacting chickens were investigated for salmonella isolation, resulting in isolation of 68 strains of salmonellae from 27 chickens. 2. By organs, the isolation frequency from liver, cecum, spleen, ovary and gall bladder showed 8.9% (18 strains), 8.9% (18 strains), 7.4% (15 strains), 4.4% (9 strains) and 3.9% (8 strains), respectively. 3. By culture medium the combination of selenite broth and MacConkey agar revealed the highest isolation rate and the enrichment culture by delayed secondary enrichment culture method was found the most effective for salmonella isolation. 4. The serotypes of 68 salmonella isolates were identified as 3 strains of S pullorum, 24 strains of S gallinarum, 15 strains of S typhimurium, 8 strains of S enteritidis, 7 strains of S paratyphi A, 5 strains of S typhimurium and 6 strains of the other salmonellae. 5. The serotypes of 8 salmonella strains isolated from 49 chickens non-reacting in pullorum-typhoid agglutination test were identified as 3 strains of S typhimurium and 5 strains of S infantis. 6. When 24 chickens of which 68 strains of salmonellae isolated were examined by microplate agglutination test, the average antibody titer for pullorum antigen was $2^{5.25}$. The chickens at antibody titer between $2^3$ and $2^5$ showed the higher frequency of isolation as compared with the chickens at the other titers. 7. When salmonella isolates were tested the antimicrobial drug sensitivity by disk diffusion method, S paratyphi A were highly sensitive by 100% to ATM and GM, S typhimurium, by 88% to AM, CIP, IMP and TN, S infantis, by 100% to AM, CRO, ENR and PIP, S enteritidis,by 100% to IMP and PIP, S pullorum, by 100% to ATM, CRO, ENR and PIP and S gallinarum, by 92% to CRO, CIP and PIP.

가금티푸스 예방을 위한 adjuvant로서 mastoparan V1을 사용한 포르말린-불활화 Salmonella Gallinarum 사균체 백신의 효능 평가 (Protective efficacy of formalin-inactivated Salmonella Gallinarum whole cells vaccine using mastoparan V1 as adjuvant against fowl typhoid)

  • Mastoparan V1 was used as adjuvant of formalin-inactivated Salmonella Gallinarum whole cells vaccine against fowl typhoid in a chicken model. The 75 brown nick chickens were equally divided into 5 groups, and all chickens of each group were immunized at 6 weeks of age (0 WPPI; weeks prime post immunization), and at 9 weeks of age (3 WPPI) (except group B). Group A chickens were intramuscularly (IM) inoculated with 500 uL of sterile phosphate-buffered saline (PBS), and group B chickens were subcutaneously immunized with 0.2 ml containing 5×107 viable vaccine strain/bird. The chickens in groups C~E were IM inoculated with approximately 3×109 cells/0.5 mL of formalin-inactivated the S. Gallinarum whole cells, approximately 3×109 cells/0.5 mL of formalin-inactivated the S. Gallinarum whole cells with mastoparan V1 as adjuvant, and 0.5 mL of PBS, respectively. S. Gallinarum outer membrane proteins-specific serum IgG titers were considerably higher in groups B~D than in groups A and E. However, the levels of IFN-γ in groups B and D only than in groups A and E were significantly higher. Following oral challenge with virulent wild-type S. Gallinarum, no chicken in groups A (no challenge group) and B was dead, and only 30% of chickens in group D was dead. However, 70% of chickens in group C and all chickens in group E were dead after oral challenge. The results of this study demonstrated that IM immunization with approximately 3×109 of the formalin-inactivated S. Gallinarum whole cells containing mastoparan V1 induced robust antibody and cell-mediated immune responses in chickens. The whole cells also conferred protection against infection with wild-type S. Gallinarum.

난황 중의 항-Salmonella gallinarum 특이 항체 생성 및 가공 특성 (Formation and Processing Properties of Anti-Salmonella gallinarum Specific lgY from Yolk)

  • 가금티푸스 유발균인 Salmonella gallinarum 대항하는 항체의 함량이 높은 계란을 생산하기 위해 면역처리 후 산란계의 난황에서 항체를 분리하여 총 IgY와 특수 IgY의 함량의 변화와 그 특성을 조사하였다. 먼저 난황으로부터 항체의 수율과 경제적인 측면을 비교하기 위하여 난황 분리된 난황 25 g을 λ-카라기난(Sigma C-3889) 0.1%, 냉수 가용성(MSC #10387) 1%, 2%, 90℃ 가용성 (MSC #10386) 1%, 2% 및 냉수 가용성(0.1%) : 90℃ 가용성(0.1%) = 1 : 1 1%, 2%로 사용하여 분리하는 하였을 때 λ-카라기난 0.1%, 냉수 가용성 1% 비교적 높은 회수율과 total IgY, specific IgY 함량을 나타내었지만 λ-카라기난 0.1%가 더 나은 것으로 나타났다. Specific IgY는 마지막 백신 접종 후 44주간까지 꾸준히 검출되고 있는 것으로 나타났다. 산도 변화에 의한 total IgY의 잔존 함량을 보면 pH 5-9 범위에서 70% 이상 잔존하며, specific IgY도 pH 4-10에서 74% 이상 존재하여 산에 대하여 대체로 안정적임을 알 수 있었다. 열처리 후 조항체 용액을 total IgY, specific IgY 역가 높게 유지되는 것을 관찰 할 수 있었다. 이로써 난황으로부터의 면역 항체 생산의 가능성이 확인되었고 이러한 IgY는 추출과 가공처리 등에 의해 비교적 안정적인 것으로 나타나 IgY를 이용한 가금티푸스의 예방 또는 치료용의 대량 생산 가능성이 높은 것으로 사료되었다.

2003년 국내 원종계 및 종계의 추백리-가금티푸스 감염실태 (The prevalence of pullorum disease-fowl typhoid in grand parent stock and parent stock in Korea, 2003)

  • Serum samples of 30 chickens per flock from 6 grand parent stock (GPS) farms and 70 parent stock (PS) farms were collected for seroprevalent study of pullorum disease-fowl typhoid (PD-FT) infection by serum plate agglutination test (SPA). The incidence of PD-FT infection in GPS flocks and PS flocks were 0% and 15.7%, respectively. Especially PS flocks infected with PD-FT showed age dependent patterns that 22.2% of flocks between 20 to 30 weeks of age and 38.9% of flocks between 30 to 40 weeks of age were positive. The incidence of GPS flocks and PS flocks using Salmonella (S.) gallinarum 9R (SG9R) live vaccine were 33.3% and 58.6%, respectively. The sero-positive rate of 11 flocks were 6.7-83.3% by SPA and 2.9-55.6% by enzyme linked immunosorbent assay (ELISA), and ELISA showed more lower antibody levels than SPA. Furthermore, specific antibodies produced by SG9R vaccination were detectable by SPA using SG9R antigen without cross-reaction with the PD-FT infection.

가금티푸스 저항성 계통 선발을 위한 산란종계의 경제능력 검정에 관한 연구 (Examination of Economic Performance of Egg Type Breeder Lines for the Selection of Fowl-Typhoid Resistant Breed)

  • 가금티푸스에 저항성을 가진 합성종을 생산하기 위하여 갈색계통인 이사(ISA), 하이라인(Hyline), 로만(Lohman) 3 계통과, 백색계통인 이사(ISA) 및 로만(Lohman) 2 계통의 종계(PS)에 대한 생존율, 사료섭취량, 체중, 성성숙일령, 시산 난중, 및 난질을 조사하였다. 성계 암컷 생존율(34주~66주)은 갈색계통과 백색계통에서 각각 93.3%, 93.5%로 차이가 없었고, 수컷 생존율에도 차이가 없었다(P>0.05). 수당사료 섭취량(21~66주)은 암컷에서 갈색계통과 백색계통이 각각 113.9g과 114.3g으로 비슷한 수준이었으며, 수컷에서도 차이가 없었다. 성계 체중은 암 수 공히 갈색 산란종계 3계통 및 백색 산란종계 2계통간에 유의한 차이가 없었다. 갈색과 백색 산란종계의 체중을 비교해 보면, 갈색 산란종계가 암컷과 수컷 모두 백색 산란종계 에 비하여 무거웠으나, 반복간에 변이가 워낙 커서 유의성은 검출되지 않았다. 성성숙일령은 갈색계통과 백색계통이 각각 147.8일과 140.5일로 갈색계통이 7일정도 늦었다(P<0.05). 산란율은 23-66주에서 갈색과 백색계통이 각각 75.0% 및 81.4%로 백색계통이 약 6% 높았다(P<0.05). 39주령에 갈색란과 백색란의 난질 성적을 비교해보면 Haugh Unit, 난형계수, 난황색, 난중에 있어서는 유의한 차이가 검출되지 않았다. 그러나 난각두께는 갈색 산란계통의 계란이 더 두꺼웠고, 난각강도도 유의하게 강한 것으로 나타났다.

기니픽을 이용한 BR92021(정제 브이아이 장티푸스 백신)의 항원성 평가 (Antigenicity Tests of BR92021, a Vi polysaccharide Typhoid Vaccine, in Guinea Pigs)

  • To study the antigenicity of BR92021(Vi polysaccharide typhoid vaccine), active systemic ana-phylaxis and passive cutaneous anaphylaxis were tested in guinea pigs. The groups were as follows: group I(low dose, 30 $\mu\textrm{l}$/kg), group II(high dose, 300 $\mu\textrm{l}$/kg), group III(300 $\mu\textrm{l}$/kg plus complete Freund's adjuvant), group IV(positive control, ovalbumin plus complete Freund's adjuvant) and group V(saline-treated control). Male Hartley guinea pigs at 7 weeks of age were sensitized subcutaneously with the test article or saline three times per week for three weeks(j.e., total 9 times). For groups III and IV, animals were sensitized subcutaneously with either the test article or ovalbumin plus complete Freund's adjuvant once per three week for 6 weeks(i.e., total 3 times). Twelve days after the last sensitization, the blood was collected from the sensitized animals for the passive cutaneous anaphylaxis test. In addition, the sensitized animals were subjected to the active systemic anaphylaxis test on fourteen days after the last sensitization by an intravenous challenge with either the test article or ovalbumin. In group I, mild(1/5) or moderate(4/5) symptoms of anaphylactic shock were observed. In group II, no sign(1/5), moderate(3/5) and severe(1/5) symptoms were observed. In group III, four animals of revealed moderate signs and one of 5 showed no signs of anaphylactic shock. In group IV, all 5 animals showed severe signs of shock. In group V, one of 5 revealed moderate and four of 5 showed no signs. The necropsy findings related to the active systemic anaphylaxis were observed in most animals of groups I to V In the passive cutaneous anaphylaxis test, the antiserum was diluted 10- to 5120- fold and was injected intradermally on the clipped back of recipient animals, followed by an intravenous challenge with either the test article or ovalbumin. No animals in groups I, II, III and V showed the positive reaction, whereas all animals in group IV, the positive control, showed the positive reaction at the dilution range of x1280 to x5120. Our results indicate that the test article, BR92021, may have weak antigenic potential in male guinea pigs.

사람성장(成長)홀몬의 방사면역측정(放射免疫測定)에 관한 연구 - 제II편 각종 발열성질환에 있어서의 사람성장(成長)홀몬의 혈중농도 (Studies on the Radioimmunoassay of Human Growth Hormone - 2. The plasma HGH concentrations in the various febrile diseases)

  • The plasma HGH concentrations were assayed in total 138 cases by the radioimmunoassay. The groups of control, typhoid fever, epidemic hemorrhagic fever, tuberculous meningitis and other febrile diseases were studied, also were the groups of hyperthyroidism, acromegaly and hypopitutarism. Insulin stimulation test was performed in control, typhoid fever and hypopituitarism. In the control group, the plasma HGH concentration in fasting (early morning) was $2.06{\pm}1.183m{\mu}g/ml$ and its upper limit was $4.5m{\mu}g/ml$. No sexual difference was observed. By the insulin stimulation, plasma HGH concentration had rised to the peak level of $24.1{\pm}15.71m{\mu}g/ml$, 60 min. after the intravenous insulin injection, then decreased to the normal level progressively. In typhoid fever, fasting HGH concentrations in febrile state and in defeverence were $2.5{\pm}1.35m{\mu}g/ml\;and\;2.2{\pm}3.32m{\mu}g/ml$ respectively, showing no significant difference with the control group. However, the levels of individual cases ranged widely, conpared with the control group. The response to the insulin stimulation test was similar to the control group. In epidemic hemorrhagic fever the HGH concentrations in oliguric phase, in diuretic phase and in convalescence were $4.2{\pm}3.71m{\mu}g/ml,\;2.2{\pm}1.30m{\mu}g/ml\;and\;3.4{\pm}3.01m{\mu}g/ml$ respectively. No significant differences were observe compared to the control, but they showed wide range of plasma HGH levels. In tuberculous meningitis, the fasting HGH concentration was $2.9{\pm}1.42m{\mu}g/ml$. In the other febrile diseases, the value was $2.5{\pm}2.23m{\mu}g/ml$. In 4 cases of hypopituitarism, the fasting HGH concentration was $2.3{\pm}0.42m{\mu}g/ml$ and ranged normally. However, the response to the insulin stimulation test was not observed. Very high plasma HGH concentrations were observed in acromegalic patients.

  • Diarrhea is one of the most common causes of morbidity and mortality in children worldwide. Rotavirus is the most common cause of infectious diarrhea both in developed and developing countries. However, bacterial causes such as Salmonella typhi and Vibrio cholerae still play an important role in developing countries. Newly developed vaccines for rotavirus, S. typhi, and V. choleae are highly immunogenic and safe in children.
