A 20-residue hybrid peptide CA(1-8)-MA(1-12) (CAMA) incorporating residues 1-8 of cecropin A (CA) and residues 1-12 of magainin 2 (MA) has high antimicrobial activity without toxicity. To investigate the effects of the total positive charges of CAMA on the antibacterial activity and toxicity, a hybrid peptide analogue (CAMA-syn) was designed with substitutions of $Ile^{10}\;and\;Ser^{16}$ with Lys. According to CD spectra, structure of CAMA-syn with increase of cationicity was very similar to that of CAMA in DPC micelle. CAMA-syn showed antimicrobial activity similar with CAMA while CAMA-syn has no hemolytic activity and much lower cytotoxicity against RAW 264.7 macrophage cells than CAMA. Also, CAMA and CAMA-syn significantly inhibited NO production by LPSstimulated RAW264.7 macrophage at 10.0∼20.0 $\mu$M. CAMA-syn displayed salt resistance on antimicrobial activity against Escherichia coli at the physiological concentrations of $CaCl_2\;and\;MgCl_2$. The combination studies of peptides and antibiotics showed that CAMA-syn has synergistic effects with synthetic compound and flavonoid against Enterococcus faecalis and VREF. CAMA-syn can be a good candidate for the development of new antibiotics with potent antibacterial and synergistic activity but without cytotoxicity.
We previously identified Streptomyces griseus as an anti-cancer agent (Kim et al., 2014). In this study, we isolated compounds from S. griseus and evaluated their anticancer effect and toxicity in vitro and in vivo. Preparative centrifugal partition chromatography (CPC) was used to obtain three compounds, cyclo($_{\small{L}}$-[4-hydroxyprolinyl]-$_{\small{L}}$-leucine], cyclo($_{\small{L}}$-Phe-trans-4-hydroxy-$_{\small{L}}$-Pro) and phenethyl acetate (PA). We chose PA, which had the highest anticancer activity, as a target compound for further experiments. PA induced the formation of apoptotic bodies, DNA fragmentation, DNA accumulation in $G_0/G_1$ phase, and reactive oxygen species (ROS) formation. Furthermore, PA treatment increased Bax/Bcl-xL expression, activated caspase-3, and cleaved poly-ADP-ribose polymerase (PARP) in HL-60 cells. Simultaneous evaluation in vitro and in vivo, revealed that PA exhibited no toxicity in Vero cells and zebrafish embryos. We revealed, for the first time, that PA generates ROS, and that this ROS accumulation induced the Bcl signaling pathway.
Guh Sung Y.L.S.-95 is one of the polyacidic solution of which main component is acetic acid. We investigated the subchronic toxicity of the Guh Sung Y.L.S.-95 using SPF ICR mouse for 4 weeks. The Guh Sung Y.L.S.-95 was administered by gastric intubation, 1.0, 2.5, 5.0 g/kg body weight. The results are as follows: 1. There are no adverse effects on the clinical obserbation and body weight changes. Also, there are some significant changes in organ weight, but it was meaningless because of the absence of dose-response relationships. 2. In the hematological patterns of administered mouse, there are no significant changes between the treated groups. Also, there are no serological enzymatic changes in the treated mouse. In the 1.0 g/kg treated group, ASP activity was increased significnatly compared with control group. But, this level of activity was fall under the normal physiological range of control mouse. 3. Histopathological findings of the brain, liver, heart, spleen, kidneys, stomach, lung, testis, ovary, uterus and thymus were not observed in the treated mouse. From the above results, the Guh Sung Y.L.S.-95 has no toxicity upto the 5.0 g/kg/day of oral dose for 4 weeks.
The toxicity of the mixture of diazinon, toxaphene(TOX) and/or endrin was studied in ICR male mice(18-22 g) by oral intubation, in corn oil, daily for up to 14 days. On day 15, the exposure was discontinued and animals were monitored for an additional period of 7 days for the possible reversibility of the toxicity. The body weight gain decreased with the mixtures, as well as with the individual pesticides, during the 14-day period. TOX and TOX containing mixtures significantly increased the liver/body weight ratio. The serum glutamic pyruvic transaminase level increased at 23~374% in diazinon, TOX, and endrin or their mixture group. The cholinesterase(ChE) activity in the serum and brain was inhibited in the animals of the group of diazinon(5, 10 mg/kg) and diazinon(5 mg/kg) containing mixtures. TOX(40, 80 mg/kg) caused initial inhibitory effects on the serum ChE Day 1. but there is little effects on the brain ChE levels. endrin(5,10 mg/kg) results in significantly elevated levels of the serum ChE, with substantial decreases in the brain ChE activity. TOX and TOX containing mixtures decreased the pentobarbital(60 mg /kg, ip., in saline) induced sleep. The effects produced by this pesticides singly, as well as by their mixtures, appeared to be reversible in nature. The toxic effects exhibited by the mixtures of diazinon(5 mf/kg), TOX(40 mg/kg), and /or endrin(5 mg/kg) were found to be the resultant of the effect showed by their components individually.
Background: Although cetrimonium bromide is widely used for its bactericidal effects, the safety of cetrimonium bromide remains controversial. Therefore, the phytotoxicity of cetrimonium bromide was tested to evaluate its acute toxicity to plants and possible toxicity to other organisms and the ecosystem. Results: The germination rates of two test species, Lactuca sativa and Brassica campestris, were significantly decreased after cetrimonium bromide treatment. Furthermore, cetrimonium bromide treatment at over 1 mg/L concentration significantly affected root elongation immediately after germination. In pot experiments with semi-mature plants, significantly decreased shoot elongation and chlorophyll content were detected in both species following cetrimonium bromide treatment. Cetrimonium bromide treatment also significantly increased the antioxidant enzyme activities of plants. Conclusion: Our results show that cetrimonium bromide is phytotoxic, and since phytotoxicity testing can imply potential toxicity in the environment, further studies of the environmental toxicity of cetrimonium bromide should be performed.
The toxicity of methyl tert-butyl ether (MTBE), tert-butyl alcohol (TBA) and formaldehyde (FA) on the indigenous microbial community in forest soil was studied. MTBE, TBA and FA with different concentrations were added into microcosms containing forest soil samples. After 10 and 30 days, total viable cell number and dehydrogenase activity in the microcosms were evaluated. Bacterial communities in the microcosms were also analyzed using a denaturing gradient gel electrophoresis (DGGE). Dehydrogenase activity and total viable cell number were decreased according to the increase of MTBE, TBA and FA concentrations (P<0.05). FA toxicity was the highest, but TBA toxicity was the lowest. The results of principal component analysis using DGGE fingerprints showed that the microbial communities contaminated MTBE, TBA and FA were grouped by exposure time not exposure concentration. Dominant species in the microcosms were as follows: Photobacterium damselae sub sp. and Bacillus sp. KAR28 for MTBE; Mycobacterium sp. and Uncultured Clostridium sp. for TBA; and Uncultured Paenibacillaceae bacterium and Anxynobacillus, Flavithermus for FA.
To investigate and evaluated the scavenging and antioxidative effects of various flavonoids on paraquat induced toxicity, in vivo and vitro tests of eight flavonoids (catechin, epocatechin, flavone, chrysin, apigenin, quercetin, morin and biochanin A) were carried out. The generation of reactive oxygen substances(ROS) in PMS-NADH system $H_2O_2$ induced hemolysis and lipidperoxidation to blood, NADPH dependent lipidperoxidation to liver and lung microsome by paraquat were studied.The results are summerized as follows; 1) In the concentration ranges from 3.3 to 9.8$\mu$M of catechin,epicatechin, quercetin and biochanin A removed the 50% of DPPH radical scavenging effects. 2) In the concentration ranges from 0.60 to 1.86 mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on superoxide anion which gernerated in PMA-NADH system. 3) In the concentration ranges from 0.12 to 0.49mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on H202 which generated in PMA-NADH system. 4) In the concentration ranges from 0.6 x10$^{-5}$ to 6.3 x 10$^{-5}$mM of catechin, epicatechin, flavone, chrysin, quercetin and morin showed the inhibitory and antioxidative activity on $H_2O_2$ induced hemolysis to blood 5) All flavonoids tested exhibited inhibitory and antioxidative effects on paraquat induced liver and tung microsomal lipidperoxidation. Therefore, all flavonoids evaluated showed the useful compounds for scavenger and antioxidant on paraquat induced toxicity.
This investigation was performed to study Cd toxicity and the influence of Ca on Cd toxicity in growth, and photosynthetic pigments and enzymes in tobacco. Cd inhibited both growth and level of chlorophyll, but the inhibition was compensated by the treatment of Ca. Especially, chlorophyll content was significantly increased by the combination of Cd and Ca treatment compared with Cd treatment alone. In addition, activity and content of rubisco by Cd treatment was also significantly lesser than the non-treated control. The highly reduced activity of rubisco was minimized by the combined treatment of Ca to Cd. Rubisco activase activity and content also showed a pattern of change similar to the rubisco level, suggesting that Cd- and Ca-induced changes of rubisco could be caused by rubisco activase.
The lectin from starfish, Asterina pectinifera, was purified and tested for its potential antitumor activity. It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of Asterina pectinifera lectin (APL) at 4mg/$5{\times}10^5$ cells resulted in 28% death of Ehrlich ascites tumor cell, 40% of L929, 60% of A549, and 52% of HeLa cells after 48 hours incubation. Toxicity of APL to L929, Ehrlich ascites, A549, and HeLa cells revealed a reduction in cell viability of approximately 70% at APL concentration of 8mg/$5{\times}10^5$ cells after 48 hours incubation. Administration of APL ($100{\mu}g/day$ or $300{\mu}g/day$) inhibited the growth of Ehrlich ascites cells in vivo. Mice given only Ehrlich cells survived an average of $15{\pm}1$ (S.E.) days. Mice given Ehrlich cells and $100{\mu}g\;or\;300{\mu}g$ APL had 58% and 67% survival, respectively, after 20 days. These results suggest that APL has antitumor activity.

Effect of chromium (Cr) stress on antioxidant enzyme activities and malondialdehyde (MDA) content were investigated in leaves and roots of mangrove (italic (L.) Druce) seedlings. Cr toxicity effects were also assessed on young seedlings. The seedlings were grown in green house condition for three months in nutrient solution with 0, 0.5, 1, 1.5, 2, 2.5, and 3 mg $L^{-1}$$CrCl_3$. This study showed that Cr led to the change of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) and activities at different concentrations. The activity of antioxidant enzymes in leaves of K. candel seedlings indicates that enzymes engaged in antioxidant defense in certain level especially in low concentration of Cr treatments. The activities of SOD and POD were activated by Cr in the root level, while CAT activity was inhibited. CAT activity decreased in response to high concentrations of Cr. In the present study indicated that SOD in root was active in scavenging the superoxide produced by Cr. Both in roots and leaves, an increase in malondialdehyde (MDA) content was observed with increase in metal concentration and exposure periods. Our finding indicated that the high concentration of excessive Cr supply may interfere with several metabolic processes of seedlings, causing toxicity to plants as exhibited by chlorosis, necrosis, photosynthetic impairing and finally, plant death.
