The Effect of Hangover Drink using Propolis on Ethanol Oxidation (프로폴리스를 이용한 숙취 해소 음료의 알콜 분해 효과)

  • The studies was conducted to investigate the effect of hangover drink using natural products propolis on ethanol oxidation. The hangover drink developed using propolis concentrates and various medicinal plants. The extractions of propolis and acanthopanax senticosus was shown the highest alcohol oxidation among the combination raw. The ethanol oxidation effect were not significantly different between propolis drink and in other company products in acute alcoholic toxic doses. In the case that it was conducted oral administration with acute alcoholic toxic doses 1/3(1.8mg per 1kg body weight of mouse), propolis drink comparative products of other company was shown 0.026% to 270 min, 0.000% to 360 min. In conclusion, propolis drink was shown excellent alcohol oxidation in fixed dose of alcohol, but was not shown in a large quantities of alcohol.

Pharmacological Studies on Butanol Fraction of Puerariae Radix (갈근 Butanol Fraction의 약리 작용에 관한 연구)

  • Effective BuOH fraction was extracted from Puerariae radix (Pueraria thunbergiana), and several pharmacological activities were examined. From several pharmacological examinations, it was found that BuOH fraction has an antipyretic activity against typhoid vaccine pyrexia in rabbit, a papaverine-like antispasmodic activity on isolated ileum and jejunum of rat, an analgesic effects on mice treated by 0.7% HAc, and an anti-inflammatory activity of carrageenin edema in rat. It was found that BuOH fraction has detoxication effects on acute and subacute toxic rat induced by alcohol in consequence of biochemical tests of serum. Therefore it was considered that BuOH fraction has preventive and therapeutic effects in alcohol intoxication.

Effect of Methyl tert-butyl Ether and Its Metabolites on the Microbial Population: Comparison of Soil Samples from Rice Field, Leek Patch and Tidal Mud Flat (다양한 토양 환경에서 Methyl tert-Butyl Ether와 그의 대사산물이 노출되었을 때 미생물 군집에 미치는 영향: 논, 밭, 갯벌 시료 비교)

  • Toxic effect of methyl tert-butyl ether (MTBE), tert-butyl alcohol (TBA) and formaldehyde (FA) on microbial activity and diversity was compared in rice field, leek patch, and tidal mud flat soil samples. MTBE, TBA and FA with different concentrations were added into microcosms containing these soil samples, and placed at room temperature for 30 days. Then the microbial activities such as dehydrogenase and viable cell numbers and microbial community using a DGGE (Denaturing gradient gel electrophoresis) fingerprinting method were measured. Among the samples, dehydrogenase activity in rice field was inhibited the most by MTBE, TBA and FA. The toxic effect was higher according to the following orders: FA > MTBE > TBA. Dominant species in the microcosms contaminated with MTBE, TBA and FA were Chloroflex, Bacilli, gamma-proteobacteria in the rice field sample, Sphingobacteria, Flavobacteria, Actinobacteria, Bacilli, gamma-proteobacteria in the leek patch sample, and Sphingobacteria, Flavobacteria, delta-proteobacteria, gamma-proteobacteria in the tidal mud flat sample.

A Review on the Report about Drug-induced Hepatitis published by the National Institute of Toxicological Research (국립독성연구원 보고서 '식이유래 독성간염의 진단 및 보고체계 구축을 위한 다기관 예비연구'에 대한 분석 및 고찰)

  • Background : A report published by the National Institute of Toxicological Research (NITR) in January 2004 about toxic hepatitis in Korea contained the result of analysis on 55 cases of severe toxic hepatitis from 7 university hospitals for 8 months. NITR claimed that the extrapolated annual frequency of severe toxic hepatitis in Korea was 1904 cases per year. They also claimed that the most frequent etiology of severe toxic hepatitis were herbal medications and similar plant preparations (61.7%), contrasted with traditional therapeutic preparations and healthy foods (29.1%). I have investigated that report to be certain of the result because it is a very important subject for public health and society in Korea. Results : The NITR report has too many problems to have faith in its results. They include the following: 1. The report uses only 55 cases to estimate annual prevalence rate of severe toxic hepatitis in Korea. 2. There was a large regional preponderancy in the NITR report (2 cases in Seoul from a population of 10.17 million, 19 cases in Gwangju from a population of 1.4 million) 3. There was another preponderancy that selected much fewer cases caused by western medication (9.1%) than other reasons. 4. The NITR report used a modified scale than that officially recognized to diagnose toxic hepatitis. 5. There was a mistake using the scale to adapt the right indications. 6. They collected cases before beginning the study, although it was a prospective study. There was also not any questionnaire or other materials concerned with alcohol, drugs, or history of past liver disease. Conclusions : NITR is one of the important official arms of the government of Korea. Nevertheless, there is a severe problem in validity because of selection bias, uncertain accuracy, and insufficiency of raw materials in the report. Therefore it seems incorrect to generalize the results of the report and there is a lack of confidence in it as a national study publishing by the NITR.

Preparation of Gas Sensor from Pitch-based Activated Carbon Fibers and Its Toxic Gas Sensing Characteristics (피치계 활성탄소섬유기반 가스센서 제조 및 유해가스 감응 특성)

  • The electrode for gas sensor was prepared by using pitch-based activated carbon fibers and polyvinyl alcohol (PVA) to investigate the toxic gas sensing characteristics. The physicochemical properties of activated carbon fibers electrode for gas sensor were analyzed with SEM and BET. Toxic gases sensing property of the electrode was also identified by different toxic gases such as $NH_3$, NO and $CO_2$. The specific surface area of activated carbon fibers electrode for gas sensor was decreased by 33% owing to PVA used as a binder compared with the activated carbon fibers. However, its pore size distribution of the ACF electrode was not greatly influenced by PVA. The activated carbon fibers electrode for gas sensor responded to toxic gases by electron hopping unlike semiconductor based gas sensors. In this study, activated carbon fibers electrode was decreased to 7.5% in resistance for the NH3 gas of the 100 ppm concentration and its $NH_3$ gas sensing property was confirmed the most excellent compared with other toxic gases.

Puerariae radix increases Alcohol-induced Suppressed Cell Proliferation and Expression of Nitric Oxide Synthase in Dentate Gyrus of Rats

  • Traditionally, aqueous extracts of Puerariae radix had been used for the treatment of alcohol-related problems. In the present study, the effect of Puerariae radix on cell proliferation and expression of nitric oxide synthase (NOS) in the dentate gyrus of alcohol-intoxicated rats were investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry and nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) histochemistry, respectively. Sprague-Dawley rats weighing 150 ± 10 g were divided into four groups: the control group, the Puerarias radix-treated group, the alcohol-treated group, and the alcohol- and Puerariae radix-treated group. The numbers of both BrdU-positive and NADPH-d-positive cells in the dentate gyrus were inhibited significantly by alcohol administration, while Puerariae radix treatment was shown to increase those numbers. In this study, it was revealed that Puerariae radix possesses protective effect against alcohol-induced suppressed new cell formation and NOS expression in the dentate gyrus. Based on the results, it is possible that NO, which might play an important role in the regulation of cell proliferation, is a major target of the toxic effects of alcohol.

Effect of a Static Magnetic Field on Susceptibility to Ethanol-Induced Hepatic Dysfunction in Rats

  • To determine whether alcohol-treated rat liver cells can be protected by a static magnetic field (SMF), we analyzed the blood chemistry and histology of hepatic tissue removed from alcohol-exposed rats that had been exposed to a static magnetic field. The rats were exposed to a 0.3 tesla (3,000 gauss) magnetic field (MF) for 24 hr daily for 5 weeks with appropriate controls. Glutamic pyruvic transaminase activity and the triglyceride levels in animals exposed to the north (N) or south (S) pole of the MF decreased significantly (p<0.01 and p<0.05, respectively) compared with negative control animals with alcohol exposure. A histological examination of hepatic tissue revealed a moderate to severe accumulation of fat vacuoles of various sizes in the cytoplasm of the hepatocytes of animals in the negative control group throughout the study; whereas in groups exposed to the MF poles, fewer fat vacuoles were seen compared with the negative control group. Electron microscopic observations showed that exposure to the N or S pole protected organelles, including the nucleus, from damage during exposure to this toxic agent, as indicated by the fact that the nucleus and the mithochondria virtually retained their shape throughout this study. These results suggest that exposure to a SMF could be an excellent way of protecting against alcohol-induced damage to the rat liver cell.

Electrospinning Fabrication and Characterization of Poly(vinyl alcohol)/Waterborne Polyurethane/Montmorillonite Nanocomposite Nanofibers (전기방사법을 이용한 폴리(비닐 알코올)/수분산 폴리우레탄/몬모릴로나이트 나노복합섬유의 제조 및 특성분석)

  • Poly(vinyl alcohol) (PVA)/waterborne polyurethane (WBPU)/montmorillonite clay (MMT) nanocomposite nanofibers were prepared using electrospinning technique of aqueous solutions. Scanning electron microscopy, transmission electron microscopy, X-ray diffraction and thermal gravimetric analyzer were used to characterize the morphology and properties of the nanocomposite nanofibers. Since PVA, WBPU and MMT are hydrophilic, non-toxic and biocompatible materials, these nanocomposite nanofibers can be used for filter and medical industries as wound dressing materials, antimicrobial filters, etc.

Studies on the Antidotal Effect of Panax ginseng The Therapeutic Effect of Ginseng on the Acute Alcoholism (인삼의 해독작용에 관한 연구 급성 알코을 중독에 대한 인삼의 치료 효과)

  • In order to investigate the influences of Panax ginseng (white ginseng and red ginseng) on the anesthetic effect and toxic effect of alcohol, experimental studies .had been carried out with albino rabbits, mongolian dogs and mice. The anesthetic effect of alcohol was observed by measuring the induction time, .anesthetic time, recovery time and duration from the beginning of induction to , the recovery of anesthesia (total time), respectively. and toxic effect ($LD_{50}$) of alcohol was measured. In addition to these experiments, al cohol concentration in .blood, blood sugar level, serum transaminase (GOT and GPT) activities and serum alkaline phosphatase activity were measured. Also in order to study the clinical effect of alcohol in healthy students, code .substitution, response time and muscle coordination were tested. The results were obtained as follows. 1. In the rabbits and mongolian Jags, the induction time of anesthesia by the administration of alcohol was delayed by the pretreatment of ginseng but recovery time and total time of anestksia were markedly shortend. 2. The bleed alcohol concentration was decreased by the pretreatment of ginseng , but not affected in mongolian dogs. 3. The blood sugar level, serum transaminase (GOT and GPT) activities and alkaline phoshatase activity in rabbits and dogs induced by the administration of alcohol were affected by the Pretreatment of ginseng. Because those were included within normal ranges, the differnces have no remarkable significance. 4. Liver alcohol dehydrogenase activity of rabbit was increased by the treatment of ginseng, especially it was markedly increased by the treatment of red ginseng 5. The average lethal dose of alcohol to mice was increased by the pretreatment. of ginseng, especially it was markedly increased by the pretreatment of red .ginseng. 6. In the clinical experiments, the blood alcohol concentration induced by alcohol administration was not affected by the pretreatment of ginseng whereas the bleed sugar level was increased. Blood alcohol concentration and bleed sugar level were measured after three hours alcohol administration. 7. The response time of healthy students administered with alcohol was markedly shortened by the pretreatment of ginseng but the experiments on the code substitution and muscle coordination were not affected.

Suppressive Effect of Lactic Acid Bacteria on the Toxicity of Bisphenol A in Rats

  • We have examined if lactic acid bacteria could suppress the toxic effect of bisphenol A. Lactobacillus casei YA-70 was chosen as representative. Thirty rats were divided into two groups (immature and mature) according to the weight. Each group was divided again into the control group (only alcohol treatment), bisphenol A treated group, and bisphenol A plus Lactobacillus casei YA-70 treated group. When 500 ppm of bisphenol A was fed everyday, 83% of immature group and 50% of mature group died within 3 weeks. Their internal organs, mainly livers and lungs, were changed in color and severely damaged. In the intestine of 5 ppm-fed group tumor-like nodules were observed. However, their number and size were markedly decreased when Lactobacillus casei YA-70 was supplemented in diet. This study strongly indicates that Lactobacillus casei YA-70 might play an important role to suppress the toxic effect of endocrine disruptor.
