Arsenic-Induced Differentially Expressed Genes Identified in Medicago sativa L. roots

  • Arsenic (As) is a toxic element that easily taken up by plants root. Several toxic forms of As disrupt plant metabolism by a series of cellular alterations. In this study, we applied annealing control primer (ACP)-based reverse transcriptase PCR (polymerase chain reaction) technique to identify differentially expressed genes (DEGs) in alfalfa roots in response to As stress. Two-week-old alfalfa seedlings were exposed to As treatment for 6 hours. DEGs were screened from As treated samples using the ACP-based technique. A total of six DEGs including heat shock protein, HSP 23, plastocyanin-like domain protein162, thioredoxin H-type 1 protein, protein MKS1, and NAD(P)H dehydrogenase B2 were identified in alfalfa roots under As stress. These genes have putative functions in abiotic stress homeostasis, antioxidant activity, and plant defense. These identified genes would be useful to increase As tolerance in alfalfa plants.

초내식성 스테인레스강의 금속이온용출특성 및 세포적합성 (Metal Ion Release Behaviour and Cytotoxicity of a Super Stainless Steel)

  • The toxic metal ion release behaviour and the cytotoxicity of a super stainless steel (S.S.S, 22cr-20Ni -6Mo-0.25N) were investigated The measurement of the amount of static and wear- induced trace metal ion released from the steels was conducted in Hank's balanced salt solution using an electrothermal atomic absdrption spectrometry equiped with Uaphite furnace. And the in vitro cytotoxicity of the materials was assesed in cell culture. The static dissolution rates of Fe and Cr ions from the S.S.S were significantly lower than those of 316L SS. However, the Ni ion release from the S.S.S during the first 4 weeks was yester than that from 316L 55 by 15-45%. Also, the wear-in- duces dissolution rates from the steels were not correlated either with their elemental composition rates or with the static metal ion release rates. The S.S.S did not deteriorate the osteoblasts viability. And no toxic response was observed from the macrophages cultured for 7 days in RFMI 1640 medium immersed with the S.S.S specimens.

Selenite Stress Elicits Physiological Adaptations in Bacillus sp. (Strain JS-2)

  • A bacterial isolate (strain JS-2) characterized as Bacillus sp. was challenged with high concentrations of toxic selenite ions. The microbe was found to transform the toxic, soluble, colorless selenite (${SeO_3}^{2-}$) oxyions to nontoxic, insoluble, red elemental selenium ($Se^0$). This process of biotransformation was accompanied by cytoplasmic and surface accumulation of electron dense selenium ($Se^0$) granules, as revealed in electron micrographs. The cells grown in the presence of selenite oxyions secreted large quantities of extracellular polymeric substances (EPS). There were quantitative and qualitative differences in the cell wall fatty acids of the culture grown in the presence of selenite ions. The relative percentage of total saturated fatty acid and cyclic fatty acid increased significantly, whereas the amount of total unsaturated fatty acids decreased when the cells were exposed to selenite stress. All these physiological adaptive responses evidently indicate a potentially important role of cell wall fatty acids and extracellular polymeric substances in determining bacterial adaptation towards selenite-induced toxicity, which thereby explains the remarkable competitiveness and ability of this microbe to survive the environmental stress.

Molecular Basis of Hexanoic Acid Taste in Drosophila melanogaster

  • Animals generally prefer nutrients and avoid toxic and harmful chemicals. Recent behavioral and physiological studies have identified that sweet-sensing gustatory receptor neurons (GRNs) in Drosophila melanogaster mediate appetitive behaviors toward fatty acids. Sweet-sensing GRN activation requires the function of the ionotropic receptors IR25a, IR56d, and IR76b, as well as the gustatory receptor GR64e. However, we reveal that hexanoic acid (HA) is toxic rather than nutritious to D. melanogaster. HA is one of the major components of the fruit Morinda citrifolia (noni). Thus, we analyzed the gustatory responses to one of major noni fatty acids, HA, via electrophysiology and proboscis extension response (PER) assay. Electrophysiological tests show this is reminiscent of arginine-mediated neuronal responses. Here, we determined that a low concentration of HA induced attraction, which was mediated by sweet-sensing GRNs, and a high concentration of HA induced aversion, which was mediated by bitter-sensing GRNs. We also demonstrated that a low concentration of HA elicits attraction mainly mediated by GR64d and IR56d expressed by sweet-sensing GRNs, but a high concentration of HA activates three gustatory receptors (GR32a, GR33a, and GR66a) expressed by bitter-sensing GRNs. The mechanism of sensing HA is biphasic in a dose dependent manner. Furthermore, HA inhibit sugar-mediated activation like other bitter compounds. Taken together, we discovered a binary HA-sensing mechanism that may be evolutionarily meaningful in the foraging niche of insects.

암모니아의 특성에 따른 활용 현황과 부식 손상에 대한 고찰 (A Study on the Utilization Status and Corrosion Damage with Ammonia Characteristics)

  • Recently, ammonia has emerged as an alternative energy source that can reduce carbon emissions in various industries. Ammonia is used as a fuel in internal combustion engines because it contains no carbon in its components and does not emit any carbon when burned. It is also used in various fields such as fertilizer production, refrigeration, cleaning and disinfection, and drug manufacturing due to its unique characteristics, such as high volatility and easy solubility in water. However, it is highly corrosive to metals and is a toxic gas that can pose a risk to human health, so caution must be exercised when using it. In particular, stress corrosion cracking may occur in containers or manufacturing facilities made of carbon-manganese steel or nickel steel, so special care is needed. As ammonia has emerged as an alternative fuel for reducing carbon emissions, there is a need for a rapid response. Therefore, based on a deep understanding of the causes and mechanisms of ammonia corrosion, it is important to develop new corrosion inhibitors, improve corrosion monitoring and prediction systems, and study corrosion prevention design.

최근 불산 저장탱크에서의 가스 누출시 공정위험 및 결과영향 분석 (Study on the Consequence Effect Analysis & Process Hazard Review at Gas Release from Hydrogen Fluoride Storage Tank)

  • 최근 발생한 경북 구미의 불산 누출 및 경남 울산의 염산 누출사고의 예와 같이 화학공장에서 발생되는 사고중대 부분은 저장탱크나 운송배관 및 플랜지호스 등의 손상에 의한 휘발성 유독성물질의 대량누출이며, 이 경우 누출된 지역의 자연환경과 대기 조건에 따른 유독성물질의 확산 거동이 인적, 물적 피해의 중요한 변수가 되기 때문에 위험성평가는 가장 중요한 관심 대상이 된다. 따라서 본 연구에서는 누출물질에 대한 대기 중 확산을 모사하기 위하여 불산 저장탱크에서 누출된 경우를 예제로 선택하여, 수치해석과 ALOHA(Areal Location of Hazardous Atmospheres)의 확산 시뮬레이션을 이용한 결과해석을 수행하였다. 먼저 공정위험분석으로 정성적 평가인 HAZOP(Hazard Operability) 결과를 살펴보면 첫째 공정흐름상(flow) 위험 요소로서 플렌지, 밸브와 호스의 균열 등 손상으로 인한 누출에 의한 운전지연 또는 독성가스누출 등이 발생할 수 있고, 둘째 온도, 압력, 부식으로부터는 화재, 질소공급과 압 그리고 탱크나 파이프 이음관의 내부 부식으로 인한 독성누출의 가능성이 높은 것으로 분석되었다. 다음 결과 영향분석 기법인 ALOHA를 운용한 결과를 살펴보면 Dense Gas Model에 대한 입력 자료값에 따라 미치는 결과 영향이 다소 차이가 있음을 발견하였으나 기상조건으로서 대기안정도 보다는 풍향 및 풍속이 가장 영향을 미치는 것으로 분석 되었다. 또한 풍속이 빠를수록 누출물질의 확산이 잘 일어났고, 수치해석결과인$LC_{50}$과 ALOHA의 AEGL-3(Acute Exposure Guidline Level)과 결과를 비교했을 때 확산길이는 다소 차이가 있지만 확산농도 측면에서는 액체와 증기누출인 경우에 있어서 거의 비슷한 결과를 보였다. 따라서 ALOHA 모델을 운영한 결과 각 시나리오별 경향은 상당히 일치함을 볼 수 있었다. 따라서 추후 수치해석과 확산모델링에 의한 예측농도를 국제적인 기준치인 IDLH(Immediately Dangerous to Life and Health), ERPG(Emergency Response Planning Guideline), AEGL(Acute Exposure Guidline Level)과 비교 함 으로서 독성 가스의 대한 완충거리를 결정 할 수 있고, 이와 같은 연구방법은 유독성물질 누출에 따른 위험성평가를 보다 효율적으로 수행하는데 도움을 줄 것이며, 지역사회 비상대응체계 수립 시 적절하게 활용할 수 있을 것이다.

반도체 공정용 차압식 질량 유량 제어 장치의 개발 및 성능 평가 (Development and Evaluation of Differential Pressure Type Mass Flow Controller for Semiconductor Fabrication Processing)

  • This paper describes the fabrication and characterization of a differential pressure type integrated mass-flow controller made of stainless steel for reactive and corrosive gases. The fabricated mass-flow controller is composed of a normally closed valve and differential pressure sensor. A stacked solenoid actuator mounted on a base-block is utilized for precise and rapid control of gas flow. The differential pressure flow sensor consisting of four diaphragms can detect a flow rate by deflection of diaphragm. By a feedback control from the flow sensor to the valve actuator, it is possible to keep the flow rate constant. This device shows a fast response less than 0.3 sec. Also, this device shows accuracy less than 0.1% of full scale. It is confirmed that this device is not attacked by toxic gas, so the integrated mass-flow controller can be applied to advanced semiconductor processes which need fine mass-flow control corrosive gases with fast response.

재난 대응 활동 시 노출가능 유해인자 종합분석 -화재 현장을 중심으로- (Comprehensive Analysis of Exposed Adverse Factors in Disaster Response Activities - Focused on Fire -)

  • 화재와 같은 재난현장에서 재난대응활동을 수행하는 소방공무원은 진압 구조 구급활동을 하는 동안 열, 연기, 유독가스에 노출되며 정신적인 스트레스, 생물학적 유해인자, 그리고 화재 진압 시 요구되는 인체공학적으로 부자연스러운 자세로 인한 육체적 과부하 등의 다양한 유해인자에 노출되고 있다. 그러나 지금까지 이에 대한 연구는 개별적 수준에서는 일부 존재해 왔지만 화재현장에서 소방관에게 노출되는 유해인자를 종합적으로 정리하고 그 대책까지 제안한 연구는 존재하지 않았다. 본 연구의 목적은 재난현장에서의 안전 및 유해인자를 종합적으로 도출하고 그 대응방안까지 제시함으로써 소방공무원의 노출로 인한 건강장애 및 사고 예방에 기여하는 데 의미가 있다고 할 수 있다. 이에 소방업무환경별로 화재진압분야, 구조 및 구급분야, 생활환경분야로 나누어 환경분석을 하고 물리적 화학적 생물학적 심리적 측면으로 구분하여 화재현장의 유해인자를 도출하고 노출현황을 제시하였다. 그리고, 물리적 유해인자 중에는 특수건강검진 결과 가장 많은 케이이스수를 차지하는 소음분야에 대하여 대처방안을 모색하였다. 화학적 유해인자는 보호장비와 클린룸 운영방안을, 생물학적 유해인자는 감염예방교육과 예방접종, 주기적 관리점검 체계확립을, 심리적 유해인자는 PTSD 경감 부스 설치와 멘토링제도의 도입을 제시하였다.

질산 화학사고 사례분석 및 독성피해 영향범위에 대한 연구 (A study on the case analysis of Nitric acid chemical accident and establishment of preventive measures)

  • 본 연구는 국내·외에서 질산화학사고를 바탕으로 사업장, 실험실, 병원, 용기파손, 화학물질 오 주입으로 이상반응으로 인한 유독가스가 발생하고, 여러 사례들을 바탕으로 질산의 위험성과 화학사고 발생 시 발생할 수 있는 상황과 안전성에 대한 사례검토를 통해서 5년간의(2014년부터 2018년까지) 질산 화학사고 사례 대상으로 각 연도별, 사고현황, 사고유형, 사고원인, 사고 장소 등의 사례를 분석하여 안전교육과 질산화학사고 발생 시 개선 방안을 검토하였다. 그 결과 36건의 질산 화학사고가 조사되었으며, 그중 16건의 작업자 부주의사고가 발생하였고, 8건의 운송사고와 12건의 시설괸리 부족과 같은 사고가 발생하였다. 질산화학사고는 지속적으로 발생하고 있으며, 이를 바탕으로 CARIS를 활용한 독성영향범위를 최악에 시나리오를 산정하여, 피해영향범위를 통해서 효과적인 사고대응 범위를 측정하였고, 질산화학사고 초기대응방법과 사고예방을 위해서 안전교육 강화와 비상조치계획과 상관관계식을 바탕으로 영향범위를 예측하여, 정량화된 자료를 파악하였다. 또한 주민대피 선정이 수월하게 할 수 있는 상관관계식을 바탕으로 영향범위의 신뢰성을 검토하고, 실제 사업장 사고 시나리오에 적용하여 사고 시 비상상황에 사고대응 효과와 예방대책을 제시하였다.

High and Low Dose Folinic Acid, 5-Fluorouracil Bolus and Continuous Infusion for Poor-Prognosis Patients with Advanced Colorectal Carcinoma

  • Objective: Evaluation and assessment of response rate, duration and toxicity in patients subjected to 5-FU based chemotherapy. Background: The therapeutic ratio shifts with different 5FU/LV regimens and none yet serve as the internationally accepted Gold Standard. A bimonthly regimen of high dose leucovorin is reported to be less toxic and more effective than monthly low dose regimens. We here compare therapeutic responses and survival benefit of the two regimens in poor prognosis patients with advanced colorectal carcinoma. Patients and Methods: A total of 35 patients with histologically confirmed colorectal carcinoma were subjected to de Gramont and Mayo Clinic regimen. Nineteen patients were treated with high dose folinic acid ($200mg/m^2$), glucose 5%, 5-FU ($400mg/m^2$) and 22 hr. CIV ($600mg/m^2$) for two consecutive days every two weeks. These patients had failed responses to previous chemotherapy and were above sixty years of age with poor general status. Sixteen patients (six below 60 years) with progressive disease were subjected to low dose folinic acid ($20mg/m^2$)for five days, 5FU($425mg/m^2$) injection bolus for 5 days, every five weeks. An initial evaluation was made in sixty days and responders were reevaluated at sixty days interval or earlier in case of clinical impairment. Based on positive prognosis, the therapy was continued. Evaluation of treatment response was made on the basis of WHO criteria. Results: The response rate was 44% in thirty four evaluable patients, with 4 complete responses (11.8%) and 11 (32.4%) partial responses. The two schedules were well tolerated, whereas, mild toxicity without WHO Grade ${\geq}2$ events was assessed. The response duration was extended (12 months) in a few patients with age above sixty years treated by high dose bimonthly regimen of 5FU/LV. Conclusion: The regimens are safe and effective in advanced colorectal carcinoma patients with poor general status.
