Objectives: This study aims to examined whether physical activity prevent the negative effect of psychological stress on cardiovascular reactivity by reducing stress induced sympathetic output and preventing norepinephrine depletion negative psych-affective responses. It is assumed that physical activity reduces the magnitude of cardiovascular responses and psychological responses to stress which threaten individuals' physical and mental health. The result of investigating the effect of physical activity on reducing negative physiological and psychological responses would suggest useful information health for practitioners who want to prevent stress-induced diseases, especially coronary heart disease. Methods: participants of this study were 30 students (10 males & 20 females), whose mean age was 21.30 (SD=2.29). Fifteen students (5 males & 10 females) were assigned for in each group, treatment and control groups. They were interviewed and given a survey that included a consent form, demographics sheet and psychological tests, such as State-Trait Anger Expression Inventory and State-Trait Anxiety Inventory. Before the application of psychological stress, participants in treatment group were going through with a course of physical exercise, running on treadmill 15minutes, while participants in control group were not physically active. After exercise, there was 15 minutes resting period before applying cognitive stress. During the experiment, all participants performed challenging cognitive tasks for 20minutes in situations that were designed to experience learned helplessness and measured their cardiovascular reactivity including blood pressure and heart rate every 5 minutes, until 10 minutes after finishing the application of psychological stress(recovering state). In the end of experiment, they were given some psychological test again. Results: Heart rates of exercise group were significantly higher than non-exercise group, especially, five minute after applying cognitive stress and at the end of recovery, in other word 10 minutes after stressful event. Systolic blood pressures of exercise group were lower than those of non-exercise group during the stressful event, but this differences in borderline level of significance. state anger level of exercise group decreased even experiencing stress, while those of non-exercise group increase. And state anxiety level of exercise group decreased in borderline level of significance. Conclusion: This study reiterate health benefits of physical activity and suggest that regular moderate exercise may regulate cardiovascular reactivity and psycho-affective responses from stress by reducing stress induced sympathetic output.
The aim of this study was to investigate the effects of exercise on intrinsic and extrinsic apoptosis signaling pathways in skeletal muscle. ICR-type white male mice were divided into a control group (CON: n=10) and an exercise training group (EX: n=10) after a 1 week adaptation period. EX performed treadmill running at 16.4 m/min with a 4% incline, 40 min/day and 5 days/week for 8 weeks. Cervical dislocation was performed at 48 hours after the last bout of exercise, after which gastrocnemius skeletal muscles were immediately collected. The results of verifying the intrinsic apoptosis pathway showed that there were no significant differences in Bcl-2, Bax, or the ratio of Bax/Bcl-2 proteins between EX and CON. On the other hand, the results of verifying the extrinsic apoptosis pathway showed that caspase-8 proteins were significantly lower in EX than in CON (p<0.05). Apoptosis suppressing protein HSP70 was higher in EX than in CON. In addition, caspase-3, which is the final factor for apoptosis, was not activated. These results indicate that apoptosis did not develop since caspase-3 is non-cleaved by the effects of caspase-8 and HSP70 extrinsic pathways rather than Bcl-2 and Bax intrinsic pathways among signal pathways for apoptosis.
This study investigated the response of GLUT -4 and GRP-78 expression in soleus muscle of streptozotocin-induced diabetic rats by imposing different exercise intensities. F344 rats were randomly divided into 4 groups (n=15 in each group): Control (Control), diabetes-operation (DO), diabetes with low intensity exercise (DLE) and diabetes with high intensity exercise (DHE). The rats in DLE and DHE groups were exercised for 8 weeks by treadmill running. Blood glucose levels in DO were significantly higher compared to that in NORMAL whereas DLE showed the most lowest level in blood glucose among diabetic groups. Diabetic groups exhibited significantly lower level in insulin change and DLE showed significantly higher insulin level among diabetic groups. GRP-78 in DO was significantly $(167.05\%)$ higher than that in Control. GRP-78 in DLE was $139.41\%$ which is significantly higher compared to Control but when compared to DO and DHE, it was significant low. GRP-78 in DHE was $194.64\%$ which doubled the protein level in Control and showed the most highest level in all groups. GLUT-4 in DO was significantly $(33.58\%)$ higher compared to Control. GLUT-4 in DLE showed $124.58\%$ which was significant high compared to Control, DO and DHE. GLUT-4 in DHE showed $26.91\%$ compared to Control and was the most lowest level among all groups. It seems clear that chiefly low intensity exercise benefits diabetic patients in controlling blood glucose. It was concluded that low intensity exercise induces translocation of GLUT-4 which results in increased blood inflow, thus GRP-78 expression is decreased.
This study investigated the response of GLUT-4 and GRP-78 protein expression in soleus muscle of Streptozotocin-induced diabetic rats with caffeine oral administration by imposing different exercise intensities. Rats were randomly divided into 5 groups (n=6 in each group): diabetic group (D), diabetic-caffeine group (DC), diabetic-caffeine group with low intensity exercise (DCL), diabetic-caffeine group with moderate intensity exercise (DCM) and diabetic-caffeine group with high intensity exercise (DCH). The rats in DCL, DCM and DCH groups were exercised acutely by treadmill running for 8 meter/m, 16 meter/m and 25 meter/m, respectively. Little difference in GLUT-4 protein expression was shown in DC and DCL compared to D. GLUT-4 protein expression was decreased in DCM and increased in DCH was observed. GRP-78 protein expressions in DCL, DCM and DCH were little lower than that of D. An increase in GRP-78 protein was observed in DC. Improved insulin sensitivity with acute high intensity exercise gives the rats important therapy that lowers insulin requirement. This improvement of insulin sensitivity for glucose transport in skeletal muscle results from translocation of the GLUT-4 protein from the endoplasmic reticilum to the cell surface and increase in total quantity of GLUT-4 protein. It is not clear what mechanism reduced GRP-78 protein level in exercise group. It is merely conjectured that caffeine-induced lipolysis provided cells with energy in abundance and this relieved stress which cells are subjected to receive when performing exercise.
The present research was conducted to investigate the effect of moderate exercise training on splenocyte inflammatory cytokines production in high fat diet-induced obese mice. To determine the effects of exercise training and low fat diet on splenocyte, C57BL/6 male mice (4 weeks aged, n=20) were fed a high fat diet (45% fat diet) for 5 weeks so that obesity was achieved intentionally. These obese mice were then divided into 2 groups; HLC (low fat diet and control n=10), and HLE (low fat diet and exercise n=10). HLE mice performed 8 weeks of exercise training on a motorized treadmill by running for 30-60 min/day at 10-22 m/min, 0% grade, five times per week. After exercise training, all the splenocyte was collected and Con A (concanvalin A, 10${\mu}g/ml$) was used to stimulate the cells, after which IL-1${\beta}$, IL-6, TNF-${\alpha}$ were measured by bio plex. Independent t-test was used and p value under 0.05 was considered a statistical significance. Splenocyte IL-1${\beta}$, IL-6, TNF-${\alpha}$ production of HLE stimulated by Con A was significantly lower than that of HLC (p<0.01). These findings suggest that moderate exercise has beneficial effects on splenocyte inflammatory cytokines in high fat diet induced obese mice.
Background and Objective: Cardiotoxicity and oxidative stress is a life-threatening side effect of doxorubicin (DOX). We investigate the effects of short-term exercise as therapeutic tool for improvement of cardioprotection against DOX-induced cardiotoxicity in the rat. Methods: Wistar males (weighing $257{\pm}28g$) were divided into six groups: (1) control+placebo (2) control+DOX $10mg.kg^{-1}$ (3) control+DOX $20mg.kg^{-1}$ (4) training+placebo (5) training+ DOX$10mg.kg^{-1}$ (6) training+DOX $20mg.kg^{-1}$. Cardiotoxicity was induced by DOX (10 and $20mg.kg^{-1}$). The rats in groups 4, 5 and 6 experienced treadmill running of 25 to $39min.day^{-1}$ and 15 to $17m.min^{-1}$, 5 days/wk for 3 wk. At the end of the endurance training program, rats in the 1 and 4 groups, in the 2 and 5 groups and in the 3 and 6 groups received saline solution, DOX $10mg.kg^{-1}$ and DOX $20mg.kg^{-1}$, respectively. Result: DOX administration (10 and $20mg.kg^{-1}$) caused significant increase in MDA and Apelin, an insignificant increase in NO and a significant decrease in SOD, as compared to the C+P group. Three weeks of the pretreatment endurance exercise resulted in a significant increase of Apelin and SOD, an insignificant increase of NO and an insignificant decrease of MDA, as compared to the C+P group. Furthermore, after three weeks of endurance training and DOX treatment with $10mg.kg^{-1}$ and $20mg.kg^{-1}$, a significant increase in apelin and SOD, and a significant decrease in MDA were detected in comparison to C+DOX10 and/or C+DOX20 groups. There was a significant difference between DOX$10mg.kg^{-1}$ and DOX$20mg.kg^{-1}$ treatments in MDA levels only. Conclusion: Pretreatment exercise may improve myocardial tolerance to DOX-induced cardiotoxicity by inhibition of oxidative stress and up-regulation of antioxidants in heart tissue.

The purpose of this study was to analyze the effect of grasshopper (Oxya chinensis sinuosa) powder ingestion with/without aerobic exercise (treadmill running) on energy metabolism. To achieve this purpose, 28 Institute of Cancer Research (ICR) mice were divided into four groups: normal diet control group (CON), a normal diet with exercise control group (COEX), a grasshopper powder-supplemented diet group (GH), and a grasshopper powder-supplemented diet with exercise group (GHEX). Duration of the powder ingestion and aerobic exercise training were 6 weeks. Body weight gain ratio was not significant. Fat mass significantly decreased in GH and GHEX. There were no changes in blood glutamic oxaloacetic transaminase and glutamic pyruvic transaminase levels between groups. Glucose transporter type 2 and glucose transporter type 4 protein levels were not significantly different between groups. Fibronectin type III domain-containing protein 5 level was the highest in GHEX. AMP-activated protein kinase level significantly increased in GHEX compared to the levels in the other groups. Glycogen synthase kinase 3 beta protein level was reduced in GHEX compared to that in CON. These results suggest that grasshopper powder ingestion and endurance exercise training influence energy metabolism.

In this study, the effects of a rice diet and bread diet on plasma triglyceride, insulin and ghrelin levels during low-intensity endurance exercise and recovery were investigated. Ten male students randomly received 2 different treatments: the rice diet and bread diet. On the first day, the participants performed 2 hours of treadmill running with 6% uphill at 50% $VO_{2max}$ after breakfast and then each consumed lunch and dinner. Blood samples were drawn 120 min before exercise and, right before exercise and, 60 min and 120 min after the start of exercise. On the second day, blood samples were drawn prior to breakfast, immediately after breakfast and, 60 min, 120 min, and 180 min after breakfast with no exercise treatment. Plasma triglyceride, ghrelin, glucose, and insulin levels were not significantly different between the two treatment groups on the first day. In addition, there was no difference in the carbohydrate and fat oxidation rate between the two treatments groups. However, plasma triglyceride levels in subjects that received the rice diet were significantly reduced by 14% when compared to subjects that received the bread diet at 180 min after consuming the breakfast diet on the second day. Ghrelin levels were significantly higher for subjects that received the rice diet than subjects that were given the bread diet. However, insulin in participants that consumed the rice diet was significantly lower than those that received the bread diet at 60 min and 120 min after consumption of the breakfast diet on the second day. Glucose levels in the subjects that were given the rice diet were significantly reduced by 10.3% when compared with participants that received the bread diet at 60 min. Therefore, the results of the study showed that a rice diet may be more effective in preventing cardiovascular diseases than a bread diet when combined with exercise.
The purpose of the present study was to determine the preventive effects of combined interventional trial of fish oil treatment and exercise training on insulin resistance of skeletal muscle in high-fat fed rats. Male Wistar rats were randomly divided into chow diet (CD), high-fat diet (HF), high-fat diet with fish oil (FO), high-fat diet with exercise training (EX), and FO+EX groups. The rats in control group were fed chow diet containing, as percents of calories, 58.9% carbohydrate, 12.4% fat, and 28.7% protein. High-fat diet provided 32% energy as lard, 18% as corn oil, 27% as carbohydrate and 23% as casein. The fish oil diet had the same composition as the high fat diet except that 100 g menhaden oil was substituted for corn oil. Insulin sensitivity was assessed by in vitro glucose transport in the soleus muscle after diet treatment and treadmill running for 4 weeks. While the FO or EX only partially prevented insulin resistance on glucose transport and visceral obesity induced by high-fat diet, these interventions completely corrected hyperinsulinemia and hyperglycemia from the high-fat diet. The rats in the FO+EX showed normalized insulin action on glucose transport, plasma chemicals and visceral fat mass. Insulin-mediated glucose transport was negatively associated with total visceral fat mass (r=-0.734; p<0.000), plasma triglyceride (r=-0.403; p<0.05) and lepin (r=-0.583; p<0.001) concentrations with significance. Multiple stepwise regression analysis showed that only total visceral fat mass was independently associated with insulin-mediated glucose transport (r=-0.668; p<0.000). In conclusion, combined interventional trial of FO+EX recovered insulin resistance on glucose transport of skeletal muscle induced by high-fat diet. Visceral fat mass might be more important factor than plasma TG and leptin to induce insulin resistance on glucose transport of skeletal muscle in high-fat fed rats.
Autophagy, a highly conserved mechanism of internal quality control, is essential for the maintenance of cellular homeostasis and for the orchestration of an efficient cellular response to stress. During aging, the efficiency of autophagic degradation declines and intracellular waste products accumulate. Therefore, the aim of this study is to investigate the effects of exercise on autophagic response in skeletal muscle. Twenty-four Young (4 month) and Old (12 month) ICR-type white male mice were divided into a control group (CON: n=6) and exercise training group (Tr: n=6) after an adaptation period of 1 week. Exercise consisted of treadmill running at 16.4 m/min with a 4% incline, 40 min/day and 5 days/week for 8 weeks. Cervical dislocation was performed at 48 hours after the last round of exercise, after which the gastrocnemius skeletal muscle were immediately collected. The results of verifying autophagy formation showed that the Sarcopenia index was decreased in the Old mice compared to the Young. However, it increased with exercise training in the Old. Lipidation LC3-II, Becline-1, and Atg7 were decreased in the Old mice compared to the Young. However, Lipidation LC3-II was significantly increased in the trained Old mice (Young:1 Vs Old:$1.32{\pm}0.042$, p<0.05). Based on these data, we suggest that autophagy regulatory events are the attenuated in Old mice, but that they are enhanced with exercise training.
