Developing the Indicator System for Diagnosing the National Status Quo of Science Culture (국가 수준의 과학문화 실태 진단을 위한 지표 체제 개발)

  • During the past two decades or so, science (or scientific or scientific & technological) culture has become one of the main themes not only of policy makers but also of science educators. Although, the idea of science culture has been taken as a desirable goal, there is little agreement about what it means and how to measure it. Particularly in Korea, there has been a rapid growth of science culture projects and programs, either by governmental or non-governmental, but with little systemic monitoring and evaluation for its practice. The purpose of this study is, thus, to explore a model of measuring science culture and develop a comprehensive indicator system for it. We reviewed many literatures on definitions of science culture and the surveys for related terms, particularly, of recent national and international surveys (e.g. US Science and Engineering Indicators, Eurobarometer, Japanese Science and Technology Indicators). Based on this review, a model for science culture is proposed and then used to define the Science Culture Indicators (SCI). This model encompasses two dimensions(i.e. individual and social), which are further divided into two aspects (i.e. potential and practice). Each dimension is expected to represent citizen literacy of and national infrastructure of science culture respectively. Each category in this $2{\times}2$ matrix is further divided into several sub-categories. The discussion concerning how the model and the indicators can be used to check the states of science culture at social as well as individual levels will be given with some concrete examples, such as indicators particularly related to science education.

  • Large scale mammalian cell culture has become, over the past two decades, the preferred method to produce therapeutic monoclonal antibodies. In this presentation, I will introduce disposable bioreactor system and analyze key factors and points for consideration during mammalian cell culture process development. Example will be provided highlighting the selection of master cell, culture media and environmental factors based on productivity and product quality.

  • Several days after culture, the parts around suspensor turned brown. In about 10 days the embryo started to form protocorm sending out hairs through seed coat. Around 20 days after culture, most of the protocorms emerged out of seed coat and some of them began to take green color. When observed two months after culture, the protocorn took the characteristic top-shape feature.

  • In vitro grown microtubers of potato (cv Jaju) were used for introduction of herbicide resistance gene using bombardment with DNA-coated particles. The apical shoot-tip area of newly sprouted microtubers were intensively bombarded. After bombardment, microtubers were germinated and transplanted in a greenhouse. Northern blot analysis indicated that bar gene was expressed in two plantlets. After 5 weeks of growing, commercial herbicide Basta was sprayed to screen the resistant plants. All untransformed potato plants died after 7 days while two transgenic plants survived.

Effects of Starter Cultures on Physicochemical Properties of Fermented Sausages

  • Fermented sausages prepared by inoculation with different starter cultures were analyzed for their physicochemical characteristics. Three types of fermented sausages were processed separately, without starter culture (control), with a commercial culture mix, and culture mix plus Lactobacillus plantarum (LP). On proximate analysis, two inoculated sausages showed an increase in moisture and fat contents (p<0.05). The inoculated sausages showed lower hardness and gumminess values (p<0.05) than control. The combination of starter culture with LP displayed the lowest chewiness and cohesiveness values and showed a more intensive red color (p<0.05). Two inoculated batches showed significantly lower pH values and water activity than control, in accordance with the increase in lactic acid bacteria (p<0.05). The inoculated sausages reduced the extent of lipid oxidation (p<0.05) and induced an increase in lauric acid, linoleic acid, eicosadienoic acid, and arachidonic acid, as well as they had a higher polyunsaturated fatty acid content and ratio of n-6 and n-3 fatty acids (p<0.05). The addition of LP to the starter culture in a suitable combination resulted in a positive effect on the physicochemical and microbiological attributes of fermented sausages.

Elicitation and In Situ Recovery of Alkaloids in Suspension Cultures of California Poppy

  • Large and rapid increases in benzophenanthridine alkaloid production occured in suspension cultures of Eschscholtzia californica cells treated with elicitors. Response to different biotic elicitors showed that elicitors prepared from yeast extract, Collectotrichum lindemuthianum and Verticillium dahliae induced alkaloid formation. Highest alkaloid accumulation was obtained with $60\;\mu\textrm{g}$ of yeast extract elicitor per gram of fresh cell weight. In time course performance after elicitor addition, more than 40 hours were required to obtain saturated alkaloid accumulation. Compounded silicone fluid, an ideal accumulation phase for two-phase culture of E. californica, accumulated a large amount of alkaloids produced in a specific manner. Elicitation in two-phase culture clearly increased net alkaloid production as well as their concentrations in the accumulation phase.

Animal Cell Culture and the Production of Monoclonal Antibody(MAb) Using Biopolymer Membrane (생물고분자 막 형성을 이용한 동물세포 배양 및 단클론항체 생산)

  • Biopolymer membrane was prepared using two oppositely charged natural biopolymers. The biopolymer membrane was used for the encapsulation of two hybridoma cell lines(ATCC CRL-1606, ATCC HB-8852) to produce monoclonal antibodies. In order to reduce the down stream steps, the pre size of the membrane was controlled to retain the monoclonal antibodies in the capsules based on the diffusion experiments with standard proteins. T-flask culture showed cell densities of 8$\times$107 cells/mL and 3$\times$107 cells/mL, and MAb concentrations of 506$\mu$g/mL and 109$\mu$g/mL for encapsulated ATCC CRL-1606 and HB-8852, respectively. Two liter perfusion cultures with encapsulated ATCC HB-8852 were performed to enhance the MAb production. The MAb production of the encapsulated hybridoma increased considerably comparing to the culture using silicon tubing for oxygen transfer.

Mycoflora and Mycotoxins of Cereal Grains in Delta, Egypt

  • Five cereal grains(wheat, barley, rice, maize and sorghum) were collected from three Egyptian provinces known to be grain producers(Daqahlia, Gharbia and Kafer el-Sheikh). Two species of Alternaria(A. raphani and A. tenusinae); two species of Aspergillus(A. flavus and A. niger); one species of Cunninghamella(C. elegans); one Dreschslera species(D. myakt); three Fusarium species(F. graminearum, F. moniliform and F. solani); one Rhizopus species(R. stolonifer) and two species of Penicillium(P. digitatum and P. notatum) were isolated from the grains. The densities of these fungi and their frequencies of occurrence have been investigated. All the fungal isolates were tested for the production of toxic metabolites in culture media and the percentages of toxigenic isolates were calculated. The biological assay of the toxigenic fungal isolates showed significant variations in toxigenic activity. Thin layer chromatography revealed that the most active isolate produces moniliformin in culture media. The effect of culture conditions on the production of moniliformin was studied.

  • IThis research meets with empirical results in pursuit of aim as below mentioned. Firstly, it gets a line on two-way effects which have an effect on both (the country image and corporate image) by using the corporate image for the study of the country image. After making an analysis of the empirical test, found out that the corporate image has influence upon the country image significantly. Also found out that the country image has an effect on the corporate image powerfully. Therefore, it effects an inspection of evidence that the country image and corporate image have significantly positive concern, each other. Secondly, it is offering the way to use the culture marketing as grasping the mediating effects, which culture difference has influence upon the country image and the corporate image. The mediating effects show that the country image has influence on the corporate image more as culture difference is bigger.

  • Applying H. Gans' taste culture theory to the comtemporary fashion phenomena this study had an object to classify the fashion phenomena in Korea and Japan from 1995 to 1999, and look into and compare them. First of all, the characters of each fashion group of the two countries show that the details of the Korean traditional costume are grafted into the most popular style in each season in Korean high fashion and oriental look used the formative method far Japanese traditional costume and Tokyo street style are reflected in the Japanese high fashion. Mass fashion in Korea equally comes under the influence of European high fashion, Korean street fashion, and Japanese mass fashion, And mass fashion in Japan reflects European high fashion and japanese street fashion. The Street fashion in Korea was affected by Korean entertainers'fashion, 'Tongdaemun market fashion' in Seoul, and Japanese street fashion. And street fashion in Japan is also affected by the pursuit of powerful personality, the absolute imitation if Japanese entertainers' fashion, and 'Tongdaemun market fashion'. All of two countries exercise considerable influence over mass fashion each other.
