  • Sequencing Batch Reactor (SBR) was operated under various experimental conditions to improve the efficiency of biological filters used for the treatment of recycled wastewater from aquaculture. This SBR system was operated for removing COD, ammonia and suspended solid that were the major pollutants in aquaculture wastewater. Aerobic and anoxic conditions after FILL mode were applied intermittently for effective removal of nitrogen. SETTLE and DRAW modes were followed by the complete aerobic and anoxic REACT mode. The total volume of the SBR was 75 liter, while the working volume in a cycle was 35 liters. When the final operating strategy of the SBR was FILL/REACT/SETTLE/DRAW of 0.5/10/1/0.5 hr. the removal efficiencies of TCODcr, $NH_{4}^{+}-N,$ and T-N were 94, 98, and $89\%,$ respectively.

Effect of Crude Protein Levels in Concentrate and Concentrate Levels in Diet on In vitro Fermentation

  • The effect of concentrate mixtures with crude protein (CP) levels 10%, 13%, 16%, and 19% and diets with roughage to concentrate ratios 80:20, 60:40, 40:60, and 20:80 (w/w) were determined on dry matter (DM) and organic matter (OM) digestibility, and fermentation metabolites using an in vitro fermentation technique. In vitro fermented attributes were measured after 4, 24, and 48 h of incubation respectively. The digestibility of DM and OM, and total volatile fatty acid (VFA) increased whereas pH decreased with the increased amount of concentrate in the diet (p<0.001), however CP levels of concentrate did not have any influence on these attributes. Gas production reduced with increased CP levels, while it increased with increasing concentrate levels. Ammonia nitrogen ($NH_3$-N) concentration and microbial CP production increased significantly (p<0.05) by increasing CP levels and with increasing concentrate levels in diet as well, however, no significant difference was found between 16% and 19% CP levels. Therefore, 16% CP in concentrate and increasing proportion of concentrate up to 80% in diet all had improved digestibility of DM and organic matter, and higher microbial protein production, with improved fermentation characteristics.

오존 전처리 공정이 생물활성탄 공정의 효율에 미치는 영향

  • 본 연구에서는 오존과 생물활성탄을 연계시킨 공정을 통하여 용존 유기물질의 제거 경향을 살펴보았다. 오존 처리를 거치면서 원수중의 난분해성 용존 유기물질과 같이 생물학적 분해 속도가 느린 화합물의 상당량이 저분자 형태로 전환되었음을 파악할 수 있었으며, 오존 처리후의 생분해성의 향상에 의해 흡착의 부담이 경감되어 활성탄의 수명이 연장되는 것으로 조사되었다. 생물활성탄 반응조 내에서 여층 깊이에 따른 DOC 제거 경향을 알아본 결과, 전체 제거량의 약 50%가 column 상단에서 제거되었으며, 따라서 짧은 EBCT 에서도 용존 유기탄소의 제거는 용이한 것을 사료된다. 또한 유기 오염에 대한 지표로서 국내외에서 일반적으로 고도정수처리의 처리 대상물질로 선정되어 있는 암모니아성 질소의 제거는 75.9%로 상당히 높은 제거율을 나타내었다.

Investigation of Dietary Lysophospholipid (LipidolTM) to Improve Nutrients Availability of Diet with In Vitro Rumen Microbial Fermentation Test

  • This study was conducted to investigate the effect of biological membrane transfer modifier, lysophospholipd (LPLs) on the parameters from in vitro rumen simulated fermentation. Commercially available LPLs product (Lipidol$^{TM}$) was supplemented into experimental diets which consisted of orchard grass and concentrate diet (60:40) in different levels (0.1%, 0.3% and 0.5%). Then in vitro rumen simulated fermentation was performed. Although, a declining trend of pH was found in treatments, all pH values were detected in a range relevant to normal rumen fermentation. Gas production, ammonia nitrogen and total VFA production were greatly influenced by the supplementation of LPLs. All parameters were increased along with increased levels of LPLs in diet. As a result, 0.1% of Lipidol$^{TM}$ is recommended based on the determined in vitro rumen fermentative parameters in this study.

Diversity of freshwater red algae at Khao Luang National Park, southern Thailand

  • Freshwater red algal diversity and the relationship with water conditions in 22 stream segments in the area around Khao Luang National Park, Nakhon Si Thammarat province, southern Thailand, were studied during a period of twelve months (May 2014 to April 2015). Sixteen species of freshwater red algae, belonging to eight genera (Audouinella, Balliopsis, Batrachospermum, Caloglossa, Compsopogon, Kumanoa, Sirodotia, and Thorea) were identified, which were all reported earlier for the country. Thorea clavata (Thoreaceae) was the most common species occurring in eight stream segments. Caloglossa beccarii sensu lato (Delesseriaceae) and Sirodotia huillensis Skuja (Batrachospermaceae) had the highest percent cover with up to 40% and 20% per stream segment, respectively. The water quality showed most sites to be unpolluted or ultra-oligotrophic to oligotrophic. Canonical Correspondence Analysis revealed some trends in occurrence of individual species with stream environmental variables: Batrachospermum sp.with strong current velocity; Kumanoa hirosei with high turbidity, total dissolved solid and alkalinity; Caloglossa beccarii with high conductivity; Kumanoa tabagatenensis with high ammonia-nitrogen and Thorea siamensis with high calcium and magnesium. In view of the scarce studies on the stream ecology of freshwater red algae in Philippines and neighbouring countries, it is expected that the data presented here would be helpful in more critical further studies in south-east Asia in general.

연안해수와 지하해수를 사용하는 제주 넙치 양식장의 수질과 미생물 변동 (Investigation of water qualities and microbials on the flow-through olive flounder, Paralichthys olivaceus farms using coastal seawater and underground seawater in Jeju)

  • This study assessed the levels of water qualities and microbials contamination of inland olive flounder farms in Jeju in the summers from 2015 to 2017. Three farms (A-C) located in a concentrated area using mixing coastal seawater and underground seawater and one farm (D) located in an independent area using only coastal seawater were selected. Total ammonia nitrogen (TAN) reached a maximum of 0.898 ± 1.024 mg/L as N in the coastal seawater of A-C, which was close to the limit of the water quality management goal of the fish farm. TAN in the influent from A-C was up to three times higher than that of D, so that the discharged water did not spread to a wide range area along the coast and continued to affect the influent. TAN of the effluent in A-C increased by 2.7-4.6 times compared to the influent, resulting in serious self-pollution in the flounder farm. Heterotrophic marine bacteria in the influent of A-C was about 600 times higher than D, and the discharge of A-C was increased by about 30 times compared to the influent.

Effect of the Addition of Antimicrobial Materials Before and After Aging on the Physicochemical Properties of Low-Salt Kochujang during Storage

  • To improve the shelf life of low-salt Kochujang, Korean hot pepper paste, antimicrobial materials were added at different times before and after aging. The kochujang was then packaged and stored at $30^{\circ}C$ for 15 weeks, and changes in microbiological and physiochemical properties were evaluated. Hunter a- and b-values decreased considerably during storage. The total color difference (${\delta}E$) was greater in the ethanol-chitosan (EC) treatment than in the control and after pasteurization (A-P) treatment. Gas was produced until the seventh week of storage. The control and the A-P treatments produced more gas than the other treatments, and these had the largest number of yeasts and aerobic bacteria. The pH of the EC treatment was higher than that of the other treatments, and the A-P treatment had the highest level of titratable acidity. During storage, the oxidation-reduction potential was lower in the EC and ethanol-mustard-chitosan (EMC) treatments. The reducing sugar content decreased remarkably in the control and A-P treatments, with high production of ethanol. There was a significant change in the content of amino-type nitrogen in the control and A-P treatments, and the content of ammonia-type nitrogen was lowest in the EMC treatment. In the sensory test of kochujang, the ethanol-mustard (EM) and ethanol (E) treatments were significantly higher than the EC, control, and A-P treatments (p <0.05). EM or E alone was effective in extending the shelf life of kochujang when added before aging.

Use of Chemical and Biological Agents to Improve Water Quality of Effluent Discharge from Abattoirs

  • Intensive animal industries create large volumes of nutrient rich effluent which, if untreated, has the potential for substantial environmental degradationand to recover valuable nutrients that would otherwise be lost. Members of the family Lemnaceae are widely used in lagoon systems, to achieve inexpensive and efficient remediation of effluent. Only limited research has been conducted into their growth in highly eutrophic media and there has been little done to systematically distinguish between different types of media. This study examined the growth characteristics of duckweed in abattoir effluent and explored possible ways of ameliorating the inhibitory factors to growth on this medium. A series of pot trials was conducted to test the tolerance of duckweed to abattoir effluent partially remediated by a sojourn in anaerobic fermentation ponds, both in its unmodified form and after the addition of acid to manipulate pH, and the addition of bentonite. Unmodified abattoir effluent was highly toxic to duckweed, even at dilutions of 3:1. Duckweed remained viable and grew sub-optimally in simplified media with total ammonia nitrogen (TAN) concentrations of up to 100 mg/L. Duckweed grew vigorously in effluent diluted 1:4 v/v, containing 56 mg TAN/L when modified by addition of acid (to decrease pH to 7) and bentonite at 0.5%. The results of this study suggest that bentonite plays an important role in modifying the toxicity of abattoir effluent to duckweed.

Comparisons of In vitro Nitrate Reduction, Methanogenesis, and Fermentation Acid Profile among Rumen Bacterial, Protozoal and Fungal Fractions

  • The objectives were to compare the ability of various rumen microbial fractions to reduce nitrate and to assess the effect of nitrate on in vitro fermentation characteristics. Physical and chemical methods were used to differentiate the rumen microbial population into the following fractions: whole rumen fluid (WRF), protozoa (Pr), bacteria (Ba), and fungi (Fu). The three nitrogen substrate treatments were as follows: no supplemental nitrogen source, nitrate or urea, with the latter two being isonitrogenous additions. The results showed that during 24 h incubation, WRF, Pr and Ba fractions had an ability to reduce nitrate, and the rate of nitrate disappearance for the Pr fraction was similar to the WRF fraction, while the Ba fraction needed an adaptation period of 12 h before rapid nitrate disappearance. The WRF fraction had the greatest methane ($CH_4$) production and the Pr fraction had the greatest prevailing $H_2$ concentration (p<0.05). Compared to the urea treatment, nitrate diminished net gas and $CH_4$ production during incubation (p<0.05), and ammonia-N ($NH_3$-N) concentration (p<0.01). Nitrate also increased acetate, decreased propionate and decreased butyrate molar proportions (p<0.05). The Pr fraction had the highest acetate to propionate ratio (p<0.05). The Pr fraction as well as the Ba fraction appears to have an important role in nitrate reduction. Nitrate did not consistently alter total VFA concentration, but it did shift the VFA profile to higher acetate, lower propionate and lower butyrate molar proportions, consistent with less $CH_4$ production by all microbial fractions.

Effect of Lactic Acid Producing Bacteria on the Performance of Male Crossbred Calves Fed Roughage Based Diet

  • To study the effect of feeding lactic acid producing bacteria on the performance of cattle calves, twenty four, day old male crossbred cattle calves (Bos indicus${\times}$Bos taurus), were distributed into two groups of 12 animals each. The animals were fed on calf starter containing wheat bran and green berseem ad libitum and milk as per requirement upto 8 weeks of age. The diet of calves of Group 2 was supplemented with 500 ml culture of Lactobacillus acidophilus-15. Total duration of the experiment was 31 weeks. There was no significant difference in intake and digestibility of dry matter (DM), organic matter (OM), neutral detergent fibre (NDF), acid detergent fibre (ADF) and crude protein (CP) between the groups. The rumen pH, protozoa numbers, concentration of volatile fatty acids (VFA), ammonia nitrogen ($NH_3-N$), trichloroacetic acid precipitable nitrogen (TCA-ppt N) and activity of microbial enzymes (carboxymethylcellulase, xylanase, amylase and protease) were not affected due to probiotic supplementation. Average live weight gain of the calves was improved (about 10%) and feed:gain ratio was reduced (about 5%) in the animals given Lactobacillus culture. The data indicated that crossbred calves could be reared on a diet devoid of cereal grain and addition of Lactobacillus culture in the diet resulted in an added advantage in growth performance of the animals.
