BACKGROUND: The buds of persimmon trees are susceptible to cold damage, often with the late frost, at the time of budburst. This study was conducted to determine effect of the cold damage on shoot and fruit growth the current season. METHODS AND RESULTS: 'Fuyu' trees, grown in 50-L pots, were placed for 1 h at $-2.2{\pm}0.5$, $-2.6{\pm}0.5$, or $-3.0{\pm}0.5^{\circ}C$ within a cold storage, at their budburst on April 5. Some trees under ambient temperature at $10-17^{\circ}C$ served as the control. Cold damage of the buds containing flower buds was 54% at $-2.2^{\circ}C$, and significantly increased to 95% at $-3.0^{\circ}C$. The bud damage included the complete death of all, complete death of main buds only, or the late and deformed shoot growth in the spring. Number of flower buds in early May dramatically decreased as the damage ratio increased. Since the thinning of flower buds in mid-May and fruitlets in early July was done in no or slightly damaged trees, the final number of fruits and yield did not decrease compared with the control when the damage increased by 60% and 70%, respectively. Average fruit weight and skin coloration tended to be better with increasing bud damage. Shoot growth was more vigorous in those trees whose buds were severely damaged by low temperature. CONCLUSION(S): Shoot growth and the yield may depend on the number of flower buds and percent fruit set after the cold damage.

This study was conducted to reveal the effects of local climatic conditions on reproductive growth in a mature stand of Korean white pine based on climatic estimates. For this, the reproductive growth such as production and characteristics of cone and seed were first measured and summarized for seven years from 1974 to 1980. The local climatic conditions in the study site were also estimated by both a topoclimatological method and a spatial statistical technique. The local climatic conditions were then correlated with and regressed on the growth factors to reveal the relationships between the climatic estimates and the reproductive growth. Average number of conelet formation per tree showed highly negative correlation with some climatic variables related to minimum temperature in the year of flower bud differentiation. Especially, the most significant negative correlation were found between average of the minimum temperature for June and July of flower bud differentiation year and the number of conelet formation. There was no significant correlation between the number of cone production and climatic variables. However, total precipitation from December of the flowering year to February of the cone production year showed the most high correlation (r=0.6036) with the number of cone production. It was found that significant climatic variables affecting the amount of cone drop and cone drop percentage were the sum of cloudy days from June of the flowering year to August of the cone production year. Positive correlation was significantly recognized between the average weight of empty seed per cone and total precipitation from December of the flowering year to February of the cone production year. For the percentage of empty seed, five climatic variables among 19 variables were significantly correlated at 10% level. The average weight of a cone showed negative correlation with total precipitation from June of the flowering year to August of the cone production year. It was also found that average weight of a seed had highly negative correlation with total precipitation from December of the flowering year to February of the cone production year. The average weight of cone coat was negatively correlated with two climatic variables derived from clear days, which are sum of clear days from November of the flowering year to March of the cone production year and sum of clear days from December of the flowering year to February of the cone production year. On the other hand, it showed positive correlation with mean temperature of May in the flowering year. The exactly same results were obtained in correlation analysis for the percentage of cone coat.

We investigated the mechanism on occurring of briny injury in four fruit species (apple, pear, grape, peach). Briny injury of apple, pear, peach, and grape was induced by the foliar splay of saline solutions. Browning symptom was initiated one hour after the spray of 3% NaCl in apple, pearl and peach trees. while two hour were needed for grape, but the latter developed the symptom faster than the formers. Foliar spray of 3% NaCl resulted in 4-fold increase in ethylene production 24 h after the spray, and the production was returned to early stage rate after 72 h. Ethylene production of control was nearly constant with slight decrease after 4 h. Browning and defoliation response to the spray of 3% NaCl on lower side were not different from the spray on both sides regardless of fruit species, while no injury symptoms occurred by the spiny on upper side. In saline damage for treated date, lateral bud was only germinated in apple at 30 July. A shoot apex was 100% grew in all fruit tree. In result for hormonal content of leaf and shoot apex, control was high t-zeatin content in leaf. However, IAA and ABA content was increasing in high saline concentration.

Thinning of apple fruitlets is one of the most laborious and important works for the improvement of fruit quality and for the promotion of sufficient flower bud formation to prevent alternate bearing in commercial cultivars. Lateral fruits of self-thinning apple cultivars fall naturally within 30 days after full bloom and only central fruit remains to mature. Differences of gene expression between central fruit and lateral fruit were investigated by differential display (DD) PCR. Partial cDNAs of 30 clones from the central fruit and 24 clones from the lateral fruit were selected for nucleotide sequence determination and homology searches. The levels of transcripts coding for proteins involved in pathogenesis related proteins, senescence, temperature stress, protein degradation, fruit browning, sorbitol metabolism were significantly higher in pedicels of lateral fruit than in pedicels of central fruit. On the other hand, the up-regulation of proteins involved in anthocyanin and flavanol biosynthesis and ethylene synthesis were observed in pedicels of central fruit. In Real time PCR analysis, cytochrome P450 gene was confirmed as showing a higher expression level in lateral fruit than in central fruit. The results of this study indicate that differentially expressed genes are related to self-thinning characteristics in apple tree.

Climate change affects plant responses on physiological characteristics and growth, and Pinus densiflora, one of the major tree species in Korea, are expected to be particularly vulnerable to rising temperature and increased precipitation. This study was conducted to investigate the effects of an open-field warming and precipitation manipulation on physiological characteristics and growth of P. densiflora seedlings. Seedlings of 2-year-old P. densiflora were planted in April, 2013, in open-field nursery located at Korea University. The air temperature of warmed plots had been set to be $3^{\circ}C$ higher than the control plots using infrared lamps. Precipitation was manipulated to be 30% lower or higher than the control, using transparent panels and drip irrigation. Net photosynthetic rate, total chlorophyll content, seedling height, root collar diameter and biomass were measured from April, 2014 to April, 2015. The increase in new shoot biomass from warming was statistically significant, with the biomass in warmed plots about 2-fold higher than in the control plots in 2014 and 2015. This result might be related to advanced bud burst and increased occurrence of abnormal new shoots in warmed plots. Meanwhile, the results of net photosynthetic rate, total chlorophyll content, seedling height, root collar diameter and total biomass from warming and precipitation manipulation were not statistically significant, but tendencies of lower net photosynthetic rate and higher seedling height and biomass in warmed plots compared to the control were shown. Such might be speculated as results of the extended growth period. When root to shoot (R/S) ratio was calculated from the biomass data obtained in April 2014 and April 2015, increased R/S ratio was observed regardless of the treatments applied. Drought tolerance of P. densiflora and particularly low annual precipitation observed in 2014 were suggested as the possible reasons.

Pear cultivar 'Sinhwa' (Pyrus pyrifolia var. culta Nakai) was originated from a cross between 'Niitaka' and Whasan' with the aims of improving the fruit quality and the traits of cultivation and of early maturing more than 'Whasan' cultivar at Pear Research Station of National Institute of Horticultural & Herbal Science, Rural Development Administration in 1995. 'Sinhwa' was preliminarily selected in 2004 and named in 2009. The tree shows vigorous growth habit and semi-spread characters like 'Niitaka'. Furthermore, it has a sufficient and well upkeep of the flower bud, so it can be more easily cultivated in orchards. In the flower characteristics, flowering time of 'Sinhwa' is $11^{th}$ April like as maternal parent 'Niitaka'. Also 'Sinhwa' has short of pollen grains, so it is need above two pollinizer cultivars. 'Sinhwa' is highly resistant to black leaf spot (Alternaria kikuchiana) and relatively strong to pear scab (Venturia nashicola) in field condition. The optimum harvest time is around Sep. $15^{th}$ in Naju, which is ahead of 'Whasan' about 10 days in the harvest period. The fruit shape is oblate and fruit skin color is yellowish-brown during harvesting time. The average weight of fruit is 627 g, and the soluble solids content is $13.0^{\circ}Brix$. The flesh is very soft and juicy, and renders good eating quality. Shelf life is about 30 days under the room temperature condition.
Environmental Specimen Banks (ESBs) are playing pivotal role in monitoring the effect of environmental pollution on the ecosystem based on the retrospective analysis of the representative samples collected regularly and stored in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009 and the standard operating procedures (SOPs) for sampling, and cryogenic milling and storage had been prepared during 2007-2010. Since then, the tentative SOPs for the seven kinds of specimens (shoots of Red Pine (Pinus densiflora) and Korean Pine (Pinus koraiensis), leaves of Mongolian Oak (Quercus mongolica) and Zelkova Tree (Zelkova serrata), eggs of Feral Pigeon (Columba livia var. domestica), muscles and organs of Common Carp (Cyprinus carpio), and Freshwater Bivalve (Unio (Nodularia) douglasiae)) have been put to test in the field and laboratory as well against the practicality and feasibility. The SOPs were improved by reflecting the findings from the research and the following discussion regarding the selection of specimen (Feral Pigeon suffering from a control management), sample size (a problem of decreasing number of sampling trees related to increasing sampling time) and period (a problem related to a bud growth), and sampling methods etc.. In addition, barcoding system for the management of the specimen information, and monitoring system of the cryogenic storage to regulate the optimum temperature and the liquid nitrogen level were also developed for the efficient and effective control of the samples. Lastly, the safety guide and emergency protocol were augmented to guarantee a safe work environment with the cryogenic facility. These improvements of the SOPs are expected to contribute to more stable operation of the NESB.
The chestnut gall wasp, Dryocosmus kuriphilus $Y^{ASUMATSU}$, has been recently distributed and chestnut trees have teen seriously damaged by this insects in Korea. However, this insect pest has not been investigated in Korea. This paper gives an account of field and Laboratory observations on the seasonal occurrence and the life history of the chestnut gall wasp, and the damage of the chestnut trees by. its insects. This study was carried out at Chungchungpukdo, Kangwon-do and Kyungi-do in Korea, from 1961 to 1963, inclusive. 1. The Chestnut gall wasp (Dryocosmus kuriphilus $Y^{ASUMATSU}$) has one generathion per year and females are only known. 2. The adult wasps were observed from late June to late July with peak of wasp emergence about early and middle July. Most of new adult wasps were coming out from, the galls in the morning rather than the afternoon. 3. Each wasp produced auerage 198.5 eggs and about 4.89 eggs was observed in each bud of the chestnut tree. 4, The egg period of wasps was about 30 days and they were hatching from July to August. 5. The larval period of wasps were from middle of August to middle of next June and pupation took places about early June to middle of July. 6. The adult wasps were ovipositing into the buds and from early May next year, galls were growing. They caused stunting of growth and no fruiting or dying of the chestnut tries. 7. The first observation of the wasp was made at Chaechun of Chungchung-pukdo in 1958. Up to 1963 this insects are distributed in Kangwon-do, Kyunggi-do, north part of Chungchung-pukdo and north part of Kyungsangpukdo
The axillary buds of 15-year-old Tilia amurensis were cultured on Saito and Ide (IS), Murashige and Skoog (MS) media and woody plant medium (WPM) to establish an effective micropropagation method. Five levels of 6-benzylaminopurine (BAP) were tested. On IS medium and WPM addition of 1.0/l BAP enhanced shoot development and shoot elongation, whereas addition of 0.5/l BAP was effective on MS medium. A better results were obtained from WPM with 1.0/l BAP and MS with 0.1/l BAP. Developed shoots were subcultured on each basal media but with 0.2/l BAP, Multiple shoots were almost doubled in a month. Root formation could be enhanced at higher concentration of indole-3-butyric acid (IBA). Better rooting rate (83.3%) was achieved on a half-strength MS medium with 3.0 /l IBA. Regenerated plantlets were successfully transferred to soil. To investigate the clonal variation in shoot development and shoot elongation by axillary bud culturing, seven plus tree clones were tested, Clonal variation in tissue culturability among plus trees was recognized by the Duncan's multiple range test at the 5% level. Kang Won No. 12 showed the best response on WPM with 1.0/l BAP.

An accurate prediction of blooming date is crucial for many authorities to schedule and organize successful spring flower festivals in Korea. The Korea Meteorological Administration (KMA) has been using regression models combined with a subjective correction by forecasters to issue blooming date forecasts for major cities. Using mean monthly temperature data for February (observed) and March (predicted), they issue blooming date forecasts in late February to early March each year. The method has been proved accurate enough for the purpose of scheduling spring festivals in the relevant cities, but cannot be used in areas where no official climate and phenology data are available. We suggest a thermal time-based two-step phenological model for predicting the blooming dates of spring flowers, which can be applied to any geographic location regardless of data availability. The model consists of two sequential periods: the rest period described by chilling requirement and the forcing period described by heating requirement. It requires daily maximum and minimum temperature as an input and calculates daily chill units until a pre-determined chilling requirement for rest release. After the projected rest release date, it accumulates daily heat units (growing degree days) until a pre- determined heating requirement for flowering. Model parameters were derived from the observed bud-burst and flowering dates of cherry tree (Prunus serrulata var. spontanea) at KMA Seoul station along with daily temperature data for 1923-1950. The model was applied to the 1955-2004 daily temperature data to estimate the cherry blooming dates and the deviations from the observed dates were compared with those predicted by the KMA method. Our model performed better than the KMA method in predicting the cherry blooming dates during the last 50 years (MAE = 2.31 vs. 1.58, RMSE = 2.96 vs. 2.09), showing a strong feasibility of operational application.
