Characterization of antimicrobial proteins produced by Bacillus sp. N32 (Bacillus sp. N32 균주가 생산하는 항균 단백질 특성)

  • An antagonistic bacterial isolate, that inhibits the growth of plant pathogens, was selected and identified from 5,000 isolates screened from the rhizosphere of various crop plants. An isolate Bacillus sp. N32, tested against Colletotrichum gloeosporioides causing anthracnose disease in hot pepper, produced both a heat resistant antifungal protein and a heat sensitive antifungal protein. The heat resistant protein was partially purified by Ammonium sulfate fractionation and gel filtration chromatography. The bioautography showed that the proteins possessed high antifungal activity. The biosynthetic gene cluster responsible for the heat resistant antifungal protein was cloned from cosmid library using DNA probe obtained from PCR product with the primers targeting the conserved nucleotide sequence of the synthetic genes reported earlier, Most of the clones obtained showed higher homology to fengycin antibiotic synthetic gene family reported earlier. On the other hand, the heat sensitive protein was isolated from SDS-PAGE and electroblotting to determine the N-terminal amino acid sequences. The heat sensitive antifungal protein gene was cloned from the ${\lambda}-ZAP$ libraries using a DNA probe based on the N-terminal amino acid sequences of the heat sensitive protein. We are contemplating to clone and sequence the whole gene cluster encoding the heat sensitive protein for further analysis.

Characteristic of Microorganism and Effect Analysis of Spent Mushroom Compost after Cultivation of Button Mushroom, Agaricus bisporus (양송이버섯 재배 후 폐상퇴비의 효과 분석 및 분리 미생물의 특성)

  • This study was carried out to investigate the feasibility for the use of environmental-friendly materials and the effective recycling of spent mushroom compost(SMC) after cultivation of Button Mushroom, Agaricus bisporus. SMC of white button mushroom contained diverse microorganisms including fluorescent Pseudomonas sp., Bacillus sp., Tricoderma sp. and Actinomycetes. These isolates showed the extensive antifungal spectrum against plant pathogen. Among of the isolates, fungal pathogen such as Alternaria brassicicola, Phytophtora melonis, Phytophthora capsici and Colletotichum gloeosporioides strong showed strong antagonistic activity. 45.8% of the isolates were actively colonized on the pepper root and 5.8% showed rhizosphere competent of >$5{\times}10^2cfu\;root^{-1}$. The plant growth promotion ability of the collected isolates were tested in pot experiments using red pepper seedling. Among them, 62.7% showed pepper growth promoting ability and growth of pepper root showed superior to the control. The germination of pepper treated with aqueous extracts of non-harvest SMC completely inhibited at concentration of more than 33%. The sterilization of SMC resulted in higher inhibition of germination and early growth of pepper. These results suggest that spent mushroom compost(SMC) of Button Mushroom may have adequately the feasibility for the use with environmental-friendly materials.

Occurrence and Biological Control of Postharvest Decay in Onion Caused by Fungi

  • Postharvest decay of onion bulbs was examined by inspecting the commercial packages in the market or in storage. Bulb rot incidence was unexpectedly high, and onion bulbs with 1st quality grade were rotten most severely by 51%, followed by 32% for 2nd and 21% for 3rd grades. This indicates that larger bulbs had higher incidences of bulb rots. Major pathogens associated with basal and neck rots were Fusarium oxysporum and Aspergillus sp. or Botrytis allii, respectively, of which basal rot was most prevalent and damaging during storage. Among the epiphytic microorgani는 from onion plants, several Bacillus and Paenibacillus spp. and previously selected Pseudomonas putida and Trichoderma harzianum had inhibitory efficacy against bulb rot pathogens. Among these B. amyloliquefaciens BL-3, Paenibacillus polymyxa BL-4, and P. putida Cha 94 were highly inhibitory to conidial germination of F. oxysporum and B. allii. P. putida Cha 94, B. amyloliquefaciens BL-3, P. polymyxa BL-4, and T. harzianum TM were applied in the rhizoplane of onion at transplanting. Initially antagonist populations decreased rapidly during the first one month. However, among these antagonists, rhizoplane population densities of BL-3, Cha 94, and TM were consistently high thereafter, maintaining about 10$^4$-10$^{5}$ cells or spores per gram of onion root up to harvest time. The other bacterial antagonist BL-4 survived only for two months. TM was the most effective biocontrol agent against basal rot, with the number of rotten bulbs recorded at 4%, while that of the control was 16%. Cha 94 was effective for the first 20 days, but basal rot increased thereafter and had about the same control efficacy as that of BL-3 and BL-4. When the antagonists were applied to the topping areas of onion bulbs at harvest, TM was the most effective in protecting the stored onion bulbs from neck rotting. The second effective antagonist was BL-3. TM and BL-3 completely suppressed the neck rot in another test, suggesting that biocontrol of postharvest decay of onion using these microorganisms either at the time of transplanting or at harvesting may be promising.

Effects of Dietary Probiotic on Performance, Noxious Gas Emission and Microflora Population on the Cecum in Broiler (복합 생균제 첨가가 육계 생산성, 유해가스 발생량 및 맹장내 균총에 미치는 영향)

  • This study was carried out to investigate the effect of probiotics on the performance, nutrients digestibility, noxious gas emission and microflora population on the cecum of broilers. A total of 120 broilers, consisted of 4 treatments X 3 replicates X 10 broilers per replicates, were fed the experimental diets containing 0, 0.1, 0.3 and 0.5% probiotics for 5 weeks. Broilers fed the diets containing 0.1 and 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the others from the 3rd to 4th week. Broilers fed 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the other levels from the 5th to 6th week. Broilers fed the diets containing 0.1% and 0.3% probiotic had higher (p<0.05) body weight gain and feed conversion than those of the others from the 2nd to 6th week. Dry matter digestibility was significantly (p<0.05) improved with 0.3% probiotic. Emission of ammonia and sulfate hydrogen gas was significantly (p<0.05) decreased at 6th week. However, there was no (p<0.05) difference at the levels of 0, 0.1, 0.3 and 0.5% at the 4th weeks. There was an increase in the lactobacillus sp, but there was a decrease in the microflora population of coliforms in the cecum of broiler with 0.1% and 0.3% probiotics. These results indicated that the compound probiotics of 0.1${\sim}$0.3% were effective in the body weight gain, feed conversion, nutrients digestibility, noxious gas emission and microflora population on the cecum in broilers.
