Evaluation of Safety for the Supplement of Herbicide-resistant Rice in Old Male Rats (노령기 흰쥐에서 제초제 저항성 쌀의 급여에 대한 안전성 평가)

  • This research was conducted to evaluate the safety of the herbicide-resistant rice, a genetically modified organism (GMO) developed by the Rural Development Administration by exposing it to 12 months old Sprague-Dawley rats for 8 weeks. The composition of herbicide-resistant brown rice with/without heating treatment was compared with those of conventional Ilpum brown rice with/without heating treatment to assess composition equivalence. Compositional analysis was performed to measure proximates, fiber, and minerals. The nutritional components of herbicide-resistant rice were similar to those of the nontransgenic control or were within the normal range of nontransgenic rice. Four groups of experimental male rats were fed one of the following diets for eight weeks: Ilpum brown rice (I) and its heated rice (IH) as non-GMO, and herbicide-resistant brown rice (G) and its heated rice (GH) as GMO- We checked clinical symptoms (anorexia, salivation, diarrhea, polyuria, anuria, fecal change), food intake, and water consumption every day, change of body weight once a week, and serum biochemistry and organ weights after 8 weeks of experimental feeding We did not find any significant differences in the above-mentioned items. These results suggested that genetically modified herbicide-resistant rice was compositionally equivalent to conventional Ilpum rice, and nutritional characteristics and safety of herbicide-resistant rice in old male rats treated for 8 weeks were not different from those of Ilpum rice, non-GMO.

The effect of tulobuterol patches on the respiratory system after endotracheal intubation

  • Background: Endotracheal intubation during anesthesia induction may increase airway resistance ($R_{aw}$) and decrease dynamic lung compliance ($C_{dyn}$). We hypothesized that prophylactic treatment with a transdermal ${\beta}2$-agonist tulobuterol patch (TP) would help to reduce the risk of bronchospasm after placement of the endotracheal tube. Methods: Eighty-two American Society of Anesthesiologists (ASA) category I or II adult patients showing obstructive patterns were divided randomly into a control and a TP group (n = 41 each). The night before surgery, a 2-mg TP was applied to patients in the TP group. Standard monitors were recorded, and target controlled infusion (TCI) with propofol and remifentanil was used for anesthesia induction and maintenance. Simultaneously, end-tidal carbon dioxide, $R_{aw}$, and $C_{dyn}$ were determined at 5, 10, and 15 min intervals after endotracheal intubation. Results: There was no significant difference in demographic data between the two groups. The TP group was associated with a lower $R_{aw}$ and a higher $C_{dyn}$, as compared to the control group. $R_{aw}$ was significantly lower at 10 min (P < 0.05) and 15 min (P < 0.01), and $C_{dyn}$ was significantly higher at 5 min (P < 0.05) and 15 min (P < 0.01) in the TP group. A trend towards a lower $R_{aw}$ was observed showing a statistically significant difference 5 min after endotracheal intubation (P < 0.01) in each group. Conclusions: Prophylactic treatment with TP showed a bronchodilatory effect through suppressing an increase in $R_{aw}$ and a decrease in $C_{dyn}$ after anesthesia induction without severe adverse effects.

Epidemiological Cut-off Values Generated for Disc Diffusion Data from Streptococcus parauberis (Streptococcus parauberis의 디스크 확산법 결과에 대한 Epidemiological Cut-off Value의 설정)

  • Streptococcosis caused by Streptococcus parauberis is a very important disease in farmed olive flounder Paralichthys olivaceus. For most fish pathogens, including S. parauberis, there are no analytical criteria to distinguish antibioticsusceptible strains from antibiotic-resistant strains. In this study, epidemiological cut-off ($CO_{WT}$) values were generated to classify 75 strains of S. parauberis isolated from 1999 to 2018 as wild type (WT) and non-wild type (NWT) using disc diffusion data and normalized resistance interpretation (NRI) analysis. The susceptibility of the isolates to 16 antibiotics was evaluated using CLSI guideline M42-A. The wild-type cut-off values for amoxicillin, erythromycin, oxytetracycline, and florfenicol for S. parauberis were ${\geq}35$, 31, 28, and 27 mm, respectively. The NWT ratios of S. parauberis strains to treatment with GEN, FFC, ENR, SXT, EFT, VAN, and CHL were 17% or less, indicating that these antibiotics may be used to treat streptococcosis caused by S. parauberis. For recent S. parauberis isolates, the NWT ratios for AMX, ERY, OTC and FFC are much higher than for strains isolated from 1999-2007. The $CO_{WT}$ data from this study will assist aquatic animal disease professionals in prescribing appropriate antibiotics for the treatment of streptococcosis caused by S. parauberis, which will help reduce the misuse and abuse of antibiotics in the aquaculture sector.

  • Obstructive sleep apnea (OSA) is a sleep disorder with no breathing symptoms due to repetitive upper airway resistance. OSA is a disease that can have significant effects on the cerebral cardiovascular system. Active treatment is needed to prevent these complications. The use of continuous positive airway pressure (CPAP), the standard therapy of OSA, has comparative therapeutic effects. On the other hand, there is no comparison report of the polysomnography (PSG) results before and after CPAP therapy without using a mask. This paper reports a patient who was diagnosed as OSA and used CPAP every night for more than 2 years. The patient showed a decrease in the apnea-hypopnea index from 64.7/h to 12.9/h. In addition, other sleep-related indicators improved significantly. The daily use of CPAP as a treatment for OSA for more than 2 years may improve the PSG results. Constant follow up of PSG will be needed to adjust the appropriate CPAP pressure to patients because there might be a change in the Apnea-Hypopnea Index and other sleep-related indicators for constant CPAP users for at least 2 years.

  • The study looked at the effect of body food on stress reduction and NK cell activity in improving natural healing for nurses, and examined whether it is effective in preventing and curing human health. The study was conducted for 30 days on 22 nurses who currently worked for more than a year at a general hospital in Gyeonggi Province. Stress measurements were conducted through pulse wave measuring instrument, and blood tests were conducted on the activity of NK cells. Treatment of collected data was performed by Paired t-testing through the Stat program SPSS 21 and providing body food suitable for the physical quality of participants after the classification of the body based on the internal diameter of the emperor and scholarship. Studies have shown that the effect on reducing the stress of body food has significant positive effects and that body food is effective for the activity of NK cells. This study has the following significance: First, the main health threat factor for modern people is the increased resistance of natural healing through body food to stress, which has the potential to prevent disease. Second, immunodeficiency in disease prevention and treatment is very important, which can increase the natural healing power of the human body by increasing the activity of NK cells through body food.

Can Hinokitiol Kill Cancer Cells? Alternative Therapeutic Anticancer Agent via Autophagy and Apoptosis (Hinokitiol에 의해 유도된 Autophagy 및 Apoptosis에 의한 대체 항암요법 연구)

  • Cancer is genetically, metabolically and infectiously induced life threatening disorder showing aggressive growing pattern with invasive tendency. In order to prevent this global menace from jeopardizing human life, enormous studies on carcinogenesis and treatment for chemotherapy resistance have been intensively researched. Hinokitiol (${\beta}$-thujaplicin) extracted from heart wood of cupressaceous is a well-known bioactive compound demonstrating anti-inflammation, anti-bacteria and anti-cancer effects on several cancer types via apoptosis and autophagy. This study proposed that hinokitiol activates transcription factor EB (TFEB) nuclear translocation for autophagy and lysosomal biogenesis regardless of nutrient condition in cancer cells. Mitophagy and ${\beta}$-catenin translocation into the nucleus under treatment of hinokitiol on non-small cell lung cancer (NSCLC) cells and HeLa cells were investigated. Hinokitiol exerted cytotoxicity on HeLa and HCC827 cells; moreover, artificially induced autophagy by overexpression of TFEB granted imperfect sustainability onto HeLa cells. Taken together, hinokitiol is the prominent autophagy inducer and activator of TFEB nuclear translocation. Alternative cancer therapy via autophagy is pros and cons since the autophagy in cancer cells is related to prevention and survival mechanism depending on nutrition. To avoid paradox of autophagy in cancer therapy, fine-tuned regulation and application of hinokitiol in due course for successful suppressing cancer cells are recommended.

Immunostimulatory Activity of Agarwood through Activation of MAPK Signaling Pathway in RAW 264.7 Murine Macrophages (RAW 264.7 대식세포에서 MAPKs 신호 전달 경로의 활성화를 통한 침향의 면역 자극 활성)

  • Deterioration of the immune function weakens the body's resistance to various infections, leading to a series of diseases. Immunomodulatory biomaterials have been used to reduce the side effects of immunosuppressants or to enhance immunity. Agarwood is the aromatic resinous portion of Aquilaria trees that has been traditionally used as a medicinal herb for the treatment of various diseases. Although previous studies have shown that agarwood can improve the body's immunity, evidence for this claim is still lacking. In this study, the immune-enhancing effects of the agarwood methanol extracts of Aquilaria malaccensis Lamk were evaluated in a RAW 264.7 macrophage model. Based on the results, the agarwood extracts markedly enhanced phagocytosis in the absence of cytotoxicity. The agarwood extract-treated RAW 264.7 cells exhibited the typical morphology of activated macrophages, which are spindle-shaped with elongated filopodia. Agarwood extract also significantly increased the production of nitric oxide (NO), which is associated with the increased expression of inducible NO synthase. Moreover, the secretion and expression levels of cytokines, such as tumor necrosis factor-α and interleukin (IL)-1β and IL-6, were increased by agarwood treatment. Notably, these are also associated with a mitogen-activated protein kinase signaling pathway. Taken together, our findings provide scientific evidence that agarwood has potential immune-enhancing effects in vitro.

Development of Steam Cleaning Technique to Improve Removal Efficiency of Membrane Fouling Matter in Water Treatment Process Using Ceramic Membrane (정수처리용 세라믹 분리막의 막오염 물질의 제거 효율 향상을 위한 스팀세정 기법 개발)

  • This research has developed a high temperature steam cleaning technology using a ceramic membrane with durability against temperature and pressure conditions. In steam cleaning, steam of $120^{\circ}C$ is injected into the ceramic membrane to induce pyrolysis by the endothermic reaction to remove fouling from the membrane. The water quality of raw water was adjusted to turbidity 10, 25 NTU and DOC 2.5 mg/L, and the membrane was uniformly fouled by constant pressure operation at 100, 200, and 300 kPa. Physical backwashing was performed with water and air at a pressure of 500 kPa and steam at $120^{\circ}C$ was injected for 0 to 5 minutes. As the turbidity concentration and the operating pressure increased, the flux decreased by 0.7 to 14.4%. It is confirmed that 10.7 to 53.8% recovery is possible than physical cleaning at the injection of steam for 3 minutes, so it is considered that the steam cleaning of the ceramic membrane is effective. Compared with CEB after NaOCl (300 mg/L) filtration at 25 NTU and 300 kPa of turbidity, the steam cleaning result for 3 minutes was similar to 46.7% of CEB for 3 hours. It has been confirmed that steam cleaning is suitable for a ceramic membrane having excellent heat resistance against high temperature. It was considered to have better cleaning efficiency as compared with general physical backwashing.

Zearalenone regulates key factors of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1-nuclear factor erythroid 2-related factor 2 signaling pathway in duodenum of post-weaning gilts

  • Objective: This study explored the mechanism of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway under conditions of zearalenone (ZEA)-induced oxidative stress in the duodenum of post-weaning gilts. Methods: Forty post-weaning gilts were randomly allocated to four groups and fed diets supplemented with 0, 0.5, 1.0, or 1.5 mg/kg ZEA. Results: The results showed significant reductions in the activity of the antioxidant enzymes total superoxide dismutase and glutathione peroxidase and increases the malondialdehyde content with increasing concentrations of dietary ZEA. Immunohistochemical analysis supported these findings by showing a significantly increased expression of Nrf2 and glutathione peroxidase 1 (GPX1) with increasing concentrations of ZEA. The relative mRNA and protein expression of Nrf2, GPX1 increased linearly (p<0.05) and quadratically (p<0.05), which was consistent with the immunohistochemical results. The relative mRNA expression of Keap1 decreased linearly (p<0.05) and quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet. The relative mRNA expression of modifier subunit of glutamate-cysteine ligase (GCLM) increased quadratically (p<0.05) in all ZEA treatment groups and the relative mRNA expression of quinone oxidoreductase 1 (NQO1) catalytic subunit of glutamate-cysteine ligase decreased linearly (p<0.05) and quadratically (p<0.05) in the ZEA1.0 group and ZEA1.5 group. The relative protein expression of Keap1 and GCLM decreased quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet, respectively. The relative protein expression of NQO1 increased linearly (p<0.05) and quadratically (p<0.05) in all ZEA treatment groups in the duodenum. Conclusion: These findings suggest that ZEA regulates the expression of key factors of the Keap1-Nrf2 signaling pathway in the duodenum, which enables resistance to ZEA-induced oxidative stress. Further studies are needed to examine the effects of ZEA induced oxidative stress on other tissues and organs in post-weaning gilts.

Cold Tolerance and Physiological Response of Camellia sinensis Cultivars by Low-Temperature Treatment (저온처리에 의한 국내 품종 차나무 저온 내성 및 생리적 반응)

  • We selected a cold-tolerant tea tree (Camellia sinensis L.) through reliable evaluation using a number of cold-tolerance indicators targeting tea tree cultivars such as "Chamnok," "Bohyang," "Sangnok," and "Myungnok" in response to climate change. We conducted a low-temperature damage investigation, "Chamnok" and "Bohyang" were damaged investigated with small amounts of green and bright green. "Sangnok" and "Myungnok" were damaged investigated red and dark red at -10℃. The extent of electrolyte leakage increased as the treatment temperature decreased in all cultivars. We predicted lethal temperatures through non-linear regression analysis, finding relatively higher tolerance to low temperature in "Chamnok" (-9.344℃) and "Bohyang" (- 8.883℃) than that in "Myungnok" (-8.092 ℃) and"Sangnok" (-7.632℃). "Bohyang" showed higher levels of antioxidant activity compared to other cultivars. The lipid peroxidation reaction revealed that "Sangnok" and "Myungnok" had higher MDA content than that of other cultivars when treated at low temperatures. Consequently, predictions of the lethal temperature through non-linear regression analysis of "Chamnok" and "Bohyang" were consistent with their tolerance to low-temperature damage, and antioxidant activity and lipid peroxidation reactions were likewise consistent. The results of this study can be used not only for evaluation and selection of cold-tolerance of tea trees in response to climate change, but also in the cultivation of cold-tolerant plants.
