Antioxidative Status, DNA Damage and Lipid Profiles in Korean Young Adults by Glutathione S-Transferase Polymorphisms (Glutathione S-transferase (GST) 유전자 다형성에 따른 우리나라 젊은 성인의 항산화 상태, DNA 손상 및 지질 양상)

  • Oxidative stress leads to the induction of cellular oxidative damage, which may cause adverse modifications of DNA, proteins, and lipids. The production of reactive species during oxidative stress contributes to the pathogenesis of many diseases. Antioxidant defenses can neutralize reactive oxygen species and protect against oxidative damage. The aim of this study was to assess the antioxidant status and the degree of DNA damage in Korean young adults using glutathione s-transferase (GST) polymorphisms. The GSTM1 and GSTT1 genotypes were characterized in 245 healthy young adults by smoking status, and their oxidative DNA damage in lymphocytes and antioxidant status were assessed by GST genotype. General characteristics were investigated by simple questionnaire. From the blood of the subjects, GST genotypes; degree of DNA damage in lymphocytes; the erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase; plasma concentrations of total peroxyl radical-trapping potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene and cryptoxanthin, as well as plasma lipid profiles, conjugated diene (CD), GOT, and GPT were analyzed. Of the 245 subjects studied, 23.2% were GSTM1 wild genotypes and 33.4% were GSTT1 wild genotype. No difference in erythrocyte activities of superoxide dismutase, catalase, or glutathione peroxidase, and the plasma TRAP level, CD, GOT, and GPT levels were observed between smokers and non-smokers categorized by GSTM1 or GSTT1 genotype. Plasma levels of ${\alpha}$- and ${\gamma}$-tocopherol increased significantly in smokers with the GSTT1 wild genotype (p < 0.05); however, plasma level of ${\alpha}$-carotene decreased significantly in non-smokers with the GSTM1 wild genotype (p < 0.05). DNA damage assessed by the Comet assay was significantly higher in non-smokers with the GSTM1 genotype; whereas DNA damage was significantly lower in non-smokers with the GSTT1 genotype. Total cholesterol and LDL cholesterol levels were significantly higher in non-smokers with the GSTT1 genotype than those with the GSTT1 wild genotype (p < 0.05). In conclusion, the GSTM1 genotype or the GSTT1 wild genotype in non-smokers aggravated their antioxidant status through DNA damage of lymphocytes; however, the GSTT1 wild type in non-smokers had normal plasma total cholesterol and LDL-cholesterol levels. This finding confirms that GST polymorphisms could be an important determinant of antioxidant status and plasma lipid profiles in non-smoking young adults. Further study is necessary to clarify the antioxidant status and/or lipid profiles of smokers with the GST polymorphism and to conduct a study with significantly more subjects.

Chemopreventive Effect of Vegetable or Fruit Extract Against Total Diesel Exhaust Particle Extract in NIH/3T3 Cells Using Alkaline Single Cell Gel Electrophoresis (총 디젤분진의 DNA 손상작용과 야채 및 과일추출물의 보호효과)

  • In urban areas, diesel exhaust particles (DEP) are probably a major component of particulate matters, especially in Korea where drive many diesel vehicles. The aim of this study was to investigate genotoxic effects of DEP using single ceil gel electrophoresis. In order to evaluate the mechanisms of DEP genotoxicity, the rat microsome mediated and DNA repair enzyme treated comet assays together with conventional comet assay were performed. Total diesel particles (DEPT) was collected without site fractionation from diesel engine bus and dichloromethane extract was obtained. The organic extract of DEPT revealed DNA damage itself in NIH/3T3 cells. The level of DNA breaks plus oxidative DNA lesions and microsome mediated DNA damage was assessed by modified single cell gel eletrophoresis. DEPT was able to induce oxidative DNA damage as well as microsome mediated DNA damage. Vitamin C as an model antioxidant reduced DNA damage in endonuclase III treated comet assay. One of flavonoid, galangin as a CYP1A1 inhibitor. reduced DNA damage in the presence of S-9 mixture. $DEP_T$ is the sources of oxidative stress, but antioxidants can significantly reduce oxidative DNA dmage. And $DEP_T$ may contain indirect mutagens which can be inhibited by CYP1A1 inhibitors. The ethanol extracts of the mixed vegetables (BV) or the mixed fruits (BF) were evaluated for their in vitro antigenotoxic effects. BV and BF showed potent Inhibitory effects against DEPT induced DNA damage with oxidative DNA lesions and in the prescence of S-9 mixture. These results indicate that BV and BF could prevent cellular DNA damage by inhibiting oxidative stress and suppressing cytochrome P4501A1 in cell culture.

A cumulative damage model for extremely low cycle fatigue cracking in steel structure

  • The purpose of this work is to predict ductile fracture of structural steel under extremely low cyclic loading experienced in earthquake. A cumulative damage model is proposed on the basis of an existing damage model originally aiming to predict fracture under monotonic loading. The cumulative damage model assumes that damage does not grow when stress triaxiality is below a threshold and fracture occurs when accumulated damage reach unit. The model was implemented in ABAQUS software. The cumulative damage model parameters for steel base metal, weld metal and heat affected zone were calibrated, respectively, through testing and finite element analyses of notched coupon specimens. The damage evolution law in the notched coupon specimens under different loads was compared. Finally, in order to examine the engineering applicability of the proposed model, the fracture performance of beam-column welded joints reported by previous researches was analyzed based on the cumulative damage model. The analysis results show that the cumulative damage model is able to successfully predict the cracking location, fracture process, the crack initiation life, and the total fatigue life of the joints.

Enhanced remote-sensing scale for wind damage assessment

  • This study has developed an Enhanced Remote-Sensing (ERS) scale to improve the accuracy and efficiency of using remote-sensing images of residential building to predict their damage conditions. The new scale, by incorporating multiple damage states observable on remote-sensing imagery, substantially reduces measurement errors and increases the amount of information retained. A ground damage survey was conducted six days after the Joplin EF 5 tornado in 2011. A total of 1,400 one- and two-family residences (FR12) were selected and their damage states were evaluated based on Degree of Damage (DOD) in the Enhanced Fujita (EF) scale. A subsequent remote-sensing survey was performed to rate damages with the ERS scale using high-resolution aerial imagery. Results from Ordinary Least Square regression indicate that ERS-derived damage states could reliably predict the ground level damage with 94% of variance in DOD explained by ERS. The superior performance is mainly because ERS extracts more information. The regression model developed can be used for future rapid assessment of tornado damages. In addition, this study provides strong empirical evidence for the effectiveness of the ERS scale and remote-sensing technology for assessment of damages from tornadoes and other wind events.

Optimal Design of PSC-I Girder Bridge Considering Life Cycle Cost (생애주기비용을 고려한 PSC-I형 교량의 최적설계)

  • This paper presents the procedure for the optimal design of a PSC-I girder bridge considering life cycle cost (LCC). The load carrying capacity curves for the concrete deck, PSC-I girder and $\pi$-type pier were derived and used for the estimate of service lives. Total life cycle cost for the service life was calculated as sum of initial cost, damage cost, maintenance cost, repair and rehabilitation cost, user cost, and disposal cost. The advanced First Order Second Moment method was used to estimate the damage cost. The optimization method was applied to the design of PSC-I girder bridge. The objective function was set to the annual cost, which is defined by dividing the total life cycle cost by the service life, and constraints were formulated on the basis of Korean Standards. The optimal design was performed for various service lives and the effects of design factors were investigated.

  • Welding fume (WF) induces pulmonary disease including pneumoconiosis. To investigate whether reactive oxygen species-induced oxidative DNA damage occurs during welding fume exposure and the upregulation of DNA repair mechanisms is accompanied, SPF SD rats were exposed to welding fumes with the concentrations of 65.6${\pm}$2.9 mg/㎥(low dose) and 116.8${\pm}$3.9 mg/㎥ (high dose) of total suspended particulate for 2 hrs per day in an inhalation chamber for a total of 2hrs, 15 or 30 days.(omitted)

A Novel Procedure for Mooring Chain Fatigue Prediction based on Maximum Principal Stress Considering Out-of-Plane and In-Plane Bending Effects (면내외 굽힘 효과를 고려한 최대 주응력 기반 계류 체인 피로 평가 기법 개발)

  • As OPB and IPB moment-induced fatigue damage on mooring chain links were reported for a offloading buoy, verification of OPB and IPB fatigue has been a key engineering item in offshore structure mooring design. Mathematical and physical features of the conventional approach which was mainly explained in BV guideline are reviewed and disadvantages of the conventional approach are addressed in terms of stress proportionality and nonlinearity of OPB and IPB moments. In order to eradicate these disadvantages, a novel approach is newly proposed which is able to dispel apprehension on stress proportionality and is not dependent of nonlinearities of OPB and IPB moments. Significant differences between two approaches are suggested by comparing relations of OPB moment versus OPB interlink angle and IPB moment versus IPB interlink angle. For periodic OPB tension angle processes having three different OPB angle ranges with a simple irregular tension process, fatigue damage calculation reveals that OPB moment-induced fatigue damage has dominant portion to total fatigue damage. Comparative studies between two approaches also show that the conventional approach based on BV guideline predicts fatigue damage far conservatively since it assume unrealistic high stress concentration factor for tension load. Meanwhile IPB moment-induced fatigue damage is negligible compared to tension-induced fatigue damage.

Psychological factors affecting nurses' perceptions of gaslighting damage (간호사의 가스라이팅 피해 인지에 영향을 미치는 심리적 요인)

  • Purpose: This study aimed to determine the influence of psychological factors such as highly sensitive person, intolerance of uncertainty, depression, and self-esteem on nurses' perceptions of gaslighting damage. Methods: The participants were 178 nurses working in hospitals nationwide. Data were collected through a non-face-to-face approach using an online survey platform (Google Forms) with self-reported questionnaires. Perceptions of gaslighting damage were measured using the victim gaslighting questionnaire (VGQ). The collected data were analyzed with statistical methods such as independent t-test, ANOVA, Pearson's correlation coefficient, and multiple regression analysis using IBM SPSS 25.0. Results: Of the participants, 94.4% were female. The participants' average age was 32.7 years old, 60.2% were unmarried, and 86.0% were staff nurses. The mean score of the VGQ was 40.49. There were statistically significant differences in the VGQ according to marital status, participants' time in their current department, total clinical career, and type of medical institution. The perception of gaslighting damage was statistically significantly positively correlated with intolerance of uncertainty, highly sensitive person, depression, and self-esteem. The most influential factor on nurses' perception of gaslighting damage was intolerance of uncertainty (β=.32, p<.001), followed by depression (β=.30, p<.001) and highly sensitive person (β=.23, p=.001). The total explanation of these variables for the perception of gaslighting damage by nurses was about 75.0%. Conclusion: This study suggests that it is necessary to develop an intervention program to reduce negative psychological factors affecting gaslighting in nurses and to cultivate nurses' strength to overcome gaslighting damage.

A new damage index for reinforced concrete structures

  • Reinforced concrete (RC) structures are likely to experience damage when subjected to earthquakes. Damage index (DI) has been recognised as an advanced tool of quantitatively expressing the extent of damage in such structures. Last 30 years have seen many concepts for DI proposed in order to calibrate the observed levels of damage. The current research briefly reviews all available concepts and investigates their relative merits and limitations with a view to proposing a new concept based on residual deformation. Currently available DIs are classified into two broad categories - non-cumulative DI and cumulative DI. Non-cumulative DIs do not include the effects of cyclic loading, whilst the cumulative concepts produce more rational indication of the level of damage in case of earthquake excitations. Ideally, a DI should vary within a scale of 0 to 1 with 0 representing the state of elastic response, and 1 referring to the state of total collapse. Some of the available DIs do not satisfy these criteria. A new DI based on energy is proposed herein and its performances, both for static and for cyclic loadings, are compared with those obtained using the most widely accepted DI in literature. The proposed DI demonstrates a rational way to predict the extent of damage for a number of case studies. More research is encouraged to address some identified issues.
