강원도 고랭지 주요 원예작물의 병해 발생 상황 (Surveys on Disease Occurrence in Major Horticultural Crops in Kangwon Alpine Areas)

  • 강원 고랭지의 대표적 지대인 평창의 대관령, 홍천의 내면 그리고 횡성의 둔내지역에서 재배되고 있는 주요 원예작물에 대해 1994년$\sim$1997년까지 4년에 걸쳐 실시한 병해조사의 결과는 다음과 같다. 1) 배추와 무에서는 순무모자이크바이러스(TuMV)가 '92년부터 고랭지에 다발생하여 '94년과 '96년에 피해가 심하였으며, 이 바이러스병과 함께 무름병의 발생이 심한 편이었으며, 최근에는 무사마귀병의 피해고 '96년부터 점증 추세에 있으며 수년 전까지 피해가 심했던 뿌리 마름병은 최근 발생이 지극히 저조한 점이 특이하다. 거의 모든 채소작물에서 무름병, 잿빛곰팡이병, 노균병, 흰가루병, 잘록병, 검은무늬병 등의 발생이 심한 것이 특징인데 저지대와 달리 고랭지 기후환경과 무관하지 않았을 것이다. 한편 조사기간 중 새로운 미기록종으로는 셀러리의 바이러스(BBWV)와 잿빛곰팡이병(Botrytis cinerea), 메론의 점무늬병(Cercospora citullina), 딸기의 흰가루병(Erysiphe polygori), 양상추의 흰가루병(Erysiphe cichoraceaium) 등이 밝혀졌으며 기주 미기록으로는 파슬리의 무사마귀병(Plasmodiophora brassicae)이 밝혀진 것이 특이하다. 2)여러 가지 화훼작물에는 주로 바이러스병, 잿빛곰팡이병, 시들음병, 흰무늬병 등의 발생이 많았으며 특히 자생 나리류에 흰무늬병(Cercospora spp.) 발생이 심하였다. 미기록종으로는 꽃도라지의 시들음병(Fusarium oxysporum f. sp. eustomae), 바이러스병(BBWV, CMV), 균핵병(Sclerotinia sclerotiorum), 잿빛곰팡이병(B. cinerea), 카네이션의 반점병(Cladosporium echinulatum), 용담의 시들음병(Fusarium oxysporum), 잎마름병(Alternavia dianthi) 등과 Phytoplasma에 의한 스타티스의 빗자루병, 리아트리스의 빗자루병, 자생나리류의 흰무늬병(Cercospora sp.), 스토크의 TuMV 등이 동정되었다.

오이 모자이크 바이러스와 수눔 모자이크 바이러스의 복합감염에 의한 스톡의 꽃잎얼룩무늬병 (Color Breaking Syndrome of Matthiola incana Caused by Double Infection of Cucumber Mosaic Virus and Turnip Mosaic Virus)

  • In 1995, we collected stock (Matthiola incana) plants causing mosaic symptoms on leaves and color breakings on flowers in Daekwallyong, Korea. Two viruses were isolated from the infected plants, and identified as cucumber mosaic virus (CMV) and turnip mosaic virus (TuMV) by experiments of host range, serology and electron microscopy. Each of the virus did not produce the same symptoms on the stock seedlings as naturally infected plants caused. When the viruses were coinoculated to the stock seedlings, however, severe mosaic symptoms were observed on leaves, and then the color breakings were expressed on flowers.

십자화과식물 바이러스의 진딧물 매개에 관한 연구 (Studies on the Aphid Transmission of Some Cruciferous Viruses)

  • 무우에서 많이 발생하고 있는 turnip mosaic 대한 성질과 이의 충매전염에 대하여 몇가지를 조사하여 다음과 같은 결과를 얻었다. 1) 수집된 TuMV의 물리적성질은 기존성적과 일치하였고, 시무대근의 이병율이 가장 높았다. 2) TuMV의 매개곤충은 Myzus persicae, Lipaphis erysimi, Aphis gossypii, Aphis craccivora의 4종이었다. 3) TuMV의 진딧물에 의한 전염에서 가장 적당한 처리시간은 Myzus persicae는 1시간절식, 3분흡즙, 1분접종이었으며, Lipaphis erysimi는 2시간절식, 5분흡즙, 3분접종이었고, Aphis gossypii는 1시간절식 3분흡즙, 3분접종이었다. 4) 진딧물의 Probing 횟수는 2시간 절식시킨 진딧물과 절식시키지 않는 것에 큰 차이가 없었고, 절식시킨 진딧물의 전매체가 4분경과시부터 섭식하기 시작하였다. 5) 복숭아 혹 진딧물을 1-2분 간격으로 인위적으로 옮겨주었을 때, 2시간 절식시킨 진딧물의 Probing 휫수가 절식시키지 않은 것의 Probing횟수보다 훨씬 많았고, 2시간 절식시킨 진딧물이 주로 3분이내의 획득흡즙시간후 4분이내에 Virus를 매개하였다.

Five Newly Collected Turnip Mosaic Virus (TuMV) Isolates from Jeju Island, Korea are Closely Related to Previously Reported Korean TuMV Isolates but Show Distinctive Symptom Development

  • For several years, temperatures in the Korean peninsula have gradually increased due to climate change, resulting in a changing environment for growth of crops and vegetables. An associated consequence is that emerging species of insect vector have caused increased viral transmission. In Jeju Island, Korea, occurrences of viral disease have increased. Here, we report characterization of five newly collected turnip mosaic virus (TuMV) isolates named KBJ1, KBJ2, KBJ3, KBJ4 and KBJ5 from a survey on Jeju Island in 2017. Full-length cDNAs of each isolate were cloned into the pJY vector downstream of cauliflower mosaic virus 35S and bacteriophage T7 RNA polymerase promoters. Their fulllength sequences share 98.9-99.9% nucleotide sequence identity and were most closely related to previously reported Korean TuMV isolates. All isolates belonged to the BR group and infected both Chinese cabbage and radish. Four isolates induced very mild symptoms in Nicotiana benthamiana but KBJ5 induced a hypersensitive response. Symptom differences may result from three amino acid differences uniquely present in KBJ5; Gly(382)Asp, Ile(891)Val, and Lys(2522)Glu in P1, P3, and NIb, respectively.

순무모자이크 바이러스 Ca계통 핵봉입체와 외피단백질 유전자의 cDNA 클로닝 및 제한효소 지도작성 (Complementary DNA Cloning and Restriction Mapping of Nuclear Inclusion Body and Coat Protein Genes of Turnip Mosaic Virus-Ca Strain Genomic RNA)

  • Viral RNA was extracted from purified Chinese cabbage strain of turnip mosaic virus (TuMV-Ca) from infected leaves of turnip. Polyadenylated genomic viral RNA was recovered by oligo (dT) cellulose column chromatography and used as a template for the synthesis of complementary DNA (cDNA). Recombinant plasmids contained cDNA ranged from about 900 bp to 2, 450 bp were synthesized. Among the selected 41 transformants, pTUCA31 and pTUCA35 had over 2 Kbp cDNA insert. Restriction endonuclease patterns of the clones examined were very similar among them. Clones pTUCA23 and pTUCA31 were overlapped with pTUA35. The longest clone pTUCA35, encoding 3'-end, showed that it contained two sites for EcoRI, and one site for BamHI, ClaI, HincII, SacI and XbaI, respectively. The restriction mapping indicated that the clone pTUCA35 contained partial nuclear inclusion body gene, complete coding region of the coat protein and 3' untranslated region of TuMV-Ca genomic RNA.

The Construction of a Chinese Cabbage Marker-assisted Backcrossing System Using High-throughput Genotyping Technology

  • The goal of marker-assisted backcrossing (MAB) is to significantly reduce the number of breeding generations required by using genome-based molecular markers to select for a particular trait; however, MAB systems have only been developed for a few vegetable crops to date. Among the types of molecular markers, SNPs (single-nucleotide polymorphisms) are primarily used in the analysis of genetic diversity due to their abundance throughout most genomes. To develop a MAB system in Chinese cabbage, a high-throughput (HT) marker system was used, based on a previously developed set of 468 SNP probes (BraMAB1, Brassica Marker Assisted Backcrossing SNP 1). We selected a broad-spectrum TuMV (Turnip mosaic virus) resistance (trs) Chinese cabbage line (SB22) as a donor plant, constructing a $BC_1F_1$ population by crossing it with the TuMV-susceptible 12mo-682-1 elite line. Foreground selection was performed using the previously developed trsSCAR marker. Background selection was performed using 119 SNP markers that showed clear polymorphism between donor and recipient plants. The background genome recovery rate (% recurrent parent genome recovery; RPG) was good, with three of 75 $BC_1F_1$ plants showing a high RPG rate of over 80%. The background genotyping result and the phenotypic similarity between the recurrent parent and $BC_1F_1$ showed a correlation. The plant with the highest RPG recovery rate was backcrossed to construct the $BC_2F_1$ population. Foreground selection and background selection were performed using 169 $BC_2F_1$ plants. This study shows that, using MAB, we can recover over 90% of the background genome in only two generations, highlighting the MAB system using HT markers as a highly efficient Brassica rapa backcross breeding system. This is the first report of the application of a SNP marker set to the background selection of Chinese cabbage using HT SNP genotyping technology.

The Detection of Plant Viruses in Korean Ginseng (Panax ginseng) through RNA Sequencing

  • Korean ginseng (Panax ginseng) is a dicotyledonous, medicinal, perennial plant belonging to the genus Panax of the family Araliaceae. We investigated the occurrence and incidence of plant viruses in Panax ginseng in Korea. A total of 656 leaf samples were combined into one and total RNA was extracted from the polled sample, using RNA sequencing (RNA-Seq), a metatranscriptome analysis of the plant virome was conducted. The virus present in Panax ginseng was confirmed by reverse transcription polymerase chain reaction (RT-PCR) assay using virus-specific primers. In RNA-Seq data analysis, the multiplication protein of four viral contigs including Aristotelia chilensis virus 1 (AcV1), Turnip mosaic virus (TuMV), Watermelon mosaic virus (WMV), and Tobamovirus multiplication protein were discovered. From our metatranscriptome analysis and RT-PCR assay, TuMV and WMV were detected, whereas the three viruses reported in China such as tomato yellow leaf curl China virus; panax notoginseng virus A; and panax virus Y were not found in this study. The distribution of domestic ginseng viruses seems different from that recorded in China. Overall, this is the first plant virome analysis of Panax ginseng in Korea.

고랭지배추 바이러스병의 발생 및 피해요인 분석 (Occurrence of Virus Disease of Chinese Cabbage and Its Influence on Cabbage Production in Alpine Area)

  • The studies on the ecology of virus disease on Chinese cabbage (Brassica campestris subsp. pekinensis) cultivated in alpine area of Kangwon province during summer season to analyse its influence on damage and develope a prediction model were performed from 1993 to 1997. Virus disease on Chinese cabbage occurring in the alpine area showed various symptom types and among there, necrotic spots and dwarf were mainly detected. The disease was increased from early August and continued mid September in every year. The occurrence of virus disease was the highest in 1994 with 20.5%, and the number of aphid vectors were also the highest during the same period. The number of aphids in the alpine areas showed twice peaks every year. For the analysis of damage by virus infection, the infection and injured ratio of all treatments were more than 90% and 80%, respectively. The most important factor for the occurrence of virus disease on Chinese cabbage was temperature. Factors influencing the development of the viral disease in the alpine area were maximum temperature and number of aphid vectors.

시금치 바이러스병에 관한 연구 I. 시금치에 발생하는 순무모자익바이러스 (TuMV)의 분류동정 (Investigations on the Virus Diseases in Spinach. (Spinacia orleraea L.) I. Identification of Turnip Mosaic Virus Occuring Spinach)

  • 모자익병징을 나타내는 이병시금치를 채집하여 순무모자익 바이러스를 분류 동정하였다. 분리된 순무모자익바이러스를 지표식물에 접종한 결과 담배(B.Y)와 명아주에는 국부병반이 형성되었고 쑥갓, 시금치, 무우에는 모자익 병징이 나타났다. 이병시금치 잎을 착즙하여 순무모자익바이러스의 항혈청과의 혈청반응 시험 결과 양성 반응이 나타났다. 이병엽을 Dip법으로 시료를 제작하여 전자현미경에서 검경한 결과 대부분이 750nm의 사상 입자가 관찰되었다. 시금치에서 순무모자익바이러스의 발생분포는 수원, 안양, 대구, 진주 등 거의 전국적으로 발생하였다
