The Effect of Varying Levels of Tryptophan on Growth Performance and Carcass Characteristics of Growing and Finishing Broilers

  • The purpose of this experiment was to study the effect of varying levels of tryptophan on the performance and carcass character of broiler. Trial 1: Ninety-six, five-week-old male Hubbard chickens, average weight 1.97 kg, were used in the trial. All birds were allocated into 3 treatments of 32 birds each. Each bird was kept in an individual cage. The trial period was 3 weeks. Treatment 1: Tryptophan content 0.198%. Treatment 2: Tryptophan content 0.228%. Treatment 3: Tryptophan content 0.258%. Trial 2: Ninety-six, three-week-old male Hubbard chickens, average weight 1.23 kg, were randomly distributed into the following two treatments. Each treatment had 48 birds. Treatment 1: Tryptophan content 0.167%. Treatment 2: Tryptophan content 0.198%. Trial 3: Ninety-six, twoweek-old Hubbard chickens, average body weight 0.72 kg, were used in this experiment. There were three treatments as follows. Treatment 1. Tryptophan content 0.136%. Treatment 2. Tryptophan content 0.167%. Treatment 3. Tryptophan content 0.198%. The result of Trial 1 showed that the feed intake, performance, and carcass characteristics were not influenced by tryptophan content in the diet which between 0.198% and 0.258% (p>0.05). There was no significant difference (p>0.05) in feed intake in either treatment in Trial 2. However, weight gain, feed conversion efficiency, and most carcass characteristics in the 0.198% treatment were significantly better (p<0.05) than in the 0.167% treatment. There was a trend that feed intake increased with increasing level of tryptophan, but there was no significant difference in Trial 3. The weight gain and feed conversion efficiency were significantly reduced for the broiler in the 0.136% treatment. This series of experiment showed that broilers need about 0.198% of tryptophan.

Tryptophan과 Arginine의 대두유에 대한 항산화 및 상승효과 (Antioxidant Effect of Tryptophan and Arginine on Soybean oil Rancidity)

  • 식용대두유의 항온저장시 tryptophan과 arginine의 첨가농도에 따른 항산화 효과 및 기존 항산화제들의 항산화력에 대한 상승효과를 조사한 결과는 다음과 같다. 유지의 항온저장시 tryptophan과 arginine을 각 농도별로 첨가한 경우 모든 시료에서 항산화 효과를 나타냈으며 특히 tryptophan 1%를 첨가하였을 때 가장 효과가 좋았고 그 정도는 TBHQ를 첨가한 경우와 비슷하였다. 또한 tryptophan과 arginine첨가시료는 ${\alpha}-tocopherol$보다 월등히 높은 항산화 효과를 보였다. tryptophan과 arginine을 ${\alpha}-tocopherol$, ascorbic acid, citric acid와 혼합하여 대두유에 첨가했을 때 ${\alpha}-tocopherol$과의 혼합물이 상승효과가 가장 높았다. 또한 citric acid나 ascorbic acid의 혼합물도 약하지만 상승효과를 보였다. 이상의 결과를 볼 때 tryptophan과 arginine은 유지에 대하여 항온저장시 모두 항산화 효과가 있었으며 특히 1%농도에서 가장 높게 나타났고 다른 항산화제와 병용했을 때 상승효과가 매우 우수하였음을 알 수 있었다.

  • In this study we fed control diet and tryptophan supplemented diets containing 0.1,0.2,0.35% tryptophan to ICR mice for 1∼3 weeks to investigate the effects of tryptophan supplemented diet on pain sensitivity and the analgesic mechanism of serotonin. Animals fed tryptophan supplemented diets displayed increased antinociception when measured with hot plate and phenylquinone-writhing tests. And animals with typtophan supplemented diet had higher brain serotonin and 5-HIAA concentration than the control animals.

Isolation of a Tryptophan-Overproducing Strain Generated by EMS Mutagenesis of Candida rugosa

  • To isolate a mutant strain which overproduces tryptophan, mutants of Candida rugosa were screened after EMS (ethyl methane sulfonate) mutagenesis. Fluorotryptophan, a tryptophan analogue, was used for selection of a tryptophan-overproducing mutant after mutagenesis. Among 50 mutants, several candidates were selected based on intracellular tryptophan content. Amino acid analysis results showed that C3 was the best strain because it had the highest amount of tryptophan among the mutants.

  • In order to increase the production of tryptophan by maximizing expression of recombinant trp plasmid, Klebsiella pneumoniae KC 105(pheA tyrA trpE trpR tyrR) was genetically modified. KC 107, inosine monophospate(IMP) auxotroph from KC 105 and KC 108, histidine(His) auxotroph from KC 107 were also derived respectively to increase phosphoribosylpyrophosphate(PRPP) production which is required for tryptophan biosynthesis. From KC 107 phosphoribosylpyrophosphate consumption which is required for tryptophan biosynthesis. From KC 107 and KC 108, KC 109 and KC 110, both arginine auxotrophs were derived respectively. To investigate the expression of recombinant trp plasmid in the selected K. pneumoniae mutants, the auxotrophic mutants were transformed with recombinant trp plasmids pSC 101-$trpE^{FBR}$, pSC 101-trpL(.DELTA.att) $trpE^{FBR}$ (pSC 101-trp-AF). Amount of tryptophan produced and activities of tryptophan synthase of $trpR^{-}$ mutant (KC 100) and $tyrR^{-}$ mutnat(KC 105) containing recombinant plasmid pSC 101-trp operon were increased by 30-40% as compared with KC 99(pheA tyrA trpE) containing recombinant plasmid pSC 101-trp operon. Activities of tryptophan synthase and production of tryptophan of KC 108 ($His^{-}$) and KC 109($Arg^{-}$) containing recombinant plasmid pSC 101-trp operon were increase by two-fold as compared with KC 107 containing pSC 101-trp operon.

  • An easy and convenient synthetic route to (S)-2-hydroxy-2'-(3-phenyluryl-benzyl)-1,1'-binaphthyl-3-carboxaldehyde (1), capable of recognizing tryptophan by fluorescence has been developed. The binol carboxaldehyde 1 exhibited a high selectivity to L-tryptophan over other examined L-${\alpha}$-amino acids such as alanine, phenylalanine, glutamine, arginine, lysine, serine, threonine, aspartat, valine, histidine and cysteine, with a fluorescence "turn-on" signal. In addition, 1 displayed chiral discrimination with good enantioselectivity toward L-tryptophan over D-tryptophan through different fluorescence enhancement factors.

초기계배 발생에 미치는 Serotonin의 영향에 관한 세포생물학적 연구 (Cell Biological Studies of the EfEect of Serotonin on Chick Embryogenesis)

  • Very early stage of chick embryo cultivated in the medium containing DLtryptophan by beaker method has been investigated in vitro morphologically using electron microscopy at cellular level and found that the development of tryptophan treated chick embryos corresponding to 18∼66 hrs incubation was impaired and york granule degradation was significantly delayed. It was also found that DNA, RNA and protein biosynthesis of tryptophan treated chick embryo was greatly lowered than those of control group. Conversion of L-tryptophan into serotonin was traced using 14C-L-tryptophan and found that 13.8cA of added radioactivity was recovered from serotonin formed during 18 hrs incubation and the amounts of serotonin formed were depend upon added amount of tryptophan in e99 yolk. It seemed that the serotonin formed from external tryptophan might inhibit the degradation of yolk granule by feedback mechanism, resulting in malformation of chick embryogenesis.

Tryptophan-derived Alkaloids from Hedera rhombea Fruits and Their Butyrylcholinesterase Inhibitory Activity

  • Alzheimer's disease (AD) is the most common age-related neurodegenerative disease in industrialized countries. It is estimated that about 47 million people living with dementia and the number of cases will be tripled by 2050. However, the exact mechanism of AD is not known, and full therapy has still not been found. Various tryptophan-derived alkaloids have been reported as promising agents for the treatment of AD. In the present study, a series of tryptophan-derived alkaloids were isolated and characterized from the methanol extract of Hedera rhombea fruit. Based on the analysis of their observed and reported spectroscopic data, their structures were identified as N-[4'-hydroxy-(E)-cinnamoyl]-L-tryptophan (1), N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-tryptophan (2), N-[4'-hydroxy-(E)-cinnamoyl]-L-tryptophan methyl ester (3), and N-[3',4'-dihydroxy-(E)-cinnamoyl]-L-tryptophan methyl ester (4). These compounds were screened for anti-Alzheimer activity via their inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes in vitro. As a result, compounds 3 and 4 showed moderate BChE inhibition with IC50 values of 86.9 and 78.4 μM, respectively, compared to those of the positive control [berberine (IC50 = 11.5 μM)]. However, all four compounds did not show significant inhibition of the AChE enzyme. This is the first time, the AChE and BChE inhibitory activities of these tryptophan-derived alkaloids were investigated and reported.

수종한국상용식품중(數種韓國常用食品中)의 Tryptophan 및 Available lysine 함량(含量)에 대(對)하여 (A Study on the Contents of Tryptophan and Available Lysine in Korean Foods)

  • In order to observe the contents of some essential amino acids in Korean foods, total and free tryptophan, and available lysine in fifty kinds of Korean foods were analyzed by the sfectrophotometry. The results obtained are summarized as follows : 1) The tryptophan contents per 100g of soybean, wet green laver and skim milk were over 250 mg, and the contents Per g nitrogen in ginger, dried persimmon and chestnut were over 150 mg. 2) The free tryptophan contents per 100 g of soybean, small red bean, dried persimmon, ginseng stem were over 100 mg, ana the contents per g nitrogen in dried persimmon was over 150 mg. 3) The available lysine contents Per 100g of soybean, wet green laver and skim milk, fish(auchovy), dried yeast, casein and silkworm pupa were over 1000 mg, and the contents per g nitrogen in potato, perilla(wild sesame), red pepper, sausage and skim milk were over 300mg. 4) The contents of tryptophan and available lysine in soybean, green laver and skim milk were higher than in other samples. 5) In general, the contents of tryptophan and available lysine were abundant in seaweeds.

아미노산 성분에서의 염소 소독부산물 생성 특성 (Characteristics of Chlorination Byproducts Formation of Amino Acid Compounds)

  • 20종의 아미노산 성분들에서의 염소 소독부산물 생성 특성을 조사한 결과, 단위 DOC당 THM 생성능은 Br 첨가 유무에 관계없이 tryptophan과 tyrosine에서 높게 나타났고 나머지 18종에서는 매우 낮은 생성능을 나타내었다. 단위 DOC당 HAA 생성능 조사결과, tryptophan, tyrosine, asparagine, aspartic acid 및 histidine에서 Br 첨가 유무에 관계없이 높은 생성능을 보였으며, asparagine, aspartic acid 및 histidine은 DCAA 생성능 조사결과, aspartic acid, histidine, asparagine, tyrosine 및 tryptophan에서 Br 첨가 유무에 관계없이 높은 생성능을 나타내었으며, aspartic acid는 660.2 ${\mu}$g/mg으로 나타나 다른 아미노산 성분들에 비해 월등히 높은 HAN 생성능을 나타내었으며, 반응성이 높은 6종의 아미노산 성분에서 생성되는 HAN 구성종의 대부분은 DCAN으로 나타났다. 단위 DOC당 chloral hydrate 와 chloropicrin의 생성능을 조사결과에서 chloral hydrate는 asparagine, aspartic acid, histidine, methionine, tryptophan 및 tyrosine에서 생성능이 높은 것으로 나타났고, chloropicrin의 경우는 20종의 아미노산에서 전체적으로 1${\mu}$g/mg 이하의 아주 낮은 생성능을 보였다. 아미노산의 작용기별 소독부산물 생성 특성은 THM의 경우 방향족(aromatic) 화합물인 tryptophan과 tyrosine에 반응성이 높았고, HAA는 aromatic, neutral, acidic, basic 순으로 반응성을 보였으며, HAN은 acidic, basic, neutral, aromatic 순으로 반응성을 나타내어 방향족 화합물보다 지방족 화합물이며 친수성인 물질에서 반응성이 더 높은 것으로 나타났다.
