Null mutants generated by targeted gene replacement are frequently used to reveal function of the genes in fungi. However, targeted gene deletions may be difficult to obtain or it may not be applicable, such as in the case of redundant or lethal genes. Constitutive expression system could be an alternative to avoid these difficulties and to provide new platform in fungal functional genomics research. Here we developed a novel platform for functional analysis genes in Magnaporthe oryzae by constitutive expression under a strong promoter. Employing a binary vector (pGOF1), carrying $EF1{\beta}$ promoter, we generated a total of 4,432 transformants by Agrobacterium tumefaciens-mediated transformation. We have analyzed a subset of 54 transformants that have the vector inserted in the promoter region of individual genes, at distances ranging from 44 to 1,479 bp. These transformants showed increased transcript levels of the genes that are found immediately adjacent to the vector, compared to those of wild type. Ten transformants showed higher levels of expression relative to the wild type not only in mycelial stage but also during infection-related development. Two transformants that T-DNA was inserted in the promotor regions of putative lethal genes, MoRPT4 and MoDBP5, showed decreased conidiation and pathogenicity, respectively. We also characterized two transformants that T-DNA was inserted in functionally redundant genes encoding alpha-glucosidase and alpha-mannosidase. These transformants also showed decreased mycelial growth and pathogenicity, implying successful application of this platform in functional analysis of the genes. Our data also demonstrated that comparative phenotypic analysis under over-expression and suppression of gene expression could prove a highly efficient system for functional analysis of the genes. Our over-expressed transformants library would be a valuable resource for functional characterization of the redundant or lethal genes in M. oryzae and this system may be applicable in other fungi.
This study assessed hazards at the harvest stage of strawberry farms which may cause risk to humans. A total of 216 samples were collected from 6 strawberry farms (soil culture farms: A, B, C; nutriculture farms: D, E, F) located in Western Gyeongnam. The collected samples were subjected for sanitary indicator bacteria (aerobic plate count, coliforms and Escherichia coli), major foodborne pathogens (E. coli O157:H7, Listeria monocytogenes, Salmonella spp., Staphylococcus aureus and Bacillus cereus), and fungi. The levels of APC and coliform in the soil culture farms were 1.0-6.9 and 0.4-4.6 log CFU/g (leaf, mL, hand or 100 $cm^2$), respectively. The samples obtained from the nutriculture farms were contaminated with the levels of 0.8-4.9, and 0.2-2.6 log CFU/g (leaf, mL, hand or 100 $cm^2$) of APC and coliform. However, E. coli was not detected in any samples. In major foodborne pathogens, S. aureus was detected at the level of ${\leq}$3.3 log CFU/hand in workers' hand samples and B. cereus was detected at the levels of 0.4-4.1 log CFU/g (hand or 100 $cm^2$) in soil, plants and workers' hygiene. L. monocytogenes, E. coli O157:H7 and Salmonella spp. were not detected. Fungi were detected at the levels of 1.0-5.2 and 0.2-4.4 log CFU/g (leaf, mL, hand or 100 $cm^2$) in soil culture and nutriculture farms, respectively.
These studies were carried out to investigate the effects of rice straw on microflora in relation to the decomposition of organic matter, and the rate of rice straw decomposition. The number of total bacteria was increased in the first stage, and the number of microorganisms in upper layer was generally larger than lower layer. The number of fungi tended to decline as rice plant grew. Aerobacter among cellulose decomposition bacteria decreased with time, and the number of microorganisms in lower layer was higher than upper layer. The number of glucose decomposition bacteria and sulfate reducing bacteria increased in the submerged soil to which rice straw was applied, but decreased by percolation. the change of manganese oxidizing bacteria seemed not to be affected by rice straw application while they tend to increase as the rice plant grew. The aspect of microorganisms in the percolated water was same that of lower layer, but the number was low as much $10^{-1}$ during the whole stages. The decomposition rate of rice straw applied to submerged soil was about 40 per cent during the rice grew. The decomposition rate of cellulose contained rice straw was about 30 per cent, and lignin was about 60 per cent. The 70-80 per cent of nitrogen remained in the rice straw applied to soil.
Influnces of Fuiarium solani and Phytophthora cactorum infection on the changes in saponin components of Korean ginseng (Panax ginseng C.A. Meyer)roots and some of the biology of those fungi in relation to ginseng root were investigated. Mycelial growth of F. solani was decreased as increasing concentration of the water extracts of fresh ginseng roots, while that of P. cactorum was increased as increasing the concentration of the water extracts and crude saponin. Mycelial growth of F. solani, however, was increased as increasing concentration of crude ginseng saponin upto 20 ppm, while it was tended to be decreased when the concentration was higher than 50 ppm. Nystatin also suppresed the growth of F. solani as increasing its concentration, but it did not affected on the growth of p. cactorum. Ginsenoside Ra and Ro components were not detected in ginseng roots inoculated with F. solani or P. cactorum. Panaxadiol gisenosides were increased by 3.0%, whereas panaxatriol ginsenosides were decreased by 34.9% in ginseng roots inoculated with F. iolani. In ginseng roots inoculated with P. cactorum panaxadiol ginsenosides were increased by 21.1%, but panaxatriol ginsenosides were decreased by 23.5%. PD/PT ratio in ginseng roots inoculated with F. solani or P. cactorum were equally increased by 58.4% in spite of differences in the change of panaxadiol and panaxatriol ginsenosides. The total saponin components of ginseng roots inoculated with F. solani or P. cactorum were decreased by 17.8% and 2.5%, respectively.
This study was carried out to estimate the economic impacts on operation cost and curtail the bulking agent between two kinds of plants in swine farms. Bulking agents and Plants have a variety of roles in the fields of the composting for livestock manure and also represent an economic problem in terms of plant operation costs and compost production. Two farms which have rotary(size of reactor : 10${\times}$35${\times}$1.5m) or bucket(size of reactor : 10${\times}$68${\times}$2m) plants were used for 24 weeks for bucket conveyor system, which are composed of refilling rice hull as a bulking agent every 3 weeks till decreasing volume, for 4 weeks for rotary conveyor system, which has continuous compost production system without refilling rice hull, respectively. Composts was produced in 24 weeks in the bucket conveyor system, in 4 weeks in the rotary conveyor system, respectively. The results are as follows : 1. The tissue change of Rice hull at the composts of 45 days pursuant to composting steps was more crumbling in bucket conveyor system than in rotary conveyor system. 2. Microbial counts of the final composts for safety and quality showed that total bacteria counts was 1.01${\times}$108 cfu/g in bucket conveyor system, 2.82${\times}$108 cfu/g in rotary conveyor system, Salmonella was 0.3${\times}$102 cfu/g in bucket conveyor system, 7.6${\times}$102 cfu/g in rotary conveyor system, colifom bacteria was 0.5${\times}$106 cfu/g in bucket conveyor system, 1.5${\times}$106cfu/g in rotary conveyor system, fungi 1.24${\times}$106 cfu/g in bucket conveyor system, 0.01${\times}$106 cfu/g in rotary conveyor system, respectively. However, Any system used in this trial could not be met the regulation of A grade compost of EPA and USA. 3. C:N ratio according to the composting was more rapidly changed in bucket conveyor system with 64.5 of 5 days compost to 25.4 of final products than in rotary conveyor system with 26.7 of 5 days compost to 25.9 of final products. 4. Based on the mechanical characteristics of plants used in trial and compared with Rotary conveyor system, the Bucket conveyor system in which has 0.72 ㎥/㎥ of bulking agent capacity per slurry could be curtailed 1.78 ㎥of rice hull for disposal of waste, 1㎥. It was proper facilities to produce composts quantitative in Rotary conveyor system, and to treat waste quantitative and obtain good results in compost quality in Bucket conveyor system.

We measured the influence of antifungal antagonists Bacillus subtilis AH18 and Bacillus licheniformis K11 on soil microbial community in microcosms. Both antifungal antagonists were confirmed to suppress hot pepper phytophthora blight. Phospholipid fatty acids (PLFA) were analyzed to investigate the soil microbial community. B. subtilis AH18 changed the total PLFA composition and bio-indicators of PLFA, compared with other treatments. B. subtilis AH18 decreased the proportion of bacteria and gram negative/gram positive bacteria, and increased the fungi/bacteria and anaerobic/aerobic microorganisms. In addition cy19:0/18:$1{\omega}7c$, which means adaptation to unfavorable environmental conditions, was increased by the application of B. subtilis AH18. On the other hand the inoculation of B. licheniformis K11 or combined inoculation of both antifungal strains did not affect soil microbial community. The suppression of phytophthora blight and preservation of indigenous soil microbial community may be achieved by the combined application of B. subtilis AH18 and B. licheniformis K11.

This study is carried out to sanitize rape (Brassica napus) pollen by gamma irradiation. Rape pollens were treated with 0, 5, 10 and 15 kGy gamma irradiations, and then analyzed for the following: general composition, microbial population, reducing sugar, Hunter color values, TBARS (2-thiobarbituric acid reactive substances) values, and VBN (volatile basic nitrogen). Mold and coliform bacteria were not detected in the samples irradiated at 5 kGy or more. Yeasts and total aerobic bacteria were not detected in the samples irradiated at 10 kGy or more (<$10^2$ CFU/g). Moisture, ash, crude protein, crude fat, carbohydrate, reducing sugar and the contents of volatile basic nitrogen in the irradiated pollen did not show any significant changes by irradiation. Hunter color values, $L^*$, $a^*$ and $b^*$ values were decreased with increment of irradiation dose. TBARS values were increased with an increment of irradiation dose. In conclusion, gamma irradiation at 5 kGy was considered to be an effective treatment to control for mycotoxin producing fungi in rape pollen to minimize changes of general composition and physicochemical properties. Further studies should be investigated to reduce the detrimental effects induced by irradiation.
Hypocrea disease is the most severe disease of oyster mushroom cultivation in Korea. Physiological and ecological studies were performed on the pathogens (Hypocrea spp.) to obtain basic information for developing the integrated disease management system. Fourteen isolates of Hypocrea were collected from oyster mushroom house in five areas. Pathogenic fungi causing disease of oyster mushroom were identified as Hypocrea sp. based on morphological characteristics and pathogenicity. Two isolates (H-1, H-12) showed the fastest growth at $15^{\circ}C$ but four isolates (H-8, H-9, H-13, H-14) showed slower growth than those of other isolates at $20^{\circ}C$ and $25^{\circ}C$. Stroma with ascocarps and ascospore were produced on PDA under fluorescent light. The five isolates produced stroma with ascocarps and ascospores. Formation of fruiting body of strains H-14 of Hypocrea were the best out of all the strains on the potato dextrose agar (PDA). Also, fruiting bodies and ascospores were completely produced under fluorescent light. The growth of the isolates was correlated with total carbon content. The stroma of the isolates was formed mainly in histidine and asparagine treatment and especially in histidine-70 and asparagine-100 treatment. In the test of pathogenicity, after and before spawning showed very fast incidence of disease.
Control system for both rapid fermentation and storage of yogurt in refrigerator was developed and its performance was investigated. Fermentation temperature for normal and Bifidus containing yogurt was maintained at maximum $40^{\circ}C$ for about 7 and 11 hours, respectively. The pH, acidity, total viable cell number of lactic acid bacteria and viscosity of both yogurts after completing the fermentation were $4.23{\sim}4.29$, $0.93{\sim}0.97%$, $4.8{\times}10^7{\sim}$2.54{\times}10^8\;cfu/ml$ and $1,700{\sim}1,810\;cp$, respectively. The rate of fermentation for normal yogurt was faster than that of Bifidus yogurt. The changes of pH, acidity, viable cell number and viscosity during storage time were $4.09{\sim}4.54$, $0.76{\sim}1.1%$, $9.4{\times}10^6{\sim}5.68{\times}10^8\;cfu/ml$ and $1,450{\sim}2,000\;cp$, respectively. Yeast and fungi were not nearly detected during storage time for both yogurts.
This studies were investigated the occurrence of sclerotinia rot at the crisphead lettuce field in Uiryeong-Gun, Gyeongsangnam-Do from January to May in 2003. Average incidence rates of sclerotinia rot on crisphead lettuce was up to 21.9% at the five plastic houses. A total of 140 isolates of Sclerotinia sp. were obtained from diseased leaves of crisphead lettuce. Among them, the fungi YR-1 was isolated, which showed highly virulent on the whole plant. the YR-1 was identified as Sclerotinia sclerotiorum based on the formation, color, shape and size of sclerotium and apothecium. For the pathogenicity test, the most suitable inoculum quantity of YR-1 strain was selected as the triturated mycelial suspension of $A_{550}$=0.8, 40 ml showing disease incidence of 94%, and the symptom showed as same as at the fields, the leaves and stem had rotten and developed white downy mycelial at the diseased lesion on the leaves and stems, and produced black and irregular sclerotinia. This is the first report on the pathogenicity test using by triturated mycelial suspension-inoculum of the pathogen for the sclerotinia rot of crisphead lettuce.
