Physiological Activities and Amino Acid Compositions of Korean Dried Laver Porphyra Products (국내산 마른 김(Porphyra)의 생리활성 및 아미노산 조성 비교)

  • The physiological activities and amino acid compositions of dried lavers of Porphyra from the Jangheung, Seochen and Haenam coasts were determined. The ethanol extract of the Haenam dried laver exhibited the greatest 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (56.1%), ferric-reducing antioxidant potential (FRAP) value(86.5 mM) and total polyphenol content (3.51 ppm) of these dried laver products. In addition, there was a strong positive correlation between the antioxidative activity and total polyphenol content of the laver products, suggesting that polyphenol compounds contribute to the antioxidant capacity in the ethanol extract of dried laver. However, there was no difference among the extracts in terms of cell proliferation activity. Fourteen combined amino acids were identified in oligopeptides from the dried laver products. Of the three extracts, that of the Haenam dried laver contained the highest levels of both free and total amino acids including alanine, glutamic acid, taurine, phosphoserine, and aspartic acid.

The Effect of Green Tea Extracts on the Fermentation Properties of Polyphenol-Enriched Beers (녹차 추출물을 첨가한 polyphenol 강화 맥주의 발효 특성에 대한 연구)

  • The purpose of this study was to investigate the effects of green tea extracts (GTE) on the fermentation properties of polyphenol-enriched beers. As such, the formation patterns of tannoid in beer with GTE were investigated at 3 different infusion times, while the ale and the lager beers fortified with GTE were analyze to ascertain effects on gravity, pH, yeast viability, total polyphenol, and tannoid during fermentation period. The results were as follows: 1) The formation of tannoid in beer with GTE in the tannometer; In reaction of GTE with polyvinylpyrrolidon (PVP), control lager beer peaked in the formation of tannoid at $70\;{\sim}\;80\;mg$ of PVP, the 1st extract exceeded the detection limit, the 2nd extract at $170\;{\sim}\;180\;mg$, while the third extract at $150\;{\sim}\;160\;mg$ of PVP. The GTE were slow in reacting with PVP, and their formation patterns were different from those of polyphenols from barley and hop. 2) Ale fermentation; The final alcohol content was 9.25% (ABV). The addition of GTE increased the yeast viability after 2 days and finally reached 52.3% from 30.9%. Total polyphenol in GTE beer increased by 1.5 times (p < .05). However, its tannoid contents increased by 6.4 times. 3) Lager fermentation; The final alcohol content was 5.93% (ABV). The effect of GTE on lager beer was minimal for all variables. However, total polyphenol of GTE beer increased by 1.4 times (p < .05). Its tannoid increased by 3.3 times (p < .05).

Nutritional Components and Physiological Activity of 4 Wild Vegetables (Salvia plebeia R. Br, Angelica acutiloba, Gynura procumbens and Saururus chinensis Baill) Cultivated in Chungbuk Province (충북산 산채 4종(배암차즈기, 일당귀, 명월초 및 삼백초)의 영양성분 및 생리활성)

  • This study investigated on the nutritional components and physiological activity of four wild vegetables namely Salvia plebeia R. Br., Angelica acutiloba, Gynura procumbens and Saururus chinensis Baill for the development of representative wild vegetables in Chungbuk. Salvia plebeia possessed the highest radical scavenging activity and beta-carotene, but exhibited the lowest α-glucosidase inhibitory activity compared to the other three vegetables. Angelica acutiloba showed high crude protein content and α-glucosidase inhibitory activity, but contained low total polyphenol content, radical scavenging ability and beta-carotene compared to the other three vegetables. Gynura procumbens showed high mineral content, beta-carotene, vitamin K1 content and α-glucosidase inhibitory activity, but showed the lowest total polyphenol content and radical scavenging ability compared to the other three vegetables. Saururus chinensis showed high crude fiber content and total polyphenol content, but contained the lowest mineral and vitamin K1 content. To conclude, it is suggested to use Salvia plebeia or Saururus chinensis as antioxidant food materials and Angelica acutiloba and Gynura procumbens as food materials and sources of α-glucosidase inhibitors. In particular, it is believed that Saururus chinensis, which possessed high content of crude fiber, is suitable for low-calorie food materials such as diet products.

  • Ramie (Boehmeria nivea) is a perennial herbaceous plant belonging to the family Urticaceae. It was used in folk remedies for diuretic or anti-pyretic purposes and as an hepatoprotective, anti-oxidant, and anti-inflammation agent. In this study, we investigated the composition of free sugars, amino acids, fatty acids, organic acids, and total polyphenol contents in parts of ramie (root, stem, and leaf) and different harvest areas (10 areas). Overall, free sugars were found as sucrose, glucose, fructose, and galactose at concentrations ranging from 24.5 to 1173.8 mg/g. Amino acids lysine, threonine, tyrosine, phenylalanine, and histidine were detected at concentrations of 33.8 to 3735.3 mg/g. Major fatty acids were linoleic acid, palmitic acid, and linolenic acid at concentrations of 7.3 to 364.4 mg/g. In organic acids, the concentration of malic acid was highest of all with 672.2 mg/g. The content of free sugars, amino acids, fatty acids, organic acids, and total polyphenol was higher in leaves than in roots and stems. In contrast, the total polyphenol content was higher in roots with 33.7 - 219.4 mg/g than in leaves or in stems. As seen in collecting region, the contents of free sugars, amino acids, fatty acids, organic acids, and total polyphenol of Bn-33, 39, 55, 65, and 90 were higher than other samples. Chemical compositions of different parts of ramie are important factors to consider in manufacturing functional foods. The results of this study provide fundamental information on the chemical compositions of ramie parts and would help develop new functional foods from ramie leaves or the whole plant.

Phytochemical constituent, percentage yield and phenolic content estimation of different solvent system of Carica papaya leaves

  • Carica papaya is an important medicinal plant used in the management of different disease conditions. Phytochemical screening was carried out using different chemical test, Percentage yield and total phenolic content was evaluated using Folin Ciocalteu method in different solvent system; methanol, ethanol, ethyl acetate, n-butanol and n-hexane respectively. The phytochemical screening of the studies showed the presence of flavonoids, saponins, tannins, terpenoids, glycosides, steroids, carbonhydrate, anthraquinone and alkaloids. The percentage yield of crude extract and total polyphenol content was high in methanol, ethanol and ethyl acetate when compared with n-butanol and n-hexane. The studies show that Carica papaya leave extracts is a potent source of secondary metabolites. The use of the plant in the management of diseases is justified.

Optimization of Aqueous Methanol Extraction Condition of Total Polyphenol from Spent $Lycium$ $chinense$ Miller to Develop Feed Additives for Pig (양돈용 사료 첨가제 개발을 위하여 구기자 부산물로부터 메탄올수용액을 이용한 총 폴리페놀 추출조건 최적화)

  • This study was conducted to develop a functional feed additive for pig with spent $Lycium$ $chinense$ Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent $Lycium$ $chinense$ Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression ($p$ = 0.001), linear ($p$ = 0.002), square ($p$ = 0.017) and interaction ($p$ = 0.047) was significant and adjusted determination coefficient ($R^2$) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.

Functional Activities of Microwave-Assisted Extracts from Lyophyllum ulmarium (마이크로웨이브 추출공정에 의한 만가닥버섯의 기능적 특성)

  • Functional activities of Lyophyllum ulmarium microwave-assisted extracts under different conditions including electron donating ability, tyrosinase inhibition activity and nitrite scavenging effect were examined. Total polyphenol content increased as increasing microwave power up to 90 W in the water extracts. Electron donating ability increased with microwave power up to 90 W in 50% ethanol extract and 99% ethanol extract. Tyrosinase inhibition activity and nitrite scavenging effect in the extract increased as microwave power increased during extraction. Total polyphenol content increased as extending extraction time up to 5 min in the water extract. But the highest electron donating ability and tyrosinase inhibition activity was obtained after 10 min extraction. Significantly higher total polyphenol content and electron donating ability were found in the water extract whereas greater tyrosinase inhibition activity and nitrite scavenging effect were observed in 99% ethanol extract. The maxium nitrite scavenging effect was found at pH 1.2 and decreased as pH increased.

Growth Characteristics and Asiaticoside Content, and Antioxidant Activities in Centella asiatica by Cultivation and Irrigation Methods (재배조건 및 관수방법에 따른 병풀 (Centella asiatica) 계통의 생육, Asiaticoside 함량과 항산화활성)

  • Background: Centella asiatica is a well-known medicinal plant having a wound healing effect. In this study, the growth, asiaticoside content, and antioxidant components and activity were investigated in C. asiatica resources under different cultivation and irrigation conditions. Methods and Results: The cultivation and irrigation methods were divided as greenhouse and open field, and sprinkler and sub irrigation. respectively. Growth characteristics were measured in aerial parts of C. asiatica. Asiaticoside content was analyzed using high performance liquid chromatography. Total polyphenol, total flavonoid and 2,2-diphenyl-1picryl hydrazyl (DPPH) radical scavenging activity were analyzed for antioxidant activities. Growth was higher under greenhouse and sub irrigation than open field and sprinkler conditions. In the leaves, asiaticoside content was 53.45 mg/g (open field) and 34.38 mg/g (sub irrigation), total polyphenol was 41.14 mg/g (open field) and 25.73 mg/g (sub-irrigation), and total flavonoid was 27.26 mg/g (open field) and 23.72 mg/g (sub-irrigation). DPPH radical scavenging activity in the leaves was 85.97 mg·ascorbic acid equivalent (AAE)/g (open field) and 54.83 mg·AAE/g (sub irrigation). Conclusions: Asiaticoside and antioxidant components and activity were not accompanied with high yield, although high growth was observed under greenhouse conditions. Therefore, cultural requirements of C. asiatica should be sufficiently considered to suit each purpose.

Comparison of Biological Activities of Korean Halophytes

  • Halophytes are expected to possess abundant secondary metabolites and various biological activities because of habitat in extreme environments. In this study, we collected 14 halophytes (Asparagus oligoclonos, Calystegia soldanella, Carex pumila, Chenopodium glaucum, Elymus mollis, Glehnia littoralis, Limonium tetragonum, Messerschmidia sibirica, Rosa rugosa, Salsola komarovii, Spergularia marina, Suaeda glauca, Suaeda maritima, and Vitex rotundifolia) native to Korea and compared their total polyphenol contents, antioxidant and anti-inflammatory activities. The total polyphenol contents of R. rugosa (27.28%) and L. tetragonum (13.17%) were significantly higher than those of the other 12 halophytes and L. tetragonum, R. rugosa, and M. sibirica showed significantly greater antioxidant activities than the other 11 halophytes, as determined by DPPH (2,2-diphenyl-1-picrylhydrazyl). A. oligoclonos, E. mollis, and C. pumila showed significantly greater anti-inflammatory activities than the other 11, as determined by NO (Nitric oxide) and $PGE_2$ (Prostaglandin $E_2$) levels. In contrast, these three extracts had normal and low total polyphenol contents among the 14 halophytes. Consequently, the total polyphenol content in the 14 studied halophytes appeared to be related to antioxidant, but not anti-inflammatory activity levels.

Analysis of Ginsenosides and Non-Saponin Components of Red Ginseng from Landraces and New Varieties

  • We quantitatively analyzed eight varieties of 6-year-old red ginseng, including four local landraces from the Inje, Geumsan, Jinan, and Punggi regions and four new varieties, Chunpoong, Yunpoong, Guempoong, and K-1, for 10 ginsenosides, acidic polysaccharide, crude polyacetylene, and total polyphenol content to find out which varieties are most suitable for producing red ginseng. Most of the new varieties contained more ginsenosides than the local landraces. While the acidic polysaccharide content of Geumsan red ginseng was lower than that of the others, its crude polyacetylene content was the highest, with a mean of 33.99%. The Inje, Geumsan, and Jinan red ginseng had a significantly higher total polyphenol content than the others.
