Morphological Characteristics and URP-PCR Analysis of Hypocrea sp., a Weed Mould of Oyster Mushroom Cultivation (느타리버섯 유해균 Hypocrea sp.의 형태 및 URP-PCR 분석)

  • Recently a serious outbreak of weed mould caused by a species of Hypocrea occurred in oyster mushroom (Pleurotus ostreatus) substrates in Korea. The disease was characterized by a rapid infestation of the oyster mushroom substrates by Hypocrea sp. and subsequent inhibition of fructification of the mushroom. In spite of it's serious losses to the oyster mushroom industry in Korea, etiology and ecology of the disease have not been studied. Morphological characteristics of the fungus were examined and molecular characteristics of the fungus were compared with those of the green moulds (Trichoderma spp.) isolated from oyster mushroom bed. Stromata formed superficially on suface of the substrates were pulvinate to effuse or irreguler, initially white but becoming yellowish brown, measuring $6.0{\sim}13.0{\times}3.0{\sim}11.0mm$. Perithecia were globose to subglobose, immersed in stroma, $223{\sim}263\;(Ave.239.9){\times}167.3{\sim}231\;(Ave.204.1){\mu}m$ in size. Asci were unitunicate, cylindrical, nonamyloid, $82.7{\sim}124.8\;(Ave.103.3){\times}4.1{\sim}5.1\;(Ave.4.9){\mu}m$ in size, 16 part-spored. Ascospores were bullet-shaped or somewhat oblong, hyaline, bicellular, roughened or warted, $5.4{\sim}7.4\;(Ave.6.5){\times}3.6{\sim}5.5\;(Ave.4.7){\mu}m$ in size. This fungus readily form the stroma on PDA. Mycelia on PDA nearly invisible and without cottony aerial mycelium. Optimum temperature for mycelial growth of this fungus was $25^{\circ}C$ on PDA and its growth rate was 15 mm per day. This species did not grow at below 10 and above $35^{\circ}C$. Phialides in culture enlarged in the middle and aggregated to penicillate type. They were very variable, shorted ampulliform and occasionally curved when matured, but cylinderical when young, measuring $11.9{\sim}24.3\;(Ave.\;14.7){\times}2.9{\sim}3.9\;(Ave.\;3.4){\mu}m$ when matured and $7.2{\sim}14.0\;(Ave.\;10.8){\times}2.8{\sim}4.9\;(Ave.\;3.5){\mu}m$ when young. Phialosopres were ovoid to ellipsoid, smooth, measuring $3.5{\sim}7.2\;(Ave.\;4.5){\times}2.6{\sim}3.3\;(Ave.\;2.9){\mu}m$. Nineteen isolates of Hypocrea sp. were analyzed on the basis of molecular characteristics and classified into phenotypic groups. On the basis of RAPD, URP-PCR, the fungus was confirm to monoclonal, and was classified as a different taxon from reported species of Hypocrea and Trichoderma and supposed to be a new species not previously reported in literature.

First Molecular Characterization of Colletotrichum sp. and Fusarium sp. Isolated from Mangrove in Mexico and the Antagonist Effect of Trichoderma harzianum as an Effective Biocontrol Agent

  • The aim of this study was to characterize potential fungal species affecting mangrove species in Mexico. The phytopathogens were identified based on morphological and molecular characteristics using internal transcribed spacer (ITS1/ITS4) primers then sequenced and compared with the other related sequences in GenBank (NCBI). Three fungal species were identified as Colletotrichum queenslandicum (Weir and Johnst, 2012) from black mangrove (Avicennia germinans); Colletotrichum ti (Weir and Johnst, 2012) from white mangrove (Laguncularia racemosa) and buttonwood mangrove (Conocarpus erectus); Fusarium equiseti (Corda) from red mangrove (Rhizophora mangle). In addition, C. ti and F. equiseti were identified from mango Mangifera indica L. sampled close by the mangrove area. This study provides first evidence of anthracnose on four mangrove species caused by Colletotrichum and Fusarium species in the "Términos" coastal lagoon in Campeche State southern Mexico. This is the first time that C. queenslandicum and C. ti are reported in Mexico. F. equiseti has not been reported affecting M. indica and R. mangle until the present work. Little is known regarding fungal diseases affecting mangroves in Mexico. These ecosystems are protected by Mexican laws and may be threatened by these pathogenic fungus. This is the first report of the effect of Trichoderma harzianum TRICHO-SIN as an effective biological control against of Colletotrichum and Fusarium species.

  • This study was carried out to investigate the cultural charateristics of Fomitopsis pinicola in log woods. The optimal conditions for log cultivation as follows; 1) Light effect, natural light 2) $CO_2$, 0.2% below 3) Low temperature shock, $15^{\circ}C$ for 5 days 4) log-length, 100~120cm 5) It takes 52~65 days for primodia induction is possible to harvest fruiting bodies for 5~6 months. Meanwhile, the total of 10 species pest including Trichoderma sp. occurred during the cultivation.

  • Studies were made to optimize the simultaneous hydrolysis-fermentation (SSF) process for the production of ethanol from rice straw. Trichoderma sp. KI 7-2 was selected to produced cellulase by solid culture for SSF. Ethanol production was highest when the SSF process utilized koji culture of the fungus grown on a medium of wheat bran-rice straw 3 : 2 mixture with moisture content of 50% adjusted to pH 4.5 for 7 days as the enzyme source. It was found that pretreatment of the substrate is not necessary. To ferment 1g of rice straw by SSF 2.47 units of cellulase were required, and the initial yeast concentration of 2.5$\times$10$^{7}$ cell/$m\ell$ was found to be sufficient. Optimum pH and temperature for the process were 4.5 and 4$0^{\circ}C$, respectively. It was also found that higher ethanol concentration in the broth can be obtained by the addition of substrate or substrate and enzyme to SSF broth.

Diversity of Fungi from Dokdo Island Soil, Korea and Their Antimicrobial and Hydrolytic Enzyme Activity

  • Dokdo island is located in the northeastern part of Ulleungdo, known as volcanic island. In total, 53 fungal isolates were isolated from Dokdo island soil sample, using dilution plate technique. The isolates were identified on the basis of morphological characteristics and rDNA ITS sequence analysis. Out of them, 41 isolates were identified at the level of species. The dominant fungal species and genera included Fusarium spp., Mucor sp., Clonostachys spp., and Trichoderma sp. The % sequence identity (the number of matches/the complete alignment length) values via NCBI BLAST searching of EML-IF9, EML-MF30-1 and EML-DDSF4 represented 97.19% (485/499) with Clonostachys cf. rosea (GenBank accession no. KC313107), 98.33% (472/480) with Metarhizium guizhouense (GenBank accession no. HM055445), and 100% (350/350) with Mortierella oligospora (GenBank accession no. JX976032), respectively. Three species of C. rosea, M. guizhouense and M. oligospora represented new records of fungi from Dokdo island, Korea. The antimicrobial activities of the fungal strains varied with tested. Two isolates (EML-MFS30-1 and EML-IF9) showed antifungal activity against several fungi including Fusarium oxysporum and Rhizotonia solani. Clonostachys rosea (EML-IF9) showed strong hydrolytic enzyme activity. Our results showed that the antagonistic fungi including Clonostachys rosea will be used as potential biocontrol agents for control of fungal diseases.

  • Mold producing cellulase were isolated from rotten woods, and identified as the three species: Aspergillus niger van Tieghem, Aspergillus schiemanni Thom and Trichoderma viride Pers. In this paper, culture conditions in the media and characteristics of these strains were investigated. Using these strains, we have conducted a research concerning the utilization of farm product waste materia's. 1. Optimum conditions for the cellulase formation were as follows. KM 10-1; pH 5.2-5.5, $35^{\circ}C$, incubation time 6 days. OL 11-1; pH 5.5, $30-35^{\circ}C$, incubation time 6 days. SH 9-2; pH 5.5, $30^{\circ}C$, incubatoin time 6 days. 2. Their cellulase activities in their optimum condition were as follows: KM 10-1; CMC-LP 78.5% CMC-SP 4.0 glucose mg/gm of the cultures/min. OL 11-1; CMC-LP 89.9%, CMC-SP 4.9 glucose mg/gm of the cultures/min. SH 9-2; CMC-I.P 77.4%, CMC-SP 3.9 glucose mg/gm of the cultures/min. 3. Hydrolysis of animal feed containing a large quantity (23-30%) of cellulose by means of the crude enzyme in the selected strains resolved 30% of the cellulose contained in the animal feed.

  • This study was carried out to investigate the antimicrobial activity of the grapefruit seed extract (GFSE) The antimicrobial activity of GFSE was strong enough against such bacteria as Vibrio vulnificus, Vibrio fluvialis, Bacillus cereus, Staphylococcus aureus and Serratia sp. Growth of the above strains was inhibited by the GFSE'S concentration of 50 ppm. The growth of Vibrio vulnificus was completely inhibited by adding the 50 ppm GFSE to the nutrient broth medium with $3\%$ NaCl. The cell counts of Vibrio uulnfficus $5.2\times10^5$ at first in $5\%$ skim milk containing GFSE 0, 10, 30, 50 and 100 ppm were reduced to 35, 48, $5.6\times10^2,\;5.3\times10^3\;and\;9.6\times10^3/ml$ after 120 hours, respectively. And growth of Aspergillus Parasiticus, Asperillus versicolor, Penicillium funiculosum, Py-renochaeta terrestris and Trichoderma viride were inhibited by the concentration of GFSE 100, 50, 100, 10 and 30 ppm, respectively. The shelf life of Mulkimchi containing GFSE 50 and 100 ppm was 20 days longer than the control during storage at $5^{\circ}C\;and\;20^{\circ}C$.

Effects of Dietary Probiotic on Performance, Noxious Gas Emission and Microflora Population on the Cecum in Broiler (복합 생균제 첨가가 육계 생산성, 유해가스 발생량 및 맹장내 균총에 미치는 영향)

  • This study was carried out to investigate the effect of probiotics on the performance, nutrients digestibility, noxious gas emission and microflora population on the cecum of broilers. A total of 120 broilers, consisted of 4 treatments X 3 replicates X 10 broilers per replicates, were fed the experimental diets containing 0, 0.1, 0.3 and 0.5% probiotics for 5 weeks. Broilers fed the diets containing 0.1 and 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the others from the 3rd to 4th week. Broilers fed 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the other levels from the 5th to 6th week. Broilers fed the diets containing 0.1% and 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the others from the 2nd to 6th week. Dry matter digestibility was significantly (p<0.05) improved with 0.3% probiotic. Emission of ammonia and sulfate hydrogen gas was significantly (p<0.05) decreased at 6th week. However, there was no (p<0.05) difference at the levels of 0, 0.1, 0.3 and 0.5% at the 4th weeks. There was an increase in the lactobacillus sp, but there was a decrease in the microflora population of coliforms in the cecum of broiler with 0.1% and 0.3% probiotics. These results indicated that the compound probiotics of 0.1${\sim}$0.3% were effective in the body weight gain, feed conversion, nutrients digestibility, noxious gas emission and microflora population on the cecum in broilers.

  • These studies were conducted to investigate nutrient sources and supplementary materials of synthetic compost media for Agaricus bisporus culture. Investigation were carried out to establish the optimum composition for compost of Agaricus bisporus methods of out-door fermentation and peakheating with rice straw as the main substrate of the media. The incidence and flora of harmful organisms in rice straw compost and their control were also studied. 1. When rice straw was used as the main substrate in synthetic compost as a carbon source. yields were remarkably high. Fermentation was more rapid than that of barley straw or wheat straw, and the total nitrogen content was high in rice straw compost. 2. Since the morphological and physico-chemical nature of Japonica and Indica types of rice straw are greatly dissimilar. there were apparent differences in the process of compost fermentation. Fermentation of Indica type straw proceeded more rapidly with a shortening the compost period, reducing the water supply, and required adding of supplementary materials for producing stable physical conditions. 3. Use of barley straw compost resulted in a smaller crop compared with rice straw. but when a 50%, barley straw and 50% rice straw mixture was used, the yield was almost the same as that using only rice straw. 4. There were extremely high positive correlations between yield of Agaricus bisporus and the total nitrogen, organic nitrogen, amino acids, amides and amino sugar nitrogen content of compost. The mycerial growth and fruit body formation were severely inhibited by ammonium nitrogen. 5. When rice straw was used as the main substrate for compost media, urea was the most suitable source of nitrogen. Poor results were obtained with calcium cyanamide and ammonium sulfate. When urea was applied three separate times, nitrogen loss during composting was decreased and the total nitrogen content of compost was increased. 6. The supplementation of organic nutrient activated compost fermentation and increased yield of Agaricus bisporus. The best sources of organic nutrients were: perilla meal, sesame meal, wheat bran and poultry manure, etc. 7. Soybean meal, tobacco powder and glutamic acid fermentation by-products which were industrial wastes, could be substituted for perilla meal, sesame meal and wheat bran as organic nutrient sources for compost media. B. When gypsum and zeolite were added to rice straw. physical deterioration of compost due to excess moisture and caramelization was observed. The Indica type of straw was more remarkable in increase of yield of Agricus bisporus by addition of supplementing materials than Japonica straw. 9. For preparing rice straw compost, the best mixture was prepared by 10% poultry manure, 5% perilla meal, 1. 2 to 1. 5% urea and 1% gypsum. At spring cropping, it was good to add rice bran to accelerate heat generation of the compost heap. 10. There was significantly high positive correlation (r=0.97) between accumulated temperature and the decomposition degree of compost during outdoor composting. The yield was highest at accumulated temperatures between 900 and $1,000^{\circ}C$. 11. Prolonging the composting period brought about an increase in decomposition degree and total nitrogen content, but a decrease in ammonium nitrogen. In the spring the suitable period of composting was 20 to 25 days. and about 15 days in autumn. For those periods, the degree of decomposition was 19 to 24%. 12. Compactness of wet compost at filling caused an increase in the residual ammonium nitrogen. methane and organic acid during peak heating. There was negative correlation between methane content and yield (r=0.76)and the same was true between volatile organic acid and yield (r=0.73). 13. In compost with a moisture content range between 69 to 80% at filling. the higher the moisture content, the lower the yield (r=0.78). This result was attributed to a reduction in the porosity of compost at filling the beds. The optimum porosity for good fermentation was between 41 and 53%. 14. Peak heating of the compost was essential for the prevention of harmful microorganisms and insect pests. and for the removal of excess ammonia. It was necessary to continue fer mentatiion for four days after peak heating. 15. Ten species of fungi which are harmful or competitive to Agaricus bisporus were identified from the rice compost, including Diehliomyces microsporus, Trichoderma sp. and Stysanus stemoites. The frequency of occurrance was notably high with serious damage to Agaricus bisporus. 16. Diehliomyces microsporus could be controlled by temperature adjustment of the growing room and by fumigating the compost and the house with Basamid and Vapam. Trichoderma was prevented by the use of Bavistin and Benomyl. 17. Four species of nematodes and five species of mites occured in compost during out-door composting. These orgnanisms could be controlled through peakheating compost for 6 hours at $60^{\circ}C$.
