Effects of Factors on Serum Lipid Levels in Suwon Adult Males (수원지역 성인 남성들의 혈청지질 수준에 영향을 미치는 요인)

  • This study was performed to find and compare the characteristics of dietary factors and obesity indices on serum lipid profiles using 407 healthy adult males that had physical examinations conducted at the general hospital located in Suwon. The subjects were divided 56.3% for the normal group, 16.7% for TG, 14.5% for TC and 12.5% for TC TG group, respectively. When comparing the blood lipid profiles to normal group, TC group was high in both total cholesterol and LDL-cholesterol, TG group was high in triglyceride and low in HDL-cholesterol, TC TG group was high in triglyceride, total cholesterol, LDL-cholesterol and low in HDL-cholesterol. In comparison of blood lipid profiles with normal group, among anthropometric factors, body weigh, BMI (Body Mass Index) and WHR (Waist Hip Ratio) showed significant differences. TC group was high in both total cholesterol and LDL-cholesterol, and TG group was high in triglyceride and low in HDL-cholesterol. TC TG group was high in triglyceride, total cholesterol and LDL-cholesterol, and was low in HDL-cholesterol. Body weigh of TG group was significantly higher than that of normal group (p<0.05). BMI and WHR of TG group and TC TG group were significantly higher than those of normal group (p<0.05). Total cholesterol was mainly related to the intake of animal lipids and alcoholic beverage. Serum triglyceride levels were closely related with lipid intake and high BMI in this group. In TC TG group, BMI and WHR were high. As well, serum LDL-cholesterol was high and HDL-cholesterol was low which indicates TC TG group had the most undesirable blood lipid profiles. Therefore, it may be necessary to manage BMI and WHR to prevent hyperlipidemia and obesity for adult males in Suwon.

Caspase-2 mediates triglyceride (TG)-induced macrophage cell death

  • Triglyceride (TG) accumulation causes macrophage cell death, which affects the development of atherosclerosis. Here, we examined whether caspase-2 is implicated in TG-induced macrophage cell death. We found that caspase-2 activity is increased in TG-treated THP-1 macrophages, and that inhibition of caspase-2 activity drastically inhibits TG-induced cell death. We previously reported that TG-induced macrophage cell death is triggered by caspase-1, and thus investigated the relationship between caspase-2 and caspase-1 in TG-induced macrophage cell death. Inhibition of caspase-2 activity decreased caspase-1 activity in TG-treated macrophages. However, caspase-1 inhibition did not affect caspase-2 activity, suggesting that caspase-2 is upstream of caspase-1. Furthermore, we found that TG induces activation of caspase-3, -7, -8, and -9, as well as cleavage of PARP. Inhibition of caspase-2 and -1 decreased TG-induced caspase-3, -7, -8, and -9 activation and PARP cleavage. Taken together, these results suggest that TG-induced macrophage cell death is mediated via the caspase-2/caspase-1/apoptotic caspases/PARP pathways.

Effects of CPs (Combined Preparations) of Radix Salvia Miltiorrhiza, Radix Notoginseng, and Borneolum on Hyperlipidemia (CPs(단삼, 삼칠, 빙편 복합제)가 고지혈증에 미치는 영향)

  • Objectives: In order to determine the effect of CPs, experimental studies were performed in hyperlipidemia humans. The contents of serum total cholesterol, TG (triglyceride), LDL cholesterol, HDL cholesterol, AST/ALT and BUN/creatinine were measured. Methods: We made a comparative study of total cholesterol, TG (triglyceride), LDL cholesterol, HDL cholesterol, AST/ALT and BUN/creatinine during one month off and on for twenty patients with hyperlipidemia who visited Gunpo Oriental Medical Center of Wonkwang University from December 2002 to July 2003. Results: The following result were obtained: 1. The serum TG and total cholesterol of patients who took CPs for one month showed significant (p<0.01) decrease. 2. The serum HDL-cholesterol and LDL-cholesterol of patients who took CPs for one month showed significantly increase in HDL, and decrease in LDL (p<0.05). 3. The serum AST/ALT and BUN/creatinine of patients who took CPs for one month showed no significant effects. Conclusions: According to the results mentioned above, the effects of CPs on serum triglyceride and total cholesterol of hyperlipidemia patients were significantly confirmed for decreasing effects.

The Effects of Dietary Carbohydrate on Serum Triglyceride Concentrations in Korea (우리나라 성인의 당질섭취가 혈청 중성지방수준에 미치는 영향)

  • The relationship between dietary carbohydrate (CHO) and fasting triglyceride (TG) concentrations was examined in 2,689 men, 1,908 premenopausal women, and 966 postmenopausal women aged 20-69 years in the 1998-Korean Health and Nutrition Survey. The serum TG concentrations in fasting blood samples were measured and dietary CHO intake was assessed by using 24-hour recall method. As CHO intake increased, the TG concentrations were significantly affected (127.4 mg/dl, 132.4 mg/dl, 134.8 mg/dl, 142.6 mg/dl, 147.2 mg/dl) in postmenopausal women, while CHO intake was not significantly associated with TG concentrations in men and in premenopausal women. For the lowest to the highest quintiles of CHO, the mean TG concentrations were 120.0 mg/dl and 140.3 mg/dl in postmenopausal women with body mass index (BMI) <25 kg/m$^2$ and 135.6 mg/dl & 155.3 mg/dl in postmenopausal women with BMI$\geq$25 kg/m$^2$. In multivariate analysis, BMI, waist hip ratio (WHR), glucose, smoking, and drinking habit were associated with serum TG concentrations in men. In premenopausal women, BMI, WHR, and glucose levels were associated with serum TG concentrations, while age, BMI, WHR, glucose levels, and CHO intake were associated with serum TG concentrations in postmenopausal women. In conclusion, CHO intake is related to serum TG concentrations in postmenopausal women in Korea.

Effects of Intaking of Red Ginseng Products on Correlationship between Obesity and Blood Lipids (홍삼류의 섭취가 비만과 혈중 지질의 상호관계에 미치는 영향)

  • Obesity is the risk factor of atherosclerosis and not only increases triglyceride concentration in blood but also decreases relatively the ratio of TG to HDL-Cholesterol in blood. In case of obesity, systolic blood pressure is also increased in responding the increase of TG in blood. Index of obesity in red ginseng-taking group (ginseng group) was lower as compared with non-red ginseng-taking group (control group). The TG concentration, the ratio of triglyceride to HDL-cholesterol in blood and systolic blood pressure were decreased in the subjects of ginseng group compared with that in control group. It is inferred that long-term intake of ginseng products may help to prevent the risk of atherosclerosis and obesity.

Serum Lipids by Gender, Age and Lifestyle in Korean Adults (일부 한국성인의 성, 연령 및 생활습관에 따른 혈청지질 농도)

  • The purpose of this study was to measure serum lipids concentrations by gender and age, and to investigated the relation of serum lipids levels to cigarette smoking alcohol intake exercise and menopausal status in Korean adults. The subjects consisted of 463 healthy adult(29- men and 173 women) volunteers who were visiting the health Promotion Center in the Asan Medical Center from July 1, 1994 to July 22, 1994 Subjects were ambulatory free-living people and aged 20- 80 years, Anthropometric measurement was performed and serum triglyceride(TG), HDL-cholesterol and total cholesterol(TChol) concentrations were measure by automatic system(Hitachi 736-40). Data on life styles were obtained using structured questionnaires issued by a trained interviewer, All data were statistically analyzed by PC-SAS(Version 6.04) Serum concentrations of triglyceride and total cholesterol was higher(p<0.05) in men than in women, and serum HDL-cholesterol was lower(p<0.05) in men than in women. The prevalence of hyperlipidemia(TG>170 mg/dL and TChol>220 mg /dL) was 12.5% in men and 4.1% in women. The levels of serum triglyceride, total cholesterol and BMI increased with age in women. The distribution curves of serum triglyceride, total cholesterol and LDL-cholesterol of women were located in lower increased more rapidly with aging than in men, The distribution curves of BMI of women increased with aging after the thirties, The BMI of women was located in higher levels than of men in their sixties. The serum concentration of triglyceride and total cholesterol of male current-smokers and non- smokers were significantly(p<0.05) higher than those of ex-smokers. The concentrations triglyceride the male current0drinkers were higher(p<0.05) than those of male non-drinkers. In non-smokers and non- drinkers, serum TG concentration were lower (p<0.05) than those of current-smokers and current-drinkers. In exercisers, the serum triglyceride concentration was significantly(p<0.05) lower than in non- exercisers. In postmenopausal woman BMI and serum concentrations of TChol, TG and LDL-Chol were significantly by age and gender and smoking alcohol drinking, exercise and menopausal staus might be a better indicator to predict serum lipids in both genders.

Inducible Nitric Oxide Synthase Mediates the Triglyceride-induced Death of THP-1 Monocytes

  • Triglyceride (TG) accumulation can cause monocytic death and suppress innate immunity. However, the signaling pathways involved in this phenomenon are not fully understood. This study aimed to examine whether inducible nitric oxide synthase (iNOS) is involved in the TG-induced death of THP-1 monocytes. Results showed that iNOS was upregulated in TG-treated THP-1 monocytes, and iNOS inhibition blocked TG-induced monocytic death. In addition, TG-induced poly (ADP-ribose) polymerase (PARP) cleavage and caspase-3 and -7 activation were suppressed by iNOS inhibition. Furthermore, the expression of X-linked inhibitor of apoptosis protein (XIAP) and survivin, which inhibit caspase-3 and -7, was reduced in TG-treated THP-1 monocytes, but iNOS inhibition recovered the TG-induced downregulation of XIAP and survivin expression. Considering that TG-induced monocytic death is triggered by caspase2 and -8, we investigated whether caspase-2 and -8 are linked to the TG-induced expression of iNOS in THP-1 monocytes. When the activities of caspase-2 and -8 were inhibited by specific inhibitors, the TG-induced upregulation of iNOS and downregulation of XIAP and survivin were restored in THP-1 monocytes. These results suggest that TG-induced monocytic death is mediated by the caspase-2/caspase-8/iNOS/XIAP and survivin/executioner caspase/PARP pathways.

A Study of Salmon Oil Type Analysis by FT-IR and Carbon Isotopes Ratio (FT-IR과 탄소동위원소 분석을 통한 연어유의 구분에 관한 연구)

  • This study analyzes the structure types of salmon oil to evaluate the purity of salmon oil products based on the 38 different types of imported salmon oil products distributed in the Republic of Korea. The major types of omega-3 foods in the salmon oil are ethyl ester (EE) and triglyceride (TG). If the salmon oil contained potential contaminants and was processed in order to remove it, EE type omega-3 fatty acids are found in concentration. This provides a good guide in assessing if products were made with EE type ingredients or re-esterified contaminated materials. The results of the FT-IR analysis showed significant difference in the C=O, C-O band positions in TG and EE. There were 19 TG type products and 19 EE type products. The analysis of carbon isotope ratio was performed on the types of TG and EE. There were different properties in the 19 TG type products. In one product, the carbon isotope ratio was -25.15 and the other 18 products showed -22.15~-23.96. The carbon isotope ratio of all 19 EE type products showed -21.91~-23.74. The results of the TLC analysis showed similar results with FR-IR. The re-esterified TG form was not detected in the TG type products, confirming that the TG type products contained natural salmon oil. This study aimed to provide the basic material in classifying the types of natural salmon oil and re-esterified salmon oil, by analyzing the pattern and proportion of FT-IR spectrum, carbon isotope ratio, and TLC.

Cytotoxic Effect of Triglycerides via Apoptotic Caspase Pathway in Immune and Non-immune Cell Lines

  • Hyperlipidemia is defined as conditions of the accumulation of lipids such as free fatty acids (FFA), triglyceride (TG), cholesterol and/or phospholipid in the bloodstream. Hyperlipidemia can cause lipid accumulation in non-adipose tissue, which is lipid-cytotoxic effects in many tissues and mediates cell dysfunction, inflammation or programmed cell death (PCD). TG is considered to be a major cause of atherosclerosis through inflammatory necrosis of vascular endothelial cells. Recently, TG have also been shown to exhibit lipid-cytotoxicity and induce PCD. Therefore, we investigated the effect of TG on the cytotoxic effect of various cell types. When exposed to TG, the cell viability of U937 monocytes and Jurkat T lymphocytes, as well as the cell viability of MCF-7, a non-immune cell, decreased in time- and dose-dependent manner. In U937 cells and Jurkat cells, caspase-9, an intrinsic apoptotic caspase, and caspase-8, an extrinsic apoptotic caspase, were increased by exposure to TG. However, in TG-treated MCF-7 cells, caspase-8 activity increased only without caspase-9 activity. In addition, the reduction of cell viability by TG was recovered when all three cell lines were treated with pan-caspase inhibitor. These results suggest that activation of apoptotic caspases by TG causes lipotoxic effect and decreases cell viability.

Triglyceride Down-regulates Expression of MSR-1 in PMA-induced THP-1 Macrophages

  • Atherosclerosis is a cardiovascular disease in which plaque builds up inside of an artery and can lead to various complications such as myocardial infarction, stroke, and thrombosis. Recently, hypertriglyceridemia has attracted significant attention as contributors to development of atherosclerosis. However, molecular mechanism of its contribution to atherosclerosis is poorly understood. Here we proposed a potential link between triglyceride (TG) and atherosclerosis. TG treatment promoted downregulation of certain scavenger receptor, macrophage scavenger receptor-1 (MSR-1) in phorbol myristate acetate (PMA)-derived human macrophages. TG treatment caused reduction of MSR-1 mRNA expression in a time- and dose-dependent manner. Using chemical inhibitors, we found that inhibition of signaling pathways associated with PI3K and PLC enhances TG-induced reduction of MSR-1 expression in THP-1 macrophages implying that PI3K and PLC is implicated in the expression of MSR-1 in macrophages. Since MSR-1 is associated with uptake and clearance of atherogenic lipoprotein, oxidized low density lipoprotein (oxi-LDL), our data suggest that increase of oxi-LDL due to TG-mediated reduction of its receptor MSR-1 can promote atherosclerosis.
