Distribution of Seven N-Nitrosamines in Food

  • N-nitrosamines, which are classified as carcinogens by IARC and US EPA, can be easily found in various foods. They are reaction products between nitrogen oxide and secondary amines, but can also be generated during fermentation. Ever since the 1960s, when nitrite, used as a preservative in processed meats, was suspected to generate N-nitrosamines, the usage of the food additive has been debated. However, the benefit of nitrite in food supply could not be ignored and the risk-benefit analysis has become a key issue in the use of the additive. For a risk analysis, an accurate estimation of the hazardous material is necessary; therefore, analytical methods for nitrosamines have continuously evolved from the 1950s. Solid supported liquid-liquid extraction and solid phase extractions have replaced the distillation for the clean-up steps, and tandem mass spectrometry is employed for higher selectivity and sensitivity. In the present study, for a better estimation of N-nitrosamine intake, the total diet study samples were prepared for the N-nitrosamines analysis. In order to obtain the most sensitive results, a partial preparation procedure was developed and modified for different food matrices. Among seven N-nitrosamines (N-nitrosodimethylamine, N-nitrosomethylethylamine, N-nitrosodiethylamine, N-nitrosodibutylamine, N-nitrosopiperidine, N-nitrosopyrrolidine, and N-nitrosomorpholine) analyzed in the present study, N-nitrosodiethylamine has shown the highest detection rate in agricultural foods, while N-nitrosodimethylamine has appeared most frequently in livestock and fishery food products. The concentration of N-nitrosodimethylamine was the highest in seasoning.

Effect of Trans Fat on Lipid Profiles and Fatty Acid Composition in Serum, Heart, Liver and Kidney in Mice (트랜스 지방 섭취가 마우스의 혈청 및 조직의 지질 및 지방산 조성에 미치는 영향)

  • The purpose of this investigation was to determine the effect of feeding trans fat on serum and tissue lipid profiles and fatty acid composition in mice. Forty-five male mice aged at 4 weeks were fed on the 10% palm oil (n-3 deficient group), 20% dried mackerel (n-3 adequate group) and 5% trans fat (trans group) for three months. Total triglyceride, cholesterol and LDL-cholesterol levels were increased in serum, liver and heart of the n-3 deficient and trans groups compared with the n-3 adequate group, while liver HDL-cholesterol levels were lowest in the trans group (p<0.05). In liver and heart, trans group showed higher levels of total cholesterol and LDL-cholesterol compared with the n-3 deficient group (p<0.05). In fatty acid compositions of serum, heart, liver and kidney, the intake of trans fat led to increased percentages of 18:1n-9trans and 18:2n-6trans. The trans group showed similar patterns to the n-3 deficient group, except an increased percentage of 18:2n-6cis. Generally, the n-3 deficient and trans groups showed increased percentages of n-6 fatty acids and decreased percentages of n-3 fatty acids, specially docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) compared to the n-3 adequate group (p<0.05). These results indicate that intake of trans fat increased total triglyceride, cholesterol and LDL-cholesterol levels, which may affect risk for cardiovascular mortality.

Application of Hybrid SNCR/SCR process for Improved N Ox Removals Efficiency of SNCR (SNCR의 N Ox 제거효율 향상을 위한 Hybrid SNCR/SCR 공정 응용)

  • The objective of this research was to test whether, under controlled laboratory conditions, hybrid SNCR/SCR process improves N $O_{x}$ removal efficiency in comparison with the SNCR only. The hybrid process is a combination of a redesigned existing SNCR with a new downstream SCR. N $O_{x}$ reduction experiments using a hybrid SNCR/SCR process have been conducted in simple NO/N $H_3$/ $O_2$ gas mixtures. Total gas flow rate was kept constant 4 liter/min throughout the SNCR and SCR reactors, where initial N $O_{x}$ concentration was 500 ppm in the presence of 5% or 15% $O_2$. Commercial catalysts, $V_2$ $O_{5}$ -W $O_3$-S $O_4$/Ti $O_2$, were used for SCR N $O_{x}$ reduction. The residence time and space velocity were around 1.67 seconds and 2,400 $h^{-1}$ or 6000 $h^{-1}$ in SNCR and SCR reactors, respectively. N $O_{x}$ reduction of the hybrid system was always higher than could be achieved by SNCR alone at a given value of N $H_{3SLIP}$. Optimization of the hybrid system performance requires maximizing N $O_{x}$ removal in the SNCR process. An analysis based on the hybrid system performance in this lab-scale work indicates that a equipment with N $O_{xi}$ =500 ppm will achieve a total N $O_{x}$ removal of about 90 percent with N $H_{3SLIP}$ $\leq$ 5 ppm only if the SNCR N $O_{x}$ reduction is at least 60 percent. A hybrid SNCR/SCR process has shown about 26∼37% more N $O_{x}$ reduction than a SNCR unit process in which a lower temperature of 85$0^{\circ}C$ turned out to be more effective.be more effective.

Vertical Distribution and Composition of Dissolved Free Amino Acids in the Northeast Pacific Ocean (북동태평양 해역의 용존 자유아미노산 분포 및 조성 연구)

  • Concentrations of total dissolved free amino acids (DFAAs) in the northeast Pacific Ocean $9^{\circ}54'-10^{\circ}27'N$, $131^{\circ}43'-131^{\circ}53'W$) ranged from 15.9 to 1778.2 17M, and the average was 407.2 nM. Concentrations of DFAAs in surface mixed layers, ranged from 60.1 to 1411.9 nM, and the average was 535.2 nM. Seasonal thermoclines with maxima were formed between the depth of 50 to 150 m. DFAAs in this layer were varied in concentrations from 91.7 to 1778.2 nM, and the average was 588.5 nM. DFAAs below the seasonal thermoclines fluctuated from 15.9 to 384.2 nM, and the average was 175.1 nM. Consequently, in average relatively abundant DFAAs were observed in the subsurface layer than the deeper layer. DFAA vertical profiles and compositions of station A showed similar to station Band C. Glycine, alanine, glutamic acid, serine and valine were predominant accounting for more than 60% of total amino acids. Isoleucine, tyrosine, methionine and phenylalanine comprised only few percents of total DFAAs in the study area. In mole % of amino acid, according to characteristics of functional group of amino acid, aliphatic neutral accounted for 59% and aliphatic hydroxy 16%, acidic 12%, respectively. Although differences in DFAA concentrations with depth were observed, the amino acid composition and mole % of deeper layers in all stations were similar to those of subsurface layers. The results indicate that individual DtAAs remains invariably in water columns relative to the compositions and distributions of DFAAs in the study area, which may be the result of close coupling between microbial activity and their water solubility.

Effect of Nitrogen Fertilization on Oxalate Content in Rhodesgrass, Guineagrass and Sudangrass

  • An experiment was conducted to evaluate the effects of nitrogen (N) level on the dry matter (DM) yield, N concentration and oxalate content of some tropical grasses, namely Rhodesgrass (Chloris gayana), Guineagrass (Panicum maximum) and Sudangrass (Sorghum vulgare). Three levels of N as urea were applied (Standard- 260, $Standard{\times}2$- 540 and$Standard{\times}4$- 1,060 kg N/ha for Rhodesgrass; Standard- 380, $Standard{\times}2$- 770 and $Standard{\times}4$- 1,570 kg N/ha for Guineagrass and Sudangrass) in a completely randomized design and grasses were harvested twice at approximately two-month intervals. Dry matter yield tended to be higher with increased rate of N fertilizer in all species, while further additional N ($Standard{\times}2$ or $Standard{\times}4$) did not significantly (p>0.05) further increase DM yield, when compared with the Standard level of N fertilizer application. There was also a trend towards higher N concentration in plants as N fertilization increased in all species and it was increased significantly in Rhodesgrass and Sudangrass (p<0.05 or p<0.01, respectively). Further additional N ($Standard{\times}2$ or $Standard{\times}4$) application showed no significant (p>0.05) differences on oxalate content in plant tissue within species, when compared with the Standard level of N. The Rhodesgrass contained 0.11, 0.13 and 0.15% soluble oxalate and 0.23, 0.25 and 0.27% total oxalate with Standard, $Standard{\times}2$ and $Standard{\times}4$ level of N application, respectively. The Guineagrass contained 0.54, 0.50 and 0.42% soluble oxalate and 1.60, 1.56 and 1.45% total oxalate with Standard, $Standard{\times}2$ and $Standard{\times}4$ level of N application, respectively. The Sudangrass contained 0.06, 0.15 and 0.12% soluble oxalate and 0.22, 0.22 and 0.21% total oxalate with Standard, $Standard{\times}2$ and $Standard{\times}4$ level of N application, respectively The results from this study suggest that these grasses do not use further addition of N fertilizer ($Standard{\times}2$ or $Standard{\times}4$) to form high content of oxalate salts, when compared with the Standard level of N. In addition, the levels of oxalate present with these grasses are quite low as far as toxicity to animals is concerned.

Osteological Development of the Larvae and Juveniles in Aucha Perch Coreoperca kawamebari (Perciformes: Centropomidae) in Korea (한국산 꺽저기(Coreoperca kawamebari) 자치어의 골격발달)

  • Samples were obtained from broodstork in May, 1998, while naturally fertilized embryos were maintained and the process of skeletal development was observed from larvae and juveniles. Prelarvae immediately after hatching showed an average total length of $5.38{\pm}0.41mm$ (n=10), premaxillary and dentary were ossified, parasphenoid was ossified in the cranium, and centrum and caudal bone did not ossify. Prelarvae showed ossification with maxillary, articular and epihyal and branchiostegal rays of hyoid arch were ossified at 5 days after hatching with an average total length of $6.40{\pm}0.39mm$ (n=10). The vertebrae began to ossify in the direction of the tail, and neural spine began to ossify above the ossified vertebra. Postlarvae showed ossification of lateral ethmoid, parietal, and caudal skeleton in the cranium when the average total length was $7.30{\pm}0.12mm$ (n=10) in 8 days after hatching. At 22 days after hatching, postlarvae ossified maxillary in the cranium, and ossified endopterygoid and ectopterygoid, etc. in the palate, when the average length of $11.1{\pm}1.27mm$ (n=10). At 32 days after hatching, with the average length was $12.8{\pm}1.97mm$ (n=10), caudal skeleton had one additional epural bone ossification, resulting in ossification of a total of 3 epural bone to complete ossification of all spicules.

Grazing on Bacteria and Algae by Metazoans in the Lake-river Ecosystem (River Spree, Germany)

  • Direct effects of zooplankton grazing activities on the natural assemblage of bacterioplankton and algae were evaluated at monthly intervals, from June to October of 2000, in the middle part of the River Spree, Germany. We quantified bacterioplankton, algae, zooplankton abundance and measured carbon ingestion rates (CIRs) by zooplankton according to two zooplankton size classes: (i) micro zooplankton (MICZ), ranging in size from 30 to $150{\mu}m$ and including rotifers and nauplii, excluding protozoans and (ii) macrozooplankton (MACZ), larger than $150{\mu}m$ and including cladocerans and copepods. CIRs were measured using natural bacterial and algae communities in the zooplankton density manipulation experiments. Algae biomass (average${\pm}$SD: $377{\pm}306{\mu}gC\;L^{-1}$, n=5) was always higher than bacterial biomass ($36.7{\pm}9.9{\mu}gC\;L^{-1}$, n=5). Total zooplankton biomass varied from 19.8 to $137{\mu}gC\;L^{-1}$. Total mean biomass of zooplankton was $59.9{\pm}52.5{\mu}gC\;L^{-1}$ (average${\pm}$SD, n=5). Average MICZ biomass ($40.2{\pm}47.6{\mu}gC\;L^{-1}$ n=5) was nearly twofold higher than MACZ biomass ($19.6{\pm}20.6{\mu}gC\;L^{-1}$ n=5). Total zooplankton CIRs on algae (average${\pm}$SD: $56.6{\pm}26.4{\mu}gC\;L^{-1}\;day^{-1}$) were $\sim$fourfold higher than that on bacteria $(12.7{\pm}6.0{\mu}gC\;L^{-1}\;day^{-1})$. MICZ CIRs on bacteria $(7.0{\pm}2.8{\mu}gC\;L^{-1}\;day^{-1})$ and algae $(28.6{\pm}20.6{\mu}gC\;L^{-1}\;day^{-1})$ were slightly higher than MACZ CIRs. On average, MICZ accounted for 55.6 and 50.5% of total zooplankton grazing on bacteria and algae, respectively. Considering the MICZ and MACZ CIRs, the relative role of transferring carbon to higher trophic levels were nearly similar between both communities in the lake-river ecosystem.

At 40 days after hatching, postlarvae ossified maxillary in the cranium, and ossified endopterygoid and ectopterygoid, etc. in the palate, when the average length of $23.3{\pm}0.28mm$ (n=5). At 120 days after hatching, with the average length was $37.5{\pm}2.83mm$ (n=5), caudal skeleton had one additional epural bone ossification, resulting in ossification of a total of 3 epural bone to complete ossification of all spicules.

Preparation and Properties of Aromatic Polybenzoxazoles with high char yields (높은 char 수득률를 갖는 방향족 polybenzoxazoles의 제조 및 특성)

  • A series of polyhydroxyamides(PHAs) having ether linkages in the polymer backbone were prepared via solution polycondensation at low temperature. These polymers were studied by FT-IR, $^1H-NMR$, DSC, TGA and PCFC. The PHAs exhibited inherent viscosities in the range of $0.5{\sim}1.1dL/g\;at\;35^{\circ}C$ in DMAc solution. Most of PHAs except PHA 3 were soluble in polar organic solvents such as N,N-dimethylacetamide(DMAc), N-methyl-2-pyrrolidone(NMP), and N,N-dimethylform-amide(DMF). Subsequent thermal treatment of PHAs afforded polybenzoxazols(PBOs). However, the PBOs were insoluble in a variety of solvents. Most of the PBOs except PBO 3 showed glass-transition temperature($T_g$) in the range of $200{\sim}246^{\circ}C$ by DSC and maximum weight loss temperature in the range of $597{\sim}697^{\circ}C$ in nitrogen by TGA. PBOs showed high char yields in the range of $51{\sim}64%$. PCFC results of the PBOs showed the heat release(HR) capacity, $8{\sim}65J/gK$ and total heat release(total HR), $2.4{\sim}4.7kJ/g$.

Antioxidative Activity of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts Using Various Solvents (자생 산사(Crataegus pinnatifida BUNGE)씨의 추출 용매에 따른 항산화 활성)

  • The purpose of this study was to examine the antioxidative activity of feral haw (Crataegus pinnatifida BUNGE) seed extracts using 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v), n-butanol and ethyl acetate (EA). The total phenol content of the five extracts ranged from 37.29 mg/g to 55.53 mg/g. Moreover, the content was high in the 70% methanol and, 70% ethanol extracts, but low in the n-butanol extract. On the contrary, the total flavonoid content decreased in the order of n-butanol (2.93 mg/g), EA (2.67 mg/g), 70% methanol (1.00 mg/g), 70% ethanol (0.88 mg/g) and CM (0.67 mg/g) extracts. The $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay and, superoxide dismutase (SOD) like ability decreased in the order of 70% methanol, 70% ethanol, CM, EA and, n-butanol extracts; further, a similar tendency was also observed in the total phenol contents. Overall, these results indicated that the antioxidative activity of feral haw seeds was closely related to the total phenol and flavonoid contents. Therefore, haw seeds might be usefully applied to natural antioxidants as well as functional foods.
