A Suggested Air Sampling Strategy for Bioaerosols in Daycare Center Settings (어린이활동공간에서의 바이오에어로졸 포집 전략)

  • Objectives: We aimed to compare the sampling performance of different flow-based impactor samplers for collecting fungal spores and bacteria and to explore the association of the level of bioaerosols with activity patterns of occupants in daycare center settings. Methods: For comparison of sampling performance, two different flow-based samplers (greater than 100 L/min or not) were selected; a low flow-based sampler (one-stage Andersen sampler) and two high flow-based samplers (DUO SAS SUPER 360 sampler, BUCK bio-culture sampler). We collected airborne mold and bacteria in 30 daycare centers with various levels of contaminated air. Three repeat samplings per each sampler were performed. Mold and bacteria were grown for 96 hours at $25{\pm}1^{\circ}C$ and 48 hours at $35{\pm}1^{\circ}C$, respectively. The Andersen and SAS samplers were used for investigating the association between the level of bioaerosols and the activity patterns of occupants in daycares. Particular matters 10($PM_{10}$), temperature, and relative humidity were monitored as well. Samplings were carried out with one-hour interval from 9 to 5 O'clock. For statistical comparisons, Kruskal-Wallis test, Wilcoxon's signed rank test, and multiple regression analysis were carried out. Results: The airborne level of molds by the low flow-based sampler were significantly higher than that of high flow-based samplers (indoor, P=0.037; outdoor, P=0.041). However, no statistical difference was observed in the airborne level of bacteria by each sampler. Also the level of bioaerosols varied by the time, particularly with different activity patterns in daycare centers. The higher level of mold and bacteria were observed in play time in indoor. Similarly, the concentrations of $PM_{10}$ were significantly associated with the level of bioaerosols (P<0.05). Conclusions: Our findings indicate that the flow rate of sampler, rather than total air volume, could be able to affect the results of sampling. Also, the level of airborne mold and bacteria vary behavior patterns of occupants in indoor of daycare settings. Therefore, different samplers with other flow rate may be selected for mold or bacteria sampling, and activity patterns should be considered for bioaerosol sampling as well.

Characterization of Microorganisms in Eoyukjang (어육장의 미생물학적 특성)

  • Fermented soybean foods are an important component of the Korean diet. Eoyukjang is a type of traditional fermented soybean source. Microbial analysis of eoyukjang was conducted during the fermentation period in this study. Microorganisms isolated from eoyukjang were identified by biochemical tests and 16S rDNA sequencing. 17 different microorganisms, including bacteria, yeast, and fungi were detected in eoyukjang during the fermentation period. Even though Aspergillus participated in the early stage of fermentation of eoyukjang, Bacillus species and Saccharomyces cerevisiae were the major microzymes in eoyukjang throughout the maturation period. Eoyukjang is generally consumed after the boiling of the final sample. Therefore, the final sample of eoyukjang was boiled and analyzed. Our results showed that no vegetative microorganisms survived under the boiling conditions for eoyukjang. Fermented soybean products in the domestic market were also assessed for comparison with the results from eoyukjang. The total cell number of kanjang (soy sauce) samples was between 0 to 42 CFU/mL. The isolated microorganisms were identified as Bacillus species. All Bacillus isolates were not found to harbor the three enterotoxin-producing and emetic toxin-producing genes.

Effect of Storage Temperature on the Physicochemical Characteristics in Kochujang(Red Pepper Soybean Paste) (저장온도에 따른 고추장의 품질변화)

  • To predict quality change of Kochujang distributed in the market, physicochemical properties were observed during storage at $13^{\circ}C$, $27^{\circ}C$ and $37^{\circ}C$ for 240 days. Moisture, crude protein and capsaicin contents were nearly constant for storage at the selected temperatures. Amino nitrogen, value of surface color and pH were decreased during storage while ammonia nitrogen and titratable acidity were increased. Storage temperature affected quality change significantly as higher temperature showed clear increase or decrease phenomena of above factors. Number of total cell count was not changed significantly, and fungi was not detected. From the correlation coefficient among physicochemical properties and sensory evaluation scores, the highest correlation was obtained in amino nitrogen content and sensory score. Degradation of amino nitrogen was a first order reaction, and the $Q_{10}$ value calculated from reaction constant was 2.98. Also, activation energy for the destruction of amino nitrogen calculated from Arrhenius equation was 15.34 Kcal/mole.

Occurrence of Stem Canker and Tuber Rot on Yam Caused by Rhizoctonia solani AG 2-2IIIB in Korea (Rhizoctonia solani AG 2-2IIIB에 의한 마 뿌리썩음병의 한국 내 발생)

  • Stem canker and tuber rot symptoms were observed on yam grown in Andong and Jinju, Korea in 2011. A total of 20 isolates of Rhizoctonia and allied fungi were obtained from the symptomatic plants. Among the isolates, 8 isolates were identified as Rhizoctonia solani and 12 isolates as Ceratobasidium sp. based on rDNA-internal transcribed spacer (ITS) sequence similarity. In the cluster analysis of rDNA-ITS sequences, 7 isolates of R. solani belonged to AG 2-2IIIB and remaining one to AG 1-1A. In addition, among the 12 isolates of Ceratobasidium sp., 7 isolates belonged to AG-Fa, three isolates to AG-A and the other two isolates to AG-Fb and AG-O, respectively. Pathogenicity tests showed that all the R. solani AG 2-2IIIB isolates are pathogenic on stem and tuber of yam but R. solani AG 1-1A and all the Ceratobasidium isolates are non-pathogenic. The results indicate that R. solani AG 2-2IIIB is an important pathogen causing stem canker and tuber rot on yams grown in the study areas. This is the first report of R. solani AG 2-2IIIB causing stem canker and tuber rot of yam in Korea.

Effects of Application Rates with Swine Liquid Manure on Rice Yield and Quality in Cheorwon Region (철원지역에서 가축분뇨 발효액비 시용수준이 벼의 수량과 품질에 미치는 영향)

  • This study was carried out to determine the effect of swine liquid manure on the rice growth and yield in 2006. Field experiment was conducted under variable application rates; 100%, 120%, 150%, 180% N level of slurry based on 11 kg N 10a recommended amount of nitrogen. The experimental sites were located at Cheorwon in Gangwondo area. The results were summerized as follows; In the plot treated with swine liquid manure of 150% and 180% N levels, the plant height and tillers were higher, and the color of leaf was darker than that of 100% N level of swine liquid manure. Rice yield in the plot applied with 120% N slurry level was increased at 11%, but those of 150%, 180% N-level application plots were reduced 10, 19% compared to 100% N level, respectively. Rice quality of the 100 and 120% application plots of swine liquid manure was significantly better than those of 150 and 180% levels of application plots. Total nitrogen content in rice plant after harvesting was increased with increasing levels of swine liquid manure. The content of K in the soil was accumulated in plot treated with 150%, 180% slurry compared to control plot. The heavy metal contents in soils were not increased treated with swine liquid manure. The density of bacteria was low in the application plot of 180%N of liquid swine manure. The bacteria/fungi ratio was highest in 120% N level of liquid manure treatment.

Preservatory effect of stored 'Setoka'(Citrus sp.) using the noncontacted low temperature atmospheric pressure surface discharged plasma (비접촉식 저온 대기압 면방전 플라즈마를 이용한 저장 '세토카' 감귤의 선도유지 효과)

  • Activity of the noncontacted low temperature atmospheric pressure surface discharged plasma (LASDP) converts stable gas to ionized gas known as discharge or plasma. This ionized gas exhibits the antimicrobial activity. We examined the effects of 3 different storage treatments for 80 days on 'Setoka' : ambient storage (AS), low-tempperature storage (LTS), and low-temperature atmospheric pressure plasma+low-tempperature storage (PLTS). Total soluble solids showed no the significant differences between the 3 treatments. Acidity gradually decreased, and was 0.5% under AS after 30 days of storage. Fruit firmness increased by a few percent until 40 days of storage. Weight loss in AS was higher than for other treatments. After 80 days of storage, the decay ratio was significantly low in PLTS treatment: (AS, 50.5%; LTS, 5.6%; PLTS, 1.9%). In AS treatment, 73% of the rotten fruits were infected particularly with green and blue mold; however, only 1% of the rotten fruits were infected in case of PLTS treatment. In conclusion, LASDP treatment can prevent postharvest decay caused by fungi and is an efficacious alternative extending the shelf-life of citrus fruits.

Interaction between the Rice Pathogens, Fusarium graminearum and Burkholderia glumae

  • Species belonging to the genus Fusarium are widely distributed and cause diseases in many plants. Isolation of fungal strains from air or cereals is necessary for disease forecasting, disease diagnosis, and population genetics [1]. Previously we showed that Fusarium species are resistant to toxoflavin produced by the bacterial rice pathogen Burkholderia glumae while other fungal genera are sensitive to the toxin, resulting in the development of a selective medium for Fusarium species using toxoflavin [2]. In this study, we have tried to elucidate the resistant mechanism of F. graminearum against toxoflavin and interaction between the two pathogens in nature. To test whether B. glumae affects the development of F. graminearum, the wild-type F. graminearum strains were incubated with either the bacterial strain or supernatant of the bacterial culture. Both conditions increased the conidial production five times more than when the fungus was incubated alone. While co-incubation resulted in dramatic increase of conidial production, conidia germination delayed by either the bacterial strain or supernatant. These results suggest that certain factors produced by B. glumae induce conidial production and delay conidial germination in F. graminearum. To identify genes related to toxoflavin resistance in F. graminearum, we screened the transcriptional factor mutant library previously generated in F. graminearum [3] and identified one mutant that is sensitive to toxoflavin. We analyzed transcriptomes of the wild-type strain and the mutant strain under either absence or presence of toxoflavin through RNAseq. Expression level of total genes of 13,820 was measured by reads per kilobase per million mapped reads (RPKM). Under the criteria with more than two-fold changes, 1,440 genes were upregulated and 1,267 genes were down-regulated in wild-type strain than mutant strain in response to toxoflavin treatment. A comparison of gene expression profiling between the wild type and mutant through gene ontology analysis showed that genes related to metabolic process and oxidation-reduction process were highly enriched in the mutant strain. The data analyses will focus on elucidating the resistance mechanism of F. graminearum against toxoflavin and the interaction between the two pathogens in rice. Further evolutionary history will be traced through figuring out the gene function in populations and in other filamentous fungi.

Optimization of monitoring methods for air-borne bacteria in the environmental conditions of pig facilities (무균 돈사 환경 모니터링을 위한 대기 중 미생물 탐지기법 확립)

  • Experimental animals have been used to biological and medical purposes and the animals must be, for these purposes, healthy and clean to microbial infection. However, the animals can be easily exposed to pathogenic microorganism via several routes. Of the routes, environmental conditions are the most important factors to keep the animals healthy and clean, especially air condition. Monitoring of air-condition has been required to keep the animal healthy and clean. However, any guideline is not available for experimental conditions with pigs. Therefore, the sampling times and points were compared in different conditions to establish an optimal protocol for monitoring of air borne bacteria. Tryptic soy agar(TSA), blood agar containing 5% defibrinated sheep blood and Sabraud dextrose agar(SDA) were used as media to capture total bacteria, pathogenic bacteria and fungi, respectively. Two methods, compulsive capture using an air-sampler and capturing fall-down bacteria were used to capture the microorganisms in the air. The points and time of capturing were different at each experiment. Air borne microorganisms were captured at three and five points in the open and closed equipments, respectively. Air was collected using an air-sampler for 1 min and 5 min and the agar plates as open status were left from 30 min to 2hr. At first, we monitored an experimental laboratory which dealt with several pathogenic bacteria and then, a protocol obtained from the investigation was applied to open or close experimental conditions with pigs. Number of bacteria was high from 10:00 to 15:00, especially on 13:30-15:30 but sharply decreased after 17:00. The tendency of the number of bacteria was similar between two methods even though the absolute number was higher with air sampler. Critical difference in the number of cells was observed at 5 min with air sampler and 2 hr with fall-down capturing method. However, 1 min with air sampler and 1 hr with fall-down capturing were the best condition to identify bacterial species collected from the air. Number of bacteria were different depending on the sampling points in closed condition but not in opened condition. Based on our results, a guide-line was suggested for screening air-borne microorganism in the experimental conditions with pigs.

An Etiologic Study of Rabbit Dermatitis at Large Rabbit Farms in South Korea (집단 사육 되는 토끼에서 호발하는 피부병에 관한 병인론적 연구)

  • This study was carried out between August and September 2007 to determine the causative agents and epidemiologic features of rabbit dermatitis in Korea. Rabbits were shipped to the laboratory in the College of Veterinary Medicine from 10 rabbit farms. A total of 520 hair, blood, and skin specimens collected from skin lesions of 40 rabbits with suspected dermatopathy were examined mycologically, bacteriologically, and parasitologically. The positive rates of dermatophytosis, bacterial skin dermatitis, and ectoparasite dermatitis were 95, 92.5, and 7.5%, respectively. The etiologic agents of dermatophytosis were identified as Trichophyton mentagrophyte (95%), non-dermatophytic filamentous fungi such as Aspergillus s(5%), and Cryptococcus humilocus (2.5%). With respect to bacteria-related skin dermatitis, Staphylococcus coagulase negative was the most common etiological agent. Staphylococcus aureus was the second most frequent causative agent. Most of the pathogenic isolates were resistant to tetracycline, and aminoglycosides such as amikacin and gentamicin were the most effective drugs against the pathologic bacteria isolated. Ectoparasites were rarely detected in this study. Only Psoroptes cuniculis was detected in 3 (7.5%) out of the 40 tested rabbits. The role of ectoparasites as a causative agent of dermatitis in rabbits in this study was minimal. Our results provide important information related to rabbit dermatitis treatments and researches.

Assessment of the Quality Characteristics of Mixed-grain Nuruk Made with Different Fungal Strains (곰팡이 균종을 달리하여 제조한 혼합 곡류 누룩의 품질특성)

  • Nuruk is a fermented ingredient used for production of traditional Korean rice wine. In this study, quality characteristics of mixed-grain nuruk was made by brewing with different fungal strains was analyzed. Quality elements including enzyme activity and organic acids constituents were measured. The fermentation time of the nuruk did not make a significant difference in terms of its pH, but the acidity and amino acid content for nuruk made from a mixture of two fungal strains was higher than that seen with a single fungal strain. Overall, the enzyme activity for two fungal strain nuruk was higher than that observed for single fungal strain nuruk, with ${\alpha}$-amylase and acidic protease activity in the mixed strain nuruk observed to be more than twice that of the single strain. The major organic acids observed in the manufactured nuruk were identified as acetic, citric, formic, fumaric, lactic, malic and oxalic acids. The total amount of organic acids contained in the nuruk made with the two fungal strain was (2,116.3 mg%). The fungal strains used were A. kawachii SC60 nuruk (1,608.5 mg%) and A. oryzae RIB1353 nuruk (1,146.7 mg%).
