Trigger Factor Interacts with DnaA Protein to Stimulate its Interaction with DnaA Box

  • While screening proteins that interact with DnaA protein, the initiator protein for Escherichia coil chromosomal DNA replication, we found a 52-kD sized protein which bound to DnaA protein in a salt-dependent manner. This protein was identified as trigger factor, a ribosome-associated peptidyl-prolyl- cisltrans isomerase with chaperone activity. Trigger factor was overproduced and purified to near homogeneity, and its effect on the function of DnaA protein was examined, Enhanced binding of DnaA protein to DnaA box with no apparent supershift in the gel-shift experiments suggested that trigger factor, by virtue of its chaperone activity, exerts a change on DnaA protein thus increasing its binding affinity for DnaA box.

A Clinical Study of Trigger Point by Blocking Methods (유발점 차단에 의한 임상적 연구)

  • The purpose of this study is the evaluation of the degree of post injection soreness, symptom duration, factor and autonomic symptoms after trigger point injection in patients with trigger points. We devided the subjects of the study into four groups Such as, only dry needling, needle-TENS, with massage-stretch, massage-stretch only, including 100 patients, and measured the visual analog scale before treatment and after treatment. Before treatment, The VAS mean scores were $6.2{\pm}1.03$ in needle-TENS with massage-stretch group ; $6.2{\pm}1.75$ in needdle-TENS group, and $6.3{\pm}1.85$ in dry needling group, and $6.8{\pm}1.03$ in massage-stretch group. In post injection 3rd day, The VAS mean score were $0.9{\pm}1.78$ in needling-TENS with massage-stretch group, $1.1{\pm}1.52$ in needling-TENS group, $1.7{\pm}1.10$ in dry needling group, and $3.9{\pm}3.01$ in massage-stretch group. As for a causative factor of activities for trigger were overload with 37.0%, overwork with 35.0% and fatigue with 13.0%. Symptoms for trigger were tenderness with 28.0%, numbness and tingling with 24.0%. ROM limit with 17.0% and tightness with 17.0%. As a result, needling-TENS with massage-stretch group showed less soreness and effect than other group.

Comparison of Ultrasonography Images on Normal Muscle and Myofascial Trigger Points Activated Muscle (정상근과 근막 유발점이 활성화된 근육의 초음파 영상의 비교)

  • Purpose: The objective of this study was to offer primary clinical data examining whether change of imaging structure and quantitative evaluation of muscle activity on myofascial trigger points can lead to implementation of an analytical technique for evaluation of myofascial pain diagnoses. In addition, we examined the effect of a variety of mediation techniques, in order to examine neuromuscular physiological characteristics of myofascial trigger points muscle by comparing differences in pressure pain threshold and ultrasound imaging. Methods: Participants in the study included 30 adults in their twenties. The subjects were divided into the normal and myofascial trigger points groups. Clinical outcomes were evaluated by pressure pain threshold for pain and ultrasound imaging was performed for evaluation of the structural characteristics of muscle. Independent t-test was used for statistical analysis. Results: The two groups showed statistical significance in the change in pressure pain threshold (p<0.05). Findings of ultrasound imaging analysis showed no significant differences, increased muscle thickness was observed (p>0.05). Findings of ultrasound imaging analysis showed significant differences, increased muscle echodensity was observed (p<0.05). Findings on ultrasound imaging analysis showed significant differences, increased muscle white area index was observed (p<0.05). Conclusion: From these results, active myofascial trigger points muscle showed quality deterioration on ultrasound imaging. Thorough evaluation of imaging structure and physiological characteristics can be useful quantitative analytical techniques for diagnosis of myofascial pain syndrome and a primary factor reflected in physical therapy intervention.

Ultrasound Imaging and Electrophysiological Characteristic According to Activity Levels of Myofascial Trigger Point (근근막 발통점의 활성도에 따른 초음파 영상구조 및 전기생리학적 특성)

  • Purpose : This study is to offer clinical primary data that examines the change of imaging structure and the quantitative evaluation of muscle activity on myofascial trigger points. This study examines neuromuscular physiological characteristic by comparing the differences in physical findings, pressure pain threshold, imaging, and electrophysiological characteristics in latent and active myofascial trigger points muscle and normal muscle through the following experimental procedures. Methods : The participants for the study were thirty-three adults in their twenties. We divided three groups into normal, latent and active myofascial trigger points groups by physical findings. We analyzed the results of measured pressure pain, threshold for pain, ultrasound imaging perform for structure characteristic of muscle, surface EMG according to type of muscle contraction for function of muscle contraction. Results : Significant differences were indicated in pressure pain threshold (p<0.05). Significant differences were discovered in the ultrasound imaging analysis. There were increases in muscle Echogenicity white area index (p<0.001). There were significant differences that decrease in %MVIC (p<0.05), increase in MDF (p<0.05). Conclusion : From these results, active rnyotascial trigger points muscle showed quality deterioration on ultrasound imaging and decreased function of muscle contraction, increased motor unit action potential of II type fiber, and electrophysiologically. Imaging structure and neuromuscular physiological characteristic can be diagnostic and quantitative analytical techniques for myofascial pain syndrome and a primary factor that reflected in physical therapy intervention.

A Study on Flash Flood Warning Trigger Rainfall in Mountainous Area (산악지역 돌발홍수 기준우량 결정에 관한 연구)

  • The purpose of this study is to estimate the critical flood discharge and flash flood trigger rainfall for alarm system providing for a flash flood in mountainous area. The flash flood need non-linear approaching method, because rainfall-runoff is nonlinear and it is difficult to explain the existing linear rainfall-runoff. Hydrological characteristics would be utilized to apply such as hydrologic modelling or basin management. This study was effectively estimated a topographic characteristic factor of basin using the GIS. Especially, decided stream order using GIS at stream order decision that is important for input variable of GCIUH. A flash floods defined as a flood which follows shortly after a heavy or excessive rainfall event, with a few hours. In this study, we gave a definition that a critical flood for alarm is the flood when valley depth judging dangerous depth is over 0.5m depth from the bottom of channel. Result that calculate threshold discharge to use GCIUH, at the Mureung valley basin, flash flood trigger rainfall was 16.34mm in the first 20minutes when the threshold discharge was $14.54m^3/sec$.

An Analysis for Goodness of Fit on Trigger Runoff of Flash Flood and Topographic Parameters Using GIS (GIS를 이용한 돌발홍수의 한계유량과 유역특성인자의 적합도 분석)

  • Recently, local heavy rain for a short term is caused by unusual changing in the weather. This phenomenon has, several times, caused an extensive flash flood, casualties, and material damage. This study is aimed at calculating the characteristics of flash floods in streams. For this purpose, the analysis of topographical characteristics of water basin through applying GIS techniques will be conducted. The flash flood prediction model we used is made with GCIUH (geomorphoclimatic instantaneous unit hydrograph). The database is established by the use of GIS and by the extraction of streams and watersheds from DEM. The streams studied are included small, middle and large scale watersheds. For the first, for the establishment or criteria on the flash flood warning, peak discharge and trigger runoff must be decided. This study analyzed the degree or aptitude of topographical factors to the trigger runoff calculated by GCUH model.

Effect of a dual trigger on oocyte maturation in young women with decreased ovarian reserve for the purpose of elective oocyte cryopreservation

  • Objective: The aim of this study was to determine whether co-administration of a gonadotropin-releasing hormone (GnRH) agonist and human chorionic gonadotropin (hCG) for final oocyte maturation improved mature oocyte cryopreservation outcomes in young women with decreased ovarian reserve (DOR) compared with hCG alone. Methods: Between January 2016 and August 2019, controlled ovarian stimulation (COS) cycles in women (aged ≤35 years, anti-Müllerian hormone [AMH] <1.2 ng/mL) who underwent elective oocyte cryopreservation for fertility preservation were retrospectively analyzed. Results: A total of 76 COS cycles were triggered with a GnRH agonist and hCG (the dual group) or hCG alone (the hCG group). The mean age and serum AMH levels were comparable between the two groups. The duration of stimulation, total dose of follicle-stimulating hormone used, and total number of oocytes retrieved were similar. However, the number of mature oocytes retrieved and the oocyte maturation rate were significantly higher in the dual group than in the hCG group (p=0.010 and p<0.001). After controlling for confounders, the dual-trigger method remained a significant factor related to the number of mature oocytes retrieved (p=0.016). Conclusion: We showed improved mature oocyte collection and maturation rate with the dual triggering of oocyte maturation in young women with DOR. A dual trigger appears to be more beneficial than hCG alone in terms of mature oocyte cryopreservation for young women with DOR.

  • From July 1th 2001 to October 31th 2001, the Clinical investigation studies were carried out 10 cases of patient with Bell' palsy using Trigger point treatment group, who had been treated in Han-seung Oriental Medical Clinic. The results were like below: 1. The distribution of sex : Male were 4cases. female were 6case and the female to male ratio was 1.5 : 1. 2. The distribution of contributing factor : Unknown reason was most predominant 4 cases, and then were revealed to labor too hard. mental stress(3 cases), trauma common cold(1 case) in turn. 3. The distribution of perid to fall ill : Under a week was the most number 6cases, and then were revealed a week under two weeks(3 cases), over two weeks(1 case) in turn. 4. About period of treatment : The Trigger Point treatment group is shorter than the Acupuncture treatment group. 5. The distribution of grade on admission in descending order : Gr I was the most number and then GrII, GrIII in turn. 6. The distribution of clinical symptoms on admission : Mastoid pain was the most number 9 cases and then lacrimal gland disorder 8 cases, dysgeusia 4cases, hyperacusis 2 cases, the others 2 cases in turn. 7. About effect of treatment : The Trigger point treatment group is more excellent than the Acupuncture treatment group. 8. The more patient age is young, the more effect of treatment was excellent, had nothing to do with both groups. 9. The more perid to fall ill is short, the more effect of treatment was excellent. had nothing to do with both groups. 10. About period of improved frontal belly : The Trigger point treatment group is more excellent than the Acupuncture treatment group.

Study on the variation of cellular physiology of Escherichia coli during high cell density cultivation using 2-dimensional gel electrophoresis

  • Physiological changes of Escherichia coli during the fed-batch fermentation process were characterized in this study. Overall cellular protein samples prepared at the different stage of fermentation were separated by 2-dimensional gel electrophoresis (2-DE), and differently expressed 15 proteins, Phosphotransferase enzyme I, GroEL, Trigger factor, ${\beta}$ subunit of ATP synthase, Transcriptional regulator KDGR, Phosphoglycerate mutase 1, Inorganic pyrophosphatase, Serine Hydroxymethyl-transferase, ${\alpha}$ subunit of RNA polymerase, Elongation factor Tu, Elongation factor Ts, Tyrosine-tRNA ligase, DnaK suppressor protein, Transcriptional elongation factor, 30S ribosomal protein S6 were identified using matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS). When bacterial cells grow to high cell density, and IPTG-inducible heterologous protein is produced, expression level of overall cellular proteins was decreased. According to their functions in the cell, identified proteins were classified into three groups, proteins involved in transport process, small-molecule metabolism, and synthesis and modification of macromolecules.
