Treatment with exopolymers of Aureobasidium pullulans SM-2001 containing $\beta-1,3/1,6-glucan$ inhibited osteoclastogenesis of bone marrow stem cells in a co-culture system with calvariae osteoblastic cells. In addition, the treatment increased mineral deposition in osteoblastic cells. These two observations prompted us to evaluate whether the exopolymers could be used as an anti-osteoporotic agent, and efficacy of the exopolymers to prevent bone loss was compared with alendronate, a bisphosphonate, in ovariectomized mice prone to osteoporosis. Administration of the exopolymers to the ovariectomized mice resulted in improved effects on femur weight and histomorphometric changes of femur such as trabecular bone volume (TBV), trabecular bone thickness (TBT), and cortical bone thickness (CBT). In conclusion, the exopolymers treatment inhibited bone loss from osteoporosis induced by ovariectomy, and the effect was comparable to alendronate administration.
Objectives: Osteoporosis is one of the most common metabolic bone disorders in the menopausal stage, a disease signified by decrease of bone mass. The purpose of this study was to learn how Lycii Fructus influences SD rats with osteoporosis caused by the removal of the ovaries. Methods: After the removal of the ovaries from 24 female Sprague-Dawley rats, they were divided into a normal group, a control group, and a Lycii Fructus (25mg/100g) administration group. After 8 weeks, the rats were sacrificed and their weights, albumin, AST, ALT, ALP, total-cholesterol, triglyceride, P, Ca, T4, E2, weight of a femur, ash content of the tibia, and area and thickness of trabecular bone were measured. Results: Regarding the density of ALT, ALP, and estradiol from serum, the Lycii Fructus group was significantly decreased compared to the control group, and the density of phosphorous and calcium also decreased slightly. Moreover, albumin, triglyceride, and T4 showed an increasing propensity, but AST and total-cholesterol showed a decreasing propensity, but not significant. Regarding the variation of bone, femur weight and ash content of tibia, the Lycii Fructus group was increased compared to the control group but significance wasn't shown. The Lycii Fructus group increased significantly over the control group in area and thickness of trabecular bone and osteoclast number. Conclusions: From the results of the above study, Lycii Fructus should be effective for osteoporosis treatment and prevention.
Purpose: This study aimed at examining the thickness of lateral cortical bone in the mandibular posterior body and the location of the inferior alveolar nerve canal as well as investigating the clinically viable bone grafting site(s) and proper thickness of the bone grafts. Subjects and Methods: The study enrolled a total of 49 patients who visited the Department of Oral and Maxillofacial Surgery at Kyung Hee University Dental Hospital to have their lower third molar extracted and received cone beam computed tomography (CBCT) examinations. Their CBCT data were used for the study. The thickness of lateral cortical bone and the location of inferior alveolar nerve canal were each measured from the buccal midpoint of the patients' lower first molar to the mandibular ramus area in the occlusal plane of the molar area. Results: Except in the external oblique ridge and alveolar ridge, all measured areas exhibited the greatest cortical bone thickness near the lower second molar area and the smallest cortical bone thickness in the retromolar area. The inferior alveolar nerve canal was found to be located in the innermost site near the lower second molar area compared to other areas. In addition, the greatest thickness of the trabecular bone was found between the inferior alveolar nerve canal and the lateral cortical bone. Conclusions: In actual clinical settings involving bone harvesting in the posterior mandibular body, clinicians are advised to avoid locating the osteotomy line in the retromolar area to help protect the inferior alveolar nerve canal from damage. Harvesting the bone near the lower second molar area is judged to be the proper way of securing cortical bone with the greatest thickness.
Purpose: The objective of this study was to investigate the relationships between primary implant stability as measured by impact response frequency and the structural parameters of trabecular bone using cone-beam computed tomography(CBCT), excluding the effect of cortical bone thickness. Methods: We measured the impact response of a dental implant placed into swine bone specimens composed of only trabecular bone without the cortical bone layer using an inductive sensor. The peak frequency of the impact response spectrum was determined as an implant stability criterion (SPF). The 3D microstructural parameters were calculated from CT images of the bone specimens obtained using both micro-CT and CBCT. Results: SPF had significant positive correlations with trabecular bone structural parameters (BV/TV, BV, BS, BSD, Tb.Th, Tb.N, FD, and BS/BV) (P<0.01) while SPF demonstrated significant negative correlations with other microstructural parameters (Tb.Sp, Tb.Pf, and SMI) using micro-CT and CBCT (P<0.01). Conclusions: There was an increase in implant stability prediction by combining BV/TV and SMI in the stepwise forward regression analysis. Bone with high volume density and low surface density shows high implant stability. Well-connected thick bone with small marrow spaces also shows high implant stability. The combination of bone density and architectural parameters measured using CBCT can predict the implant stability more accurately than the density alone in clinical diagnoses.
Aging is characterized by an age-dependent reduction in bone density. Pronounced bone loss by postmenopausal estrogen deficiency induces a bad effect on dynamic balance of bone metabolism. When the bone density is reduced to the level below the fracture threshold, the risk for fracture is greatly increased. The purpose of this study is to examine what are the effects of the Guisinhwan(GSH : 歸腎丸) on the ovariectomized rat model of postmenopausal osteoporosis. The results of the experiment are as follows: Body weight in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum albumin in control group showed significant decrease in comparison with sham. That in GSH-treated was slightly decreased in comparison with control. The level of serum ALP activity in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. The level of serum phosphorous, calcium, GOT, GPT, T3 did not significant change among the three groups. The level of serum estrogen in control group showed slightly decreased in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum T4 in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. Trabecular bone area as well as trabecular thickness in control group showed significant decrease In comparison with sham. Those in GSH-treated showed significant increase in comparison with control. Trabecular number did not significant change among the three groups. Trabecular separation only in GSH-treated showed significant decrease in comparison with control. Osteoclast number in control group showed significant increase in comparison with sham. That in GSH-treated showed significant decrease in comparison with control. Oseoblast surface in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. According to the above these results, GSH has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.

Objective : Osteoporosis is a disease of unbalanced bone metabolism that results in low bone mineral density with increased bone fragility and propensity for fractures. The increased rate of bone fracture due to osteoporosis places a significant burden on public health care expenditures. Therefore, numerous studies have been designed and performed to identify the drugs or health foods that can improve the bone quality or quantity. This study was designed to evaluate and analyze the therapeutic effects of rutin on histomorphometric values of the spine and femur in an osteoporotic mouse model induced by bilateral ovariectomy. Methods : Thirty female ICR mice (8 weeks old) underwent either a sham operation (only abdominal incision, sham group, n=10) or bilateral ovariectomy (n=20). The ovariectomized (OVX) animals were randomly divided into two groups : untreated OVX group (OVX-C, n=10), or rutin-administered group (OVX-R, n=10). The OVX-C group received weight-adjusted doses of saline vehicle and the OVX-R group received 50 mg/kg of rutin intraperitoneally, starting 1 day after surgery. At 4 and 8 weeks after surgery, serum estrogen, osteocalcin, alkaline phosphatase (ALP), and the telopeptide fragment of type I collagen C-terminus (CTX-1) were analyzed. Interleukin (IL)-1β, IL-6, IL-10, and tumor necrosis factor (TNF)-α were also analyzed. Bone histomorphometric parameters of the 4th lumbar vertebra and femur were determined by micro-computed tomography. Results : In OVX-C group, ALP, osteocalcin, CTX-1, IL-1β, IL-6, and TNF-α levels were significantly increased at 4 and 8 weeks compared to sham operation group. Rutin administration after OVX statistically significantly reduced ALP, CTX-1, IL-1β, IL-6, and TNF-α levels at 4 and 8 weeks. Rutin administration also improves bone histomorphometric parameters including trabecular bone volume fraction, trabecular thickness, and trabecular number. Trabecular separation was also decreased in OVX-R group compared to OVX-C group. Conclusion : The present study demonstrated that rutin has therapeutic effects on improving bone histomorphometric values in an OVX mouse model. The improvement in histomorphometric values may be associated with the reduction of osteoclastic activity via inhibition of IL-1β, IL-6, and TNF-α. In future studies, the mechanism for the effect of rutin on osteoporosis should be demonstrated more clearly to use rutin in human osteoporosis.
Purpose : To evaluate the relationship between morphometric analysis of bone microstructure from digital radiographic image and trabecular bone strength. Materials and Methods : One hundred eleven bone specimens with 5 mm thickness were obtained from the mandibles of 5 pigs. Digital images of specimens were taken using a direct digital intraoral radiographic system. After selection of ROI (100 × 100 pixel) within the trabecular bone, mean gray level and standard deviation were obtained. Fractal dimension and the variants of morphometric analysis (trabecular area, periphery, length of skeletonized trabeculae, number of terminal point, number of branch point) were obtained from ROI. Punch sheer strength analysis was performed using Instron (model 4465, Instron Corp., USA). The loading force (loading speed 1 mm/min) was applied to ROI of bone specimen by a 2 mm diameter punch. Stress-deformation curve was obtained from the punch sheer strength analysis and maximum stress, yield stress, Young's modulus were measured. Results: Maximum stress had a negative linear correlation with mean gray level and fractal dimension significantly (p<0.05). Yield stress had a negative linear correlation with mean gray level, periphery, fractal dimension and the length of skeletonized trabeculae significantly (p < 0.05). Young's modulus had a negative linear correlation with mean gray level and fractal dimension significantly (p < 0.05). Conclusions : The strength of cancellous bone exhibited a significantly linear relationship between mean gray level, fractal dimension and morphometric analysis. The methods described above can be easily used to evaluate bone quality clinically.
Objectives & Methods : The purpose of this study is to observe the effects of herbal-acupuncture with $Artemisiae$$Vulgaris$$Folium$ extract(AaH-HA) at $KI_{10}$($Eumgok$) on osteoporosis in ovariectomized(OVX) ddy mice. We carried out several experimental items to analyze the changes in body weight, urine, weight, uterus index, tibial length, the ash bone weight, tibial BMD, serum ALP, serum osteocalcin, serum Ca, and the levels of Ca, P, Ca/P ratio in tibia, and we performed histological and histomorphological analysis as well. Results : 1. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction phosphorus level in serum in ovariectomized mice. 2. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction creatinine level in serum in ovariectomized mice. 3. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the increase of tibial osteoclast cells in ovariectomized mice. 4. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the reduction of TBV(trabecular bone volume) and TBT(trabecular bone thickness) in ovariectomized mice. 5. Herbal-acupuncture with AaH-HA at $KI_{10}$ significantly inhibited the overgrowth of tibial growth plate length(GPL) in ovariectomized mice. Conclusions : In conclusion, our study suggested that Herbal-acupuncturing with AaH-HA at $KI_{10}$ can be effective treatment for osteoporosis.

Methods : We devide mice to 3 Group(Sham operated group, control group, WGY treated group) and analyze each serum component. Results : 1. Body weight in control group showed significant increase in comparison with sham, but that in WGY-treated showed no change in comparison with control. 2. The level of serum albumin, ALP in control group showed significant decrease in comparison with sham. That in WGY-treated was decreased in comparison with control. 3. Trabecular bone area as well as trabecular thickness in control group showed significant decrease in comparison with sham. Those in WGY-treated showed significant increase in comparison with control. 4. Osteoclast number and oseoblast surface in control group showed significant increase in comparison with sham. Those in WGY-treated showed significant decrease in comparison with control. Conclusion : WGY has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.
