A Study on the Effect of Garlic on the Liver Lipids of the Mouse (마늘이 마우스 간조직(肝組織)의 지질(脂質)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • (1) Control values of the hepatic lipids of mouse analysed by means of thin-layer chromatography were as follows: Total cholesterol $5.23{\pm}0.46\;mg/g$ Free cholesterol $4.29{\pm}1.23\;mg/g$ Esterified cholesterol $0.94{\pm}0.30\;mg/g$ Triglycerides $15.56{\pm}1.84\;mg/g$ Free fatty acids $1.26{\pm}0.56\;mg/g$ Phospholipids $29.84{\pm}6.11\;mg/g$ Total lipids $62.44{\pm}12.13\;mg/g$ (2) The garlic administration brought significant elevations in the hepatic contents of free and esterified cholesterds, triglycerides, free fatty acids, and phospholipids in the mouse, when it was injected in excess (2%, 10% suspension) intraperitoneally. (3) The total hepatic lipids of mouse were increased in corelation with the amount of the garlic administered to the animals. (4) A brief discussion was made on the results obtained.

  • In order to compare the effects of epinephrine, cortisone or insulin on the hepatic storage of triglycerides, the fatty liver was induced in mice by the subcutaneous injection for 30 days of either epinephrine, cortisone or insulin. The morphological characteristics of fatty liver in relation to the inducing agents was observed with the following results. 1. The fatty liver induced by epinephrine or cortisone was characterized by the centrilobular accumulation of triglycerides although the degree of accumulation by epinephrine appeared to be much higher than that by cortisone. 2. The fatty liver produced by insulin, however, was characterized by the infiltration of small-sized lipid droplets scattering all over the hepatic lobules.

Effect of Platycodon Grandiflorum Aqueous Extract on Lipid Levels in Rats

  • This study investigated the effects of Platycodon grandiflorum aqueous extract on lipid concentration of serum and liver in rats fed high cholesterol diet Male Sprague-Dawley rats were assigned to three groups (control, low dose of extract, high dose of extract) for four weeks. The serum total cholesterol concentration was significantly lower in the low and high doses of extract groups than in the control group. There was a significant decrease in the free cholesterol, cholesterol ester, LDL-cholesterol, and triglycerides concentrations in serum, and the total cholesterol and the triglycerides contentin liver in the low and high doses of extract groups compared to the control group. When the serum phospholipid concentration was compared among the groups, it was significantly lower in high dose of extract group than in control and low dose of extract group. It can be postulated that Platycodon grandiflorum aqueous extract may possess substantial hypolipidemic properties in rats.

  • The lipid composition of total serum lipids were investigated by latroscan TH-10 TLC analyzer in 69 healthy subjects and 62 patients with hypercholesterolemia. Total serum lipids level in healthy subjects was higher in females than males. There was a increase in total serum lipids level with increasing total serum cholesterol level in patients with hypercholesterolemia, regardless sex. Total serum lipids obtained from healthy subjects and patients with hypercholesterolemia were separated into phospholipids, free cholesterol, free fatty acids, triglycerides and cholesterol esters. Increasing the level of total serum cholesterol in the patients resulted in a decrease in the level of phospholipids, which was a major lipid fraction, indicating that HDL level in the patients was also decreased. However, cholesterol esters, triglycerides and free cholesterol levels in the patients were higher than those found in healthy subjects. The FC/CE ratio for male patients was lower than for healthy males, while in female patients, this ratio was similar to that observed in healthy females.

A study of serum lipid level and relation between the serum lipid level and coronary aneurysm after kawasaki disease (가와사끼병에서 혈청 지질 대사의 변화 및 관상동맥류에 관한 연구)

  • We measured plasma concentrations of high density lipoprotein, total cholesterol and triglycerides in 31 patients with history of kawasaki disease during acute stage and convalescence stage. High density lipoprotein cholesterol and total cholesterol concentrations were significantly lower(P<0.001) in samples taken within 11 days of the onset of illness($28.7{\pm}12.4mg/dl$ and $145.20{\pm}29.60mg/dl$) than in the second samples taken 1-2months after onset of disease($51.5{\pm}15.2mg/dl$ and $175.4{\pm}29.0mg/dl$). Change of triglycerides was not significant. There was no correlation between the serum cholesterol cencentration and coronary aneurysm and continued long term surveillance of much population is necessary to monitor lipid level and their relation to development of premature coronary atherosclerosis.

An Update on Hypertriglyceridemia-Induced Acute Pancreatitis (고중성지방혈증 급성췌장염의 최신 지견)

  • Hypertriglyceridemia a major cause of acute pancreatitis, accounting for up to 10% of all cases. The pathophysiological mechanism of hypertriglyceridemia-induced acute pancreatitis (HTGP) is presumed to involve the hydrolysis of triglycerides by pancreatic lipase resulting in an excess of free fatty acids and elevated chylomicrons, which are thought to increase plasma viscosity and induce ischemia and inflammation in pancreatic tissue. Although the clinical course of HTGP is similar to other forms of acute pancreatitis, the clinical severity and associated complications are significantly higher in patients with HTGP. Therefore, an accurate diagnosis is essential for treatment and prevention of disease recurrence. At present, there are no approved guidelines for the management of HTGP. Different treatment modalities such as apheresis/plasmapheresis, insulin, heparin, fibric acids, and omega-3 fatty acids have been successfully implemented to reduce serum triglycerides. Following acute phase management, lifestyle modifications including dietary adjustments and drug therapy are important for the long-term management of HTGP and the prevention of relapse. Additional studies are required to produce generalized and efficient treatment guidelines for HTGP.

Effects of Taurine on Lipid Metabolism and Protein Synthesis in Poultry and Mice

  • In this study, we have attempted to understand the effects of taurine on serum and liver concentrations of cholesterol and triglycerides in broiler chickens and mice in the post-absorptive state, and on in vitro protein synthesis in the livers of broiler chickens and laying hens, as well as the effects of taurine on in vivo protein synthesis in the liver of mice. The experimental animals were subjected to 24 h of starvation in order to perpetuate a post-absorptive state. Serum concentrations of high density lipoprotein cholesterol and triglycerides were significantly (p<0.05) higher in the taurine groups than in the controls in both the broilers and the mice. However, taurine resulted in a significant (p<0.05) reduction in liver concentrations of total cholesterol and triglycerides, relative to what was seen in the control groups of both animals. Taurine stimulated the in vitro synthesis of 57-kDa, 40-kDa and 23-kDa proteins in the liver of broilers, but inhibited the in vitro synthesis of 54-kDa, 37-kDa and 24-kDa proteins. Taurine in the liver of laying hens exerted effects on in vitro protein synthesis, with the exception of the 26-kDa protein which was not detected in broiler liver, but was inhibited by taurine in the liver of laying hens. Unlike the findings regarding in vitro protein synthesis in the liver of broilers or laying hens, taurine appeared to stimulate the synthesis of only two proteins, a 47-kDa and a 40-kDa protein, in the liver of mice. Overall, theses findings indicate that taurine treatment results in a reduction in cholesterol and triglyceride concentrations, and also affects protein synthesis in the livers of broilers, laying hens, and mice.

Relationship between Metabolic Syndrome and Cataract in a Middle-Aged Men over the Age of 40's -Based on the 5th Korean National Health and Nutrition Examination Survey- (40세 이상 중장년의 대사증후군과 백내장과의 관련성 -제5기 국민건강영양조사 자료중심으로-)

  • Objectives: This study aimed to analyze the relationship between metabolic syndrome and cataract in a middle-aged men over the age of 40's by using the data from the 5th Korean National Health and Nutrition Examination Survey(2010~2012). Methods: The subjects for the analysis were comprised of 5,808 patients who were at least 40 years or more with metabolic syndrome and cataract. The comparison between the metabolic syndrome and ophthalmic diseases according to the health demographic characteristics, health behavior and health conditions was analyzed with ${\chi}^2-test$. The complex samples logistic regression analysis was used to calculate the odds ratio of cataract according to the health demographic characteristics, health behavior and health conditions and the combination of the comprising factors of the metabolic syndrome. Results: After controlling the confounding factors, the odds ratio of cataract in the metabolic syndrome group was OR 1.30, which was significantly high (p<0.01). Furthermore, the odds ratio of cataract was significantly high in the case of males, as the age increased, educational level decreased, with exercising habits of less than 3 days a week, sun exposure of at least 5 hours, with hypertension and diabetes (p<0.01). The results of analyzing the odds ratio for the occurrence of cataract with the components of metabolic syndrome were OR 1.36 (concurrent high fasting blood glucose, high triglycerides, low HDL cholesterol), OR 1.63 (concurrent high fasting blood glucose, high triglycerides, high waist circumference) and OR 1.33 (high triglycerides, low HDL cholesterol, high waist circumference), which showed significantly high odds ratio for cataract occurrence (p<0.05). Conclusions: We come to know that the relation between metabolic syndrome and components are associate with cataract. Therefore the thorough management of metabolic syndrome and components is needed to prevent cataract.

Influence of Periodicity of Eating on Body Fat Accumulation and Lipases in Rat Adipose Tissue (급식횟수가 체내의 지방축적과 지방조직의 lipase에 미치는 영향)

  • Activities of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) in adipose tissue, accumulation of carcass fat, and serum triglyceride have been determined in meal-fed (MF) and ad libitum-fed (AD) rats. At each feeding frequency, the animals received diets providing total fat as 15% or 30% of calories and polyunsaturated fatty acids (PUFA) as 2.5% or 11% of calories. The food intake of the MF rats was 75% of that consumed by the AD rats but MF rat utilized their food more efficiently, as evidenced by weight gain per 100 Kcal consumed. Meal feeding, as contrasted to ad libitum feeding, resulted in greater activities of both LPL and HSL. This suggested a higher turnover of fat in the adipose tissue of MF rats. In AD rats, body fat was significantly correlated with LPL and the ratio of LPL/HSL. Meal feeding significantly increased the ratio of LPL/HSL, indicating a greater capacity for energy storage and fat deposition in the MF rat. However, at the limited caloric intake, MF rats failed to realize this potential; there was no significant difference in percentage of body fat at the two feeding frequencies. Body fat deposition was greater in rats fed the 30% fat diet, as compared with the 15% diet, regardless of the rate of food ingestion. This was coupled with a higher ratio of LPL/HSL. The significant correlation of serum triglycerides with body fat and with the ratio of LPL/HSL in AD rats suggests that LPL activity and fat deposition may be controlled by the concentration of circulating triglycerides. Both serum triglycerides and adipose LPL activity were significantly reduced when the diet contained high levels of PUFA. The percentage of body fat was also lower in animals whose intake of PUFA was high.

Autoxidative Stability of Triglyceride Molecular Species (트리글리세리드 분자종의 산화안정성에 관한 연구)

  • The influence of triglyceride molecular species on autoxidation was investigated by determining the residual molecular species after incubating soybean oil triglycerides. The molecular species of soybean oil triglycerides were analyzed by capillary column gas chromatography and electron impact ionization mass spectrometry utilizing selected ion monitoring. The autoxidative stability of each molecular species in soybean oil triglycerides appeared to decrease in proportion to the increase in the number of double bonds present in the acyl residues, and it was affected by degree of unsaturation of fatty acid when the number of double bonds in triglyceride were the same. And it appeared to be enhanced by a decreases in the length of the saturated acyl chain present in the glycerides.
