Expression of Aquaporin-4 and -8 Genes in Mouse Uterus during the Estrous Cycle (발정주기 동안 생쥐 자궁에서의 Aquaporin-4와 -8 유전자의 발현)

  • Aquaporins(AQPs) are a family of transmembrane water channel proteins that are widely distributed in various tissues throughout the body and play a major role in Oanscellular and Oansepithelial water movement. Uterine endometrium undergoes recurrent uterine stromal edema in response to hormonal stimuli, however, the mechanism regulating the fluid transport during the estrous cycle has not been fully understood. To investigate the possible role of AQPs in water movement in uterus during the estrous cycle, expression patterns of AQP -1, -3, -4, -5, -8, and -9 UMh in mouse uterus were analyzed by using semiquantitative reverse transcription- polymerase chain reaction(RT-nR). We employed a combination of laser capture microdissection(LCM) and RT-PCR to examine the expression patterns in specific uterine cell types luminal epithelial cells(LE) and stromal cells(S). Our results showed that the level of AQP-4 mRNA was significantly increased while the level of AQP-3 mRNA was significantly decreased during the proestous through the estrus stage. In addition LCM revealed that AQP-4 and -8 mRNAs were highly expressed in LE compared with S. Taken together, these results suggest that AQPs may have an important function in physiological changes of mouse uterus during the estrous cycle.

Effect of a Phospholamban Peptide on the Skeletal Sarcoplasmic Reticulum $Ca^{2+}$ Transport (골격근 근장그물 칼슘이동에 대한 Phospholamban 펩타이드의 조절)

  • Phospholamban is the regulator of $Ca^{2+}-ATPase$ in cardiac sarcoplasmic reticulum(SR). The mechanism of regulation appears to involve inhibition by dephosphorylated phospholamban. Phosphorylation of phospholamban relieves this inhibition. Recently, there has been a report that the cytoplasmic domain (amino acids 1-25) of phospholamban is insufficient to inhibit the $Ca^{2+}$ pump. To explore the domains of phospholamban responsible for $Ca^{2+}-ATPase$ inhibitory activity, we examined the effect of a synthetic phospholamban peptide consisting of amino acid residues 1-25 on $Ca^{2+}$ uptake by reconstituted skeletal SR $Ca^{2+}-ATPase$. The $Ca^{2+}-ATPase$ of skeletal SR was purified and reconstituted in proteoliposomes containing phosphatidylcholine (PC) or phosphatidylcholine: phosphatidylserine (PC:PS). Inclusion of a phospholamban peptide in PC proteoliposomes was associated with significant inhibition of the initial rates of $Ca^{2+}$ uptake at pCa 6.0, and phosphorylation of this peptide by the catalytic subunit of cAMP-dependent protein kinase reversed the inhibitory effect on the $Ca^{2+}$ pump. Similar effects of phospholamban peptide were also observed using PC:PS proteoliposomes. Based on these results, we could conclude that the cytoplasmic domain of phospholamban, containing the phosphorylation sites, by itself is sufficient to inhibit the $Ca^{2+}$ pump of SR.

  • High frequency ventilation (HFV) is a new ventilatory technique that uses very small tidal volume(less than the anatomic dead space) and high frequency, and classified 4 distinct types according to frequency and mode of gas delivery. The mechanism of gas transport of high frequency oscillation ventilation(HFOV) is somewhat different to other types of HFV. To evaluate the determinants of $PaCO_2$ in HFOV, a study was done with a HFOV on 9 cats, and the results are : 1) $PaCO_2$ was not correlated with frequency at the constant stroke volume(6 voltage) and bias flow (6L/minutes). 2) $PaCO_2$ was correlated with stroke volume but not with bias flow under the constant frequency(15Hz/min) and bias flow(3 to 6L/min). From above results, the main determinant of $PaCO_2$ on artificial ventilation with HFOV was stroke volume, but further study between flow, the site of delivery to the airway and humidification of bias flow and $CO_2$ elimination is required in future research.

Involvement of Oxidative Stress and Poly(ADP-ribose) Polymerase Activation in 3-Nitropropionic Acid-induced Cytotoxicity in Human Neuroblastoma Cells

  • 3-Nitropropionic acid (3-NP) inhibits electron transport in mitochondria, leading to a metabolic failure. In order to elucidate the mechanism underlying this toxicity, we examined a few biochemical changes possibly involved in the process, such as metabolic inhibition, generation of reactive oxygen species (ROS), DNA strand breakage, and activation of Poly(ADP-ribose) polymerase (PARP). Exposure of SK-N-BE(2)C neuroblastoma cells to 3-NP for 48 h caused actual cell death, while inhibition of mitochondrial function was readily observed when exposed for 24 h to low concentrations (0.2${\sim}$2 mM) of 3-NP. The earliest biochemical change detected with low concentration of 3-NP was an accumulation of ROS (4 h after 3-NP exposure) followed by degradation of DNA. PARP activation by damaged DNA was also detectable, but at a later time. The accumulation of ROS and DNA strand breakage were suppressed by the addition of glutathione or N-acetyl-L-cysteine (NAC), which also partially restored mitochondrial function and cell viability. In addition, inhibition of PARP also reduced the 3-NP-induced DNA strand breakage and cytotoxicity. These results suggest that oxidative stress and activation of PARP are the major factors in 3-NP-induced cytotoxicity, and that the inhibition of these factors may be useful in protecting neuroblastoma cells from 3-NP-induced toxicity.

The Effect of Compressed Video Traffic over ABR on Satellite ATM Networks (위성 ATM 망에서 압축된 비디오 트래픽의 ABR 서비스에 미치는 영향)

  • In this paper we consider the performance of TCP video traffic over ABR with Long-Range Dependent VBR traffic. As compressed coded video traffics are increasing rapidly over Internet, lots of studies are being done for transmitting those traffics efficiently using limited network resources. We consider here the transmitting video service over ABR service in ATM networks, especially satellite networks. CBR or VBR services are suggested in transmitting the video traffic in ATM Forum TM 4.0. But ABR service connection, which is considered as appropriate service for data traffic, can be established with a small amount of bandwidth, MCR (Minimum cell rate). Furthermore ABR service can control the source's transmitting rate using feedback mechanism. Using this feature ABR service can be used in some applications which can control their quality of services corresponding to network loads. Compressed video sources with MPEG-2 are used for Long-Range Dependent VBR traffic here. We model the compressed video source to resemble the MPEG-2 transport streams. These compressed video traffic streams are consisted of three different frames, I-frame, P-frame, and B-frame. So when a network are overloaded, we can control the quality of service using this traffic features. TCP Traffics over ABR need large buffers in ATM switch to satisfy their QoS with background VBR traffics, which have high deviations in bandwidth. Furthermore satellite ATM networks with large feedback delay need large buffers corresponding RTT delay. The performance comparisons among EFCI and ER switch (ERICA+) switches in the network circumstances described above were shown in this paper. We also considered the case with ON-OFF VBR traffics.

  • The main objective of this study is to determine the methodology to increase the practical use of future Marine GIS based on the user's point of view of the analysis of maritime & fishery government agencies's roles. GIS related job duty of two Busan District offices of MOMAF(Ministry of Maritime and Fisheries), NFRDI(National Fisheries Research & Development Institute), Korea Institute of Maritime and Fisheries Technology and marine & fisheries local government offices of Busan Metropolitan city were analyzed for the establishment of MGIS, future extended service targeting user's increased practical utilization. Several suggestions in the area of production mechanism and technical policy for marine geographic information among the government agencies are developed to settle down successfully MGIS in Busan. Furthermore, 10 fields of Marine Geographic issues are rearranged for intending the desirable system and service.

$RpoB_{127-135}$ Peptide Derived from Mycobacterium tuberculosis is Processed and Presented to HLA-$A^*0201$ Restricted CD8+ T Cells via an Alternate HLA-I Processing Pathway

  • Mycobacterium tuberculosis (MTB) resides and replicates inside macrophages. In our previous report, we reported that CD8+ T cell-mediated immune responses specific for the peptide derived from MTB RNA polymerase beta-subunit ($RpoB_{127-135}$) could be induced in TB patients expressing HLA-$A^*0201$ subtype. In order to examine whether $RpoB_{127-135}$ specific CD8+ T cells can recognize MTB infected macrophages in vitro, CD8+ T cell lines specific for $RpoB_{127-135}$ peptide were generated from peripheral blood mononuclear cells (PBMCs) of healthy HLA-$A^*0201$ subjects by in vitro immunization technique. In this study, we observed $RpoB_{127-135}$ specific CD8+ T cells could recognize and destroy macrophages infected with MTB for 2 to 4 days. $RpoB_{127-135}$ specific CD8+ T cell immune response was inducible from PBMC of healthy subjects expressing HLA-$A^*0206$ subtype, one of HLA-A2 supertype members. Next, we investigated the HLA-I processing mechanism of $RpoB_{127-135}$ peptide in MTB infected macrophages. As a result, the presentation of the MTB derived epitope peptide, $RpoB_{127-135}$, to CD8+ T cells was not inhibited by the treatment with brefeldin-A (ER-Golgi transport inhibitor) or lactacystin (proteasome inhibitor), which blocks the classical HLA-I processing pathway. However, $RpoB_{127-135}$ specific CD8+ T cell activity was blocked either by the blocking agent for the endocytosis (cytochalasin D) or by the blocking antibody (W6/32) for HLA-I molecules. Therefore, the $RpoB_{127-135}$ peptide may be processed by accessing the alternate HLA-I processing pathway. Understanding the processing and presentation mechanisms of the MTB derived proteins will help to improve the efficacy of vaccines and the efficiency of therapeutic agents for TB.

  • In spite of extensive knowledge of the surface chemistry and the transport mechanism in filtration systems, there is still insufficient understanding of the physical characteristics of suspensions and the system components. Because of this, no filtration mechanisms are mathematically generalized to the full extent. The purpose of this paper is to propose experimental equations for the filtration process. using the tracer study in filter layer. Some of results are as follows. (1) The Volume of the specific deposit (${\sigma}$) in filtration was directly measurable using the tracer study without interrupting the filtration. (2) It was also confirmed that the head loss in filtration was greatly in fluenced by the micro-air babbles. (3) The correction coefficient(f) was introduced into the Kozeny-Carman equation in order to apply it for the clogging filter media. The coefficient(f) was experimentally obtained. The total head loss of the filter media is given by next equation. $${\frac{h}{h_0}}={\frac{1}{L}}{\int}^{z=L}_{z=0}f({\sigma})g({\varepsilon}_0,{\sigma})dz$$ $$f=aexp(-b{\sigma})$$ The above equation was applicable without regard to the variation of the suspension concentration, the filter medium diameter, the filter depth, the filtration velocity, and the amount of aluminum in all continuous filtration experiments. (4) The total head loss was graphically generalized assuming mathematical filtration models I II (see fig. 7,8) (5) The total head loss was obtained from the filtration model in the field filtration conditions.   • A model for the numerical simulation of lithium-ion batteries (LIBs) is developed for use in battery cell design, with a view to improving the performances of such batteries. The model uses Newman-type electrochemical and transfer $theories^{(1,2)}$ to describe the behavior of the lithium-ion cell, together with the Levenberg-Marquardt optimization scheme to estimate the performance or design parameters in nonlinear problems. The mathematical model can provide an insight into the mechanism of LIB behavior during the charging/discharging process, and can therefore help to predict cell performance. Furthermore, by means of least-squares fitting to experimental discharge curves measured at room temperature, we were able to obtain the values of transport and kinetic parameters that are usually difficult to measure. By comparing the calculated data with the life-test discharge curves (SB LiMotive cell), we found that the capacity fade is strongly dependent on the decrease in the reaction area of active materials in the anode and cathode, as well as on the electrolyte diffusivity.

Effects of Different Exercise Intensities on GLUT-4 and GRP-78 Protein Expression in Soleus Muscle of Streptozotocin-Induced Diabetic Rats with Caffeine Oral Administration (카페인 경구투여가 운동강도 차이에 따른 당뇨유발 흰쥐 가자미근의 GLUT4 및 GRP78 단백질 발현에 미치는 영향)

  • This study investigated the response of GLUT-4 and GRP-78 protein expression in soleus muscle of Streptozotocin-induced diabetic rats with caffeine oral administration by imposing different exercise intensities. Rats were randomly divided into 5 groups (n=6 in each group): diabetic group (D), diabetic-caffeine group (DC), diabetic-caffeine group with low intensity exercise (DCL), diabetic-caffeine group with moderate intensity exercise (DCM) and diabetic-caffeine group with high intensity exercise (DCH). The rats in DCL, DCM and DCH groups were exercised acutely by treadmill running for 8 meter/m, 16 meter/m and 25 meter/m, respectively. Little difference in GLUT-4 protein expression was shown in DC and DCL compared to D. GLUT-4 protein expression was decreased in DCM and increased in DCH was observed. GRP-78 protein expressions in DCL, DCM and DCH were little lower than that of D. An increase in GRP-78 protein was observed in DC. Improved insulin sensitivity with acute high intensity exercise gives the rats important therapy that lowers insulin requirement. This improvement of insulin sensitivity for glucose transport in skeletal muscle results from translocation of the GLUT-4 protein from the endoplasmic reticilum to the cell surface and increase in total quantity of GLUT-4 protein. It is not clear what mechanism reduced GRP-78 protein level in exercise group. It is merely conjectured that caffeine-induced lipolysis provided cells with energy in abundance and this relieved stress which cells are subjected to receive when performing exercise.
