Inhibitory Actions of Mycotoxins on Brain $\gamma$-Aminobutyrate Transaminase ($\gamma$-Aminobutyrate Transaminase에 대한 Mycotoxin의 저해작용)

  • GABA transminase (4-aminobutyrate aminotransferase), which catalyzes the breakdown of the major inhibitory neurotransmitter, GABA, in mammalian brain, was inactivated by preincubation with the mycotoxin patulin. The time course of the reaction was significantly affected by the substrate .alpha.-ketoglutarate, which aforded complete protection against the loss of catalytic activity. The recovery from the inhibition of patulin by the addition of dithiothreitol (DTT) supports that patulin reacts with the sulfhydryl residue in the catalytic domain of the enzyme. The reconstitution of the reduced enzyme and apoenzyme with pyridoxal-5-P(PLP) was inhibited by another mycotoxin, penicilic acid. This mycotoxin may interact with lysyl residue of the enzyme. Therefore, it is postulated that the critical sulfhydryl and lysyl residues in the catalytic domain of the enzyme react with mycotoxin patulin and penicillic acid, respectively.

Anti-hepatotoxic Activity of Icariside II, a Constituent of Epimedium koreanum

  • Icariside II, a flavonol glycoside, was isolated from the aerial part of Epimedium Koreanum Nakai by the anti-hepatotoxic acitivity guided fractionation technique employing $CCl_4-in-toxicated$ primary cultured rat hepatocytes as an assay system. Its anti-hepatotoxic activity was evaluated by measuring activity of glutamic pyruvic transaminase released from the $CCl_4-in-toxicated$ primary cultured rat hapatocytes. Icariside II significantly reduced the activity of glutamic pyruvic transaminase released from the $CCl_4-in-toxicated$ primary cultured rat hepatocytes and resulted in 78% recovery of the toxicity at the concentration of $200{\;}\mu\textrm{m}$. The anti-hepatotoxic activity of icariside II on the $CCl_4-in-toxicated$ primary cultured rat hepatocytes was as potent as that of silybin.

Effects of Methanol Extracts of Black Soybean on Enzymes Activities of Serum in Rats Fed Pb and Cd Solution (검정콩 추출물이 납과 카드뮴을 투여한 흰쥐의 혈청 효소 활성도에 미치는 영향)

  • The effects of black soybena extracts on enzymes activies of rat were evaluated in present study. Eighty-four male Sprague-Dawley rats weighing 100$\pm$10g were divided into twelve groups which consisted of black soybean extract, Pb and Cd solution, and black soybean extract plus Pb or Cd soln groups. The weight gain was increased in black soybean extracts and Pb soln solution group but decreased in Cd soln solution group. The results obtained form the experiment were as follows: Glutamate pyruvate trasaminase (GPT) and glutamate oxaloacetate oxaloacetate transaminase (GOT) activities were not significantly different among experimental groups. The lactate dehydrogenase (LDH) activities of black soybean extract administered groups were decreased than those of Pb and Cd solution group. Black soybean group increased cholinesterase (ChEase) activity as compared to administration of Pb and Cd soln group.

  • In order to investigate the inter-relation with the growth of the rice-root and its transaminase-activity, by measuring the growth of its root and transaminase-activity supplying this root with various nitrogen Compounds($NO_3$-N, $NH_4$-N and Amino acid.). The obtained results are summarized as follows; 1. Growth of rice-root supplied with $NO_3$-N is generally increased in length and weight, compared with that of the root fertilized by $NH_4$-N. 2. The above-metnioned root with $NH_4$-N is not only decreased in its weight and length but also is apt to inhibited its growth as the nitrogen concentruration is increased, in compared with the root provided with $NO_3$-N. 3. The activity of G.O.T. and G.P.T. for the root fertilized by $NH_4$-N, the badly grown root is generally increased, while of the root supplied with $NO_3$-N is decreased. 4. The activity of G.O.T. and G.P.T. for the root provided with amino acid known as the considerable growth inhibiting compound is generally decreased, while that of the badly-grown root is increased. 5. The activity of G.O.T. and G.P.T. in the supernatant fraction of the rice-root is for the most part, high and low in the mitochondrial fraction.

A comparative Study on the Activities of Glutamic Dehydrogenase and Glutamic Transaminase in Livers of the Crucian carp, Pigeon, and Rat (붕어, 비둘기, 흰쥐의 肝臟의 Glutamic Dehydrogenase 및 Glutamic Transaminase 의 活性에 관한 比較 硏究)

  • The present investigation involves a comparative study of enzymatic activities in various animals. The levels of the liver protein of rat (22.0$\pm$0.01mg/ml and pigeon (22.0$\pm$ 0.16) are twice as high as that of crucian carp (13.0$\pm$ 0.09)(p < 0.01). Generally, the specific acitivity (3.77 $\pm$0.18 unit/mg) of rat glutamic pyruvic transaminase (GPT) is highest, pigeon intermediate (1.93 $\pm$0.01), and crucian carp lowest (0.71$\pm$0.07). On the other hand, the specific activity (8.23$\pm$0.09 unit/mg)of rat glutamic oxaloacetic transaminase (GOT) is highest, pigeon intermediate (3.95$\pm$0.09), and crucian carp lowest (0.92$\pm$0.01) (p < 0.01). Ratios of GOT activity to GPT activity appear no remarkable difference from the levels of various animals. Specific activity of glutamic dehydrogenase(GDH) in pigeon tissue exceeds those of rat and crucian carp. In liver, rat GOT specific activity is greater than crucian carp and pigeon. On the other hand, pigeon GDH specific activity is greater than those of rat and crucian carp. This wouls seem to be in accord with protein metabolic intensity. The patterns for GDH isozyme were remarkably appeared in various animals. Glutamic dehydrogenase isozymes gave different electrophoretic mobilities in various animals. It is interesting that crucian carp, pigeon, and rat would show this difference, which may be indicative of an evolutional pattern. The fact that livers in various animals show quite different enzyme activities would suggest the existence of such a general phylogenetic relationship.

  • Effect of various fractions from the roots of Patrinia scabiosaefolia (Valerianaceae) and whole plants of Melandryum firmum (Caryophyllaceae) on enzyme activities in mice was investigated. The butanol fractions from both plants caused a significant elevation of serum transaminase activities when administered intraperitoneally, but did not, orally. Prolonged exposure by oral administration of both plants elevated hepatic cytochrome p-450 content, indicating the induction of drug metabolizing enzymes in liver.

  • The root of Polygonum cuspidatum (Polygonaceae) has been used as treatments of hyperlipidemia, dermatitis, gonorrhea, favus athlete's foot, inflammation in traditional medicine. In order to examine anti-lipidperoxidation activity, hexane, EtOAc, BuOH and water fractions of its methanol extract were administered to carbon tetrachloride intoxicated rats. Ethylacetate fraction exhibited antilipidperoxidative effect on liver lipid homogenate and the radical scavenging effect on DPPH. Serum transaminase, AlP, triglyceride and total cholesterol contents significantly decreased by administrations of ethylacetate fraction.

  • Effects of forced physical exercise on the CCl4 hepatotoxicity were examined in adult female rats. Rats were treated with CCl4 (2 mmol/kg, ip) and introduced into a cylindrical cage rotating at 9 rpm for 20 min/hr for 6 hr. Eighteen hr following the termination of exercise serum sorbitol dehydrogenase (SDH), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT) activities and glucose-6-phosphatase activity were determined as parameters for hepatotoxicity. Physical exercise inhibited the $CCI_{4-}$induced increases in SDH, GOT, GPT activity, and glucose-6-phosphatase activity were determined as parameters for hepatotoxicity.

  • The effects of dichloromethane (DCM) on carbon tetrachloride (CT) toxicity were examined in adult male rats. A concomitant treatment of rats with DCM (0.3, 0.6, 1.2 g/kg, po) significantly potentiated the hepatotoxicity of CT (1.0 g/kg, po) as determined by increase in serum GPT (glutamic pyruvic transaminase), GOT (glutamic oxaloacetic transaminase), and SDH (sorbitol dehydrogenase) activity 24 hr following the treatments. Serum LDH (lactate dehydrogenase) activity was increased by either DCM or CT treatment.

  • Polyphenolic compounds have been reported to exhibit a wide range of pharmacological properties. In this study. we investigated the hepatoprotective effect of skin and seed of grape which contain abundant polyphenol compounds on dimethylnitrosamine(DMN)-induced liver damage in rats. Ingestion of skin and seed of grape (10% diet, daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels.
