Linkage Analysis of both RAPD and I-SSR Markers using Haploid Genome from a Single Tree of Pinus densiflora S. et Z. (소나무 단일(單一) 모수(母樹)의 반수체(半數體) 게놈을 이용(利用)한 RAPD 및 I-SSR 표식자(標識子)의 연관분석(連關分析))

  • A linkage map for Japanese red pine (Pinus densiflora) was constructed on the basis of two DNA marker systems of random amplified polymorphic DNAs (RAPDs) and inter-simple sequence repeats (I-SSR). Haploid genomic DNAs were extracted from megagametophyte tissues of 96 individual seeds in a single tree. A total of 98 DNA markers including 52 RAPD markers amplified by 25 primers and 46 I-SSR markers amplified by 18 primers were verified as Mendelian loci showing 1 : 1 segregation in 96 megagametophytes which were ${\chi}^2$-tested at 5% significance level. Of them, 63 segregating loci turned out to be linked into 20 linkage groups by the two-point analysis. However, 35 loci (17 RAPD and 18 I-SSR) of the 98 segregating loci did not coalesced into any linkage groups at a LOD of 3.0. The linked 63 loci were separated by an average distance of about 25.5 cM, which were spanned 1097.8 cM as a whole. The minimum and maximum map distances of the linkage groups were 4.3 cM and 54.9 cM, respectively. Incorporation of I-SSR loi into linkage map of RAPD loci resulted in extended and partially more saturated linkage blocks.

Acetyl Eburicoic Acid from Laetiporus sulphureus var. miniatus Suppresses Inflammation in Murine Macrophage RAW 264.7 Cells

  • The basidiomycete Laetiporus sulphureus var. miniatus belongs to the Aphyllophorales, Polyporaceae, and grows on the needleleaf tree. The fruiting bodies of Laetiporus species are known to produce N-methylated tyramine derivatives, polysaccharides, and various lanostane triterpenoids. As part of our ongoing effort to discover biologically active compounds from wood-rotting fungi, an anti-inflammatory triterpene, LSM-H7, has been isolated from the fruiting body of L. sulphureus var. miniatus and identified as acetyl eburicoic acid. LSM-H7 dose-dependently inhibited the NO production in RAW 264.7 cells without any cytotoxicity at the tested concentrations. Furthermore it suppressed the production of proinflammatory cytokines, mainly inducible nitric oxide synthase, cyclooxygenase-2, interleukin (IL)-$1{\beta}$, IL-6 and tumor necrosis factor ${\alpha}$, when compared with glyceraldehyde 3-phosphate dehydrogenase. These data suggest that LSM-H7 is a crucial component for the anti-inflammatory activity of L. sulphureus var. miniatus.

  • In this study, when we make a diagnosis of constitution using QSCC Ⅱ(Questionnaire of Sasang Constitution Classification). data mining techniques are applied to seek the classification function for improving the accuracy. Data used in the analysis are the questionnaires of 1051 patients who had been treated in Dong Eui Oriental Medical Hospital and Kyung Hee Oriental Medical Hospital. The criteria for data cleansing are the response pattern in the opposite questionnaires and the positive proportion of specific questionnaires in each constitution. And the criteria for variable selection are the test of homogeneity in frequency analysis and the coefficients in the linear discriminant function. Discriminant analysis model and decision tree model are applied to seek the classification function into Sasang constitution. The accuracy in learning sample is similar in two models, the higher accuracy in test sample is obtained in discriminant analysis model.

Analysis on On-line Q&A Cases regarding Landscape Trees Management - Focused on Online Consultation Board at Tree Diagnostic Center - (조경수 관리에 관한 온라인 질의응답 사례 분석 - 수목진단센터 온라인 상담 사례를 대상으로 -)

  • The persons in charge of management request diagnosis and prescription to tree hospitals in order to get consultation about the problems like blight that occur in landscape tree management. This study aims to analyze what the main problems and questions raised by landscape gardeners are and those concerned in landscape tree management. This is done by investigating landscape tree-related questions and answers uploaded on the online consultation boards of the plant diagnostic centers approved in Korea including the Seoul National University Plant Clinic, the Chungbuk National University Plant Hospital, and the Kangwon Diagnostic Center. As a result, those concerned in landscape occupied the most as 81.4% among the questioners. However, only 11.5% did explain the plant management history or surrounding environment, which is essential for landscape tree diagnosis when asking questions. This shows that those concerned in landscape lack basic knowledge or interest about plant diagnosis. Among 263 questions about landscape trees, questions about physiological damage included 94 cases that were the most taking up 35.8%. Moreover, the next were damage by insects and damage by disease in order. It is thought that due to the characteristics of physiological problems that occur by various sorts of stress and with no signs, they tend to request diagnosis or prescription the most. The most frequent reasons for physiological damage are water stress and temperature stress. About damage by disease, there exist many types of diseases, and there are many complex damages accompanied by physiological causes. About damage by insects, the most common include damage by moths. In consideration of this result, universities or technician training centers should provide education for landscape tree management so that landscape technicians and students can acquire essential knowledge and information about landscape tree management and increase their interest in it. In particular, it is necessary to provide profound learning opportunities for plant physiology, and the technicians should make efforts themselves. In addition, it is needed to build organizations to which they can ask technical questions about landscape planting and management in order to understand landscape industry in general and the actual status of landscape planting technique and the actual field. Moreover, to elevate systemicity and expertise in the area of landscape tree management not yet equipped with the foundation, it is needed to cultivate the technicians intensively and conduct research by those concerned both in academic and industrial circles.

  • The effects on birch (Betula spp.) of elevated carbon dioxide ($CO_2$) and ozone ($O_3$), which are both increasing in the troposphere, are surveyed in detail based on the literature. Birches establish themselves in the open field after disturbances, and then become dominant trees in temperate or boreal forests. Ecophysiological approaches include the measurement of photosynthesis, biomass, growth, and survival of seedlings and trees. Elevated $CO_2$ levels give rise to a net enhancement of the growth of birch trees, whereas high $O_3$ generally reduces growth. Although the effects of the two are opposed, there is also an interactive effect. Basic physiological responses of the single genus Betula to $CO_2$ and $O_3$ are set out, and some data are summarized regarding ecological interactions between trees, or between trees and other organisms.

Methanogenic Archaeal Census of Ruminal Microbiomes (반추위 마이크로바이옴 내 메탄생성고세균 조사)

  • The objective of the study was to undertake a phylogenetic diversity census of ruminal archaea based on a meta-analysis of 16S rRNA gene sequences that were publicly available in the Ribosomal Database Project. A total of 8,416 sequences were retrieved from the Ribosomal Database Project (release 11, update 5) and included in the construction of a taxonomy tree. Species-level operational taxonomic units (OTUs) were analyzed at a 97% sequence similarity by using the QIIME program. Of the 8,416 sequences, 8,412 were classified into one of three phyla; however, the remaining four sequences could not be classified into a known phylum. The Euryarchaeota phylum was predominant and accounted for 99.8% of the archaeal sequences examined. Among the Euryarchaeota, 65.4% were assigned to Methanobrevibacter, followed by Methanosphaera (10.4%), Methanomassillicoccus (10.4%), Methanomicrobium (7.9%), Methanobacterium (1.9%), Methanimicrococcus (0.5%), Methanosarcina (0.1%), and Methanoculleus (0.1%). The 7,544 sequences that had been trimmed to the V2 and V3 regions clustered into 493 OTUs. Only 17 of those 493 OTUs were dominant groups and accounted for more than 1% of the 7,544 sequences. These results can help guide future research into the dominant ruminal methanogens that significantly contribute to methane emissions from ruminants, research that may lead to the development of anti-methanogenic compounds that inhibit these methanogens regardless of diet or animal species.

Effects of CEPA on the Secretion of Lacquer and Bark Physiology of Lactree(Rhus verniciflua Stokes) (CEPA 처리(處理)가 옻나무의 칠액분비(漆液分泌) 및 수피생리(樹皮生理)에 미치는 영향(影響))

  • This study was conducted to investigate the effects of CEPA(2-chloroethyl phosphonic acid) on bark physiology and anatomy of lactree(Rhus verniciflua Stokes). Sample trees of similar size and growth rate were selected from 7-year-old lactree plantation located in Hyengsung-kun, Kangwon-do. Lanolin pastes containing 0.1, 1, or 10% CEPA were put into the bark-removed hole made by corer(${\phi}1cm$) on the main stem at 1.2m above the ground on June 16, 1995. Five weeks after application of CEPA, bark thickness was markedly increased as a result of the increase in the amount of phloem and intercellular spaces, and correlated with the increased production of urushiol. By the application of 10% CEPA, bark thickness was increased approximately 2.5 times, and the urushiol content within bark was increased 2.8 times compared to that of untreated trees because CEPA stimulated the accumulation of urushiol within bark. Treatment of 10% CEPA also increased the size and the total number of secretory canals, and induced an increase in ray width. The phloem parenchyma cells of CEPA-treated trees were well-developed and closely packed with little intercellular space.

Effect of Occurrence of Scion Root on the Growth and Root Nutrient Contents of 'Shiranuhi' Mandarin Hybrid grown in Plastic Film House (자근발생이 부지화 감귤나무의 수체 생육과 뿌리내 양분함량에 미치는 영향)

  • BACKGROUND: 'Shiranuhi' mandarin is a major cultivar among all late ripening type of citrus, and is widely cultivated in Korea. However, many farmers have reported scion root problems in their orchard resulting in reduced flowering and fruiting. It is necessary that the physiology of scion-rooted 'Shiranuhi' mandarin trees is further understood. METHODS AND RESULTS: This experiment was conducted to understand the growth response and physiology of scion-rooted 'Shiranuhi' mandarin hybrids. In our study, 'Shiranuhi' mandarin trees were divided into two groups: trees without scion roots (control) and trees with scion roots. The experiment was conducted in Seogwipo of Jeju, with ten replicates for each group. Growth of trees with scion roots was more vigorous and the trees were taller than the controls. Tree height and trunk diameter of scion-rooted trees were significantly higher than those of control trees. Exposed length of rootstocks of scion-rooted trees was significantly lower (by about 2 cm) than that of control trees (8.6 cm). In terms of root nutrition, carbon contents of scion-rooted trees was significantly lower than that of control trees, but nitrogen and potassium concentrations in scion roots were significantly higher than those in control roots. CONCLUSION: Based on the results, we infer that growth of scion-rooted trees was very vigorous and the content of nitrogen in these roots was higher than that in the control tree roots. Thus, the carbon/nitrogen ratio of scion roots was significantly lower than that of the control roots.

Relationship between Maximum Stem Volume and Density during a Course of Self-thinning in a Cryptomeria japonica Plantation

  • Cryptomeria japonica plantation was monitored every year during 15 years from 1983 to 1997 for stem diameter and volume. The reciprocal equation, 1/Y = A + B/N, was applied to the relationship between cumulative volume Y and cumulative number N from the largest tree in the stand each year. The parameters A and B, which means respectively the reciprocal of an asymptotic value of total stand stem volume and the reciprocal of the maximum stem volume, are related by a power function. The power functional relationship between A and B derived a linear relationship of B-points ( $N_{B}$, $V_{B}$; $N_{B}$ = B/A, $Y_{B}$ = 1/2A) of each Y-N curve on log-log coordinates. The gradient of B-point line was so steep that the Y-N curve moved parallel upward year by year. The time trajectory of mean stem volume (W) and density ($\rho$) provided evidence in favor of the 3/2 power law of self-thinning, because the gradient of W - $\rho$ trajectory on log-log coordinates approximated to -3/2 at the final stage of stand development. On the basis of the results of Y-N curves and W - $\rho$ trajectory, the time trajectory of maximum stem volume $W_{max obs}$ and $\rho$ was derived theoretically. The gradient of $W_{max obs}$ - $\rho$ trajectory on log-log coordinates is calculated to be -0.6105 at the final stage. The gradient of $W_{max obs}$ - $\rho$ trajectory was steeper than that of W - $\rho$ trajectory at the early stage, while the former is gentler than the latter at the later stage.stage.e.age.e.

Growth Inhibition of Human Lung Carcinoma Cells by ${\beta}>-lapachone$ through Induction of Apoptosis (Tabebuia avellanedae에서 유래된 ${\beta}>-lapachone$의 인체폐암세포 apoptosis 유발에 관한 연구)

  • The DNA topoismerase I inhibitor ${\beta}-lapachone$, the product of a lapacho tree (Tabebuia avellanedae) from South America, activates a novel apoptotic response in a number of cell lines. In the present report, we investigated the effects of ${\beta}-lapachone$ on the growth of human lung in human non-small-cell-lung-cancer A549 cells. Upon treatment with ${\beta}-lapachone$, a concentration-dependent inhibition of cell viability and cell proliferation was observed as measured by hemocytometer counts and MTT assay. The ${\beta}-lapachone-treated$ cells developed many of the hallmark features of apoptosis, including membrane shrinking, condensation of chromatin and DNA fragmentation. These apoptotic effects of ${\beta}-lapachone$ in A549 cells were associated with a marked induction of pro-apoptotic Bax expression, however the levels of anti-apoptotic Bcl-2 expression were decreased in a dose-dependent manner. Accordingly, elevated amount of cyclin-dependent kinase inhibitor p21 expression accompanied by up-regulation of tumor suppressor p53 was observed. By RT-PCR analyses, decrease in gene expression level of telomerase reverse transcriptase and telomeric repeat binding factor were also observed. Thus, these findings suggest that ${\beta}-lapachone$ may be a potential anti-cancer therapeutics for the control of human lung cancer cell model.
