Soil Microarthropods at the Kwang Yang Experiment Plantation -1. Composition of Oribatid Mites (Acari: Cryptostigmata)- (서울대 광양연습림내 토양 미소 절지동물에 관한 연구 -1. 날개응애의 종조성)

  • This study was carried out to investigate by the MGP analysis the composition of oribatid mite in coniferous and broad-leaved forests, Kwang Yng experiment plantation, Chola Nam Do Province, southern part of Korea. In these study area, 157 species of oribatid mites (Acari: Cryptogimata) were identified. Among them 6 species such as; Brachychochthonius jugatus JACOT, B hungaricus BALOGH, Eremulus translamealtus BALOGH et. MAHUNKA, Brachioppiella ctenifera GOLOSOVA, Striatoppia opuntiseta BALOGH et. MAHUNKA, Suctobelba perdentata baculifer BALOGH et. MAHUNKA have not been described in Korea. Species compositions were as follows; Among them 69 species were found in all the six sampling sites, 33 species scattered sporadically in all the area, 12 species found in coniferous forest, 10 species in broad-leaved forest only, and 33 species sampled at one site. According to the MGP analysis I, site B-1, B-2, and B-3 were found to be "Type G" and site C-1, C-2, and C-3 were "Type MG". According to the MGP analysis II, site B were found to be all "Type G". But site C-1 was "Type MG", C-2 was "Type M" and C-3 was "Type G" suggesting that "Group M" increased in the site C.e G" suggesting that "Group M" increased in the site C. in the site C.

Isoenzyme pattern of Aldaline and Acid Phosphatase in the Culture of Saccharomyces uvarum (Saccharomyces uvarum의 Alkaline 및 Acid Phosphatase의 Isoenzyme 양상에 대하여)

  • The present study was designed to investigate isoenzyme (ACPase, ALPase) pattern and its refulatory function between catabolically repressed and derepressed states in yeast, Saccharomyces uvarum. As the results, no other isoenzyme was detectable in acid phosphatase, but there were three isoenzyme types in aldaline phosphatase. Type "B" isoenzyme among alkaline phosphatases in catabolically repressed cell was derepressed, but in normally cultivated cell, type "C" isoenzyme was derepressed while type "B" activity was lowered. Type "B" isoenzyme could be postulated as repressible enzyme, type "A" as constityityve enzyme and type "C" as L-histidinol phosphatase, respectively, Also, it could be shown that type "B" ALPase, repressible enzyme, compensated for phosphate group supplier under catabolically repressed states. Protein profile in cytoplasmic soluble fraction of exponential phase cell was characterized by negative charged protein.

  • 1. Aflatoxin free and highly active new type kokja was made with strong two strains of yeast (Y-16, Y-21) and two strains of highly saccharifying active Aspergillus (A-32, A-11). And they were isolated from many kinds of kokja (over hundred) collected from all over Korea. 2. Activities of new type kokja was compared with old type kokja, and results were as following. (i) Saccharifying activity; New type kokja B, C and D were excellent (over 8. 0%), and home made old type kokja were poor quality. (ii) Alcohol fermentation; New type kokja C and D were over 12% and the old type K-12 and K-161 were low activity (less than 10%). (iii) Acid fermentation; Old type kokja K-2o and K-156 (industrial use) were better than the new type C and D. (iv) Fusel oil fermentation; Old type kokja K-23, K-26 (industrial use) and K-12 (home made) fermented high fusel oil (0.07%) and new type kokja C, D and B were the lowest (0.04%). 3. Adding food preservatives (AF-2, D.E.P.C.) and heat sterilizing of wheat bran for kokja had little concerning with the activity of new type kokja. 4. In brewing test, takju made from kokja C and D were more excellent in alcohol and fusel oil concentration than those from old type kokja K-1 and K-23. Taste and odour of takju from new type kokja was good enough to drink.

Performance Evaluation of Bending Strength of Curved Composite Glulams Made of Korean White Pine (잣나무 만곡 복합집성재의 휨강도 성능평가)

  • In this study, to improve bending strength performance of Korean white pine, we made the curved composite glulam that was reinforced with glass fiber materials and larch lamina. Five types of Korean white pine curved glulams were made depending on whether they had been reinforced or not and how they had been reinforced. Type-A, reference specimen, was produced only with Korean white pine lamina, and Type-B was with larch lamina in the same thickness. Type-C was made by inserting a glass fiber cloth of textile shape between the each layer. Type-D was reinforced with two glass fiber cloths, which were placed inside and outside of the outermost lamina. Type-E was reinforced with GFRP sheet in the same way as Type-D. As a result of this bending strength test, the modulus of rupture (MOR) of Type-B, Type-C and Type-E were increased by 29%, 6%, and 48% in comparison with Type-A. However, MOR of Type-D was decreased by 2% in comparison with Type-A. In the failure modes, Type-A, Type-B and Type-C were totally fractured at the maximum load. However, load values of Type-D and Type-E decreased slowly because of reinforcement of fracture suppression, and the GFRP sheet (Type-E) had better reinforcing effect on compressive stress and tensile stress than the glass fiber cloth (Type-D).

  • In the previous studies, we have demonstrated that prorenin converting enzyme (PRECE) was identical to the epidermal grouch factor-binding protein (EGF-BP) type B, which was a member of the mouse glandular kallikrein family, To examine whether or not EGF-BP type A and C are involved in the processing of prorenin, we have cloned the CDNAS of the EGF-BP type h and C from a library of male ICR mouse submandibular gland (SMGI. And then CHO cells were transfected with the EGF-BP expression plasmids. and stable cell lines expressing a high level of the EGF-BPS precursor were obtained. The conditioned medium was then treated with trypsin, which has been knotvn to effectively convert the EGF-BP type A and C precursor to the active forms. 수ubsequentlv, the prorenin converting activity of the trypsin-treated or untreated medium was examined. PRECE converted exactly prorenin to renin, but the prorenin converting activities of EGF-BP type A and C were not detected. From these results, it seems that only type B of these EGF-BPs is involved in processing Ren 2 prorenin in mouse SMG.

  • The purpose of this study is to identify female 500m sprint start motion by the center of gravity position in short track speed skating. The center of gravity position ratio was divided into three type(type A front : 80%-back : 20%, type B front : 70%-back 30%, type C front: 50%-back : 50%). Three video cameras were used for 3D motion analysis with DLT method and the results were as follows: The elapsed time in starting motion was appeared that type B was the shortest and type A was the longest. It was appear that the stroke length of type A was longer than that type B and C during starting phase. This result was similar to displacement of center of gravity. It was appeared that skill type of center of gravity position ratio type B' ankle and knee joint angle were lower than that of type A and C. Observing these results it was conclusion that skill type B of center of gravity position ratio was more faster than that of type A and C. But it is important that these skill type needed to verifying more subjects.

Changes of the Physicochemical Characteristics of Korean Peanuts with Roasting and Storaging(I) (한국산 낙화생의 가열 및 저장 중 품질의 이화학적인 변화 (I))

  • This study was carried out to determine the proximate composition, amino acids and fatty acids contents and changes of physicochemical characteristics of each oil extracted from Spanish and Virginia type peanuts grown in Korea roasted at 110, 120, 130 and 14$0^{\circ}C$ for 2 minutes. 1. The moisture contents of raw Spanish and Virginia type peanuts were 6.5~6.8% respectively. The crude ash and reduced sugar contents of raw Spanish and Virginia type peanuts were 2.3% and 16.5% and the crude protein content was 27.0% in Spanish type peanuts and was aproximately 1% higher than in Virginia type peanut. The protein content was 25.7%~26.7% in Virginia type peanut roasted at 110, 120, 130 and 14$0^{\circ}C$. The crude fat content of Virginia type peanut was 46.0% which was aproximately 1% higher than that of Spanish type. But four kinds of oils content were 51.3%~51.8% in Spanish type peanut roasted at 110, 120, 130 and 14$0^{\circ}C$, which was about 2% higher than those of Virginia type. 2. Amino acids existed in peanut were glutamie acid, arginine, aspartic acid, leucine, glycine, phenylalanine, proline-lysine, tyrosine, valine and isoleucine, etc mainly. But methionine and threonine contents were very low. The content of glutamic acid was the highest in 71.6-81.7mg among amino acids. Glutamic acid content of Virginia type peanut was about 12% higher than that of Spanish type peanut. Total amino acid content was 441.8mg/g in Virginia type peanut and that was 16% higher than that of Spanish type peanut. The lysine content of Spanish and Virginia type peanuts roasted at 14$0^{\circ}C$ were 24% and 13%, these were lower than those of peanuts roasted at 11$0^{\circ}C$. 3. Main fatty acids of raw Spanish and Virginia type peanut oils were oleic(40.99-46.58%), linoleic(33.21-38.82%) and palmitic acid(9.72-11.58). Linoleic acid content of raw Virginia type peanuts was 5.6% higher than that of raw Spanish type peanut. And the oleic acid content of Spanish and Virginia type peanuts roasted at 11$0^{\circ}C$, 12$0^{\circ}C$, 13$0^{\circ}C$ and 14$0^{\circ}C$ was 50-53% and 41-43% respectively. Linoleic acid content of Spanish and Virginia type peanuts roasted at same temperatures as the former was about 28-31% and 37-38% respectively. That linoleic acid content of roasted peanuts was lower than that of raw peanuts. Linoleic acid content of raw and roasted Virginia type peanut, were higher than that of Spanish type peanuts. 4. Acid value and peroxide value of oils extracted from roasted Spanish and Virginia type peanuts were much higher than those of oils extracted from raw peanuts. The maximum AVs of oils extracted from Spanish and Virginia type roasted peanuts were samples roasted at 12$0^{\circ}C$and those AVs were 0.50 and 0.63 respectively. And the maximum POVs of oils extracted from Spanish and Virginia type roasted peanuts were samples roasted at 12$0^{\circ}C$ also and those POVs were 26.8 and 32.8 meq/kg. oil respectively. Acid value and peroxide value of oils extracted from roasted peanuts were increased with increasing the roasting temperatures from 11$0^{\circ}C$ to 12$0^{\circ}C$, then decreased, while TBA values were increased continuously with increasing the roasting temperatures.

Inhibition of mitochondrial activity induces muscle fiber type change from slow to fast in C2C12 myotubes

  • Mitochondrial activity affects skeletal muscle energy metabolism and phenotype. To address whether mitochondrial activity can modulate muscle phenotype in vitro, protein expression of myosin heavy chain (MyHC) in C2C12 muscle cell lines was investigated after treated with antimycin A, an inhibitor of oxidative phosphorylation in mitochondria. Fully differentiated C2C12 myotubes were administrated with different concentration of antimycin A including 0, 100, 200, 500, 700, and 1000 ng/mL. After 72 h treatment, myosin heavy chain isoform expression and related enzyme activity (lactate dehydrogenase; LDH and creatine kinase) were analyzed. Administration of antimycin A changed expression of MyHC in C2C12 myotubes showing a shift from slow to fast twitching muscle type. Protein expression of MyHC type 2b (fast twitching muscle type) was decreased (P < 0.05) by antimycin A treatment (500, 700, and 1000 ng/mL) when compared with control group. Administration of antimycin A (1000 ng/mL), however, decreased (P < 0.05) MyHC type I (slow twitching muscle type). Interestingly, LDH activity was increased (P < 0.05) by antimycin A treatment. Results from our current study proposed a possibility that skeletal muscle phenotype, including MyHC and LDH activity, can be shifted from slow to fast twitching type by inhibiting the mitochondrial activity in C2C12 myotubes.

Thermal Environment Transition of Response Climate Change and Heat Wave Application Evaporative Cooling System (기후변화 및 폭염대응 증발냉각시스템 적용에 따른 내·외부 열환경 변화 연구)

  • This study evaporative cooling system a heat wave climate change and reduction of the inside and outside thermal environment change research. Measurement items included micro meteorological phenomena and measured comfort indices. A micro meteorograph of temperature, relative humidity, surface temperature, and the comfort indices of WBGT, UTCI, and PMV were measured. The difference in inside and outside temperatures were compared for different land types, with the largest difference found in Type A ($4.81^{\circ}C$), followed by Type B ($4.40^{\circ}C$) and Type C ($3.12^{\circ}C$). Relative humidity was about 10.43% higher inside due to water injection by the evaporative cooling system. Surface temperature was inside about $6.60^{\circ}C$ higher than the outside all types. WBGT were Type A ($3.50^{\circ}C$) > Type B ($2.71^{\circ}C$) > Type C ($1.88^{\circ}C$). UTCI was low heat stress inside than outside all types. PMV was analysed Type C for inside predicted percentage of dissatisfied 75%, other types was percentage of dissatisfied 100% by inside and outside. Correlation analysis between land cover type and temperature, surface temperature, pmv, utci. T-test analysed inside and outside temperature difference was significant in all types of land.
