Effects of Popped Soybean on Concentration of Ruminal Peptide and Blood Amino Acids in Holstein Calves

  • This study conducted to evaluate effects of popped soybean on levels of ruminal peptides and blood amino acids in Holstein calves fed sudan grass hay as a forage source and popped (PSB) soybean as a concentrate supplement. At 0, 2, 4 and 6 h after feeding, rumen fluid and blood samples were collected from the rumen and jugular vein, respectively, and amino acids, peptides and other nitrogen-containing compounds in the rumen were analyzed. Ruminal pH tended to be higher in the RSB than in the PSB treatments, and declined upto 4 h after feeding, since then increased in both treatments. The concentrations of ammonia-N in all treatments increased upto 2 h after feeding, and then decreased gradually with time after feeding. The concentrations of ammonia N in the rumen were not significantly different between the treatments, however, those in RSB treatment appeared to be higher. Also, protein concentrations in the rumen were not significantly different between the treatments. Peptide productions were the highest at 2 h after feeding in the group fed RSB which is rapidly degradable in rumen, whereas those in the group fed PSB which is slowly degradable in rumen were maximized at 4 h after feeding. The concentration of total free essential amino acids in plasma was higher in the RSB treatment than in the PSB, but disappearance rates of these amino acids out of plasma was higher in the PSB treatment than in the RSB treatment. Disappearance rates of free non-essential amino acids in plasma were not significantly different between the treatments. Consequently, this study implies that the production of peptide and utilization of blood amino acid may be controlled by the modification of protein degradability.

Effect of Encapsulating Nitrate in Sesame Gum on In vitro Rumen Fermentation Parameters

  • Encapsulation is a method used to protect material from certain undesirable environments, for controlled release at a more favorable time and place. Animal productivity would be enhanced if feed additives are delivered to be utilized at their site of action, bypassing the rumen where they are likely to be degraded by microbial action. A novel method of encapsulation with sesame gum was used to coat nitrate, a known enteric methane mitigating agent, and tested for the effect on methane reduction and other in vitro fermentation parameters using rumen fluid from cannulated Hanwoo steers. Orchard grass was used as basal diet for fermentation. The treatments were matrix (1.1 g sesame gum+0.4 g sesame oil cake) only, encapsulated nitrate (matrix+nitrate [21 mM]), free nitrate (21 mM), and a control that contained no additive. Analyses of fermentation parameters were done at 0, 3, 6, 9, 12, 24, and 48 h time periods. In comparison to control, both free and encapsulated nitrate produced significantly reduced (p<0.01) methane (76% less) and also the total volatile fatty acids were reduced. A significantly higher (p<0.01) concentration of ammonia nitrogen was obtained with the encapsulated nitrate treatment (44%) compared to the free form (28%) and matrix only (20%) (p = 0.014). This might suggest slow release of encapsulated nitrate so that it is fully reduced to ammonia. Thus, this pioneering study found a significant reduction in methane production following the use of sesame gum encapsulated nitrate that shows the potential of a controlled release system in enhancing sustainability of ruminant production while reducing/eliminating the risk of nitrite toxicity.

NUTRITIVE VALUE OF SILAGES PREPARED FROM FIBER AND LIQUID RESIDUES AFTER THE SEPARATION OF LEAF NUTRIENT CONCENTRATE FROM ITALIAN RYEGRASS IN GOATS

  • Italian ryegrass (Lolium multiflorum Lam.) was fractionated into leaf nutrient concentrate, fiber and deproteinised juice (DPJ). The fresh and fermented DPJs were concentrated and referred to as fresh deproteinised juice concentrate (FDPJC) and fermented deprotenised juice concentrate (FMTD DPJC). The FDPJC and FMTD DPJC were separately mixed with dried fiber and ensiled. Wilted crop silage and fresh fiber silage were also prepared from the same material crop. The nutritive value of these four silages were compared using four goats by $4{\times}4$ Latin square design. Green crop fractionation resulted lesser amount of crude protein and ash, and higher amount of neurtal detergent fiber, acid detergent fiber, cellulose and hemicellulose in fresh fiber. The pH of fresh fiber silage was lower than that of the other silages. Addition of FDPJC or FMTD DPJC to the dried fiber at ensiling did not improve the silage qualities; but all the silages were satisfactorily preserved. Goats fed these silages showed similar ruminal pH and total volatile fatty acid concentrations. But addition of FMTD DPJC was effective on increasing ruminal acetic acid concentration reducing propionic acid concentration. Ruminal n- and iso- butyric acid concentrations were proportional to that of propionic acid. High ammonia content of the silage containing FMTD DPJC was reflected to the ruminal ammonia concentration, urinary nitrogen excretion and serum urea level of goats. Inclusion of FDPJC or FMTD DPJC added 15 to 25% dry matter to the fiber silages with a little reduction in the digestibilities of most components of the silages.

  • Ginseng (Panax ginseng CA. Meyer) hairy root cultures, which are established via the infection of ginseng root discs with Rhizobium rhizogenes, have been used to construct profiles of both biomass growth and nutrient consumption in flask cultures. In a 250 mL shake flask culture, the maximum biomass was observed on the 59th day of the culture period, at 216.8 g (fresh wt) per liter or 11.4 g (dry wt) per liter. The hairy roots were determined to have a growth rate of 0.355 g-DW/g cells/day during the exponential growth phase and a maximum specific growth rate on day 7. Total ginseng saponin and phenolic compound contents were noted to have increased within the latter portion of the culture period. Linear correlations between increases in biomass weight and nutrient uptake were used to imply the conductivity yield $2.60g-DW/(L{\cdot}mS)$ and carbon yield 0.45 g-DW/(g sugar) in the 250 mL flask cultures. The biomass yield when two different nitrogen sources were used (ammonia and nitrate) was shown to remain approximately constant. at $0.47g-DW/(L{\cdot}mM\;NH_4$) and $0.33g-DW/(L{\cdot}mM\;NO_3$); it remained at these levels for 16 days with the ammonia. and for 24 days with the nitrate. The biomass yield when a phosphate source was used was also shown to remain approximately constant for 9 days, at $3.17g-DW/(L{\cdot}mM\;PO_4$), with an $R^2$ of 0.99.

Effects of Dioscoreae Rhizoma on Excercise Practice Ability and Blood Fatigue Variable Factor (산약의 운동수행능력 증진 및 항피로 효과)

  • Objectives In this study, we analyzed the effect of Dioscoreae Rhizoma on exercise practice ability and blood fatigue variable factor in the forced swimming test. Methods Male mice were divided into four group. Mice were administrated with saline, Discorae Rhizoma extracted by water (DRW), Discorae Rhizoma extracted by 30% ethanol (DRE), red ginseng (RG), orally once per day, for 1 week. Exercise performance was evaluated by exhaustive swimming time and weights. Glycogen contents of liver and soleus muscle were measured at the end of the experiment. In addition, the biochemical parameters such as asparate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen, creatine, lactate dehydrogenase (LDH), ammonia, triglycerides (TG), total cholesterol (TC) and free fatty acid (FFA) were analyzed. Results Both DRW and DRE showed an improvement of swimming endurance compared with control group. Levels of ALT, ammonia, TG, TC and FFA were significantly decreased in both DRW and DRE compared with control group. Levels of AST, Creatine were significantly reduced in DRE group compared with control group. Moreover LDH showed only a tendency to increase in the DRW and DRE groups. Conclusions In summary, the present study showed that Dioscoreae Rhizoma improved in physical fatigue and exercise practice ability and DRE improved the superior than those of DRW.

Performance of sequencing batch reactor under aeration-limited condition and characteristics of microbial community change (폭기 에너지 저감 연속회분식반응조 운전과 미생물 군집 변화 특성)

  • Recent focus on wastewater treatment includes energy-saving and renewable energy generation for energy-independence of water infrastructures. Aeration and pumping in biological wastewater removal processes account for nearly 30-60% of the total electricity cost in real wastewater treatment plants. In this study, the performance and microbial characteristics were investigated in sequencing batch reactor under typical oxygen and oxygen limited condition. Under typical DO ($7.55{\pm}0.99mg/L$) and low DO ($0.23{\pm}0.08mg/L$) conditions, COD removal was stable over 91 % during SBR operation. Ammonia removal efficiency was reduced from 95.6 % to 89.2 % when DO concentration was dropped sharply. Phosphorus removal efficiency also reached 77% at oxygen-limited condition. The results indicated that removal efficiency both ammonia and phosphorus was influenced by DO condition. Microbial analysis revealed that Proteobacteria and Bacteroidetes at phylum level was dominant in typical DO and low DO conditions and DO concentration did not much affect phylum distribution. Population decrease of genera of nitrifying bacteria(Dokdonella) and Dechloromonas spp. affect removal efficiency of nitrogen and phosphorus at low DO condition.

Hepatic Encephalopathy in a Connemara Pony

  • A 3-year-old female Connemara pony was referred to Jeju National University Equine Hospital because of weight loss and prolonged anorexia. On admission, heart and respiratory rates were slightly elevated while body temperature was within the normal range. The color of the conjunctival and oral mucosa was yellowish pink. The blood chemistry results showed that total bilirubin, aspartate aminotransferase, and gamma-glutamyl transferase levels were remarkably elevated while blood urea nitrogen was within the reference range. Transcutaneous abdominal ultrasound revealed an enlarged right lobe of the liver with prominently increased parenchymal echogenicity, reduced hepatic vessels, and rounded caudal border. The pony was tentatively diagnosed with chronic hepatitis and icterus; rest and supportive treatments were provided. Clinical signs aggravated on day 2 with hind quarter paresis and cranial nerve signs such as circling, drooping, jerking, and head pressing against walls. Recumbency and generalized ataxia (grade 5/5) were shown on day 3. Plasma ammonia concentration on day 3 was as high as 656 µmol/L. Necropsy and histopathologic examinations strongly supported a diagnosis of hepatic encephalopathy. This case of hepatic encephalopathy exhibited rapid progress from low to terminal grade within 4 days in a Connemara pony. The results provide well-established clinical and pathological data for future application.

Effect of Grape Pomace Powder, Mangosteen Peel Powder and Monensin on Nutrient Digestibility, Rumen Fermentation, Nitrogen Balance and Microbial Protein Synthesis in Dairy Steers

  • This study was designed to investigate the effect of grape pomace powder (GPP), mangosteen peel powder (MPP) and monensin on feed intake, nutrients digestibility, microorganisms, rumen fermentation characteristic, microbial protein synthesis and nitrogen balance in dairy steers. Four, rumen fistulated dairy steers with initial body weight (BW) of $220{\pm}15kg$ were randomly assigned according to a $4{\times}4$ Latin square design to receive four treatments. The treatments were as follows: T1 = control, T2 = supplementation with monensin at 33 mg/kg diet, T3 = supplementation with GPP at 2% of dry matter intake, and T4 = supplementation with MPP at 30 g/kg diet. The steers were offered the concentrate diet at 0.2% BW and 3% urea treated rice straw (UTRS) was fed ad libitum. It was found that GPP supplemented group had higher UTRS intake and nutrient digestibility in terms of neutral detergent fiber and acid detergent fiber than those in control group (p<0.05). Ammonia nitrogen ($NH_3-N$) and blood urea-nitrogen concentration were higher in monensin, GPP and MPP supplemented groups (p<0.05). Total volatile fatty acids and propionate in the GPP group were higher than those in the control group (p<0.05) while acetate concentration, and acetate to propionate ratio were decreased (p<0.01) when steers were supplemented with GPP, monensin, and MPP, respectively. Moreover, protozoal populations in GPP, MPP, and monensin supplementation were significantly lower than those in the control group (p<0.05), while cellulolytic bacterial population was significantly higher in the control group (p<0.05). Nitrogen retention, microbial crude protein and efficiency of microbial nitrogen synthesis were found significantly higher in steers that received GPP (p<0.05). Based on this study it could be concluded that the GPP has potential as an alternative feed supplement in concentrate diets which can result in improved rumen fermentation efficiency, digestibility and microbial protein synthesis in steers fed on treated rice straw.

Seasonal Changes of Microflora in Paddy Soil with Long-term Application of Organic Matter (유기물(有機物) 연용답토양(連用畓土壤)에 있어서 미생물상(微生物相)의 계절적(季節的) 변화(變化))

  • This experiment was carried out to investigate the effects on the seasonal population change of microflora of long-term application of organic matters in Fluvio-Alluvial plain of Jeonbug series. As organic matters, rice straw and compost of 5 and 10ton/ha, which were applied with the different nitrogen fertilizer level of 0, 150kg/ha into the soil 15cm deep, respectively. A number of total aerobic bacteria were gradually increased from just after water-logging before rice transplanting to pancle formations stage, afterthat decreased at harvest. The other side, a number of actinomycetes, fungi and cellulose-decomposers were slightly fluctuated until panicle formation stage and increased at havesting stage. In general, microorganism numbers were higher in organic matter with long-term nitrogen fertilizer applied plot, while cellulose-decomposers were higher in only organic matter applied plot. The microorganisms of ammonia-oxidizing, nitrate-reducing and nitrite-oxidizing, and denitrifying bacteria showed the maximum number at harvest stage, at panicle formation stage and at early tillering stage, respectively, while that of ammonifying bacteria were variable if nitrogen fertilizer applied or not at the respective periods in nitrogen cycle under water-logging. These bacteria were numerous in the organic matter plots combined with nitrogen fertilizer, especially, denitrifying bacteria in rice straw, others no difference.

An Influence of Point-Source and Flow Events on Inorganic Nitrogen Fractions in a Large Artificial Reservoir (대형 인공호에서 무기 질소원에 대한 점오염원 및 유입수의 영향)

  • This paper evaluated the influence of point source and flow events on inorganic nitrogen fractions at 17 sites of Taechung Reservoir during 1993${\sim}$1994. Total nitrogen (TN) averaged 1.53 mg/L during the study and ranged between 0.70 and 2.56 mg/L. Dissolved inorganic nitrogen(DIN) accounted for >90% of TN regardless of season and location, indicating a nitrogen-rich system showing eutrophic${\sim}$hypereutrophic conditions. Some 67${\sim}$94% of DIN was NO$_{3}$-N, whereas mean level of NH$_{4}$-N was less than 5% of DIN. During monsoon 1993, dilution of NO$_{3}$-N was evident in the headwaters as a result of mixing of lake water with rain water, while NH$_{4}$-N increased>100% compared to the premonsoon. Values of NH$_{4}$-N had a positive correlation with rainfall (r=0.85; p<0.001) and negative correlations with theoretical water residence time(r=-0.90; p<0.001) and conductivity(r=-0.78, p<0.001), respectively. These outcomes suggest that NH$_{4}$-N came from external input from the watershed during the monsoon. In both years, mean TN was greater in the mid-lake sites than any other sites. A great amount of TN in the mid-lake was most pronounced in monsoon 1994 because of an accumulated influence of the point sources during low inflow. Overall data suggest that concentrations of TN in this system did not show large differences along the longitudinal gradients and its distributions is likely determined by point-sources rather than inflow regime.
