  • The three-dimensional structure of the carboxyl-terminal domain of the E.coli RNA polymerase $\alpha$ subunit, which is regarded as the contact site for transcription activator proteins and the promoter UP element, was determined by NMR spectroscopy. Its compact structure of four helices and two long arms enclosing its hydrophobic core shows a folding topology distinct from those of other DNA-binding proteins. (omitted)

Magneto-optical Measurements of Semiconductor Quantum Structures in Pulsed-magnetic Fields

  • Semiconductor quantum structures are often characterized by their energy gaps which are modified by the quantum size effect. Energy levels in semiconductors can be realized by optical transitions within confined structures. Photoluminescence spectroscopy in magnetic fields at low temperatures has proved to be a powerful technique for investigating the electronic states of quantum semiconductor heterostructures and offers a complimentary tool to electrical transport studies. In this review, we examine comprehensive investigations of magneto-excitonic and Landau transitions in a large variety of undoped and doped quantum-well structures. Strong magnetic fields change the diamagnetic energy shift of free excitons from quadratic to linear in B in undoped single quantum well samples. Two-dimensional electron gas induced by modulation doping shows pronounce quantum oscillations in integer quantum Hall regime and discontinuous transition at ${\nu}=1$. Such discontinuous transition can be explained as the formation of spin waves or Skyrmions.

  • Using antifungal activity-guided fractionation, an actinomycete, Streptomyces padanus strain TH-04, which was isolated from mummified peaches infected with Monilinia fructicola, was found to produce the valinomycin (1). The structure of 1 was established using spectroscopic data, which including one- and two-dimensional NMR experimental and mass spectroscopy. Valinomycin (1) showed antifungal activity against Phytophthora capsici, with an IC$_{50}$ value of 15.9 ${\mu}$g/mL.

  • Hexagonal boron nitride (h-BN) is a two dimensional material which has high band-gap, flatness and inert properties. This properties are used various applications such as dielectric for electronic device, protective coating and ultra violet emitter so on. 1) In this report, we were growing h-BN sheet directly on sapphire 2"wafer. Ammonia borane (H3BNH3) and nickel were deposited on sapphire wafer by evaporate method. We used nickel film as a sub catalyst to make h-BN sheet growth. 2) During annealing process, ammonia borane moved to sapphire surface through the nickel grain boundary. 3) Synthesized h-BN sheet was confirmed by raman spectroscopy (FWHM: ~30cm-1) and layered structure was defined by cross TEM (~10 layer). Also we controlled number of layer by using of different nickel and ammonia borane thickness. This nickel film supported h-BN growth method may propose fully and directly growing on sapphire. And using deposited ammonia borane and nickel films is scalable and controllable the thickness for h-BN layer number controlling.

Lanostane Triterpenoids from Ganoderma tropicum Collected in Vietnam and Their Nitroblue Tetrazolium Reductive Activity In Vitro

  • A new compound, 3��-acetoxylanosta-7,9(11),24-triene-26-al (3), and seven known compounds (1 - 2 and 4 - 8) were isolated from Ganoderma tropicum (Jung.) Bres. collected in Tay Nguyen, Vietnam. The structures of these compounds were determined by one- and two-dimensional nuclear magnetic resonance spectroscopy, electrospray ionization mass spectrometry (ESI-MS), and high-resolution ESI-MS, and by comparison with literature data. All of the isolated compounds were tested for nitroblue tetrazolium (NBT) reduction activity in Saccharomyces cerevisiae-stimulated RAW 246.7 cells. Among them, compounds 2 - 4 and 6 - 8 enhanced the NBT reduction in a dose-dependent manner.

저온 성장 AlN 층이 삽입된 Al0.55Ga0.45N/AlN/GaN 이종접합 구조의 구조적 특성 및 이차원 전자가스의 광학적 특성 (Structural properties and optical studies of two-dimensional electron gas in Al0.55Ga0.45/GaN heterostructures with low-temperature AlN interlayer)

  • 저온에서 성장된 AlN (LT-AlN)층이 삽입된 $Al_xGa_{1-x}N/LT$-AlN/GaN 이종접합 구조를 금속유기 화학기상 증착법 (metal-organic chemical vapor deposition)을 사용하여 사파이어 기판 위에 제작하였다. Rutherford backscattering spectroscopy 실험을 통하여 $Al_xGa_{1-x}N$층의 Al의 조성비 x가 55% 임을 확인하였고, X-선 역격자 공간 mapping을 통하여 층간 변형력을 조사하였다. LT-AlN층의 삽입 여하에 따른 $Al_{0.55}Ga_{0.45}N$ 층의 깨짐 현상을 광학현미경과 주사전자현미경을 통하여 조사하였는데, LT-AlN 층이 삽입된 시료의 경우에 깨짐 현상이 현저히 줄어든 $Al_{0.55}Ga_{0.45}N$ 층을 얻을 수 있었다. 뿐만 아니라 LT-AlN 층이 삽입된 $Al_{0.55}Ga_{0.45}N$/LT-AlN/GaN 이종접합 구조에 대하여 이차원 전자가스 (two-dimensional electron gas, 2DEG) 관련된 photoluminescence (PL) 신호를 관찰하였다. 이 시료에 대하여 온도 변화에 따른 PL 실험을 수행하여 100 K 근방까지 2DEG 관련된 PL 신호를 관찰하였다. 여기광 세기에 따른 PL 실험을 통하여 ~3.411 eV에서 나타난 2DEG PL 신호와 함께 ${\sim}3.437eV$에서도 PL 신호가 관측되었는데, 이는 AlGaN/LT-AlN/GaN 계면에 형성된 2DEG 버금띠와 Fermi 에너지 준위에서의 재결합 특성으로 각각 해석되었다.

Combining Information of Common Metabolites Reveals Global Differences between Colorectal Cancerous and Normal Tissues

  • Metabolites of colorectal cancer tissues from 12 patients were analyzed and compared with those of the normal tissues by two-dimensional NMR spectroscopy. NMR data were analyzed with the help of the metabolome database and the statistics software. Cancerous tissues showed significantly altered metabolic profiles as compared to the normal tissues. Among such metabolites, the concentrations of taurine, glutamate, choline were notably increased in the cancerous tissues of most patients, and those of glucose, malate, and glycerol were decreased. Changes in individual metabolites varied significantly from patient to patient, but the combination of such changes could be used to distinguish cancerous tissues from normal ones, which could be done by PCA analysis. The traditional chemometric analysis was also performed using AMIX software. By comparing those two results, the analysis via $^1H-^{13}C$ HSQC spectra proved to be more robust and effective in assessing and classifying global metabolic profiles of the colorectal tissues.

Layer Controlled Synthesis of Graphene using Two-Step Growth Process

  • Graphene is very interesting 2 dimensional material providing unique properties. Especially, graphene has been investigated as a stretchable and transparent conductor due to its high mobility, high optical transmittance, and outstanding mechanical properties. On the contrary, high sheet resistance of extremely thin monolayer graphene limits its application. Artificially stacked multilayer graphene is used to decrease its sheet resistance and has shown improved results. However, stacked multilayer graphene requires repetitive and unnecessary transfer processes. Recently, growth of multilayer graphene has been investigated using a chemical vapor deposition (CVD) method but the layer controlled synthesis of multilayer graphene has shown challenges. In this paper, we demonstrate controlled growth of multilayer graphene using a two-step process with multi heating zone low pressure CVD. The produced graphene samples are characterized by optical microscope (OM) and scanning electron microscopy (SEM). Raman spectroscopy is used to distinguish a number of layers in the multilayer graphene. Its optical and electrical properties are also analyzed by UV-Vis spectrophotometer and probe station, respectively. Atomic resolution images of graphene layers are observed by high resolution transmission electron microscopy (HRTEM).

Fibrobacter succinogenes, a Dominant Fibrolytic Ruminal Bacterium: Transition to the Post Genomic Era

  • Fibrobacter succinogenes, a Gram-negative, anaerobic ruminal bacterium is a major fibre digesting species in the rumen. It intensively degrades plant cell walls by an erosion type of mechanism, burrowing its way through the complex matrix of cellulose and hemicellulose with the release of digestible and undigested cell wall fragments. The enzymes involved in this process include a combination of glucanases, xylanases, arabinofuranosidase(s) and esterases. The genome of the bacterium has been sequenced and this has revealed in excess of 100 putative glycosyl hydrolase, pectate lyase and carbohydrate esterase genes, which is greater than the numbers reported present in other major cellulolytic organisms for which genomes have been sequenced. Modelling of the amino acid sequences of two glycanases, CedA and EGB, by reference to crystallized homologs has enabled prediction of the major features of their tertiary structures. Two dimensional gel electrophoresis in conjunction with mass spectroscopy has permitted the documentation of proteins over expressed in F. succinogenes grown on cellulose, and analysis of the cell surfaces of mutant strains unable to bind to cellulose has enabled the identification of candidate proteins with roles in adhesion to the plant cell wall substrate, the precursor to cellulose biodegradation.

An NMR Study on Complexation of Cesium Ion by p-tert-Butylcalix[6]arene Ethyl Ester

  • Complexation of cesium ion by p-tert-butylcalix[6]arene ethyl ester was studied by NMR spectroscopy in nonpolar $CDCl_3$ and polar acetone-$d_6$ and the results were compared with each other. Analysis of temperature dependent $^1H$ spectra and titration curves reveals that both solvents result in a 1 : 1 cone-form complex with nonpolar $CDCl_3$yielding a more tightly bound one than acetone-$d_6$. Unexpectedly, at very low temperature, we have found that two phenyl ring proton peaks of equal intensity appear both in $CDCl_3$and in acetone-$d_6$ solution which gradually collapse and eventually coalesce into a single line as temperature is raised. This observation could be interpreted in terms of the chemical exchange through direct and/or indirect interconversion between two equivalent conformations possible the complex in both solvents over the temperature range observed. And broadening of $^{133}Cs$ (I = 7/2) nmr line with increasing temperature has also been observed, indicating the exchange of $^{133}Cs$ ion between the complex and the solvent. From numerical fitting of lineshape changes for one-dimensional $^1H$ and $^{133}Cs$ spectra, the exchange rate constants and other relevant parameters for this conformational interconversion and the complex-solvent exchange were deduced.
