Combustion-Retardation Properties of Pinus rigida Treated with Ammonium Salts (암모늄염으로 처리된 리기다 소나무의 난연성)

  • This study was performed to test the combustion-retardation properties of Pinus rigida-based materials by the treatment of ammonium salts. Pinus rigida plate was soaked by the treatment with three 20 wt% ammonium salt solutions consisting ammonium sulfate (AMSF), monoammonium phosphate (MAPP), and diammonium phosphate (DAPP), respectively, at the room temperature. After the drying specimen treated with chemicals, combustion properties were examined by the cone calorimeter (ISO 5660-1). When the ammonium salts were used as the retardant for Pinus rigida, the flame retardancy improved due to the treated ammonium salts in the virgin Pinus rigida. However the specimen shows increasing CO over virgin Pinus rigida and It is supposed that toxicities depend on extents. Also, the specimen with ammonium sulfate showed both the lower total smoke release (TSR) and lower total smoke production (TSP) than those of virgin plate. Among the specimens, the sample treated with diammonium phosphate showed a strong inhibitory effect of combustion.

Incorporation of phosphate into protein and other nitrogenous compounds in Chlorella cells (Chlorella 세포에서의 $^{32}P$-인산의 단백질 및 다른 질소화합물로의 전환)

  • In the process of the incorporation of orthophsphate into protein and other cell constituents, the role of inorganic polyphosphate and RNA-polyphosphate complex and the correlation between them were pursued by analyzing the contents of $^{32}P$ and total P in various fractions of Chlorella cells, which had been uniformly labeled with $^{32}P$ before the inoculation in a normal "cold" medium or P-free medium during the culture. The effects of ionizing radiation and various micronutritional-element deficiencies on the phosphate incorporation into, and biosynthesis of, protein and other introgenus compounds in the cells were also observed. When the uniformly $^{32}P$-labeled algae were grown in a normal "cold" medium the contents of $^{32}$ P in the fractions of protein, DNA and RNA-polyphosphate complex increased, but those in the fraction of acid-insoluble polyphosphate decreased. On the other hand, amount of $^{32}P$in the fraction of RNA was almost unchanged in spite of rapid increase of the total P. In the growing period of $^{32}P$-labeled algae in a P-free medium, amounts of $^{32}P$ in the fractions of DNA, protein and lipid increased, while those in the fractions of RNA-polyphosphate and inorganic polyphosphates decreased. When the algal cells were irradiated with about 70, 000r of gamma-rays before the inoculation in the medium, amounts of phosphate in the fractions of DNA, RNA, nucleotides and protein decreased during the culture, compared with those of the control. However, the phosphate content in the fraction of acid-insoluble polyphosphate of the irradiated cells increased than those of the control. In the growing period of the algae in a Mo-free, medium, amounts of acid-soluble total phosphate and nucleotides of the cells increased, while the amounts of residual protein and RNA decresed compared with those of the normal cells. Amounts of alkali-labile protein and phospholipid of the cells grown in a B-free medium decreased, whereas amount of phosphate in acid-soluble fraction increased compared with the control. In general, the contents of protein and RNA in each microelement deficient cells decreased more or less, compared with those in the normal cells.in the normal cells.

Characterization of Phosphate-solubilizing Microorganisms in Upland and Plastic Film House Soils (밭과 시설재배지 토양의 인산가용화 미생물의 특성)

  • With the aim to explore the possible role of phosphate-solubilizing bacteria in soil, we conducted a survey of phosphate-solubilizing microorganisms colonizing in upland and plastic film house soils. Soil EC, pH, organic matter, available phosphate, exchangeable cation such as potassium, calcium and magnesium, and total P of plastic film house soils were higher than those of upland soils. Phosphate-solubilizing bacteria population was higher in plastic film house soils than upland soils, but species of phosphate-solubilizing bacteria was more diverse in the upland soils than the plastic film house soils. There was significant positive correlation between phosphate solubilization and phosphate-solubilizing bacteria in soils. Bacillus, Cedecea, Brevibacillus, Paenibacillus, Pseudomonas, Serratia spp. were isolated from upland soils and Bacillus and Cellulomonas spp. were from plastic film house soils.

The Effect of Dietary Calcium and Phosphate Levels on Calcium and Bone Metabolism in Rats (흰쥐에서 칼슘과 인의 섭취비율이 체내 칼슘 및 골격대사에 미치는 영향)

  • This study was designed to investigate the effect of dietary calcium and phosphate levels on calcium and bone metabolism in rats. The rats were divided into six groups and each of the groups was fed diets with different Ca/P ratios. The experimental periods were 5 weeks . There was no significant different difference in dietary intake, body weight gain, and organ weight among the groups with different calcium and phosphate intake levels. Fecal calcium excretion was not significantly different among the groups, but urinary calcium excretion was increased by the increase in Ca/P ratio. Fecal phosphate excretion was not different but urinary phosphate excretion was increased by the increase in dietary phosphate intake. There was no significant difference in serum alkaline phophatase activity and urinary hydroxyproline levels were not significantly different among the groups. The low calcium-high phosphate(0.25Ca-1.2% P) group showed the lowest total calcium content in femur and scapula. This may be due to it having the lowest Ca/P ratio among groups. The low calcium-high phosphate(0.2%Ca-1.2%P) group showed that mandible is almost lost and osteolyzed Harversian canal was expanded in femur. Results suggest that phosphate intake affects calcium and bone metabolism more with inadequate calcium nutrition that with adequate calcium intake. Thus , for normal bone growth and metabolism , adequate calcium intake and/or high Ca/P ratio are important.

Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate (Chlorella의 인산대사에 관한 연구)

  • Yung Nok Lee (Dept. of Biology, Korea University) : Studies on the phosphate metabolism in Chlorella, with special reference to polyphosphate. Kor. J. Microbiol., Vol.2, No.1, p1-11 (1964). 1. Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction.   • Phosphate based inhibitor is playing a decisive role in inhibiting the corrosion of steel rebar in chloride condition. We have used different amount of ammonium phosphate monobasic (APMB) as corrosion inhibitor in mortar with different amount of chloride ions. The compressive strength, flexural strength, open circuit potential (OCP), electrochemical impedance spectroscopy (EIS), potentiodynamic polarization resistance (PPR), scanning electron microscopy (SEM) and Raman spectroscopy were performed to access the effect of inhibitor on corrosion resistance. As the amount of inhibitor is increased, the compressive strength increased. The electrochemical results show that as the amount of inhibitor and chloride ions are increased, the total impedance and corrosion resistance of steel rebar increased attributed to the formation of the stable oxide films onto the steel rebar surface. It is suggested that APMB can work in high concentration of chloride ions present in concrete where phosphate ion helps in formation of stable and protective phosphate based oxide film.

Removal of Total Phosphate by Bipolar Packed Bed Electrolytic Cell (충전복극전해조에 의한 총 인 제거)

  • To evaluate the performance of a bipolar packed bed cell (BPBC) filled with granular aluminium, the experiments were carried out in two groups as batch and continuous processes. In a batch process, removal efficiency of total phosphate (T-P) was 88% in case of electrolytic treatment of phosphate solution, T-P 10 mg/L at 6 V during 3 h by BPBC filled with granular aluminium. In a continuous process, residual T-P concentration was about 2 mg/L in case of electrolytic treatment of phosphate solution, 10 mg/L at 6 V, HRT 3 h by BPBC filled with granular aluminium. Break-through point was observed after running for 120 h at hydraulic retention time (HRT) of 3 h.

  • The physiological responses of microorganisms to specific nutrient limitation can be regulated at the transcriptional levels. In this study, in order to develop the Pichia pastoris-derived promoter inducible by nutrient-limited condition, we constructed cDNA libraries using RT-PCR of total RNA from P. pastoris in steady-states of phosphate-limited chemostat with different dilution rates. Various genes were detected from cDNA library. Among these genes, the gene encoding putative sodium/phosphate ($Na^+$/Pi) symporter (NPS), high affinity transporter of phosphate, was detected. It was observed that expression of NPS increased in a manner specific to phosphate-limited condition through Northern blot. Therefore, it is thought that the promoter from NPS gene may have the potential as auto-inducible promoter by phosphate-limited culture condition without inducer.

Effects of Lime and Phosphate Applications on Growth and Nitrogen Fixation of Alfalfa in Acid Soil (산성토양에서 석회와 인산시용이 Alfalfa의 생장 및 질소고정에 미치는 영향)

  • We investigated the effects of applications of various levels of lime(0, 250, 500 and 1,000 kg/10a) and phosphate (0, 17 and 34 kg/10a) on growth and nitrogen fixation of alfalfa (Medicago sativa L.). Effects of lime and phosphate applications were significantly different on dry matter (DM) weight of each part and on acetylene reduction activity (ARA) of alfalfa at 9 weeks alter sowing (p<.05). The effect of lime on DM of shoot and root was not significantly different at 14 weeks after sowing (early bloom stage), but that of phosphate on DM was significantly improved as increasing of phosphate levels (p<.01). The effects of lime and phosphate on ARA were significantly increased (p<.05). Application of lime and phosphate decreased total nitrogen (TN) content of each part of alfalfa at 9 weeks after sowing (p<.05). The effects of lime application on TN was higher but that of phosphate application on TN was lower than no application of lime or phosphate at 14 weeks after sowing (p<.05).

Quality Enhancement of Frozen Chicken Meat Marinated with Phosphate Alternatives

  • The effects of phosphate alternatives on meat quality in marinated chicken were investigated with the application of chilling and freezing. Breast muscles were injected with solution of the green weight containing 1.5% NaCl and 2% sodium tripolyphosphate (STPP) or phosphate alternatives. Treatment variables consisted of no phosphate [control (-)], 0.3% sodium tripolyphosphate [control (+)], 0.3% prune juice (PJ), 0.3% oyster shell, 0.3% nano-oyster shell, and 0.3% yeast and lemon extract (YLE) powder. One-third of the meat samples were stored at 4℃ for 1 d, and the rest of the meats were kept at -18℃ for 7 d. In chilled meat, a lower drip loss was noted for control (+) and YLE, whereas higher cooking yield in YLE compared to all tested groups. Compared with control (+), the other treatments except PJ showed higher pH, water holding capacity, moisture content, lower thawing and cooking loss, and shear force. Natural phosphate alternatives except for PJ, improved the CIE L* compared to control (-), and upregulated total protein solubility. However, phosphate alternatives showed similar or higher oxidative stability and impedance measurement compared to control (+), and an extensive effect on myofibrillar fragmentation index. A limited effect was observed for C*, h°, and free amino acids in treated meat. Eventually, the texture profile attributes in cooked of phosphate alternatives improved except for PJ. The results indicate the high potential use of natural additives could be promising and effective methods for replacing synthetic phosphate in chilled and frozen chicken with quality enhancement.
