Effect of Thermophilic Ammonium Tolerant Bacteria on Malodors Emission of Composting of Pig Manure (돈분 퇴비화 과정중 악취물질에 대한 고온성 암모니움 내성균 접종 효과)

  • In order to investigate microbiological control of malodors, particularly including ammonia, the effect of three thermophilic ammonium tolerant bacteria strains. TAT112. TAT117 and TAT119, were tested during composting of pig manure in the laboratory scale composters. The total weight, volatile solids and BOD of the pig manure compost were decreased during composting process in all treatments. The temperature in all treatments rose in first 3 days dramatically, but that in control without inoculation reached its maximum most lately among the treatments. The nitrogen content of drain water accumulated inside and outside composter, and trapped in 6N $H_2SO_4$ was lower in TAT112 inoculated composter than in control. However, it was not lower in the treatment of TAT117 and TAT119 inoculated. Ammonia concentration in the exhaust gas monitored everyday during composting also demonstrated that it was lowest at TAT112 inoculated among all treatments. It was appeared to have an effect on reducing ammonia emission at the treatment of TAT112 inoculated than the control.

A Field Research on Mud Flat Remediation by Biological Treatments (생물학적 처리에 따른 갯벌 복원을 위한 현장 적용성 연구)

  • A field test on mud flat remediation was carried out in order to observe the effects of the treatments such as microbial dose and an oxygen releasing compound like $CaO_2$. The size of each treatment site was $100m^2$ and the dosage was 3.6 kg per site. The 6 week monitoring showed that pH on two sites was below 7 and ORP increased from .178~-188 mV to .121~-142 mV. In Ignition loss and COD there were no significant changes. Meanwhile nitrogen and phosphorus concentrations changed: ammonia concentration decreased both on control and treatment sites. Nitrate nitrogen decreased more on combined treatment site than on single microbial treatment (11.3% vs. 7.3%) probably because the extra oxygen supplied by $CaO_2$ formed more oxic environment so that the facilitated nitrification might produce more nitrate but the nitrate would be much rapidly released into the water layer out of the sediment. That also explains the total nitrogen reduction(6.1%). Similarly, T-P and $PO_4-P$ reduced by 29% and 31.8%, respectively on combined treatment sites, resulting from the phosphorus release effect though the initial concentrations of the two factors were considerably high.

Determination of the Origin of Particulate Organic Matter at the Estuary of Youngsan River using Stable Isotope Ratios (${\delta}^{13}C$, ${\delta}^{15}N$) (탄소 및 질소 안정동위원소 비를 이용한 영산강하구역 유기물 기원 추정 연구)

  • Organic carbon and total nitrogen stable isotope ratios of suspended materials were determined at 3 stations (from Mongtan Bridge to Youngsan river estuary barrage) (n=6, each) from November 2008 to August 2009, in order to understand the origin of particulate organic matter at the Estuary of Youngsan River. Allochthonous organic matter, ammonia-N and silicate were increased after heavy rain (in August). Carbon isotope ratios were significantly different between stations in November and August, and it was possible to determine the origin of organic matter. The heavier nitrogen isotope ratios, as well as higher phosphate concentrations, were found in November than other sampling times. Livestock wastewater and farmland input was likely the main causes of these high values. In addition, YS3 station, the nearest site to estuary barrage, appears to be affected by a substantial amount of livestock wastewater and farmland input, considering that nitrogen isotope ratios were heavier than those at the upper sites. These results suggest that the analysis of stable isotope ratios is a simple but useful tool for the determination of organic matter origin in aquatic environments.

Eutrophication in the Downstream of Nakdong River (낙동강 하류역의 부영양화현상에 관한 연구)

  • A study on eutrophication in the downstream of the Nakdong River was conducted from October 1983 to September 1984. Nutrients concentration of the samples in this area was usually high and the nitrogen ratio to the phosphorus was as very high as $63.5{\sim}119.6$. The concentration of nutrients was in the range of $0.22{\sim}173{\mu}g-at/l$ for ammonia nitrogen, $26.7{\sim}187{\mu}g-at/l$ for nitrate nitrogen, $1.07{\sim}8.22{\mu}g-at/l$ for nitrite nitrogen, $58.7{\sim}231{\mu}g-at/l$ for total inorganic nitrogen and $0.44{\sim}4.43{\mu}g-at/l$ for phosphate phosphorus. The concentration of chlorophyll-a was in the range of $1.8{\sim}75.2\;mg/m^3$. Correlationship between concentration of nutrients and chlorophyll-a was not found. The concentrations of total suspended solid were in the range of $10.7{\sim}45.9\;ppm$, and $16.2{\sim}35.2\%$ of the total suspended solid was consisted of volatile suspended solid. According to the state of lacustrine nutrition, the state of the downstream of Nakdong River was equivalent to the eutrophication state in term of chlorophyll-a and inorganic nitrogen, and mid-nutritional state in term of phosphate.

Effects of Feeding Aspergillus oryzae Ferments on Performance, Intestinal Microflora, Blood Serum Components and Environmental Factors in Broiler (Aspergillus oryzae 배양물의 급여가 육계의 생산성, 장내미생물, 혈청성분 및 계사환경 요인에 미치는 영향)

  • Two experiments were carried out to evaluate the effects of feeding Aspergillus oryzae(AO) ferment on performance, intestinal microflora, serum components, ammonia generation and litter dampness in broiler chicks. In experiment I, three hundred sixty, one day old broiler chicks, Abor Acres, were fed 0 and 0.1% of Aspergiilus oryzae short conidia ferment(AOS) and 0.1% of Aspergillus oryzae long conidia ferment(AOL) for five weeks. In experiment II, three hundred sixty, one day old broiler chicks, Abor Acres were fed 0, 0.1 and 0.2% of Aspergillus oryzae long conidia ferment(AOL) for five weeks. In experiment I, growth rates were not statistically different among dietary treatments. AOS and AOL showed increased tendency in weight gain and feed intake compared to those of control, whereas feed conversion was not different. Litter dampness of AOS and AOL was also tended to decrease compared to that of control, but was not significantly different. Fecal ammonia gas generation was decreased in feeding AOS and AOL, and maintained 1/2 to 3/4 compared to the control. In serum metabolites, AOS and AOL increased glucose and calcium, and decreased total protein, blood urea nitrogen and total cholesterol. In experiment II, body weight of chicks fed 0.1 and 0.2% AOL were heavier than the control(P<0.05). Feed intake of chicks fed 0.1 and 0.2% AOL also were higher than the none, but feed conversion ratio was not different among treatments. Ileal and cecal microflora showed increased tendency in lactic acid bacteria compared to those of the control. Salmonella and E. coli were decreased in ileum of chicks fed 0.1 and 0.2% AOL. In conclusion, feeding AO ferment increased growth performance and improved intestinal microflora of broiler chicks and environments of broiler house.

Effects of Corn and Soybean Meal Types on Rumen Fermentation, Nitrogen Metabolism and Productivity in Dairy Cows

  • Twelve multiparous Holstein dairy cows in mid-lactation were selected for a replicated $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement to investigate the effects of corn and soybean meal (SBM) types on rumen fermentation, N metabolism and lactation performance in dairy cows. Two types of corn (dry ground [DGC] and steam-flaked corn [SFC]) and two types of SBM (solvent-extracted and heat-treated SBM) with different ruminal degradation rates and extents were used to formulate four diets with the same basal ingredients. Each period lasted for 21 days, including 14 d for adaptation and 7 d for sample collection. Cows receiving SFC had a lower dry matter (DM) and total N intake than those fed DGC. However, the milk yield and milk protein yield were not influenced by the corn type, resulting in higher feed and N utilization efficiency in SFC-fed cows than those receiving DGC. Ruminal acetate concentrations was greater and total volatile fatty acids concentrations tended to be greater for cows receiving DGC relative to cows fed SFC, but milk fat content was not influenced by corn type. The SFC-fed cows had lower ruminal ammonia-N, less urea N in their blood and milk, and lower fecal N excretion than those on DGC. Compared with solvent-extracted SBM-fed cows, cows receiving heat-treated SBM had lower microbial protein yield in the rumen, but similar total tract apparent nutrient digestibility, N metabolism measurements, and productivity. Excessive supply of metabolizable protein in all diets may have caused the lack of difference in lactation performance between SBM types. Results of the present study indicated that increasing the energy degradability in the rumen could improve feed efficiency, and reduce environmental pollution.

Multivariate Analysis among Leaf/Smoke Components and Sensory Properties about Tobacco Leaves Blending Ratio

  • This study focused on the relationships among leaf and smoke components and sensory properties following tobacco leaf blending. A completely randomized experimental design was used to evaluate components of leaf and smoke and sensory properties for sample cigarettes with four mixtures of flue cured and burley tobacco (40:60, 60:40, 80:20 and 100:0). Eleven leaf components, six smoke components, and eight sensory properties of smoking taste were analyzed. A sensory evaluation method known as quantitative descriptive analysis was used to evaluate perceptual strength on a fifteen score scale. Raw data from ten trained panelists were obtained and statistically analyzed. Based on the MANOVA, clustering analysis, correlation matrix and partial least square (PLS) method were applied to find out which smoke component most affected sensory properties. The PLS method was used to remove the influence between explanatory variables in the leaf, smoke components derived from the results. High correlations (p<0.0l) were found among ten specific leaf and smoke components and sensory attributes. Total nitrogen, ammonia, total volatile base, and nitrate in the leaf were significantly correlated (p<0.05) with impact, bitterness, tobacco taste, irritation, smoke volume, and smoke pungency. From the results of PLS analysis, influence variables are used to explain about the correlation. In terms of bitterness, with only two explanatory variables, Leaf $NO_3$ and Leaf crude fiber were enough for guessing their correlation. In the distance weighted least square fitting analysis, carbon monoxide highly influenced bitterness, hay like taste, and smoke volume.

A Study on the Removal of Pollutants from Wastewater by Aquatic Macrophytes (수생식물에 의한 폐수의 오염물질제거에 관한 연구)

  • Macrophyte plays an important role in purification of wastewater. They have capacity to improve the water quality by absorbing nutrients, with their effective root system. In this study, removal of nutrient as well as organic matter was observed by some important macrophytes i.e. Pistia stratoites, Hydrocharis dubia and Salvinia sp. indepe ndently as well as in mixed culture under the laboratory condition. The highest total nitrogen removal was observed for Pistia stratoites (86.47%) in monoculture and Salvinia sp. + P. stratoites (76.11%) in mixed culture system. Corresponding figures for total phosphorous were observed for P. stratoites (75.60%) in monoculture and Salvinia sp. + P. stratoites (71.11%) in mixed culture system. Similar result was observed for ammonia removal in both systems. Additionally, P. stratoites showed the highest removal of organic matter, in monoculture system (68.46%) where as Salvinia sp. + P. stratoites showed the highest removal of organic matter in mixed culture system (82.73 %).

Development of Appropriate Fibrolytic Enzyme Combination for Maize Stover and Its Effect on Rumen Fermentation in Sheep

  • In vitro studies were undertaken to develop an appropriate fibrolytic enzymes cocktail comprising of cellulase, xylanase and ${\beta}$-D-glucanase for maize stover with an aim to increase its nutrient utilization in sheep. Cellulase and xylanase added individually to ground maize stover at an increasing dose rates (0, 100, 200, 400, 800, 1,600, 3,200, 6,400, 12,800, 25,600, 32,000, 38,400, and 44,800 IU/g DM), increased (p<0.01) the in vitro dry matter digestibility and in vitro sugar release. The doses selected for studying the combination effect of enzymes were 6,400 to 32,000 IU/g of cellulase and 12,800 to 44,800 IU/g of xylanase. At cellulase concentration of 6,400 IU/g, IVDMD % was higher (p<0.01) at higher xylanase doses (25,600 to 44,800 IU/g). While at cellulase doses (12,800 to 32,000 IU/g), IVDMD % was higher at lower xylanase doses (12,800 and 25,600 IU/g) compared to higher xylanase doses (32,000 to 44,800 IU/g). At cellulase concentration of the 6,400 to 32,000 IU/g, the amount of sugar released increased (p<0.01) with increasing levels of xylanase concentrations except for the concentration of 44,800 IU/g. No effect of ${\beta}$-D-glucanase (100 to 300 IU/g) was observed at lower cellulase-xylanase dose (cellulase-xylanase 12,800 to 12,800 IU/g). Based on the IVDMD, the enzyme combination cellulase-xylanase 12,800 to 12,800 IU/g was selected to study its effect on feed intake and rumen fermentation pattern, conducted on 12 rams (6 to 8 months; $20.34{\pm}2.369$ kg body weight) fed 50% maize stover based TMR. The total volatile fatty acids (p<0.01) and ammonia-N concentration was higher in enzyme supplemented group, while no effect was observed on dry matter intake, ruminal pH and total nitrogen concentration.

Effect of Air Distributor Pore Size in Foam Separator of Sea Water (해수의 포말분리시 공기분산기 기공크기 영향)

  • Effect of the air distributor pore size for the removal of aquacultural waste, such as protein, total suspended solids (TSS), chemical oxygen demand (COD), turbidity and total ammonia nitrogen (TAN) from sea water was investigated by using foam separator. With the increase of pore size of air distributor, removal rates and efficiency of protein decreased. Removal rate by commercial air stone was in the range between the removal rates by G2 and G4 sintered glass discs. Within the range of pore size distributor from Gl to G4, the removal efficiency of protein were ranged from 21 to $42\%.$ The changes of removal rates and efficiencies of TSS, COD and turbidity were similar to proteins. TAN was removed by stripping. The pore size of air distributor for a higher overall oxygen mass transfer coefficient and saturation efficiency provided the condition for higher protein removal rate. Also the foam separator could be used as an aerator.
