Effects of Alkylating Agent on the Sertoli Cell of the Seminiferous Tubule in the Mouse (Alkylating agent가 생쥐 정소의 Sertoli Cell에 미치는 영향)

  • This paper aims to probe that the effect of high dose of cyclophosphamide to the Sertoli cells of the mouse was examined by transmission electron microscope. In the normal group, Sertoli cells contact each other around the basal aspect of the seminiferous tubule, forming numerous row of tight junction, blood-testis barrier. Sertoli cells contain smooth endoplasmic reticulum, well developed Golgi comples, a number of round mitochondria and microfilament. The cytoplasmic necrosis are observed from the 1-time treated group. In the 2-times treated group, smooth endoplasmic reticulum are more developed than normal group, but cisternae are partially dilated. In the 3-times treated group, the smooth endoplasmic reticulum are not developed. In the 2-times treated group, the inner membrane of the mitochondria are partially disrupted, and cristae are all disrupted in the 3-times treated group. The microfilaments are not observed in the all treated groups. According to the results above, it seems that smooth endoplasmic reticulum, mitochondria, and microfilament are disrupted by toxic effects of the cyclosphamide to the Sertoli cells of the mouse.

Synthesis of Copper Nanoparticles by a Chemical Reduction Method (화학적 환원법에 의한 구리 나노분말 합성)

  • Copper nanoparticles attract much attention as substitutes of noble metals such as silver and can help reduce the manufacturing cost of electronic products due to their lower cost and good conductivity. In the present work, the chemical reduction is examined to optimize the synthesis of nano-sized copper particles from copper sulfate. Sodium borohydride and ascorbic acid are used as reducing and antioxidant agents, respectively. Polyethylene glycol (PEG) is used as a size-control and capping agent. An appropriate dose of PEG inhibits the abnormal growth of copper nanoparticles, maintaining chemical stability. The addition of ascorbic acid prevents the oxidation of nanoparticles during synthesis and storage. Transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FTIR) are used to investigate the size of the synthesized nanoparticles and the coordination between copper nanoparticles and PEG. For chemical reduction, copper nanoparticles less than 100 nm in size without oxidized layers are successfully obtained by the present method.

XTEM Study of 1 MeV Argon Ion Implantation Induced Defects in Si and Their Annealing Behavior (1MeV Argon 이온주입에 의해 유기되 결합 및 회복기구의 XTEM 분석)

  • Ar ions were implanted at 1 MeV into (100)Cz Si wafers with dose of 1 * 10$^{15}$ ions/cm$^{2}$. Damage induced by high energy implantation and its annealing behavior during rapid thermal annealing for 10sec at temperatures from 550 to 1100${\circ}C$ were investigated by crosssection transmission electron microscopy study. It can be clearly seen from the observation that the SPE(Solid Phase Epitaxy) regrowth of the buried amorphous layer induced by ion implantation proceeds from both upper and lower amorphous/crystalline (a/c) interfaces, and the activation energy for SPE from interfaces were both 1.43eV. Misfit dislocation where two interfaces met was formed and it coalesced into the hair pin dislocation in the upper regrown region. At the higher temperature after annealing out of the misfit dislocation, hair pin dislocations showed considerable drop in its bandwidth. However, they were not disappeared even at the temperature 1100${\circ}C$ with the end of range dislocation loops which were formed at the original lower a/c interface.

High alloyed new stainless steel shielding material for gamma and fast neutron radiation

  • Stainless steel is used commonly in nuclear applications for shielding radiation, so in this study, three different types of new stainless steel samples were designed and developed. New stainless steel compound ratios were determined by using Monte Carlo Simulation program Geant 4 code. In the sample production, iron (Fe), nickel (Ni), chromium (Cr), silicium (Si), sulphur (S), carbon (C), molybdenum (Mo), manganese (Mn), wolfram (W), rhenium (Re), titanium (Ti) and vanadium (V), powder materials were used with powder metallurgy method. Total macroscopic cross sections, mean free path and transmission number were calculated for the fast neutron radiation shielding by using (Geant 4) code. In addition to neutron shielding, the gamma absorption parameters such as mass attenuation coefficients (MACs) and half value layer (HVL) were calculated using Win-XCOM software. Sulfuric acid abrasion and compressive strength tests were carried out and all samples showed good resistance to acid wear and pressure force. The neutron equivalent dose was measured using an average 4.5 MeV energy fast neutron source. Results were compared to 316LN type stainless steel, which commonly used in shielding radiation. New stainless steel samples were found to absorb neutron better than 316LN stainless steel at both low and high temperatures.

Optimization of the multi-chamber perforated muffler for the air processing unit of the fuel cell electric vehicle (연료전지 자동차용 흡기 소음기의 설계 변수 최적화에 관한 연구)

  • Fuel cells convert a fuel together with oxygen in a highly efficient electrochemical reaction to electricity and water. Since the electrochemical reaction in the fuel cell stack dose not generate any noise, Fuel cell systems are expected to operated much quieter than combustion engines. However, the tonal noise and the broad band noise caused by a centrifugal compressor and an electric motor cause which is required to feed the ambient air to the cathode of the fuel cell stack with high pressure. In this study, the multi-camber perforated muffler is used to reduce noise. We propose optimized muffler model using an axiomatic design method that optimizes the parameters of perforated muffler while keeping the volume of muffler minimized.

  • Spillage of cyst contents during surgical operation is the major cause of recurrence after hydatid cyst surgery. Instillation of a scolicidal agent into a hepatic hydatid cyst is the most commonly employed measure to prevent this complication. SB202190 is a pyridinyl imidazole derivative and is known to be a specific inhibitor of p38 MAPK. In the present study, the scolicidal effect of SB202190 was investigated. Freshly isolated Echinococcus granulosus protoscolices were subjected to SB202190 treatment (10, 20, 40, and $80{\mu}M$), and the effects on parasite viability were monitored by trypan blue staining. Corresponding effects were visualized by scanning and transmission electron microscopy. Dose-dependent protoscolex death within a few days of SB202190 treatment was observed. Although the in vitro scolicidal effect of SB202190 was satisfactory, the in vivo efficacy of this drug and also possible side effects remain to be further investigated.

Age-dependent resistance to Cryptosporidium muris (strain MCR) infection in golden hamsters and mice

  • An age-dependent aspect of resistance to Cryptosporidium muris (strain MCR) infection was monitored in Syrian golden hamsters. Mesocricetus auratus. at 1-, 5- and 10-week of age and in ICR mice, Mus musculus, at 3-, 12-, and 15-week of age orally inoculated with a single dose of $2{\times}10^6$ oocysts. respectively. The prepatent periods for both animals were similar, independent of age, but the patency was significantly longer in younger hamsters (P<0.001) and a long tendency in younger mice. Hamsters infected at 1-week of age excreted about 10 times higher oocysts than those at 5- and 10-week of age. However, the total oocyst output was similar among mice of different ages. There was a good correlation between the length of the patency and the total oocyst output in hamsters (R=0.9646), but not in mice (R=0.456l). The immunogenicity of the parasite to homologous challenge infections was very strong in hamsters and relatively strong in mice. These results indicate that acquired resistance to C. muris infection is age-related and the innate resistance is independent of age of hamsters, and that both innate and acquired resistance, on the contrary, are irrespective of age of mice.

  • The purpose of this study was to investigate the irradiation effects on the secretory granules of the rat submandibular gland acinar cell. For this study, Sprague-Dawley strain rats were irradiated to their neck region with the dose of 5Gy and l0Gy by 6MV X-radiation, and sacrificed on the experimental periods after irradiation. The authors observed the ultrastructural changes of the secretory granules of the rat submandibular gland acinar cell under a trasmission electron microscope. The results were as follows: In the transmission electron microscopic examination, secretory granules were blurred in limiting membrane on the 3hours groups after irradiation. And they showed decrease in number, irregularities in shape and distributional pattern, and inhomogeneous internal electron density on the Iday and 3days groups. After then, these changes were recovered in shape and distributional pattern on the 14days groups, and changes of internal electron density and limiting membrane were recovered on the 28days groups after irradiation. Among the intracellular organelles, rough endoplasmic reticulum was scattered, but golgi complex was not changed. And such pathologic changes were earlier and more prominent in 10Gy irradiated groups than in 5Gy irradiated groups.

The Antinociceptive Effect of Intraperitoneally Administered Nonselective Nitric Oxide Synthase Inhibitor on the Rat Formalin Test (흰쥐의 포르말린시험에서 복강 내로 투여한 비선택적 산화질소합성효소 억제제의 항통각효과)

  • Background: Nitric oxide (NO) is involved in the transmission and modulation of nociceptive information at the peripheral, spinal cord and supraspinal levels. We conducted this experiment to assess the antinociceptive effects of a nonselective nitric oxide synthase (NOS) inhibitor, N-nitro-L-arginine methyl ester (L-NAME), on the modulation of pain in rats subjected to the formalin test. Methods: Formalin 5% was injected in the right hind paw after intraperitoneal (IP) injection of various doses of L-NAME (0.5 mg/kg, 1.5 mg/kg with and without L-arginine 100 mg/kg, 5.0 mg/kg). The number of flinches was measured. Results: Formalin injected into the rat hind paw induced a biphasic nociceptive behavior. IP injected L-NAME diminished the nociceptive behaviors in a dose-dependent manner during phases 1 and 2. The concomitant injection of L-arginine reversed the antinocipetive effect of L-NAME. Conclusions: The data demonstrates that a nonselective NOS inhibitor, L-NAME, possesses antinociceptive properties in rats subjected to the formalin test, and the antinociceptive effect of L-NAME is reversed by the concomitant administration of L-arginine.

Selection and Characterization of Peptides Specifically Binding to $TiO_2$ Nanoparticles

  • We have screened phage display peptide libraries to select for peptides binding to various sized $TiO_2$ nanoparticles. Phage libraries displaying random 7mer, 12mer, and C-7-Cmer peptides were used for screening. The size of target $TiO_2$ particles used were 7 nm, 15 nm, and 25 nm in diameter. We could select peptides binding each nanoparticles from all 3 libraries. Their binding was confirmed by transmission electron microscopy (TEM). Each peptide investigated was also shown to bind the other sized particles, meaning that the binding was specific for the nature of the particle rather than for the size of it. One of the 7mer peptides (PEP9, SVSPISH) was chosen for further analysis. The binding was shown to be in a dose-dependent manner, suggesting a specific interaction.
