Effects of the NADPH Oxidase p22phox C242T Polymorphism on Endurance Exercise Performance and Oxidative DNA Damage in Response to Aerobic Exercise Training

  • We examined the effects of the NADPH oxidase p22phox C242T polymorphism on endurance exercise performance and oxidative DNA damage in response to acute and chronic exercises. One hundred three subjects were recruited, among which 26 healthy subjects (CC: 12, TC: 12, and TT: 2) were studied during rest, exercise at 85% $VO_2max$, and recovery before and after 8 weeks of treadmill running. Lymphocyte DNA damage increased significantly in response to exercise (p < 0.05). There were no significant differences in plasma MDA, SOD concentrations and lymphocyte DNA damage between CC genotype and T allele group, but significant endurance training differences were observed. Endurance training increased exercise time to exhaustion in both the CC genotype and T allele groups (p < 0.05) but no significant difference was found between groups. The results of the current study with young, healthy, Korean men are interpreted to mean that 1) the majority had the CC genotype of the NADPH oxidase p22phox C242T polymorphism (82.5%: CC, 15.5%: TC, 1.9%: TT), 2) acute exercise increased lymphocyte DNA damage, 3) endurance training significantly increased exercise time to exhaustion, and alleviated lymphocyte DNA damage, and 4) The NADPH oxidase p22phox C242T polymorphism, however, did not alter lymphocyte DNA damage or exercise performance at rest, immediately after exercise, or during recovery.

Effects of 4 Week Exercise on Activation of Extracellular Signal-regulated Kinases and c-Jun N-terminal Kinase Pathways in Rat Tibialis Muscle (4주간 달리기 운동이 흰쥐의 전경골근에서 ERK 및 JNK의 활성화에 미치는 영향)

  • The effect of either low or high intensity four weeks exercise treadmill running on the activation of the extracellular-signal regulated protein kinase (ERK1/2) and the c-Jun N-terminal kinase(JNK) pathways was determined in rat tibialis muscle. Sprague-Dawley rats were assigned to one of three groups: (i) sedentary group(NE; n=10); (ii) low intensity exercise group (8m/min; LIE; n=10); and (iii) high intensity exercise group(28m/min; HIE; n=10). The training regimens were planned so that animals covered the same distance and had similar glycogenutilization for both LIE and HIE exercise sessions. After four weeks exercise, 48 h after the last exercise bout obtained samples. pERK1 increased 1.5 times comparing with the sedentary group in the low intensity group while it increased 11.7 times in high intensity group, in the tibialis of rats. In the low intensity group, pERK2 increased 1.4 times comparing with the sedentary group while it increased 3.3 times in high intensity group. While pJNK1 decreased 0.9 times, comparing with the sedentary group, pJNK2 was increased to 0.5 times in the low intensity group. But in high intensity group, pJNK2 decreased 0.7 times while pJNK1 didn't show any change. In conclusion, Four weeks exercise of different intensities results in tibialis muscle activation of intracellular signal pathways, which may be one mechanism regulating specific adaptations induced by different exercise intensities.

Pretreatment Effects of Regular Aerobic Training on the IGF System and Hepatotoxicity Induced by Doxorubicin in Rats

  • Aims: To examine the pretreatment effects of regular aerobic training on the IGF system (IGF-I, IGFBP-3 and IGF/IGFBP) and doxorubicin(DOX) induced hepatotoxicity in rats. Materials and Methods: Forty-eight male rats were divided into groups:(1) control+placebo (2) $control+DOX_{10}mg{\cdot}kg^{-1}$ (3) $control+DOX_{20}mg{\cdot}kg^{-1}$ (4) training+placebo (5) $training+DOX_{10}mg{\cdot}kg^{-1}$ (6) $training+DOX_{20}mg{\cdot}kg^{-1}$. Hepatotoxicity was induced by DOX with dosages of 10 and 20 $mg{\cdot}kg^{-1}$. The rats in groups 4, 5 and 6 performed treadmill running of 25-54 min/day and 15-20 m/min, 5 days/wk for 6 wks. At the end of the aerobic training protocol, rats in the 1 and 4 groups, in the 2 and 5 groups and in the 3 and 6 groups received saline solution, $DOX_{10}mg{\cdot}kg^{-1}$ and $DOX_{20}mg{\cdot}kg^{-1}$, respectively. Results: Administration of $DOX_{20}mg{\cdot}kg^{-1}$ caused a significant increase in IGF-1 and IGF-1/IGFBP-3, an insignificant decrease in IGFBP-3, as compared to the control+placebo group. However, after six weeks of aerobic training and DOX treatment with $10mg{\cdot}kg^{-1}$ and or/ $20mg{\cdot}kg^{-1}$ an insignificant decrease in IGF-1, an insignificant increase in IGFBP-3 and a significant decrease in IGF-1/IGFBP-3 were detected, in comparison to $C+DOX_{10}$ and $C+DOX_{20}$. Conclusions: Hepatotoxicity of doxorubicin is dose-dependent and pretreatment with regular aerobic training may improve DOX-induced hepatotoxicity by up-regulation of IGFBP3.

  • Peripheral insulin resistance in obese/type II diabetes animals results from an impairment of insulin-stimulated glucose uptake into skeletal muscle. Insulin stimulate the translocation of GLUT4 from intracellular location to the plasma membrane. Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) is implicated in mediation of fusion of GLUT4-containing vesicle with the plasma membrane. Present study investigated regulatory effects of Rhodiola sachalinensis administration and exercise training on the expression of GLUT4 protein and SNAREs protein in skeletal muscles of obese Zucker rats. Experimental animals were randomly assigned into one of five groups ; lean control(LN), obese control(OB), exercise-treated(EXE), Rhodiola sachalinensis-treated(Rho), combine of Rho & EXE (Rho-EXE). All animals of exercise training (EXE, Rho-EXE) performed treadmill running for 8 weeks, and animals of Rho groups (Rho, Rho-EXE) were dosed daily by gastric gavage during the same period. After experiment, blood were taken for analyses of glucose, insulin, and lipids levels. Mitochondrial oxidative enzyme (citrate synthase, CS ; $\beta$-hydroxyacyl-CoA dehydrogenase, $\beta$-HAD) activity were analysed. Skeletal muscles were dissected out for analyses of proteins (GLUT4, VAMP2, syntaxin4, SNAP23). Results are as follows. Exercise and/or Rhodiola sachalinensis administration significantly reduced body weight and improved blood lipids (TG, FFA), and increased insulin sensitivity. Endurance exercise significantly increased the activity of mitochondrial enzymes and the expression of GLUT4 protein, however, administration of Rhodiola sachalinensis did not affect them. The effect of exercise and/or Rhodiola sachalinensis administration on the expression of SNARE proteins was unclear. Our study suggested that improvement insulin sensitivity by exercise and/or Rhodiola sachalinensis administration in obese Zucker rats is independent of expression of SNARE proteins.

Effects of Endurance Training on the Serum Levels of Tumour Necrosis Factor-${\alpha}$ and Interferon-${\gamma}$ in Sedentary Men

  • Physical activity could be considered one of the factors that affect the immune system status and function. To find the relation between exercise and cytokines, we examined the possible effects of an 8-week endurance training program on the serum levels of cytokines, including tumour necrosis factor-alpha (TNF-${\alpha}$) and interferon-gamma (IFN-${\gamma}$) in sedentary men. A total of 30 healthy young male volunteers were randomly divided into an endurance training group and a control group. The training group followed a specific exercise protocol (running on a treadmill for 15~30 min at 50~70% maximal heart rate) for 8 weeks and the control group did not participate in any exercise program. Venous blood samples were collected from both the groups 24 h before and 24 h and 48 h after the exercise. Repeated ANOVA was used for statistical purposes. The serum levels of TNF-${\alpha}$ and IFN-${\gamma}$ were determined by ELISA. Significant (p<0.05) and non-significant (p>0.05) decreases were observed in the serum levels of IFN-${\gamma}$ and TNF-${\alpha}$, respectively, after the 8-week endurance training program. Our findings indicated that an 8-week endurance exercise may affect the serum levels of some inflammatory cytokines, suggesting the beneficial role of this training protocol in elderly population and people with certain conditions (inflammation of the vertebrae or other inflammatory diseases).

  • Estimating algorithm of physical activity energy expenditure was implemented by using a tri-axial accelerometer motion detector of the SVM(Signal Vector Magnitude) of 3-axis(x, y, z). A total of 33 participants(15 males and 18 females) that performed walking and running on treadmill at 2 ~ 11 km/h speeds(each stage increase 1km/h). Algorithm for energy expenditure of physical activities were implemented with $VO_2$ consumption and SVM correlation between the data. Algorithm consists of three kinds and hip, wrist, waist and hip can be used to apply.

Wearing Comfort Evaluation of a Summer Flight Suit to Improve Ventilation (통기성 향상을 위한 하계비행복 설계 및 착용쾌적성 평가)

  • This study verified the effect of summer flight ventilation developed in a previous study based on wearing comfort evaluation. Seven healthy males in their twenties volunteered for this experiment conducted in aclimatic chamber. The experiment consisted of three consecutive periods of rest (20 minutes), running on a treadmill (10 minutes) and recovery (20 minutes). A comparative evaluation was conducted on the general flight suit which had no ventilation holes and summer flight suit that use subjective satisfaction measures and objective measures. The subjective satisfaction was evaluated according to the criteria of temperature sensation, wet sensation, thermal comfort and fatigue sensation. The objective satisfaction was measured by skin temperature, microclimate (temperature and humidity), sweat rate and thermography. The comparative wearing evaluation identified the summer flight suit decreased the temperature between skin and suit by $0.42^{\circ}C$ (upper arm), $0.9^{\circ}C$ (calf) and the skin temperature by $0.3^{\circ}C$ (shoulder), $0.4^{\circ}C$ (upper arm), $0.5^{\circ}C$ (calf) as compared to the general flight suit. The humidity inside the summer flight suit decreased at head (7.73%), shoulder (5.86%), upper arm (5.26%), and calf (8.73%) compared to the one inside the general flight suit. Thermography showed that the air flowed through ventilation holes (neck and armpit). The design of ventilation holes applied to the summer flight suit can be applicable to overall clothing that requires thermal comfort such as dust-free garments, mechanical clothing and combat uniforms.

Pretreatment Hepatoprotective Effect of Regular Aerobic Training Against Hepatic Toxicity Induced by Doxorubicin In Rats

  • Background: Doxorubicin is an anthracycline antibiotic commonly used to treat a variety of cancers as a most effective antitumor. However, its clinical use is associated with the toxic effects in numerous healthy tissues. Here we investigated the pretreatment effect of regular aerobic exercise on oxidative stress in rats acutely exposed to DOX-induced hepatotoxicity. Materials and Methods: Forty-eight Wistar male rats were randomly divided into 2 groups: control and training. The training protocol included treadmill running between 25 to 54 min/day and 15 to 20m/min, 5 days/week for 6 weeks. At the end of the exercise training protocol, rats from the control and trained groups were again randomly separated into 3 subgroups: DOX10mg/kg, DOX20mg/kg and saline. All treatments were carried 24 h after the last exercise bout and animals were sacrificed 24 h after DOX and saline injections. Results: Administration of DOX (10 and 20 $mg.kg^{-1}$) resulted in imbalance in biomarkers related to oxidants and antioxidants in liver tissue, as compared to control groups. Six weeks of pretreatment training led to a significant increase in nitric oxide (NO), superoxide dismutase (SOD) and glutathione peroxidase (GPX) as compared to the control+DOX 10 mg/kg group. Training before DOX 20 mg/kg administration also led to a significant increase in NO and SOD, and a significant decrease in malondialdehyde (MDA). In addition, there was a significant difference between DOX 10 mg/kg and DOX 20 mg/kg treatments in MDA levels, only. Conclusions: The results of the present study indicate that pretreatment with aerobic exercise induces positive adaptations and has a potential protective effect against doxorubicin (DOX)-induced hepatotoxicity with doses of 10 and 20 mg.kg.

Effect of Dietary Supplementation of Eleutherococcus Senticosus, Taurine and Carnitine on Endurance Exercise Performance in Rats (가시오갈피, 타우린 및 카르니틴 보충식이가 흰쥐의 지구력운동 수행능력에 미치는 영향)

  • The effects of dietary supplementation of Eleutherococcus senticosus, taurine and carnitine on maximal endurance exercise performance along with other related parameters were evaluated in rats that underwent aerobic exercise training for 6 weeks. Thirty-two male rats (4 weeks old) were randomly divided into 4 groups, and fed experimental diets and/or aerobic exercise trained according to the protocol: SC (sedentary control group), EC (exercise-trained control group), EE (exercise-trained Eleutherococcus senticosus-supplemented group), and EETC (exercise-trained Eleutherococcus senticosus, taurine and carnitine-supplemented group). The food efficiency ratio of EC rats was significantly lower than the value for SC rats (p < 0.01). Exercise-trained control animals (92 $\pm$ 8.8 min) could run significantly longer until exhausted on the treadmill than sedentary control rats (11 $\pm$ 0.8 min) (p < 0.001). Animals fed an Eleutherococcus senticosus-supplemented diet, and an Eleuthherococcus sonticosus, taurine and carnitine- supplemented diet while undergoing aerobic exercise training for 6 weeks exhibited, respectively, 8 and 5 minutes longer running performance until exhausted than the rats fed the control diet. The gastrocnemius muscle glycogen concentration of the rats, measured at 48 hours post maximal exercise performance test, was 43% higher in EC rats than the value for SC rats (p < 0.05), but was not different among EC, EE, and EETC rats. The mitochondrial citrate synthase activity of the soleus muscle was significantly higher in EC rats compared to the value for SC rats (p < 0.01), and showed a tendency to increase, without statistical significance, in EE or EETC rats compared to the value for EC rats. These results indicate that aerobic exercise training for 6 weeks significantly improved maximal exercise performance, muscle glycogen content along with citrate synthase activity, which are important in the energy metabolism of muscle under aerobic exercise. Dietary supplementation of Eleutherococcus senticosus in rats while undergoing aerobic exercise training improved maximal endurance exercise performance without significantly affecting muscle glycogen content and enzyme activities involved in energy metabolism during exercise. Taurine and carnitine supplementation failed to show an additive effect on maximal endurance exercise performance when consumed along with Eleutherococcus senticosus.

  • The purpose of this study was to analyze the physiology changes by circuit weight training(CWT). The subject of this study consists of 16 student on ship(experimental groups(SE) N=8, control group(SCa) N=8) and 16 students on land (experimental groups(LE) n=8, control groups(LCb) N=8). The items of measurement were the changes of CWT time and heart rate per week, musclear functino and cardiopulmonary function. The conclusions are as follows : 1. Muscular fuction 1) Back strength in SE and LE group was increased significantly (p<.01, p<.05, 8.86% and 7.08%).Grip strength was increased slightly in 4 groups but there was no significance. 2) In push-ups, all 4 groups were increased significantly (p<.05, p<.01), sit-ups also were increased significantly in SE and LE group (p<.05, 6.71% and 9.62%). 3) In SE and LE group , standing long jump was in significantly (p<.01, p<.05, 4.49% and 6.09%), but only in LE group , side step was increased significantly (p<0.1, 5.84%). 2. Cardiopulmonary function 1) HRrest was decreased slightly in all 4 groups but was not significant changes. HRmax was increased only in LE group significantly (p<.05, 2.81%), treadmill running time was increased significantly in SE and LE group respectively (p<.01, p<.05, 10.78% and 11.07%). 2) VEmax was increased significantly in SE and LE group(p<.05, p<.01, 10.59% and 13.68%), but only in LE group Rfmax was increased significantly (p<.01, 4.83%). 3) In VO2max, LCb group was increased significantly (p<.05, 6.835), but SCa group was decreased significantly (p<.05, 4.32). VO2max/kg$.$min was increased significantly in LE and LCb group respectively (p<.01 p<.05. 4.75% and 3.98%).
