Effects of Sediment Harvesting on Bacterial Community Structure (골재채취가 세균군집구조에 미치는 영향)

  • The dynamics of bacterial populations belonging to $\alpha\;\beta\;\gamma-subclass$ proteobacteria, Cytophaga-Flavobacterium (CF) group and sulfate reducing bacteria (SRB) in water column of the middle reaches of Nakdong River depending on sediment harvesting were analyzed by fluorescent in situ hybridization (FISH) at sediment harvesting site (near the Seongju bridge) and non-sediment harvesting site (near the Gumi bridge). In addition, some physico-chemical parameters such as temperature, pH, $chi-\alpha$ and electrical conductivity were measured. Regarding the number of total cell counts, cells stained by DAPI, there were no substantial quantitative differences between both sites, but those fluctuation at sediment Harvesting site was greater. And also the ratios of CFgroup and SRB to total cell counts tend to increase at sediment harvesting site with higher $chl-\alpha$, maybe due to the resuspension of sediment into water column. But the total proportion of all determined bacterial populations to total cell counts were greater at non-sediment harvesting site, compared with those at sediment harvesting site. Since the detectibility of bacteria by FISH depends on their metabolic activity, those lower proportion at the sediment harvesting site implies that sediment harvesting may lead to malfunction of those bacteria respect to nutrient recycling and subsequently negative effects on microbial food web.

Dynamics of Total Phosphorus and Attached Bacteria in a Porous Medium Concentrating Nutrients from Low-Nutrient Water (저농도 영양염류를 농축하는 여재에서 총인과 부착세균의 변화)

  • A nutrient-concentrating system was operated to retrieve total phosphorus efficiently from a non-point pollution source. Attached bacteria were expected to play an important role in the system. Phosphorous was concentrated by formation of bacterial biofilms on rubberized coconut fiber media of the system. While concentration of total phosphorus (TP) ranged merely 0.12~0.35 mg/L in the stream water, TP levels in pore water and the media were 0.45~0.86 mg/L and 40.91~242.71 mg/kg, respectively. Total bacterial number (TBN) ranged $0.3\sim2.3\times10^6$ cells/ml in stream water, $0.4\sim4.4\times10^6$ cells/ml in pore water and $0.8\sim1.9\times10^9$ cells/g in media. There was a close correlation between TP and TBN. Based on band profiles in DGGE analyses, bacterial communities in the media were different from that in the stream water. Clostridium spp. were abundant in the stream water while Aquabacterium spp. were dominant species in early stages of biofilm formation in the media. The genera predominant in matured biofilms of the media were Clostridium and Enterococcus.

Changes in Microbiological and Chemical Properties of Natural Water with the Storage Time and Temperature (시판 먹는 샘물의 저장 기간 및 온도에 따른 세균학적 및 화학적 품질 변화)

  • This study was carried out to investigate the microbiological and chemical properties of natural water during storage. The water samples were taken at the time of purchase and the opened bottles and unopened bottles stored at the temperature of 4$^{\circ}C$, 18$^{\circ}C$, and 3$0^{\circ}C$. The bacterial content normally rose to 2.06$\times$102 CFU/$m\ell$ for the unopened bottles and 2.91$\times$102 CFU/$m\ell$ for the opened bottles after 2 weeks of storage, and 1.21$\times$102 CFU/$m\ell$ and 2.64$\times$102, respectively, after 24 weeks of storage. The number of viable cells of bacteria peaked more rapidly at the storage temperature of 3$0^{\circ}C$ than 18$^{\circ}C$. But the total samples were found to be negative for coliforms test during the study period. The average range of pH value was from 7.39 to 7.76. The results showed that the nitrates and chlorides satisfied the Korea Drinking Water Quality Standards during the storage period of 24 weeks. However, the undesirable changes of the taste and odor were detected within 2 weeks and 3 weeks, respectively.

The Effect of Blanching and Lactic Acid Bacterial Inoculation on the Quality of Kimchi (열처리 및 젖산균 접종이 김치의 품질에 미치는 영향)

  • The object of this study was to investigate the effects of blanching and lactic acid bacterial inoculation on the quality of kimchi. The pHs of the group added Leuconostoc mesenteroides were rapidly decreased, and then kept almost steady states. However, the pHs of the groups added Bifidobacterium bifidum were gradually decreased. Blanching treatment reduced the number of viable cells. At the beginning of the fermentation, the total organic acid contents of the blanched groups were lower than those of the non-blanched groups, but later on they were higher. With fermentation, the contents of malic, citric and fumaric acid were decreased in the control group, but increased in the cultured groups and all blanched groups. The cutting forces of the blanched groups were higher than those of the non-blanched groups during the whole fermentation period. The inoculation of Leu. mesenteroides was effective on the preservation of ascorbic acid. Blanching and the inoculation of Leu. mesenteroides gave good effect on the sensory acceptability. The acceptability of the groups added Bifidobacterium bifidum was low in initial fermentation period, but increased during the late fermentation period.

Blending Three Probiotics Alleviates Loperamide-Induced Constipation in Sprague-Dawley (SD)-Rats

  • BIOVITA 3 bacterial species (BIOVITA 3), a probiotic blend powder containing Clostridium butyricum IDCC 1301, Weizmannia coagulans IDCC 1201, and Bacillus subtilis IDCC 1101, has been used as a food ingredient for gut health. However, its efficacy in improving constipation has not been reported. Therefore, we aimed to investigate the functional effects of oral administration of BIOVITA 3 as well as its component strains alone (at 1.0×109 CFU/day) in Sprague-Dawley (SD) rats with loperamide-induced constipation. The study included fecal analysis, gastrointestinal transit ratio, histopathological analysis, short chain fatty acids (SCFAs), and metagenome analysis. As results, the BIOVITA 3 group showed significant improvements in fecal number, water content, gastrointestinal transit ratio, and thickening of the mucosal layer. In the SCFAs analysis, all probiotic-treated groups showed an increase in total SCFAs compared to the loperamide-constipated group. Changes in microbial abundance and the diversity index of three groups (normal, constipated, and BIOVITA 3) were also defined. Of these, the BIOVITA 3 showed a significant improvement in loperamide-constipated SD-rats. This study suggests the possibility that BIOVITA 3 can be applied as an ingredient in functional foods to relieve constipation.

Fine mapping of rice bacterial leaf blight resistance loci to major Korean races of Xoo (Xanthomonas oryzae)

  • Bacterial leaf blight(BLB), caused by X. oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice due to its high epidemic potential. Understanding BLB resistance at a genetic level is important to further improve the rice breeding that provides one of the best approaches to control BLB disease. In the present investigation, a collection of 192 accessions was used in the genome-wide association study (GWAS) for BLB resistance loci against four Korean races of Xoo that were represented by the prevailing BLB isolates under Xoo differential system. A total of 192 accessions of rice germplasm were selected on the basis of the bioassay using four isolated races of Xoo such as K1, K2, K3 and K3a. The selected accessions was used to prepare 384-plex genotyping by sequencing (GBS) libraries and Illumina HiSeq 2000 paired- end read was used for GBS sequencing. GWAS was conducted using T ASSEL 5.0. The T ASSEL program uses a mixed linear model (MLM). T he results of the bioassay using a selected set of 192 accessions showed that a large number of accessions (93.75%) were resistant to K1 race, while the least number of accessions (34.37%) resisted K3a race. For races K2 and K3, the resistant germplasm proportion remained between 66.67 to 70.83%. T he genotypic data produced SNP matrix for a total of 293,379 SNPs. After imputation the missing data was removed, which exhibited 34,724 SNPs for association analysis. GWAS results showed strong signals of association at a threshold of [-log10(P-value)] more than5 (K1 and K2) and more than4 (K3 and K3a) for nine of the 39 SNPs, which are plausible candidate loci of resistance genes. T hese SNP loci were positioned on rice chromosome 2, 9, and 11 for K1 and K2 races, whereas on chromosome 4, 6, 11, and 12 for K3 and K3a races. The significant loci detected have also been illustrated, NBS-LRR type disease resistance protein, SNARE domain containing protein, Histone deacetylase 19, NADP-dependent oxidoreductase, and other expressed and unknown proteins. Our results provide a better understanding of the distribution of genetic variation of BLB resistance to Korean pathogen races and breeding of resistant rice.

Comparison of the Bacterial and Fungal Colonies from Rana dybowskii which Collected from Inside and Outside Frog Farms and Identification of the Bacteria from the Tadpoles (개구리 증양식장 내·외부에서 채집된 북방산개구리(Rana dybowskii)로부터 검출된 세균과 곰팡이 콜로니 수의 비교 및 유생으로부터 확인된 세균 규명)

  • There are many ongoing studies of infectious diseases as the major factor responsible for global declining of the amphibian population. Although some point out the amphibian rearing facilities like frog farms as one of the important sources of harboring and spreading amphibian infectious pathogens in the wild, there have been few related studies in South Korea. In this study, we investigated the bacterial and fungal colonies on the skin and in the internal organs of frogs and tadpoles collected inside and outside of Dybowski's brown frog farms in Inje, Goesan, and Gongju to compare the difference according to the region and between inside and outside the farm. We also intended to classify the bacteria collected from the tadpoles into species by analyzing 16s rDNA gene sequences. The result showed that the number of bacterial colonies found in the skin and gut of frogs and the number of fungal colonies found in the skin and liver of frogs collected in Goesan was significantly greater than those in the frogs in Inje. However, there was no difference between the frogs collected inside and outside of farms in both regions. In the case of tadpoles, the number of fungal colonies in the tadpoles collected from Gongju was greater than that in the tadpoles collected from Inje. The comparison of inside and outside frog farms showed that there were more bacterial colonies on the skin of the tadpoles collected from inside than outside the frog farm in Inje and more bacterial colonies in the organs of the tadpoles collected from outside than inside the farm in Gongju. The frogs with higher condition factor (body weight/snout-vent length*100) showed fewer bacterial colonies on the skin and fewer fungal colonies in the heart, but there were no significant relationships in tadpoles. We identified the total of 15 genera and four phyla of bacteria, but the difference according to regions and between inside and outside farm was not evident. The result of this study indicates that the different conditions according to the locality of farm and between inside and outside farm cause the difference in the population sizes of bacterial and fungal colonies and that it can affect the overall health condition of Dybowski's brown frogs in the farm. Moreover, the result suggests that effective disease control in the facility is greatly necessary to ensure successful operation of amphibian rearing facility and to prevent the possible spread of diseases from the facility to the wild.

Quantitative analysis of oral disease-causing bacteria in saliva among bacterial culture, SYBRgreen qPCR and MRT-PCR method (타액내 구강질환 원인 균의 세균배양법, SYBR green qPCR법, MRT-PCR법 간의 정량분석)

  • Objectives: The purpose of this study was to compare SYBR Green qPCR, TaqMan, and bacterial selective medium cultures for accurate quantitative analysis of oral microorganisms. Methods: The SYBR Green method is widely used to analyze the total amount of oral microorganisms in oral saliva. However, in this study, MTR-PCR method based on TaqMan method was performed using newly developed primers and probes. In addition, it was designed to confirm the detection agreement of bacteria among bacteria detection method. Results: As a result of MRT-PCR and SYBR Green qPCR analysis, more than 40 times (0.9-362.9 times) bacterium was detected by MRT-PCR. In addition, more bacteria were detected in saliva in the order of MRT-PCR, SYBR Green qPCR, and bacterium culture, and the results of MRB-PCR and SYBR Green qPCR showed the highest agreement. The agreement between the three methods for detecting P. intermedia was similar between 71.4 and 88.6%, but the agreement between MRT-PCR and SYBR Green qPCR was 80% for S. mutans. Among them, the number of total bacteria, P. intermedia and S. mutans bacteria in saliva was higher than that of SYBR Green qPCR method, and bacterium culture method by MRT-PCR method. P. intermedia and S. mutans in saliva were detected by MRT-PCR and MRT-PCR in 88.6% of cases, followed by the SYBR Green qPCR method (80.0%). Conclusions: The SYBR Green qPCR method is the same molecular biology method, but it can not analyze the germs at the same time. Bacterial culturing takes a lot of time if there is no selective culture medium. Therefore, the MRT-PCR method using newly developed primers and probes is considered to be the best method.

Long-Term Starvation Induces the Viable-but-Nonculturable Condition in Lactobacillus crispatus KLB46

  • In a previous study, we have isolated a number of lactobacilli from Korean women, and one of them (KLB46) was identified as Lactobacillus crispatus by 16S rRNA gene sequencing. For the ecological treatment of bacterial vaginosis (BV) cell suspension of L. crispatus KLB46 was instillated into BV patients. L. crispatus KLB46 was found to persist for several days in cell suspension with no nutrients. In this study, in order to assess the influence of starvation on physiological activity, we compared the viability and culturability of KLB46 following suspension in various buffer solutions. A pair of in situ fluorescent dye was used to assess viability (i.e. membrane integrity) and the culturability was examined by plate count assay. A rapid epifluorescence staining method using the LIVE/DEAD Bacterial Viability Kit $(BacLight^{TM})$ was applied to estimate both viable and total counts of bacteria in cell suspension. $BacLight^{TM}$ is composed of two nucleic acid-binding stains ($SYTO\;9^{TM}$ and propidium iodide). $SYTO\;9^{TM}$ penetrates all bacterial membranes and stains the cells green while propidium iodide only penetrates cells with damaged membranes, therefore the combination of the two stains produces red fluorescing cells. Optimal staining conditions for $BacLight^{TM}$ were found to be with 0.0835M $SYTO\;9^{TM}$ and 0.05M propidium iodide for 15 min incubation at room temperature in dark. When cells were microscopically examined during 140 hours of starvation, the culturability decreased markedly while the viability remained relatively constant, which suggests that large fraction of KLB46 cells became viable but non-culturable (VBNC) upon starvation.

STUDIES ON SANITARY QUALITY OF JARHI-DOM CHROMIS NOTATUS (자리돔의 식품위생학적 연구)

  • This study was carried out to evaluate the sanitary quality of Jarhi-dom, Chromis notatus, and its products, during the period from May to October, 1975. And bacterial density of the sea water in the estuary of Sogwipo was compared. The number of general bacteria and coliform group in the sea water appeared lower than that of other areas. Both Jarhimuchim-hoe and Jarhimul-hoe showed higher bacterial densities than the fresh fish when caught and sold in the market although Jarhimuchim-hoe showed slightly higher density than Jarhimul-hoe. Vibrio parahaemolyticus was detected in some samples of sea water and Jarhi-dom, but it was not regarded for food poisoning. Bacterial densities in total plate counts, coliform group and Vibrio parahaemolyticus was highest in August and September.
